Marine Recirculating Aquaculture Systems (RAS) produce great volume of wastewater, which may be reutilized/recirculated or reutilized after undergoing different treatment/remediation methods, or partly discharged into neighbour water-bodies (DWW). Phosphates, in particular, are usually accumulated at high concentrations in DWW, both because its monitoring is not compulsory for fish production since it is not a limiting parameter, and also because there is no specific treatment so far developed to remove them, especially in what concerns saltwater effluents. As such, this work addresses two main scientific questions. One of them regards the understanding of the actual (bio)remediation methods applied to effluents produced in marine RAS, by identifying their advantages, drawbacks and gaps concerning their exploitation in saltwater effluents. The second one is the development of a new, innovative and efficient method for the treatment of saltwater effluents that potentially fulfil the gaps identified in the conventional treatments. Thereby, the aims of this thesis are: (i) to revise the conventional treatments targeting major contaminants in marine RAS effluents, with a particular focus on the bioremediation approaches already conducted for phosphates; (ii) to characterize and evaluate the potential of oyster-shell waste collected in Ria de Aveiro as a bioremediation agent of phosphates spiked into artificial saltwater, over different influencing factors (e.g., oyster-shell pre-treatment through calcination, particle size, adsorbent concentration). Despite the use of oyster-shells for phosphorous (P) removal has already been applied in freshwater, its biosorptive potential for P in saltwater was never evaluated, as far as I am aware. The results herein generated showed that NOS is mainly composed by carbonates, which are almost completely converted into lime (CaO) after calcination (COS). Such pre-treatment allowed obtaining a more reactive material for P removal, since higher removal percentages and adsorption capacity was observed for COS. Smaller particle size fractions for both NOS and COS samples also increased P removal. Kinetic models showed that NOS adsorption followed, simultaneously, Elovich and Intraparticle Difusion kinetic models, suggesting that P removal is both a diffusional and chemically rate-controlled process. The percentage of P removal by COS was not controlled by Intraparticle Diffusion and the Elovich model was the kinetic model that best fitted phosphate removal. This work demonstrated that waste oyster-shells, either NOS or COS, could be used as an effective biosorbent for P removal from seawater. Thereby, this biomaterial can sustain a cost-effective and eco-friendly bioremediation strategy with potential application in marine RAS.

Serines proteinases inhibitors (PIs) are widely distributed in nature and are able to inhibit both in vitro and in vivo enzymatic activites. Seed PIs in than leguminous are classified in seven families, Bowman-Birk and Kunitz type families that most studied representing an important role in the first line of defense toward insects pests. Some Kunitz type inhibitors possess activities serine and cysteine for proteinases named bifunctional inhibitor, as ApTKI the inhibitor isolate from seed of Adenanthera pavonina. The A. pavonina inhibitor presenting the uncommon property and was used for interaction studies between proteinases serine (trypsin) and cysteine (papain). In order to determinate the in vitro interaction of ApTKI against enzymes inhibitor purification was carried cut by using chromatographic techniques and inhibition assays. The 3D model of the bifunctional inhibitor ApTKI was constructed SWISS-MODEL program by homology modeling using soybean trypsin inhibitor (STI, pdb:1ba7), as template which presented 40% of identity to A. pavonina inhibitor. Model quality was evaluated by PROCHECK program. Moreover in silico analyzes of formed complex between the enzymes and ApTKI was evaluated by HEX 4.5 program. In vitro results confirmed the inhibitory assays, where the inhibitor presented the ability to simultaneously inhibit trypsin and papain. The residues encountered in the inhibitor model of folder structural three-dimensional that make contact to enzymes target coud explain the specificity pattern against serine and cysteine proteinases

O objetivo desta tese foi avaliar a dinâmica do fósforo em cultivo heterotrófico e produção de compostos celulares por Aphanothece microscopica Nägeli visando avaliar a perspectiva de implementação de uma biorrefinaria microalgal. Desta forma, foi avaliado o comportamento do micro-organismo em estudo no cultivo heterotrófico, utilizando como meio de cultivo o efluente de laticínios. O trabalho foi desenvolvido em 3 etapas. Em um primeiro momento foi avaliada a influência da temperatura (20 e 30°C) e os valores máximos e mínimos de nutrientes, em especial do fósforo dissolvido reativo (PDR), disponíveis no efluente de laticínio, na remoção de nutrientes. Os resultados demonstraram que a concentração inicial de fósforo dissolvido e a temperatura exerceram influência no crescimento celular e na eficiência de remoção de nutrientes. Em termos de otimização de processo os cultivos conduzidos a 20°C e maiores concentrações de PDR (5,5 mg.L-1 ) no efluente de laticínio, foram os mais eficientes na conversão de poluentes em biomassa e remoção de nutrientes. A segunda etapa foi desenvolvida com o objetivo de avaliar a dinâmica de distribuição de fósforo na fase líquida e sólida do reator heterotrófico, quando o efluente de laticínio foi tratado pela Aphanothece microscopica Nägeli, a 20°C e nas máximas concentrações de fósforo dissolvido encontradas no efluente. Foi demonstrado que as formas fosforadas na fase líquida do reator se caracterizam pela predominância da fração dissolvida em comparação à particulada e por apresentar como fração predominante a de fósforo orgânico. No que se refere à fase sólida, ficou demonstrado que a Aphanothece microscopica Nägeli, quando cultivada heterotroficamente apresenta 3,8 vezes mais fósforo que o requerido para o crescimento celular. Ficando demonstrado ainda que a remoção biológica de fósforo por Aphanothece microscopica Nägeli pode resultar em substanciais aportes financeiros para as estações de tratamento de efluentes. Uma terceira etapa foi desenvolvida, a qual teve como objetivo avaliar a estimativa de produção de compostos celulares por Aphanothece microscopica Nägeli, a partir do efluente de laticínio, bem como o efeito da redução de temperatura de cultivo no teor de lipídios , no momento em que é obtida a máxima concentração deste componente celular, nas condições otimizadas.Foi obtido na fase logarítmica de crescimento, concentrações de 41,8 % de proteinas, carboidratos 28,5 %, lipídios 10,4 % e minerais 10,8 %. O maior teor de lipídio registrado a 20°C correspondeu a biomassa analisada na fase logarítmica.Com a redução da temperatura para 5°C por um período de 30 h é possível obter concentrações de lipídios 2,4 vezes superior ao registrado na fase logarítmica a 20 °C. No entanto, não foram indicadas diferenças significativas (p≤0,05) em função da temperatura entre as concentrações de lipídios obtidas para a biomassa a 10°C em 40 h. O perfil de ácidos graxos da biomassa gerada a 20°C, apresentou como ácidos graxos majoritários, os ácidos graxos: palmítico, oléico, γ-linolênico, palmitoleico e esteárico, resultando um aumento na concentração de ácidos graxos saturados as espensa dos insaturados, quando a temperatura é reduzida. Em paralelo,um reator heterotrófico descontinuo foi definido, ficando demonstrado que a extrapolação da operação em batelada para contínua requer um biorreator heterotrófico com volume útil de trabalho de 240,51 m 3 , permitindo tratar 950 m3 diários de efluente, gerando 11,8 kg.d-1 de biomassa útil para produção de compostos celulares por Aphanothece microscopica Nägeli, visando à simultânea remoção de matéria orgânica, nitrogênio total e fósforo total, gerando insumos que podem suportar a implementação de uma biorrefinaria microalgal.

Este trabalho tem como objetivo desenvolver uma metodologia de seletividade cinética, para os pseudocomponentes do petróleo em escoamento gás-liquido em colunas de bolhas usando a Fluidodinâmica Computacional (CFD). Uma geometria cilíndrica de 2,5m de altura e 0,162m de diâmetro foi usada tanto na validação fluidodinâmica com base em dados experimentais da literatura, como na análise cinética do reator operando em dois modos distintos em relação a fase líquida: batelada e contínuo. Todos os casos de estudo operam em regime heterogêneo de escoamento, com velocidade superficial do gás igual a 8 cm/s e diâmetro médio de bolhas de 6 mm. O modelo fluidodinâmico validado apresentou boa concordância com os dados experimentais, sendo empregado como base para a implementação do modelo cinético de rede de Krishna e Saxena (1989). A análise da hidroconversão foi realizada a 371ºC, e os resultados mostraram o comportamento esperado para o processo reativo estudado, definindo-se os tempos (batelada) e posições axiais (contínuo) de coleta ideal para os pseudocomponentes leves. Em síntese, ressaltase o uso da ferramenta CFD no entendimento, desenvolvimento e otimização de processos.

The most common method of achieve the required fire resistance is by the use of passive fire protection systems, being intumescent coatings the fire protection material frequently used. These are usually considered thin film coatings as they are applied with a dry film thickness (DFT) between 0.3-3 [mm]. The required DFT is obtained by experimental fire resistance tests performed to assess the contribution of this reactive fire protection material to the steel member fire resistance. This tests are done after dry coating and a short time period of atmospheric conditioning, at constant temperature and humidity. As the coatings formulation is mainly made from polymeric basis compounds, it is expected that the environmental factors, such temperature, humidity and UV radiation (UVA and UVB) significantly affect the intumescent coating fire protection performance and its durability. This work presents a research study about the effects of aging on the fire protection performance of intumescent coatings. A commercial water based coating is submitted to an accelerated aging cycle, using a QUV Accelerated Weathering Tester. This tests aim to simulate 10 years of the coating natural aging. The coating durability is tested comparing the fire protection of small steel samples submitted to a radiant heat flux exposure from a cone calorimeter. In total, 28 tests were performed on intumescent coating protected steel specimens, of which 14 specimens were tested before the hydrothermal aging test and other 14 after accelerated aging. The experimental tests results of the steel temperature evolution shows that increasing the intumescent dry coating film thickness, the fire resistance time increases. After the accelerated aging cycles, the coating lose their ability to expand, resulting in an increase of the steel temperature of approximately 200 [ºC], compared to the samples without aging.

Plodia interpunctella (Indian meal moth) is a cosmopolitan pest that attacks not only a wide range of stored grain as well other food products. Due to its economic importance several researches have focused in a method with ability to control this pest with few or no damage to the environment. The study of digestive enzymes inhibitors, lectins and chitin-binding proteins, has often been proposed as an alternative to reduce insect damage. In this study we report the major classes of digestive enzymes during larval growth in P. Interpunctella, being those proteinases actives at pH 9.5 and optimum temperature of 50 oC to both larvae of the 3rd instar and pre-pupal stage of development. In vitro and zymogram assays presented the effects of several inhibitors, such as SBTI, TLCK and PMSF to intestinal homogenate of 3rd instar larvae of 62%, 92% and 87% of inhibition and In pre-pupal stage of 87%, 62 % and 55% of inhibition, respectively. Zymograms showed inhibition of two low molecular masses protein bands by TLCK and that in presence of SBTI were retarded. These results are indicative of predominance of digestive serine proteinases in gut homogenate from Plodia interpunctella larvae. This serine proteinase was then used as a target to evaluate the effect of SBTI on larvae in in vivo assay. Effect of SBTI on mortality and larval mass was not observed at until 4% of concentration (w/w) in diets. Chitin, another target to insecticidal proteins, was observed by chemical method. Moreover, optic microscopy confirmed the presence of a peritrophic membrane. Established this target, in vivo effect of EvV, a chitin binding vicilin, evaluated during the larval development of P. interpunctella and was obtained a LD50 of 0,23% and WD50 of 0,27% to this protein. Mechanism of action was proposed through of the in vivo digestibility of EvV methodology. During the passage through the larval digestive tract was observed that EvV was susceptible to digestive enzymes and a reactive fragment, visualized by Western blotting, produced by digestion was recovered after dissociation of the peritrophic membrane. The bound of EvV to peritrophic membrane was confirmed by immunohystochemical assays that showed strong immunofluorescent signal of EvV-FITC binding and peritrophic membrane. These results are a indicative that vicilins could be utilized as potential insecticide to Plodia interpunctella and a control methods using EvV as bioinsecticide should be studied to reduce lost caused by storage insect pests

A basic requirement of a plasma etching process is fidelity of the patterned organic materials. In photolithography, a He plasma pretreatment (PPT) based on high ultraviolet and vacuum ultraviolet (UV/VUV) exposure was shown to be successful for roughness reduction of 193nm photoresist (PR). Typical multilayer masks consist of many other organic masking materials in addition to 193nm PR. These materials vary significantly in UV/VUV sensitivity and show, therefore, a different response to the He PPT. A delamination of the nanometer-thin, ion-induced dense amorphous carbon (DAC) layer was observed. Extensive He PPT exposure produces volatile species through UV/VUV induced scissioning. These species are trapped underneath the DAC layer in a subsequent plasma etch (PE), causing a loss of adhesion. Next to stabilizing organic materials, the major goals of this work included to establish and evaluate a cyclic fluorocarbon (FC) based approach for atomic layer etching (ALE) of SiO2 and Si; to characterize the mechanisms involved; and to evaluate the impact of processing parameters. Periodic, short precursor injections allow precise deposition of thin FC films. These films limit the amount of available chemical etchant during subsequent low energy, plasma-based Ar+ ion bombardment, resulting in strongly time-dependent etch rates. In situ ellipsometry showcased the self-limited etching. X-ray photoelectron spectroscopy (XPS) confirms FC film deposition and mixing with the substrate. The cyclic ALE approach is also able to precisely etch Si substrates. A reduced time-dependent etching is seen for Si, likely based on a lower physical sputtering energy threshold. A fluorinated, oxidized surface layer is present during ALE of Si and greatly influences the etch behavior. A reaction of the precursor with the fluorinated substrate upon precursor injection was observed and characterized. The cyclic ALE approach is transferred to a manufacturing scale reactor at IBM Research. Ensuring the transferability to industrial device patterning is crucial for the application of ALE. In addition to device patterning, the cyclic ALE process is employed for oxide removal from Si and SiGe surfaces with the goal of minimal substrate damage and surface residues. The ALE process developed for SiO2 and Si etching did not remove native oxide at the level required. Optimizing the process enabled strong O removal from the surface. Subsequent 90% H2/Ar plasma allow for removal of C and F residues.

O presente estudo, no âmbito da Pragmática Linguística, resulta de uma investigação, que observa a realização dos atos ilocutórios expressivos de agradecimento nas interações comerciais. O objetivo é tentar compreender os processos linguísticos subjacentes à realização dos referidos atos ilocutórios, nomeadamente através do recenseamento dos diferentes tipos de agradecimento, da descrição dos atos de fala predominantes e das funções semântico-pragmáticas do agradecimento. Um dos aspetos mais importantes será o recensear dos tipos de agradecimento mais usados nas interações comerciais. Para tal, parte-se da observação detalhada de um corpus recolhido no âmbito específico de interações comerciais que ocorreram entre 10 de fevereiro de 2014 a 30 de setembro de 2014. Os resultados obtidos convergem no sentido de que o agradecimento é um ato expressivo reativo que pode ser verbal ou não verbal, com a finalidade de estabelecer a relação custo-benefício entre locutor e ouvinte, tendo em conta e respeitando os vários contextos socioculturais. De facto, o ato de agradecer, tal como os restantes atos ilocutórios expressivos, depende da forma como é expressado, dos gestos que o acompanham e das emoções que lhe são inerentes.

Brazil has vast amounts of hydric resources, whose quality has been deteriorating due to pollutant dumping. Household waste disposal is one of the main sources of water pollution, stimulating bacteria proliferation and introducing microorganisms, including those from fecal matter. Conventional water disinfection methods are a solution, but on the downside, they lead to the formation byproducts hazardous to human health. In this study, aiming to develop bactericidal filters for the disinfection of drinking water; silver nanoparticles were deposited on alumina foams through three routes: sputtering DC, dip coating and in situ chemical reduction of silver nitrate. The depositions were characterized through X-ray diffraction, scanning electron microscopy and EDS element mapping. The influence of the depositions on permeability and mechanical properties of the ceramic foams was assessed and, in sequence, a preliminary antibacterial efficiency analysis was carried out. Characterization results indicate that the chemical reduction routes were efficient in depositing homogeneously distributed silver particles and that the concentration of the metallic precursor salt affects size and morphology of the particles. The antibacterial efficiency analysis indicates that the chemical reduction filters have potential for water disinfection

A post-complementary metal oxide semiconductor (CMOS) compatible microfabrication process of piezoelectric cantilevers has been developed. The fabrication process is suitable for standard silicon technology and provides low-cost and high-throughput manufacturing. This work reports design, fabrication and characterization of piezoelectric cantilevers based on aluminum nitride (AlN) thin films synthesized at room temperature. The proposed microcantilever system is a sandwich structure composed of chromium (Cr) electrodes and a sputtered AlN film. The key issue for cantilever fabrication is the growth at room temperature of the AlN layer by reactive sputtering, making possible the innovative compatibility of piezoelectric MEMS devices with CMOS circuits already processed. AlN and Cr have been etched by inductively coupled plasma (ICP) dry etching using a BCl3–Cl2–Ar plasma chemistry. As part of the novelty of the post-CMOS micromachining process presented here, a silicon Si (1 0 0) wafer has been used as substrate as well as the sacrificial layer used to release the microcantilevers. In order to achieve this, the Si surface underneath the structure has been wet etched using an HNA (hydrofluoric acid + nitric acid + acetic acid) based solution. X-ray diffraction (XRD) characterization indicated the high crystalline quality of the AlN film. An atomic force microscope (AFM) has been used to determine the Cr electrode surface roughness. The morphology of the fabricated devices has been studied by scanning electron microscope (SEM). The cantilevers have been piezoelectrically actuated and their out-of-plane vibration modes were detected by vibrometry.

The aim of this work is to simulate and optically characterize the piezoelectric performance of complementary metal oxide semiconductor (CMOS) compatible microcantilevers based on aluminium nitride (AlN) and manufactured at room temperature. This study should facilitate the integration of piezoelectric micro-electro-mechanical systems (MEMS) such as microcantilevers, in CMOS technology. Besides compatibility with standard integrated circuit manufacturing procedures, low temperature processing also translates into higher throughput and, as a consequence, lower manufacturing costs. Thus, the use of the piezoelectric properties of AlN manufactured by reactive sputtering at room temperature is an important step towards the integration of this type of devices within future CMOS technology standards. To assess the reliability of our fabrication process, we have manufactured arrays of free-standing microcantilever beams of variable dimension and studied their piezoelectric performance. The characterization of the first out-of-plane modes of AlN-actuated piezoelectric microcantilevers has been carried out using two optical techniques: laser Doppler vibrometry (LDV) and white light interferometry (WLI). In order to actuate the cantilevers, a periodic chirp signal in certain frequency ranges was applied between the device electrodes. The nature of the different vibration modes detected has been studied and compared with that obtained by a finite element model based simulation (COMSOL Multiphysics), showing flexural as well as torsional modes. The correspondence between theoretical and experimental data is reasonably good, probing the viability of this high throughput and CMOS compatible fabrication process. To complete the study, X-ray diffraction as well as d33 piezoelectric coefficient measurements were also carried out.

No Estado do Pará, na maioria dos solos onde se cultiva milho, o fósforo é um dos nutrientes que mais limita a produtividade dessa cultura. Objetivou-se avaliar a resposta do milho a doses de fertilizantes fosfatados. O experimento foi conduzido no campo, em Latossolo Amarelo distrófico textura muito argilosa, no Município de Belterra, Pará. Foi utilizado o delineamento em blocos ao acaso com três repetições, em esquema fatorial 3x5, correspondendo a três fontes de adubos fosfatados, sendo superfosfato simples, superfosfato triplo e fosfato natural reativo de Bayóvar, e cinco doses de P2O5 (0 kg.ha-1,80 kg.ha-1, 160 kg.ha-1, 240 kg.ha-1 e 320 kg.ha-1). O milho foi colhido aos 120 dias após o plantio para avaliação da produtividade de grãos. Os maiores teores de nutrientes na folha do milho foram obtidos nos tratamentos com SFT em relação ao SFS e Bayóvar. A cultura do milho respondeu em produtividade à adubação fosfatada, independente da fonte de P e do método de aplicação do fertilizante. A dose mais adequada para as fontes de adubos fosfatados foi 160 kg.ha-1 de P2O5.

CONTEXTO: O papel de cuidador assumido pelo familiar nem sempre é bem aceite pela pessoa que vive depressão, o que tem repercussões na vida familiar, principalmente nas relações interpessoais. OBJETIVO(S): Caraterizar a depressão na perspetiva de quem a vive; Caraterizar o papel de cuidador familiar na perspetiva do doente; Descrever reações do doente na relação com os cuidadores familiares. METODOLOGIA: Desenho de natureza qualitativa e indutiva com recurso à Grounded Theory. Seleção de participantes, não probabilística intencional. Realização de entrevista semiestruturada. RESULTADOS: Encontradas quatro categorias na codificação axial. A pessoa deprimida identifica o início, causas, caraterísticas e manifestações da doença. A depressão é vivida de forma intimista pelo doente, desenvolvendo estratégias de enfrentamento e de gestão da sua situação clínica. Na perspetiva do doente, o familiar falha o papel de cuidador por controlo excessivo, falta de compreensão, incapacidade para escutar, chantagem emocional e por vezes agressividade verbal. A pessoa deprimida não reconhece capacidade ao familiar para o ajudar, não o aceita como cuidador, ignora as suas intervenções e conselhos, e o seu sentimento de solidão aumenta. CONCLUSÕES: Quando há uma pessoa com depressão na família, os familiares constroem o papel de cuidador na interação quotidiana e as estratégias de cuidados desenvolvem-se de modo reativo em função do comportamento do doente, adquirindo contornos particulares de conteúdo nem sempre adequado à situação de saúde vivida pela pessoa. O doente não se sente cuidado e não reconhece o familiar como parceiro ativo no seu processo de recuperação.

Os altos custos dos fosfatos industrializados têm contribuído para restringir o uso desses insumos na Amazônia, o que condiciona a busca de fontes alternativas de fósforo. Diante disso, a tarefa de estudar a eficiência de fontes de fósforo é importante para orientar a indústria de transformação, bem como para recomendar ao produtor rural o melhor manejo para cada fonte. Visando avaliar a eficiência agronômica do termofosfato de alumínio, como fonte de P, um experimento foi conduzido em um Latossolo Amarelo distrófico, textura muito argilosa, em Paragominas-PA. Utilizou-se o delineamento experimental em blocos ao acaso, com três repetições em arranjo fatorial de 3x4 correspondendo a três fontes de P (Bayóvar, Superfosfato triplo-SFT, termofosfato de alumínio) e quatro doses de P (0, 50, 100 e 200 kg ha-1 de P2O5). Os teores de fósforo no solo e produção de grãos de milho apresentaram um comportamento linear em resposta à aplicação de doses crescentes de termofosfato de alumínio, apresentando um comportamento muito semelhante ao fosfato reativo de Bayóvar. A aplicação de doses crescentes de termofosfato de alumínio promove aumento na produção de grãos de milho, apresentando comportamento muito semelhante ao fosfato reativo de Bayóvar. A maior eficiência agronômica do termofosfato de alumínio se verifica na maior dose de P aplicada (200 kg ha-1 de P2O5).
