In the microvillar microdomain of the kidney brush border, sodium hydrogen exchanger type 3 (NHE3) exists in physical complexes with the serine protease dipeptidyl peptidase IV (DPPIV). The purpose of this study was to explore the functional relationship between NHE3 and DPPIV in the intact proximal tubule in vivo. To this end, male Wistar rats were treated with an injection of the reversible DPPIV inhibitor Lys [Z(NO(2))]-pyrrolidide (I40; 60 mg center dot kg(-1)center dot day(-1) ip) for 7 days. Rats injected with equal amounts of the noninhibitory compound Lys[ Z(NO(2))]-OH served as controls. Na(+) -H(+) exchange activity in isolated microvillar membrane vesicles was 45 +/- 5% decreased in rats treated with I40. Membrane fractionation studies using isopycnic centrifugation revealed that I40 provoked redistribution of NHE3 along with a small fraction of DPPIV from the apical enriched microvillar membranes to the intermicrovillar microdomain of the brush border. I40 significantly increased urine output ( 67 +/- 9%; P < 0.01), fractional sodium excretion ( 63 +/- 7%; P < 0.01), as well as lithium clearance ( 81 +/- 9%; P < 0.01), an index of end-proximal tubule delivery. Although not significant, a tendency toward decreased blood pressure and plasma pH/HCO(3)(-) was noted in I40-treated rats. These findings indicate that inhibition of DPPIV catalytic activity is associated with inhibition of NHE3-mediated NaHCO(3) reabsorption in rat renal proximal tubule. Inhibition of apical Na(+) -H(+) exchange is due to reduced abundance of NHE3 protein in the microvillar microdomain of the kidney brush border. Moreover, this study demonstrates a physiologically significant interaction between NHE3 and DPPIV in the intact proximal tubule in vivo.

The functional versatility of the distal nephron is mainly due to the large cytological heterogeneity of the segment. Part of Na(+) uptake by distal tubules is dependent on Na(+)/H(+). exchanger 2 (NHE2), implicating a role of distal convoluted cells also in acid-base homeostasis. In addition, intercalated (IC) cells expressed in distal convoluted tubules, connecting tubules and collecting ducts are involved in the final regulation of acid-base excretion. IC cells regulate acid-base handling by 2 main transport proteins, a V-type H(+)-ATPase and a Cl/HCO(3)(-) exchanger, localized at different membrane domains. Type A IC cells are characterized by a luminal H(+)-ATPase in series with a basolateral Cl/HCO(3)(-) exchanger, the anion exchanger AE1. Type B IC cells mediate HCO(3)(-) secretion through the apical Cl(-)/HCO(3)(-) exchanger pendrin in series with a H(+)-ATPase at the basolateral membrane. Alternatively, H(+)/K(+)-ATPases have also been found in several distal tubule cells, particularly in type A and B IC cells. All of these mechanisms are finely regulated, and mutations of 1 or more proteins ultimately lead to expressive disorders of acid-base balance.

Nitric oxide (NO) derived from inducible nitric oxide synthase (iNOS) plays an important role in host defense, as well as in inflammation-induced tissue lesions. Here we evaluated the role of NO in bone loss in bacterial infection-induced apical periodontitis by using iNOS-deficient mice (iNOS(-/-)). The iNOS(-/-) mice developed greater inflammatory cell recruitment and osteolytic lesions than WT mice. Moreover, tartrate-resistant acid-phosphatase-positive (TRAP(+)) osteoclasts were significantly more numerous in iNOS-/- mice. Furthermore, the increased bone resorption in iNOS(-/-) mice also correlated with the increased expression of receptor activator NF-kappaB (RANK), stromal-cell-derived factor-1 alpha (SDF-1 alpha/CXCL12), and reduced expression of osteoprotegerin (OPG). These results show that NO deficiency was associated with an imbalance of bone-resorption-modulating factors, leading to severe infection-stimulated bone loss.

Objective. The objective of this study was to evaluate the disinfection degree of dentine caused by the use of diode laser after biomechanical procedures. Study design. Thirty teeth were sectioned and roots were autoclaved and incubated for 4 weeks with a suspension of Enterococcus faecalis. The specimens were randomly divided into 3 groups (n = 10): G1, instrumented with rotary files, irrigated with 0.5% sodium hypochlorite and 17% EDTA-T, and then irradiated by 830-nm diode laser at 3 W; G2, the same procedures as G1 but without laser irradiation; and G3, irrigation with saline solution (control). Dentin samples of each third were collected with carbide burs and aliquots were sowed to count viable cells. Results. The disinfection degree achieved was 100% in G1 and 98.39% in G2, when compared to the control group (G3). Conclusion. Diode laser irradiation provided increased disinfection of the deep radicular dentin in the parameters and samples tested.

Introduction: The aim of the present study was to determine the disinfection of preparations carried out by using the Protaper or MTwo system in canals infected with Enterococcus faecalis. Methods: Twenty-eight distobuccal canals of upper molars were used, in which the canals were sterilized after being enlarged to #20 file and then contaminated with an inoculation of a culture of E. faecalis. After the incubation period, bacterial samples were collected and were seeded on plates for analysis of colony-forming units (CFU)/mL. The teeth were divided into 2 groups according to the rotary system used for instrumentation; 2 noninstrumented teeth served as the control group. Then bacterial samples were collected and were seeded on plates for analysis of CFU/mL again. The data obtained were evaluated by the Wilcoxon and Mann-Whitney U tests. Results: Bacterial reduction was 81.94% and 84.29%, respectively, in Pro Taper and Mtwo systems, and there was no statistically significant difference (P > .05). Conclusions: Both systems, Pro Taper and Mtwo, reduced the amount of bacteria in the mechanical disinfection of the root canal system, demonstrating that they are suitable for this purpose. Introduction: Periapical lesions are chronic inflammatory disorders of periradicular tissues caused by etiologic agents of endodontic origin. The inflammatory chemokines are thought to be involved in the latter observed osteolysis. With a murine model of experimental periapical lesion, the objective of this study was to evaluate the role of the chemokine receptor CCR2 in the lesion progression, osteoclast differentiation and activation, and expression of inflammatory osteolysis-related mediators. Methods: For lesion induction, right mandibular first molars were opened surgically with a (1)/(4) carbine bur, and 4 bacterial strains were inoculated in the exposed dental pulp; left mandibular first molars were used as controls. Animals were killed at 3, 7, 14, and 21 days after surgeries to evaluate the kinetics of lesion development. Results: CCR2 KO mice showed wider lesions than WT mice. CCR2 KO mice also expressed higher levels of the osteoclastogenic and osteolytic factors, receptor activator of nuclear factor kappa B ligand (RANKL) and cathepsin K, of the proinflammatory cytokine tumor necrosis factor alpha, and of the neutrophil migration related chemokine, KC. Conclusions: These results suggest that CCR2 is important in host protection to periapical osteolysis. (J Endod 2010;36:244-250)

Merozoite surface proteins (MSPs) of the malaria parasites are major candidates for vaccine development targeting asexual blood stages. However, the diverse antigenic repertoire of these antigens that induce strain-specific protective immunity in human is a major challenge for vaccine design and often determines the efficacy of a vaccine. Here we further assessed the genetic diversity of Plasmodium vivax MSP4 (PvMSP4) protein using 195 parasite samples collected mostly from Thailand, Indonesia and Brazil. Overall, PvMSP4 is highly conserved with only eight amino acid substitutions. The majority of the haplotype diversity was restricted to the two short tetrapeptide repeat arrays in exon 1 and 2, respectively. Selection and neutrality tests indicated that exon 1 and the entire coding region of PvMSP4 were under purifying selection. Despite the limited nucleotide polymorphism of PvMSP4, significant genetic differentiation among the three major parasite populations was detected. Moreover, microgeographical heterogeneity was also evident in the parasite populations from different endemic areas of Thailand. (C) 2009 Elsevier B.V. All rights reserved.

The activity of the Na(+)/H(+) exchanger NHE3 is regulated by a number of factors including parathyroid hormone (PTH). In the current study, we used a renal epithelial cell line, the opossum kidney (OKP) cell, to elucidate the mechanisms underlying the long-term effects of PTH on NHE3 transport activity and expression. We observed that NHE3 activity was reduced 6 h after addition of PTH, and this reduction persisted almost unaltered after 24 h. The decrease in activity was associated with diminished NHE3 cell surface expression at 6, 16, and 24 h after PTH addition, total cellular NHE3 protein at 16 and 24 h, and NHE3 mRNA abundance at 24 h. The lower levels of NHE3 mRNA were associated to a small, but significant, decrease in mRNA stability. Additionally, by analyzing the rat NHE3 gene promoter activity in OKP cells, we verified that the regulatory region spanning the segment -152 to +55 was mildly reduced under the influence of PTH. This effect was completely abolished by the presence of the PKA inhibitor KT 5720. In conclusion, long-term exposure to PTH results in reduction of NHE3 mRNA levels due to a PKA-dependent inhibitory effect on the NHE3 promoter and a small reduction of mRNA half-life, and decrease in the total amount of protein which is preceded by endocytosis of the apical surface NHE3. The decreased NHE3 expression is likely to be responsible for the reduction of sodium, bicarbonate, and fluid reabsorption in the proximal tubule consistently perceived in experimental models of PTH disorders.

The objective of this study was to evaluate the stress distribution in the resin in contact with the spirals of cylindrical and conical mini-implants, when submitted to lateral load and insertion torsion. A photoelastic model was fabricated using transparent gelatin to simulate the alveolar bone. The model was observed with a plane polariscope and photographically recorded before and after activation of the two screws with a lateral force and torsion. The lateral force application caused bending moments on both mini-implants, with the uprising of fringes or isochromatics, characteristics of stresses, along the threads of the mini-implants and in the apex. When the torsion was exerted in the mini-implants, a great concentration of stress upraised close to the apex. The conclusion was that, comparing conical with cylindrical mini-implants under lateral load, the stresses were similar on the traction sides. The differences appear (1) on the apex, where the cylindrical mini-implant showed a greater concentration of stress, and (2) along the spirals, in the compression side, where the conical mini-implant showed a greater concentration of stress. The greater part of the stress produced by both mini-implants, after torsion load in insertion, were concentrated on the apex. With the cylindrical mini-implant, the greater concentration of tension was close to the apex, while with the conical one, the stresses were distributed along a greater amount of apical threads.

The purpose of this study was to evaluate the effectiveness of different light-curing units on the bond strength (push-out) of glass fiber posts in the different thirds of the root (cervical, middle and apical) with different adhesive luting resin systems (dual-cure total-etch; dual-cured and self-etch bonding system; and dual-cure self-adhesive cements), Disks of the samples (n = 144) were used, with approximately 1 mm of thickness of 48 bovine roots restored with glass fiber posts, that were luted with resin cements photo-activated by halogen LCU (QTH, Optilux 501) and blue LED (Ultraled), with power densities of 600 and 550 mW/cm(2), respectively. A universal testing machine (MTS 810 Material Test System) was used with a 1 mm diameter steel rod at cross-head speed of 0.5 mm/min until post extrusion, with load cell of 50 kg, for evaluation of the push-out strength in the different thirds of each sample. The push-out strength values in kgf were converted to MPa and analyzed through Analysis of Variance and Tukey`s test, at significance level of 5%. The results showed that there were no statistical differences between the QTH and LED LCUs. The self-adhesive resin cement had lower values of retention. The total-etch and self-adhesive system resin cements seem to be a possible alternative for glass fiber posts cementation into the radicular canal and the LED LCU can be applied as an alternative to halogen light on photo-activation of dual-cured resin cements.

The reaction Of Cu(ClO(4))(2)center dot 6H(2)O with dimethylglyoxime (H(2)dmg) in a 1:1 mole ratio in aqueous methanol at room temperature affords the dinuclear complex [Cu(2)(mu-Hdmg)(4)] (1). Reaction of 1 with [Cu(bpy)(H(2)O)(2)](ClO(4))(2) (bpy = 2,2`-bipyridine) in a 1:1 mole ratio in aqueous methanol at room temperature yields the tetranuclear complex [Cu(2)(mu-HdMg)(2)(mu-dMg)(2)(bpy)(2)(H(2)O)(2)](ClO(4))(2) (2). The direct reaction of Cu(ClO(4))(2)center dot 6H(2)O with H(2)dmg and bpy in a 2:21 mole ratio in aqueous methanol at room temperature also yields 2 quantitatively. The complexes 1 and 2 were structurally characterized by X-ray crystallography. Unlike the binding in Ni/Co-dmg, two different types of N-O bridging modes during the oxime based metallacycle formation and stacking of square planar units have been identified in these complexes. The neutral dinuclear complex 1 has CuN(4)O coordination spheres and complex 2 consists of a dicationic [Cu(2)(mu-HdMg)(2)(mu-dMg)(2)(bpy)(2)(H(2)O)(2)](2+) unit and two uncoordinated ClO(4)(-) anions having CuN(4)O and CuN(2)O(3) coordination spheres. The two copper(II) ions are at a distance of 3.846(8) angstrom in 1 for the trans out of plane link and at 3.419(10) and 3.684(10) angstrom in 2 for the trans out of plane and cis in plane arrangements, respectively. The average Cu-N(oxime) distances are 1.953 and 1.935 angstrom, respectively. The average basal and apical Cu-N(oxime) distances are 1.945, 2.295 and 2.429 angstrom. The UV-Vis spectra of 2 is similar to the spectrum of the reaction mixture of 1 and [Cu(bpy)(H(2)O)(2)](2+). Variable temperature magnetic properties measurement shows that the interaction between the paramagnetic copper centers in complex I is antiferromagnetic in nature. The EPR spectra of frozen solution of the complexes at 77 K consist of axially symmetric fine-structure transitions (Delta M(S) = 1) and half-field signals (Delta M(S) = 2) at ca. 1600 G, suggesting the presence of appreciable Cu-Cu interactions. A new vanadium (IV) complex with the monoanion of 2,3-dihydroxypyridine (H(2)dhp), or 3-hydroxy-2(1H)-pyridone, was synthesized, characterized by physicochemical techniques and tested biologically. The EPR data for the [VO(Hdhp)(2)] complex in DMF are: g(x) = 1.9768, g(y) = 1.9768 and g(z) = 1.9390; A values (10(-4) cm(-1)): A(x), 59.4; A(y//), 59.4; A(z), 171.0. The vV=O band in the IR spectrum of the complex is at 986 cm(-1). The complex is paramagnetic, with mu(eff) = 1.65 BM (d(1), spin-only) at 25 degrees C. The irreversible oxidation process [V(V)/V(IV)] of the [VO(Hdhp)(2)] complex, as revealed in a cyclic voltammogram, occurs at 876 mV. The calculated molecular structure of [VO(Hdhp)(2)] shows the vanadium(IV) center in a distorted square pyramidal environment, with the oxo ligand in the apical position and the oxygen donor atoms of the Hdhp ligands in the basal positions. The ability of [VO(Hdhp)(2)] to mimic insulin, and its toxicity to hepato-biliary functions, were investigated in streptozotocin-induced diabetic rats and it was concluded that the length of treatment and the amount of [VO(Hdhp)(2)] administered were effective in reducing experimental diabetes.

A partir de uma amostra de 30 radiografias mesiorradiais prémolares superiores, contendo um instrumento endodôntico em cada um de seus condutos, obtidas com a finalidade de odontometria, foi avaliado se a manipulação digital pela aplicação de filtros grráficos de inversão, pseudocolorização e relevo, favorece ou não a localização do ápice radiográfico.As imagens convencionais foram digitalizadas através de scanner em 300 dpi e 256 tons de cinca, tamanho original e ajuste automático de brilho e contraste, reproduzidas três vezes e submetidas aos filtros gráficos. Uma seqüência aleatória de todas as imagens foi apresentada em monitor, em condições padronizadas de visualização, a um observador habituado à utilização de imagens digitais que, através de régua eletrônica da barra de ferramentas do programa PhotoshopR, efetuou a medida da distância entre a borda superior da imagem e o limite apical para ambas as raízes, três vezes em oportunidades distintas. As quatro imagens de cada dente foram montadas em apresentação do programa PowerPointR e exibidas, com os mesmos critérios de padronização, para três observadores especialistas em endodontia, os quais qualitativamente em qual delas o limite apical de cada raiz era melhor visualizado. Através dos resultados do teste não-paramétrico de Friedman verificou-se que os valores médios para as imagens manipuladas não diferem significativamente daqueles obtidos sobre a imagem (p=0,001), onde se observa o maior coeficiente de variação. A imagem e sua versão em negativo foram classificadas como as melhores de forma mais consistente. A modalidade pseudocores e relevo receberam os piores escores, porém não de modo consistente.

Holymenia clavigera (Herbst, 1784) e Anisoscelis foliacea marginella (Dallas, 1852) são hemípteros pouco estudados que ocorrem no sul do Brasil, sendo pertencentes a tribo Anisoscelidini (Coreidae). Observações preliminares indicam uma alta coexistência no uso de suas plantas hospedeiras (passifloráceas), bem como uma total semelhança morfológica dos ovos e ninfas. Este estudo objetivou descrever a morfologia genérica dos imaturos destes sugadores, bem como suas trajetórias de crescimento, uma vez que a única diferença aparente entre as espécies é uma crescente dilatação da tíbia do terceiro par de pernas de A. foliacea marginella. Por não apresentarem as formas das tíbias distintas visualmente nos primeiro e segundo ínstares, foi feita a morfometria geométrica destas. Concomitantemente, alguns aspectos relativos à interação com suas plantas hospedeiras foram investigados. Para tanto, foi avaliada a performance em três maracujás existentes no estado do Rio Grande do Sul, a partir do seguinte delineamento experimental: criação em Passiflora suberosa Linnaeus (tratamento um), em Passiflora misera Linnaeus (tratamento dois), em Passiflora edulis Sims (tratamento três) e nas três hospedeiras em conjunto (tratamento quatro). Os dois primeiros maracujás são espécies nativas e silvestres, além de serem mais semelhantes em tamanho que o terceiro maracujá, nativo e cultivado, que apresenta maior porte. A performance foi mensurada através do tempo de desenvolvimento e sobrevivência ninfal, e tamanho dos adultos. A preferência alimentar destes coreídeos foi testada em três níveis: 1) em relação às estruturas de P. suberosa (região apical, folha, caule, botão, fruto verde); 2) em relação aos parâmetros espécie e idade dos frutos de P. suberosa e P. misera, uma vez que o fruto foi a estrutura preferida e contém duas fenofases marcadamente distintas e 3) em relação às três espécies de passifloráceas utilizadas no experimento de performance. Os frutos verdes e violáceos de P. suberosa e P. edulis foram também avaliados quimicamente quanto ao pH, teor de água, nitrogênio total, carbono orgânico, fenóis totais e antocianinas. Paralelamente, um trabalho de campo de 09 de janeiro a 22 de março de 2003 (intervalos amostrais de quinze dias) visou a determinar as partes de P. suberosa mais utilizadas para alimentação e outras atividades. Por fim, caracterizou-se a morfologia genérica do aparelho bucal e analisou-se por meio de técnicas histológicas os tecidos da folha de P. suberosa (parênquima, xilema e floema) e as regiões dos frutos (pericarpo e semente) utilizados por ninfas de quinto instar e adultos de ambas as espécies. Os ovos foram idênticos em sua morfologia e ultraestrutura, diferindo apenas na magnitude, sendo maiores aqueles pertencentes a H. clavigera. Proporcionalmente, um número maior de processos micropilares foram encontrados nesta espécie. A exceção do alargamento da tíbia, que tornou-se conspícuo a partir do terceiro instar e do aspecto das ninfas de quinto instar de um modo geral, os ínstares foram também idênticos na morfologia, ultraestrutura e coloração. Porém, as trajetórias de crescimento e os coeficientes alométricos das estruturas mensuradas diferiram significativamente entre as espécies. A forma das tíbias de H. clavigera e A. foliacea marginella não foram diferentes no primeiro, mas sim no segundo instar ninfal. Para ambas as espécies, a performance foi superior em P. suberosa quando comparada com P. misera e P. edulis, apenas não diferindo do tratamento misto. A criação em apenas P. edulis resultou na pior performance para ambos os coreídeos. Não houve efeito do sexo e da espécie de coreídeo nas performances. As ninfas de primeiro instar de ambos os sugadores utilizaram mais a região apical. H. clavigera utilizou preferencialmente os frutos nos demais ínstares e no estágio adulto, o que apenas ocorreu em A. foliacea marginella do quarto instar em diante. Os frutos verdes foram selecionados por ambos os coreídeos quando em comparação com os violáceos, tanto em P. suberosa quanto em P. misera. Contudo, estes não foram selecionados segundo o atributo espécie. Os fenóis totais diminuíram à medida em que o fruto amadurece, ocorrendo o contrário com as antocianinas. O teor de água foi também maior nos frutos verdes. Quando comparados com P. suberosa, os frutos de P. misera de ambas as idades apresentaram maior teor de carbono orgânico, ocorrendo o contrário em relação ao nitrogênio total. H. clavigera não demonstrou preferência por nenhuma passiflorácea, e A. foliacea marginella utilizou mais P. misera e P. suberosa em detrimento de P. edulis. Em campo, os frutos verdes e as folhas maduras de P. suberosa foram os substratos mais utilizados para alimentação e descanso, respectivamente, independente da constante abundância de todas as estruturas. O rostro não apresentou diferenças morfológicas entre espécies e idades. Os imaturos e os adultos de ambas as espécies utilizaram o xilema na quase totalidade dos casos, raramente fazendo uso do floema. Registrou-se um uso de todas as partes do fruto, incluindo as sementes para ambos os coreídeos e estágios. Diante o exposto, o panorama atual aponta para uma grande semelhança morfológica e ecológica entre H. clavigera e A. foliacea marginella, que são provavelmente espécies simpátricas. A extrema semelhança dos estágios imaturos, adicionada ao semelhante padrão de uso de suas hospedeiras aponta para uma alta coexistência devido à parcimônia nas fases imaturas após a especiação, convergência evolutiva ou mimetismo Mülleriano.

O objetivo do presente estudo foi avaliar, através de Microscopia Eletrônica de Varredura (MEV), a capacidade de remoção da lama dentinária nas regiões cervical, média e apical do canal radicular após a utilização das soluções de EDTA dissódico, trissódico e tetrassódico a 17% e do gel de EDTA a 24% durante a preparação do canal radicular. A amostra consistiu em 100 dentes humanos, monorradiculares, extraídos, aleatoriamente divididos em cinco grupos. Nos grupos experimentais foram utilizados soluções de EDTA dissódico, trissódico e tetrassódico a 17 % e gel de EDTA trissódico a 24%. No grupo controle foi utilizado água destilada. Cada dente foi dividido longitudinalmente e analisado no MEV. As paredes dos canais radiculares foram examinadas nas regiões cervical, média e apical quanto à remoção da lama dentinária, presença de detritos e abertura dos túbulos dentinários. Durante este processo, foram realizadas fotomicrografias com aumento de 2000 vezes. O sistema de classificação utilizado na análise foi: 0 – ausência de lama dentinária, túbulos dentinários abertos e livres de detritos; 1 – lama dentinária moderada, contorno dos túbulos visíveis ou parcialmente preenchidos por detritos; 2 – abundante lama dentinária, sem distinção do contorno dos túbulos. Para a análise estatística foram utilizados os testes não paramétricos de Friedman e Kruskall-Wallis. Os resultados mostraram diferença estatística entre os grupos teste e controle (p=0,01). No entanto, quando comparados entre si, os grupos teste não apresentaram diferença significativa. Foi verificado na comparação entre as regiões cervical, média e apical diferença estatística no grupo que foi utilizado EDTA gel (p=0,01). Neste grupo foi observado que a eficácia de remoção da lama dentinária foi menor na região apical do que nas regiões cervical e média, que não apresentaram diferenças entre si. A maior presença de lama dentinária na região apical foi confirmada ao se reunir todos os grupos em que se utilizou EDTA e comprovar que a região apical apresentava significativamente mais lama dentinária do que as regiões cervical e média (p=0,01). Dessa forma pode-se concluir que as soluções de EDTA dissódico, trissódico e tetrassódico a 17% e o gel de EDTA trissódico a 24% foram eficientes na remoção da lama dentinária e que a região apical do canal radicular apresentou maior presença de lama dentinária do que as regiões cervical e média.
