The physiological state of yeast cells changes during culture growth as a consequence of environmental changes (nutrient limitations, pH and metabolic products). Cultures that grow exponentially are heterogeneous cell populations made up of cells regulated by different metabolic and/or genetic control systems. The strain of baker's yeast selected by plating commercial compressed yeast was used for the production of glycerol-3- phosphate dehydrogenase. Glycerol-3-phosphate dehydrogenase (GPD) has been widely used in the enzyme assays with diverse compounds of industrial interest, such as glycerol or glycerol phosphate, as well as a number of important bioanalytical applications. Each cell state determines the level of key enzymes (genetic control), fluxes through metabolic pathways (metabolic control), cell morphology and size. The present study was carried out to determine the effects of environmental conditions and carbon source on GPD production from baker's yeast. Glucose, glycerol, galactose and ethanol were used as carbon sources. Glycerol and ethanol assimilations required agitation, which was dependent on the medium volume in the fermentation flask for the greatest accumulation of intracellular GPD. Enzyme synthesis was also affected by the initial pH of the medium and inoculum size. The fermentation time required for a high level of enzyme formation decreased with the inoculum size. The greatest amount of enzyme (0.45 U/ml) was obtained with an initial pH of 4.5 in the medium containing ethanol or glycerol. The final pH was maintained in YP-ethanol, but in the YP-glycerol the final pH increased to 6.9 during growth.

Oxidative stress is a key component in the immunosuppression of chronic kidney disease (CKD), and neutrophil function may be impaired by oxidative stress. To test the hypothesis that in uremic dogs with CKD, oxidative stress is increased and neutrophils become less viable and functional, 18 adult dogs with CKD were compared with 15 healthy adult dogs. Blood count and urinalysis were done, and the serum biochemical profile and plasma lipid peroxidation (measurement of thiobarbituric acid reactive substances) were determined with the use of commercial reagents. Plasma total antioxidant capacity (TAC) was measured with a spectrophotometer and commercial reagents, superoxide production with a hydroethidine probe, and the viability and apoptosis of neutrophils with capillary flow cytometry and the annexin V-PE system. The plasma concentrations of cholesterol (P = 0.0415), creatinine (P < 0.0001), and urea (P < 0.0001) were significantly greater in the uremic dogs than in the control dogs. The hematocrit (P = 0.0004), urine specific gravity (P = 0.015), and plasma lipid peroxidation (P < 0.0001) were significantly lower in the dogs that were in late stages of CKD than in the control group. Compared with those isolated from the control group, neutrophils isolated from the CKD group showed a higher rate of spontaneous (0.10 ± 0.05 versus 0.49 ± 0.09; P = 0.0033; median ± standard error of mean) and camptothecin-induced (18.53 ± 4.06 versus 44.67 ± 4.85; P = 0.0066) apoptosis and lower levels of superoxide production in the presence (1278.8 ± 372.8 versus 75.65 ± 86.6; P = 0.0022) and absence (135.29 ± 51.74 versus 41.29 ± 8.38; P = 0.0138) of phorbol-12-myristate-13-acetate stimulation. Thus, oxidative stress and acceleration of apoptosis occurs in dogs with CKD, the apoptosis diminishing the number of viable neutrophils and neutrophil superoxide production.

The lectin Artin M has been shown to accelerate the wound-healing process. The aims of this study were to evaluate the effects of Artin M on wound healing in the palatal mucosa of rats and to investigate the effects of Artin M on transforming growth factor beta (TGF-β) and vascular endothelial growth factor (VEGF) secretion by rat gingival fibroblasts. A surgical wound was created on the palatal mucosa of 72 rats divided into three groups according to treatment: C - Control (nontreated), A - Artin M gel, and V - Vehicle. Eight animals per group were sacrificed at 3, 5, and 7 days postsurgery for histology, immunohistochemistry and determination of the levels of cytokines, and growth factors. Gingival fibroblasts were incubated with 2.5 μg/mL of Artin M for 24, 48, and 72 hours. The expression of VEGF and TGF-β was determined by enzyme-linked immunosorbent assay. Histologically, at day 7, the Artin M group showed earlier reepithelialization, milder inflammatory infiltration, and increased collagen fiber formation, resulting in faster maturation of granular tissue than in the other groups (p < 0.05). Artin M-induced cell proliferation in vivo and promoted a greater expression of TGF-β and VEGF in both experiments (p < 0.05). Artin M was effective in healing oral mucosa wounds in rats and was associated with increased TGF-β and VEGF release, cell proliferation, reepithelialization, and collagen deposition and arrangement of fibers. © 2013 by the Wound Healing Society.

The açai palm is a Brazilian native species and economically important, grown for the hearts of palm and fruit extraction, as well as the seedling production for ornamental purposes. The weed control is one of the concerns observed in seedling production, which has the artillery plant (Pilea microphylla) with the major and widespread occurrence. Thus, the aim of this trial was to evaluate the selectivity and efficacy of oxyfluorfen in artillery plant control in açai (Euterpe oleracea) seedlings. The experimental design was completely randomized. The treatments were arranged in a 2 x 3 factorial scheme, using 2 types of herbicide application: broadcast spraying (sprayed over total area) and spot spraying (directed to the target) and 3 oxyfluorfen dosages (0, 72 and 144 g of active ingredient ha-1) with four replications. Each experimental unit consisted by one açai palm pot infested by artillery plant. They ere visually evaluated for seedling toxicity and the weed control percentage, at 7, 15, 30, 45 and 60 days after application (DAA). There was 100% control of artillery plant from 15 to 60 DAA for the two tested dosages, regardless of application type. However, 100% control was observed at 7 days, using 1.5 L ha-1 in the spot type. The effects of phytotoxicity from 30 to 60 days was moderate, regardless the application method or dosage.

A search for supersymmetry is presented based on events with large missing transverse energy, no isolated electron or muon, and at least three jets with one or more identified as a bottom-quark jet. A simultaneous examination is performed of the numbers of events in exclusive bins of the scalar sum of jet transverse momentum values, missing transverse energy, and bottom-quark jet multiplicity. The sample, corresponding to an integrated luminosity of 19.4fb-1, consists of proton-proton collision data recorded at a center-of-mass energy of 8TeV with the CMS detector at the LHC in 2012. The observed numbers of events are found to be consistent with the standard model expectation, which is evaluated with control samples in data. The results are interpreted in the context of two simplified supersymmetric scenarios in which gluino pair production is followed by the decay of each gluino to an undetected lightest supersymmetric particle and either a bottom or top quark-antiquark pair, characteristic of gluino mediated bottom- or top-squark production. Using the production cross section calculated to next-to-leading-order plus next-to-leading-logarithm accuracy, and in the limit of a massless lightest supersymmetric particle, we exclude gluinos with masses below 1170GeV and 1020GeV for the two scenarios, respectively. © 2013 CERN.

Measuring shikimic acid accumulation in response to glyphosate applications can be a rapid and accurate way to quantify and predict glyphosate-induced damage to sensitive plants. The objective of this paper was to evaluate the effect of cover crop termination timing by glyphosate application on rice (Oryza sativa L.) yield in a no-till system. A factorial experiment, arranged in a split-plot design, was conducted for 2 yr. Treatments consisted of cover crops (main plots) and timed herbicide applications (subplots) to these cover crops (30, 20, 10, and 0 d before rice planting). There was a decrease in rice yield from 2866 kg ha-1 to 2322 kg ha-1 when the herbicide was applied closer to the rice planting day. Glyphosate application on cover crops increased shikimate concentrations in rice seedlings cultivated under palisade grass (Brachiaria brizantha), signal grass (B. ruziziensis), guinea grass (Panicum maximum), and weedy fallow (spontaneous vegetation) but not under millet (Pennisetum glaucum), which behaved similarly to the control (clean fallow, no glyphosate application). Glyphosate applications in the timing intervals used were associated with stress in the rice plants, and this association increased if cover crops took longer to completely dry and if higher amounts of biomass were produced. Millet, as a cover crop, allowed the highest seedling dry matter for upland rice and the highest rice yield. Our results suggest that using millet as a cover crop, with glyphosate application far from upland rice planting day (10 d or more), was the best option for upland rice under a no-tillage system. © Crop Science Society of America.

Listeria monocytogenes, considered as one of the most important foodborne pathogens, is easily found on surfaces, particularly in the form of a biofilm. Biofilms are aggregates of cells that facilitate the persistence of these pathogens in food processing environments conferring resistance to the processes of cleaning and may cause contamination of food during processing, thus, representing a danger to public health. Little is known about the dynamics of the formation and regulation of biofilm production in L.monocytogenes, but several authors reported that the luxS gene may be a precursor in this process. In addition, the product of the inlA gene is responsible for facilitating the entry of the microorganism into epithelial cells that express the receptor E-cadherin, also participates in surface attachment. Thus, 32 strains of L.monocytogenes isolated from different foods (milk and vegetables) and from food processing environments were analyzed for the presence of these genes and their ability to form biofilms on three different surfaces often used in the food industry and retail (polystyrene, glass and stainless steel) at different temperatures (4, 20 and 30°C). All strains had the ilnA gene and 25 out of 32 strains (78.1%) were positive for the presence of the luxS gene, but all strains produced biofilm in at least one of the temperatures and materials tested. This suggests that genes in addition to luxS may participate in this process, but were not the decisive factors for biofilm formation. The bacteria adhered better to hydrophilic surfaces (stainless steel and glass) than to hydrophobic ones (polystyrene), since at 20°C for 24h, 30 (93.8%) and 26 (81.3%) produced biofilm in stainless steel and glass, respectively, and just 2 (6.2%) in polystyrene. The incubation time seemed to be an important factor in the process of biofilm formation, mainly at 35°C for 48h, because the results showed a decrease from 30 (93.8%) to 20 (62.5%) and from 27 (84.4%) to 12 (37.5%), on stainless steel and glass, respectively, although this was not significant (. p=0.3847). We conclude that L.monocytogenes is capable of forming biofilm on different surfaces independent of temperature, but the surface composition may be important factor for a faster development of biofilm. © 2013 Elsevier Ltd.

This study aimed at evaluating total or partial replacement of corn by sorghum in broiler diets and at estimating the effect of the pigment supplementation on broiler performance, carcass and cuts yield, and possible breast and leg pigmentation. We used 1680 one-d-old Ross® 308 broilers. Birds were sexed and distributed according to a completely randomized design (2 x 3 + 1). Treatments consisted of a control diet based on corn and diets with two levels corn replacement by sorghum (50 and 100%) and three pigments levels, with four replicates of 60 birds per treatment. There was no effect (p > 0.05) of the dietary replacement of corn by sorghum on performance, carcass and parts yield, and no changes in breast and leg meat pH (p >0.05). Meat redness (a*), yellowness (b*) and luminosity (L*) increased (p < 0.05) as pigment inclusion levels increased. It was concluded that the use of sorghum instead of corn did not affect broiler performance or carcass and cuts yield. When adequate pigments were used, meat color significantly improved.

Objectives: To evaluate the antimicrobial activity of Arctium lappa L. extract on Staphylococcus aureus, S. epidermidis, Streptococcus mutans, Candida albicans, C. tropicalis and C. glabrata. In addition, the cytotoxicity of this extract was analyzed on macrophages (RAW 264.7).Design: By broth microdilution method, different concentrations of the extract (250-0.4 mg/mL) were used in order to determine the minimum microbicidal concentration (MMC) in planktonic cultures and the most effective concentration was used on biofilms on discs made of acrylic resin. The cytotoxicity A. lappa L. extract MMC was evaluated on RAW 264.7 by MTT assay and the quantification of IL-1 beta and TNF-alpha by ELISA.Results: The most effective concentration was 250 mg/mL and also promoted significant reduction (log(10)) in the biofilms of S. aureus (0.438 +/- 0.269), S. epiderrnidis (0.377 +/- 0.298), S. mutans (0.244 +/- 0.161) and C. albicans (0.746 +/- 0.209). Cell viability was similar to 100%. The production of IL-beta was similar to the control group (p > 0.05) and there was inhibition of TNF-alpha (p < 0.01).Conclusions: A. lappa L. extract was microbicidal for all the evaluated strains in planktonic cultures, microbiostatic for biofilms and not cytotoxic to the macrophages. A high prevalence of nematodes, especially Haemonchus contortus and Trichostrongylus colubriformis, that exhibit multiple anthelmintics resistance has been reported in sheep in several South American countries. For this reason, the development of strategies that are less dependent on anthelmintic treatments is imperative for the prophylaxis of gastrointestinal nematode infections in small ruminants. Integrated grazing using cattle and sheep can be used for pasture decontamination with considerable reduction of H. contortus and T. colubnformis infective larvae after cattle grazing. Several breeds of sheep exhibit genetically related resistance against nematode infections, as is the case of crioulo, native or naturalised breeds of sheep. These breeds descend from livestock introduced by Portuguese and Spanish settlers and have been submitted to a long process of natural selection in various environmental conditions. In the South, the Crioula Lanada breed is more resistant to H. contortus than are Corriedale sheep. In tropical areas, where the minimum temperatures are usually higher than 20 degrees C, hair sheep flourish, especially the Santa Ines breed, which also display a higher level of resistance to nematode infections compared with certain breeds of European origin. However, Santa Ines sheep have inferior carcass quality compared with other commercial breeds. Recent studies showed that the crossbreeding of Santa Ines ewes with sires of breeds with high potential for growth and meat production, results in crossbred animals with high productivity and a satisfactory degree of resistance against nematode infections. Several studies have indicated that improvement in nutrition has a beneficial effect on the development of resistance in lambs that were naturally or artificially infected with nematodes. Therefore, supplementary feeding and breeding strategies to improve resistance to nematodes are feasible options in the effort to reduce dependence on anthelmintic drugs to control worm infections in sheep.
