Bioprocesses use microorganisms or cells in order to produce and/or obtain some desired products. Nowadays these strategies appear as a fundamental alternative to the traditional chemical processes. Amongst the many advantages associated to their use in the chemical, oil or pharmaceutical industries, their low cost, easily scale-up and low environmental impact should be highlighted. This work reports two examples of bioprocesses as alternatives to traditional chemical processes used by the oil and pharmaceutical industries. In the first part of this work it was studied an example of a bioprocess based on the use of microorganisms in enhanced oil recovery. Currently, due to high costs of oil and its scarcity, the enhanced oil recovery techniques become very attractive. Between the available techniques the use of microbial enhanced oil recovery (MEOR) has been highlighted. This process is based on the stimulation of indigenous microorganisms or by the injection of microorganism consortia to produce specific metabolites and hence increase the amount of oil recovered. In the first chapters of this work the isolation of several microorganisms from samples of paraffinic Brazilian oils is described, and their tensioactive and biodegradability properties are presented. Furthermore, the chemical structures of the biosurfactants produced by those isolates were also characterized. In the final chapter of the first part, the capabilities of some isolated bacteria to enhance the oil recovery of paraffinic Brazilian oils entrapped in sand-pack columns were evaluated. In the second part of this work it was investigated aqueous two-phase systems or aqueous biphasic systems (ABS) as extractive strategies for antibiotics directly from the fermented broth in which they are produced. To this goal, several aqueous two-phase systems composed of ionic liquids (ILs) and polymers were studied for the first time and their phase diagrams were determined. The novel ATPS appear as effective and economic methods to extract different biomolecules or/and biological products. Thus, aiming the initial antibiotics extraction purpose it was studied the influence of a wide range of ILs and polymers in the aqueous two-phase formation ability, as well as their influence in the partitioning of several type-molecules, such as amino acids, alkaloids and dyes. As a final chapter it is presented the capacity of these novel systems to extract the antibiotic tetracycline directly from the fermented broth of Streptomyces aureofaciens.

Esta tese constitui uma reflexão sobre uma realidade, a praia do Francês, Alagoas, Brasil, que após mais de uma década é agora revisitada demonstrando os impactos socioambientais evidenciados ao longo de 30 anos, e por sua vez, fazem com que esta praia se transforme em laboratório cuja análise desenvolvida poderá servir como exemplo para ações futuras, e sustentáveis, em outras localidades. Sobretudo para áreas encontradas no mundo periférico, subdesenvolvido ou no Sul do Mundo, caracterizadas pela presença de populações tradicionais com seu rico e variado conhecimento, oralmente transmitido, adquirido através de suas observações práticas e cotidianas com seu meio circundante. Porém, este conhecimento encontra-se ameaçado pelo atual padrão de modernização, homogeneização do mundo, que em seu processo de desenvolvimento exclui crenças, valores e percepções de comunidades tradicionais inteiras, uma vez que se caracteriza por ocorrer com ausência de um olhar atento as especificidades locais, onde estas populações são as primeiras a sofrer com o atual modelo imposto que as ignora por completo e, ao mesmo tempo, por ocorrer com anuência do poder público local. Ao ignorá-las, tais ações minam suas formas de subsistência e via de consequência, às exclui das áreas onde sempre viveram e preservaram fazendo com que um importante e aprofundado conhecimento se perca, juntamente com a desintegração de seus laços de sociabilidade. A exemplo do Arquipélago dos Bijagós, Guiné-Bissau, costa Atlântica da África, lugar de existência de importante população tradicional que, a exemplo de outras localidades do mundo periférico, começa a experimentar transformações em seu patrimônio natural e cultural, decorrentes de ações desconectadas da realidade local. Este estudo, acena para uma importante reflexão sobre os impactos observados nesta realidade e os evidenciados na Praia do Francês. Portanto, procura-se aqui demonstrar a importância do conhecimento e das práticas existentes nas populações tradicionais (autóctones) para ações futuras, onde a inclusão deste saber seja prioridade para outras formas de pensar o desenvolvimento; e em seu processo de construção/planejamento este ocorra: com base no respeito à diversidade e com a participação dos mais diferentes e importantes atores. Enfim, um processo de desenvolvimento de base local, endógeno, sustentável.

This thesis aims at improving the knowledge on the post-fire vegetation regeneration. For that, forests and shrublands were studied, after forest fires and experimental fires. Maritime Pine (Pinus pinaster) recruitment after fire was studied. Fire severity was evidenced as a major effect on this process. High crown fire severity can combust the pines, destroying the seed bank and impeding post fire pine recruitment. However, crown combustion also influences the post-fire conditions on the soil surface, since high crown combustion (HCC) will decrease the postfire needle cast. After low crown combustion (LCC) (scorched rather than torched crowns), a considerable needle cover was observed, along with a higher density of pine seedlings. The overall trends of post-fire recruitment among LCC and HCC areas could be significantly attributed to cover by needles, as well by the estimation of fire severity using the diameters of the burned twigs (TSI). Fire increased the germination from the soil seed bank of a Pinus pinaster forest, and the effects were also related with fire severity. The densities of seedlings of the dominant taxa (genus Erica and Calluna vulgaris) were contrastingly affected in relation to the unburned situation, depending on fire severity, as estimated from the degree of fire-induced crown damage (LCC/HCC), as well as using a severity index based on the diameters of remaining twigs (TSI). Low severity patches had an increase in germination density relatively to the control, while high severity patches suffered a reduction. After an experimental fire in a heathland dominated by Pterospartum tridentatum, Erica australis and E. umbellata, no net differences in seedling emergence were observed, in relation to the pre-fire situation. However, rather than having no effect, the heterogeneity of temperatures caused by fire promoted caused divergent effects over the burned plot in terms of Erica australis germination – a progressive increased was observed in the plots were maximum temperature recorded ranged from 29 to 42.5ºC and decreased in plots with maximum temperature ranging from 51.5 to 74.5ºC. In this heathland, the seed density of two of the main species (E. australis and E. umbellata) was higher under their canopies, but the same was not true for P. tridentatum. The understory regeneration in pine and eucalypt stands, 5 to 6 years post fire, has been strongly associated with post-fire management practices. The effect of forest type was, comparatively, insignificant. Soil tilling, tree harvesting and shrub clearance, were linked to lower soil cover percentages. However, while all these management operations negatively affected the cover of resprouters, seeders were not affected by soil tilling. A strong influence of biogeographic region was identified, suggesting that more vulnerable regions may suffer higher effects of management, even under comparatively lower management pressure than more productive regions. This emphasizes the need to adequate post-fire management techniques to the target regions.

Ionic liquids are a class of solvents that, due to their unique properties, have been proposed in the past few years as alternatives to some hazardous volatile organic compounds. They are already used by industry, where it was possible to improve different processes by the incorporation of this kind of non-volatile and often liquid solvents. However, even if ionic liquids cannot contribute to air pollution, due to their negligible vapour pressures, they can be dispersed thorough aquatic streams thus contaminating the environment. Therefore, the main goals of this work are to study the mutual solubilities between water and different ionic liquids in order to infer on their environmental impact, and to propose effective methods to remove and, whenever possible, recover ionic liquids from aqueous media. The liquid-liquid phase behaviour of different ionic liquids and water was evaluated in the temperature range between (288.15 and 318.15) K. For higher melting temperature ionic liquids a narrower temperature range was studied. The gathered data allowed a deep understanding on the structural effects of the ionic liquid, namely the cation core, isomerism, symmetry, cation alkyl chain length and the anion nature through their mutual solubilities (saturation values) with water. The experimental data were also supported by the COnductor-like Screening MOdel for Real Solvents (COSMO-RS), and for some more specific systems, molecular dynamics simulations were also employed for a better comprehension of these systems at a molecular level. On the other hand, in order to remove and recover ionic liquids from aqueous solutions, two different methods were studied: one based on aqueous biphasic systems, that allowed an almost complete recovery of hydrophilic ionic liquids (those completely miscible with water at temperatures close to room temperature) by the addition of strong salting-out agents (Al2(SO4)3 or AlK(SO4)2); and the other based on the adsorption of several ionic liquids onto commercial activated carbon. The first approach, in addition to allowing the removal of ionic liquids from aqueous solutions, also makes possible to recover the ionic liquid and to recycle the remaining solution. In the adsorption process, only the removal of the ionic liquid from aqueous solutions was attempted. Nevertheless, a broad understanding of the structural effects of the ionic liquid on the adsorption process was attained, and a final improvement on the adsorption of hydrophilic ionic liquids by the addition of an inorganic salt (Na2SO4) was also achieved. Yet, the development of a recovery process that allows the reuse of the ionic liquid is still required for the development of sustainable processes.

Photodegradation is considered to be one of the most important processes of elimination of pharmaceutical drugs from natural water matrices. The high consumption and discharge of these substances, in particular antidepressants, to the aquatic environment supports the need to study degradation processes. This dissertation aimed at studying the direct and indirect photodegradation of sertraline, an antidepressant known for its persistence in the environment, and the evaluation of the influence of environmentally relevant factors in its photodegradation. The photodegradation experiments were developed under simulated solar light and the irradiation times converted to summer sunny days (SSD), an equivalent time in natural environmental conditions. The direct photodegradation was evaluated in solutions of sertraline prepared in ultrapure water and the indirect photodegradation was studied through the addition of photosensitizers (humic substances, Fe(III), nitrates and oxygen). Further irradiation studies were perfomed in aqueous samples collected from two wastewater treatment plants, Vouga river and Ria de Aveiro. The samples were chemically characterized (dissolved organic carbon, nitrates and nitrites and iron determination and UV/Vis spectroscopy). The quantification of sertraline was done by HPLC-UV and photoproducts from direct photodegradation were identified by electrospray mass spectrometry. An observed direct photodegradation rate of sertraline of 0.0062 h-1 was determined, corresponding to a half-life time of 111 h (equivalent to 29 SSD). A significant influence of photosensitizers was observed, the best results being achieved in irradiations of sertraline with humic acids, obtaining a half-life time of 12 h. This was attributed to the hydrophobicity of this substance and higher absortivity in the UV/Vis wavelength, which promote processes of indirect photodegradation. The degradation of sertraline in natural samples was also enhanced comparatively to the direct photodegradation, achieving half-life times between 10 and 25h; the best results were achieved in samples from the primary treatment of a wastewater treatment plant and Ria de Aveiro, with half-life times of 10 and 16 h, respectively. A total of six photoproducts formed during the direct photodegradation of sertraline were identified, three of which were not yet identified in the literature. The main factors contributing to the degradation of sertraline were analysed but this was not fully accomplished, requiring further studies of the composition of the natural matrices and the combined influence of distinct photosensitizers during the irradiation. Nevertheless, it was concluded that the photodegradation of sertraline is greatly influenced by indirect photodegradation processes, promoted by the presence of photosensitizers.

Although overfishing is a concern for many fish stocks, it was for a long time only associated with commercial fishing exploitation, with less or no attention being given to the recreational fisheries. Recent research has shown however that the impact of recreational fishing on particular species can be considerable, and that the recreational harvest needs to be taken into account if fisheries are to be accurately assessed and effectively managed. In Portugal, the first recreational fishing regulations were only recently implemented. However, mirroring other European countries, regulations lacked scientific support, and specific knowledge of the activity was limited to a few studies with limited coverage. This thesis aimed to characterize the biological and socioeconomic aspects of the recreational shore angling activity in southern Portugal, to investigate whether the regulations in place were adequate and effective, and to provide recommendations for improved management and conservation of the inshore fisheries resources. A combined aerial-roving survey was conducted to gather data on fishing effort, catch, fishing trips and socioeconomic aspects (including anglers’ perceptions of regulations) of the recreational angling activity. The analysis of anglers’ catches suggested that compliance with daily bag limits was high, with less than 0.5% of creels exceeding the 10 kg angler-1 day-1 bag limit. Overall, 11.5% of the retained fishes were undersized, but non-compliance with minimum size limits was found to be high for some species (e.g. seabass, 73% undersized). In terms of the impact of recreational shore angling, the total estimated catches corresponded to less than 1% of the commercial landings for the same period (shared species). However, shore angling catches for white sea bream (Diplodus sargus) were found to be considerable, corresponding to 65% of the commercial landings (39.4% of total catch). In terms of anglers’ perceptions about the recreational fishing regulations in Portugal, the present study has shown that the majority of anglers accepted the existence of some kind of SRF regulations, but in general there was a partial or total disagreement with the recreational fishing restrictions recently put in place. Most anglers perceived themselves as not being involved in the decision-making process and claimed that some restrictions lacked a meaningful rationale (e.g. prohibition of fishing from piers/jetties). Fishers’ awareness with regard to specific aspects of the restrictions (such as the rationale for minimum size limits) was found to be very limited. During the same period, catches from sport fishing competitions were examined to test for differences with the recreational activity in terms of catches, and evaluate long term trends in catch and mean size of fish. Catches of the sport fishing competitions were found to be different from those observed for recreational fishing, being dominated by different species (e.g. garfish, mullets), and suggesting different fishing strategies of the the two types of anglers. High percentages of undersized fish were observed to be captured (and retained) during the competitions (in particular seabass, with 100% undersized), probably as a result of a single allowable minimum size (AMS) of 15 cm for all species in use in competitions. Lastly, catch and release fishing experiments were carried out to assess post-release mortality of three recreationally important species: two banded sea bream Diplodus vulgaris; black sea bream Spondyliosoma cantharus; and gilthead sea bream Sparus aurata. Post-release mortalities were found to be low (0-12%). The main predictor of mortality for Sparus aurata was anatomical hooking location, with 63% of the fishes that died being deeply hooked. The results support the release of fish, either from mandatory (e.g. minimum landing sizes) or voluntary practices. In summary, this thesis has demonstrated that the impact of recreational fishing for particular species is significant and needs to be taken into account for more effective management and stock assessment purposes. It has also highlighted several management issues that should be addressed in order to promote more adequate regulations in the future and prevent noncompliance issues. A periodic monitoring of the recreational fishing activity including all fishing modes (i.e. spear fishing, boat, and shore angling) would also be beneficial to ensure a timely knowledge on the global recreational fishing activity and support future management actions.

A crescente saturação dos mercados com produtos similares aos já existentes ou cuja adaptação ao utilizador se revelou ineficiente, torna pertinente a elaboração de projectos que se traduzam em produtos que, efectivamente, tragam vantagem competitiva e, consequentemente, contribuam para o desenvolvimento e bem estar tanto da sociedade como do ecossistema. Como tal, a actividade do design deve assumir uma posição estratégica por forma a conseguir conceber soluções que preservem tanto quanto possível, os recursos naturais ao mesmo tempo que melhoram os contextos físicos e sociais dos seus utilizadores. Assim, com este trabalho pretende-se elaborar uma estratégia para conseguir um processo de desenvolvimento de produto optimizado que englobe, de igual forma, as preocupações ambientais e o utilizador. Portanto, numa primeira fase são revistos os princípios de ecodesign, segue-se o estudo das áreas vocacionadas para o utilizador, que dizem respeito a ergonomia e, consequentemente, a usabilidade. E por fim é formulada uma metodologia projectual adaptada dos estudos revistos, que pretende dar resposta a este sistema de projecto optimizado. A metodologia é depois validada, com a sua implementação num projecto real, de redesign de um aspirador de uso quotidiano. Esta metodologia foi dividida em duas fases principais de pesquisa: (i) o reconhecimento das necessidades reais dos utilizadores, bem como problemas associados ao produto, por meio de pesquisas e testes de usabilidade (N=120), e (ii) a quantificação do impacto ambiental do produto (através do método do eco-indicador 99). Os dados globais permitem definir os requisitos do produto e delinear os critérios a serem incorporados no redesign do produto e também as medidas para melhorar a usabilidade que foram consistentes com as soluções de concepção ecológica.
