Complete blood counts and hemoglobin isoform data were gathered from 36 specimens of the turtle species Phrynops geoffroanus from the northwestern region of São Paulo State, Brazil. They were collected in an urban area. The hemoglobin profiles were obtained after red blood cell lysis and by electrophoretic migration in alkaline pH, acid pH, and neutral pH buffer. The hemoglobin components were confirmed using high-performance liquid chromatography (HPLC). Erythrogram analysis included hematocrit, total hemoglobin concentration, total red blood cell count, and red blood cell indices. The leukogram included a total white blood cell count and a calculation of the percent values of neutrophils, lymphocytes, monocytes, basophils, eosinophils, heterophils, and azurophils. HPLC analysis revealed three hemoglobin components; the first with a concentration of 5.5%, the second was a major component with an average concentration of 67.1%, and the third with a concentration of 28.5%. The hematological profile obtained for these specimens allowed us to establish a pattern for P. geoffroanus in São Paulo State Northwestern region. The average hematocrit values were 22.5% for females and 24.0% for males. For total hemoglobin, we found average values of 6.66 g/dL in females and 7.22 g/dL in males. The number of white blood cells was 2725 x 103/μL for females and 2775 x 103/μL for males. There was a predominance of heterophils, eosinophils, and monocytes in both sexes. No significant differences were found between males and females for hematological profile. The hematological results were compared to literature data for other Chelonia. They were similar to what is known for fresh water turtles. © FUNPEC-RP.

Doxorubicin (DOX) is an anthracycline antibiotic with a broad antitumor spectrum. However, the clinical use of DOX is limited because of its cardiotoxicity, a dose-dependent effect. Colloidal drug delivery systems, such as microemulsions (MEs), allow the incorporation of drugs, modifying the pharmacokinetic (PK) profile and toxic effects. In this study, we evaluated the PK profile and cardiotoxicity of a new DOX ME (DOX-ME). The PK profile of DOX-ME was determined and compared with that of the conventional DOX after single-dose administration (6mg/kg, intravenous) in male Wistar rats (n = 12 per group). The cardiotoxicity of DOX formulations was evaluated by serum creatine kinase MB (CKMB) activity in both animal groups before and after drug administration. The plasma DOX measurements were performed by high-performance liquid chromatography with fluorescence detection, and the CKMB levels were assayed using the CKMB Labtest® kit. The ME system showed a significant increase in plasma DOX concentrations and lower distribution volume when compared with conventional DOX. Serum CKMB activity increased after conventional DOX administration but was unchanged in the DOX-ME group. These results demonstrate modifications in drug access to susceptible sites using DOX-ME. DOX-ME displayed features that make it a promising system for future therapeutic application. © 2012 Wiley Periodicals, Inc.

The aim of this study was to evaluate the effects of exercise prior to or during pregnancy on maternal reproductive outcome, biochemical profile, and on fetal anomaly frequency in a rat pregnancy model utilizing chemically induced diabetes. Wistar rats (minimum n = 11 animals/group) were randomly assigned the following groups: group 1 (G1), sedentary, nondiabetic; G2, nondiabetic, exercised during pregnancy; G3, nondiabetic, exercised prior to and during pregnancy; G4, sedentary, diabetic; G5, diabetic, exercised during pregnancy; and G6, diabetic, exercised prior to and during pregnancy. A swimming program was utilized for moderate exercise. On day 21 of pregnancy, all rats were anesthetized to obtain blood for biochemical measurements. The gravid uterus was weighed with its contents, and the fetuses were analyzed. The nondiabetic rats exercised prior to pregnancy presented a reduced maternal weight gain. Besides, G2 and G3 groups showed decreased fetal weights at term pregnancy, indicating slight intrauterine growth restriction (IUGR). In the diabetic dams, the swimming program did not have antihyperglycemic effects. The exercise applied only during pregnancy caused severe IUGR, as confirmed by reduced fetal weight mean, fetal weight classification, and ossification sites. Nevertheless, exercise was not a teratogenic factor and improved the rats' lipid profiles, demonstrating that the exercise presented possible benefits, but there are also risks prior and during pregnancy, especially in diabetic pregnant women. © The Author(s) 2012.

Objective Experimental studies have shown that exposure to cigarette smoke has negative effects on lipid metabolism and oxidative stress status. Cigarette smoke exposure in nonpregnant and pregnant rats causes significant genotoxicity (DNA damage). However, no previous studies have directly evaluated the effects of obesity or the association between obesity and cigarette smoke exposure on genotoxicity. Therefore, the aim of the present investigation was to evaluate DNA damage levels, oxidative stress status and lipid profiles in obese Wistar rats exposed to cigarette smoke. Design and Methods Female rats subcutaneously (sc) received a monosodium glutamate solution or vehicle (control) during the neonatal period to induce obesity. The rats were randomly distributed into three experimental groups: control, obese exposed to filtered air, and obese exposed to tobacco cigarette smoke. After a 2-month exposure period, the rats were anesthetized and killed to obtain blood samples for genotoxicity, lipid profile, and oxidative stress status analyses. Results The obese rats exposed to tobacco cigarette smoke presented higher DNA damage, triglycerides, total cholesterol, free fatty acids, VLDL-c, HDL-c, and LDL-c levels compared to control and obese rats exposed to filtered air. Both obese groups showed reduced SOD activity. These results showed that cigarette smoke enhanced the effects of obesity. Conclusion In conclusion, the association between obesity and cigarette smoke exposure exacerbated the genotoxicity, negatively impacted the biochemical profile and antioxidant defenses and caused early glucose intolerance. Thus, the changes caused by cigarette smoke exposure can trigger the earlier onset of metabolic disorders associated with obesity, such as diabetes and metabolic syndrome. O depósito mineral de Sapucaia, situado no município de Bonito, região nordeste do Estado do Pará, é parte de um conjunto de ocorrências de fosfatos de alumínio lateríticos localizados predominantemente ao longo da zona costeira dos estados do Pará e Maranhão. Estes depósitos foram alvos de estudo desde o início do século passado, quando as primeiras descrições de “bauxitas fosforosas” foram mencionadas na região NW do Maranhão. Nas últimas décadas, com o crescimento acentuado da demanda por produtos fertilizantes pelo mercado agrícola mundial, diversos projetos de exploração mineral foram iniciados ou tiveram seus recursos ampliados no território brasileiro, dentre estes destaca-se a viabilização econômica de depósitos de fosfatos aluminosos, como o de Sapucaia, que vem a ser o primeiro projeto econômico mineral de produção e comercialização de termofosfatos do Brasil. Este trabalho teve como principal objetivo caracterizar a geologia, a constituição mineralógica e a geoquímica do perfil laterítico alumino-fosfático do morro Sapucaia. A macrorregião abrange terrenos dominados em sua maioria por rochas pré-cambrianas a paleozóicas, localmente definidas pela Formação Pirabas, Formação Barreiras, Latossolos e sedimentos recentes. A morfologia do depósito é caracterizada por um discreto morrote alongado que apresenta suaves e contínuos declives em suas bordas, e que tornam raras as exposições naturais dos horizontes do perfil laterítico. Desta forma, a metodologia aplicada para a caracterização do depósito tomou como base o programa de pesquisa geológica executada pela Fosfatar Mineração, até então detentora dos respectivos direitos minerais, onde foram disponibilizadas duas trincheiras e amostras de 8 testemunhos de sondagem. A amostragem limitou-se à extensão litológica do perfil laterítico, com a seleção de 44 amostras em intervalos médios de 1m, e que foram submetidas a uma rota de preparação e análise em laboratório. Em consonância com as demais ocorrências da região do Gurupi, os fosfatos de Sapucaia constituem um horizonte individualizado, de geometria predominantemente tabular, denominado simplesmente de horizonte de fosfatos de alumínio ou crosta aluminofosfática, que varia texturalmente de maciça a cavernosa, porosa a microporosa, que para o topo grada para uma crosta ferroalumino fosfática, tipo pele-de-onça, compacta a cavernosa, composta por nódulos de hematita e/ou goethita cimentados por fosfatos de alumínio, com características similares aos do horizonte de fosfatos subjacente. A crosta aluminofosfática, para a base do perfil, grada para um espesso horizonte argiloso caulinítico com níveis arenosos, que repousa sobre sedimentos heterolíticos intemperizados de granulação fina, aspecto argiloso, por vezes sericítico, intercalados por horizontes arenosos, e que não possuem correlação aparente com as demais rochas aflorantes da geologia na região. Aproximadamente 40% da superfície do morro é encoberta por colúvio composto por fragmentos mineralizados da crosta e por sedimentos arenosos da Formação Barreiras. Na crosta, os fosfatos de alumínio estão representados predominantemente pelo subgrupo da crandallita: i) série crandallita-goyazita (média de 57,3%); ii) woodhouseíta-svanbergita (média de 15,8%); e pela iii) wardita-millisita (média de 5,1%). Associados aos fosfatos encontram-se hematita, goethita, quartzo, caulinita, muscovita e anatásio, com volumes que variam segundo o horizonte laterítico correspondente. Como os minerais pesados em nível acessório a raro estão zircão, estaurolita, turmalina, anatásio, andalusita e silimanita. O horizonte de fosfatos, bem como a crosta ferroalumínio-fosfática, mostra-se claramente rica em P2O5, além de Fe2O3, CaO, Na2O, SrO, SO3, Th, Ta e em terras-raras leves como La e Ce em relação ao horizonte saprolítico. Os teores de SiO2 são consideravelmente elevados, porém muito inferiores aqueles identificados no horizonte argiloso sotoposto. No perfil como um todo, observa-se uma correlação inversa entre SiO2 e Al2O3; entre Al2O3 e Fe2O3, e positiva entre SiO2 e Fe2O3, que ratificam a natureza laterítica do perfil. Diferente do que é esperado para lateritos bauxíticos, os teores de P2O5, CaO, Na2O, SrO e SO3 são fortemente elevados, concentrações consideradas típicas de depósitos de fosfatos de alumínio ricos em crandallita-goyazita e woodhouseítasvanbergita. A sucessão dos horizontes, sua composição mineralógica, e os padrões geoquímicos permitem correlacionar o presente depósito com os demais fosfatos de alumínio da região, mais especificamente Jandiá (Pará) e Trauíra (Maranhão), bem como outros situados além do território brasileiro, indicando portanto, que os fosfatos de alumínio de Sapucaia são produtos da gênese de um perfil laterítico maturo e completo, cuja rocha fonte pode estar relacionada a rochas mineralizadas em fósforo, tais como as observadas na Formação Pimenteiras, parcialmente aflorante na borda da Bacia do Parnaíba. Possivelmente, o atual corpo de minério integrou a paleocosta do mar de Pirabas, uma vez que furos de sondagem às proximidades do corpo deixaram claro a relação de contato lateral entre estas unidades.

BACKGROUND: Chronic leg ulcer may have an impact on patients' quality of life.OBJECTIVES:This study aimed to identify the impact of leg ulcers on patient's quality of life using the Dermatology Life Quality Index and to define the main factors correlated with this perception.METHOD:Cross-sectional, non-probabilistic sampling study. We included patients with chronic leg ulcers being treated for at least 3 months. A sociodemographic and clinical survey was conducted to assess the profile of the ulcers. We administered a screening for depressive symptoms and the Dermatology Life Quality Index. We performed a descriptive statistical analysis, chi-square test and Mann-Whitney test for categorical data, Pearson for numeric variables, and multiple regression for categorical data.RESULTS:Forty-one patients were assessed. Their mean age was 61.78 years. Venous ulcers (48.8%) were the most prevalent. Seventy-three percent of the sample perceived no impact/low impact on quality of life in the past week, and 26.8% perceived moderate/high impact. A multiple regression analysis identified the causes of lesion, pain related to the ulcers, time of onset, and severity of the depressive symptoms as the variables that had an influence on quality of life.CONCLUSIONS:The majority of the sample perceived low or no impact of the condition on the quality of the life. The variables etiology of the lesion (p<0.001), pain related to the ulcers (p=0.001), time of onset (p=0.006), and severity of the depressive symptoms (p<0.001) had an influence on the quality of life, suggesting the need for further studies with more robust designs to confirm the causal relationship between these characteristics and quality of life.

This manuscript aims proposing a methodology for correlating soil porosity to the respective geological units using geostatistical analysis techniques, including interpolation data by kriging. The site studied was in Lorena municipality, Paraíba do Sul Valley, southeastern Brazil. Specifically all studies were carried out within an area of 12 km2 located at Santa Edwirges farm. The database comprehended 41 soil samples taken at different geological and geomorphologic units at three different depths: surface, 50 cm and 100 cm depth. The geostatistical analyses results were correlated to a geological mapping specifically elaborated for the site. This mapping accounts for two different geological formations and a geological contact characterized by a shearing zone. The results indicate the existence of a significant relationship between the soil porosity and the respective geological units. The studies revealed that the residual soils from weathered granitic rocks tend to have higher porosities than the residual soils from weathered biotite gneiss rocks, while the soil porosity within the shearing zone is relatively un-sensitive to the respective geological formation. The spatial patterns observed were efficient to evaluate the relationship between the soil porosity, geology unit and the and geomorphology showing a good potential for correlating with others soil properties such as hydraulic conductivity, soil water retention curves and erosion potentials.

One of the main challenges in the mining sector at present, related to the optimization of mineral assets, consists in to find the best proportion of each geological material to be mined and homogenized to ensure high productivity and stability in ore processing plants. The stability of a mineral processing plant is factor for achieving operational excellence, as a stable performance of the plants ensures economy and adherence to financial planning businesses. Is already well Know that the feed rate of the processing plants is dependent on the hardness and quality of the ore. Therefore, the ore behavior predictability is the solution to ensure the stability of mineral processing plants. This work was developed in Anglo American Phosphate Brazil Ltda., at the Fazenda Chapadão Phosphate Mine, located in the southeastern portion of the state of Goiás, in the municipalities of Catalão and Ouvidor. The phosphate ore is the result of supergene apatite concentration in the weathering profile of the alkaline ultramafic phlogopitized rocks, intruded by foscoritic and carbonatitic rocks series. The weathering profile has about 100 meters thick and is divided into three basic types, from top to bottom, coinciding with decreasing intensity of weathering: oxidized ore, micaceous ore and weathered rock. In each portion of this weathering profile, mainly near the contacts between these three divisions, are hard materials, cemented by minerals such as apatite, quartz, goethite, monazite and barite. The different features presented by these materials makes to be vital the correct identification, quantification and location of different hard materials in the mine area, to maximize its use. The main objective of this study was to define the type of cementation of hard materials which predominates in each portion of the weathering profile in the mine, as well as their relationship with the protolith. Proper ion channels’ functioning is a prerequisite for a normal cell and disorders involving ion channels, or channelopathies, underlie many human diseases. Long QT syndromes (LQTS) for example may arise from the malfunctioning of hERG channel, caused either by the binding of drugs or mutations in HERG gene. In the first part of this thesis I present a framework to investigate the mechanism of ion conduction through hERG channel. The free energy profile governing the elementary steps of ion translocation in the pore was computed by means of umbrella sampling simulations. Compared to previous studies, we detected a different dynamic behavior: according to our data hERG is more likely to mediate a conduction mechanism which has been referred to as “single-vacancy-like” by Roux and coworkers (2001), rather then a “knock-on” mechanism. The same protocol was applied to a model of hERG presenting the Gly628Ser mutation, found to be cause of congenital LQTS. The results provided interesting insights about the reason of the malfunctioning of the mutant channel. Since they have critical functions in viruses’ life cycle, viral ion channels, such as M2 proton channel, are considered attractive targets for antiviral therapy. A deep knowledge of the mechanisms that the virus employs to survive in the host cell is of primary importance in the identification of new antiviral strategies. In the second part of this thesis I shed light on the role that M2 plays in the control of electrical potential inside the virus, being the charge equilibration a condition required to allow proton influx. The ion conduction through M2 was simulated using metadynamics technique. Based on our results we suggest that a potential anion-mediated cation-proton exchange, as well as a direct anion-proton exchange could both contribute to explain the activity of the M2 channel.

Summary PhD Thesis Jan Pollmann: This thesis focuses on global scale measurements of light reactive non-methane hydrocarbon (NMHC), in the volatility range from ethane to toluene with a special focus on ethane, propane, isobutane, butane, isopentane and pentane. Even though they only occur at the ppt level (nmol mol-1) in the remote troposphere these species can yield insight into key atmospheric processes. An analytical method was developed and subsequently evaluated to analyze NMHC from the NOAA – ERSL cooperative air sampling network. Potential analytical interferences through other atmospheric trace gases (water vapor and ozone) were carefully examined. The analytical parameters accuracy and precision were analyzed in detail. It was proven that more than 90% of the data points meet the Global Atmospheric Watch (GAW) data quality objective. Trace gas measurements from 28 measurement stations were used to derive the global atmospheric distribution profile for 4 NMHC (ethane, propane, isobutane, butane). A close comparison of the derived ethane data with previously published reports showed that northern hemispheric ethane background mixing ratio declined by approximately 30% since 1990. No such change was observed for southern hemispheric ethane. The NMHC data and trace gas data supplied by NOAA ESRL were used to estimate local diurnal averaged hydroxyl radical (OH) mixing ratios by variability analysis. Comparison of the variability derived OH with directly measured OH and modeled OH mixing ratios were found in good agreement outside the tropics. Tropical OH was on average two times higher than predicted by the model. Variability analysis was used to assess the effect of chlorine radicals on atmospheric oxidation chemistry. It was found that Cl is probably not of significant relevance on a global scale.

The object of this work has been the analysis of natural processes controlling the geological evolution of the Montenegro and Northern Albania Continental Margin (MACM) during the Late Quaternary. These include the modern sediment dispersal system and oceanographic regime, the building and shaping of the shelf margin at the scale of 100 kyr and relative to the most recent transition between glacial and interglacial periods. The analysis of the new data shows that the MACM is a shelf-slope system formed by a suite of physiographic elements, including: an inner and an outer continental shelf, separated by two tectonically-controlled morphological highs; a lobated drowned mid-shelf paleodelta, formed during the last sea level fall and low stand; an upper continental slope, affected by gravity-driven instability and a system of extensional faults with surficial displacement, featuring an orientation coherent with the regional tectonics. The stratigraphic study of the MACM shows a clear correspondence between the Late Pleistocene/Holocene mud-wedge and the low reflectivity sectors of the inner shelf. Conversely, most of the outer shelf and part of the continental slope expose deposits from the last sea level low stand, featuring a general sediment starving condition or the presence of a thin postglacial sediments cover. The MACM shows uplift in correspondence of the Kotor and Bar ridges, and subsidence in the outer shelf and upper slope sectors. In fact, seaward of these tectonic ridges, the sparker seismic profile show the presence of four well-defined seismo-stratigraphic sequences, interpreted as forced regression deposits, formed during the last four main glacial phases. In this way, the MACM records the 100 kyr scale sea level fluctuations on its seismo-stratigraphic architecture over the last 350 kyr. Over such time range, through the identification of the paleoshoreline deposits, we estimated an average subsidence rate of about 1.2 mm/yr.

Chlorinated solvents are the most ubiquitous organic contaminants found in groundwater since the last five decades. They generally reach groundwater as Dense Non-Aqueous Phase Liquid (DNAPL). This phase can migrate through aquifers, and also through aquitards, in ways that aqueous contaminants cannot. The complex phase partitioning to which chlorinated solvent DNAPLs can undergo (i.e. to the dissolved, vapor or sorbed phase), as well as their transformations (e.g. degradation), depend on the physico-chemical properties of the contaminants themselves and on features of the hydrogeological system. The main goal of the thesis is to provide new knowledge for the future investigations of sites contaminated by DNAPLs in alluvial settings, proposing innovative investigative approaches and emphasizing some of the key issues and main criticalities of this kind of contaminants in such a setting. To achieve this goal, the hydrogeologic setting below the city of Ferrara (Po plain, northern Italy), which is affected by scattered contamination by chlorinated solvents, has been investigated at different scales (regional and site specific), both from an intrinsic (i.e. groundwater flow systems) and specific (i.e. chlorinated solvent DNAPL behavior) point of view. Detailed investigations were carried out in particular in one selected test-site, known as “Caretti site”, where high-resolution vertical profiling of different kind of data were collected by means of multilevel monitoring systems and other innovative sampling and analytical techniques. This allowed to achieve a deep geological and hydrogeological knowledge of the system and to reconstruct in detail the architecture of contaminants in relationship to the features of the hosting porous medium. The results achieved in this thesis are useful not only at local scale, e.g. employable to interpret the origin of contamination in other sites of the Ferrara area, but also at global scale, in order to address future remediation and protection actions of similar hydrogeologic settings.

Outcome-dependent, two-phase sampling designs can dramatically reduce the costs of observational studies by judicious selection of the most informative subjects for purposes of detailed covariate measurement. Here we derive asymptotic information bounds and the form of the efficient score and influence functions for the semiparametric regression models studied by Lawless, Kalbfleisch, and Wild (1999) under two-phase sampling designs. We show that the maximum likelihood estimators for both the parametric and nonparametric parts of the model are asymptotically normal and efficient. The efficient influence function for the parametric part aggress with the more general information bound calculations of Robins, Hsieh, and Newey (1995). By verifying the conditions of Murphy and Van der Vaart (2000) for a least favorable parametric submodel, we provide asymptotic justification for statistical inference based on profile likelihood.
