O presente trabalho tem como objetivo determinar a influência das interações das gomas xantana e guar com o amido de milho de alto teor de amilose na textura de gel durante o armazenamento. Foram utilizadas amostras de amido milho Hylon VII® (71% de amilose, National Starch, goma guar (Higum 55I®, Rhodia) e goma xantana (Rhodigel 200®, Rhodia). Foram utilizadas diferentes concentrações das gomas guar e xantana, que variaram de 0 a 1%, de acordo com o delineamento experimental central composto rotacional. Em cada tratamento foram utilizadas 50g de amido com alto teor de amilose (Hylon VII), adicionadas das gomas. Estas amostras foram diluídas em água destilada e submetidas à agitação mecânica até completa dissolução. As soluções foram aquecidas até 95°C por 5 minutos para formação dos géis, os quais foram acondicionados em recipientes plásticos de 50mL e mantidos em temperatura de 5-10°C até 120h. Nos tempos T1 (24h), T2 (48h), T3 (72h), T4 (96h) e T5 (120h) de armazenamento foram feitas medidas da força máxima do gel de amilose em texturômetro (Stable Micro-System, Modelo TAX-T2). No período inicial, de 24 horas, a goma guar não apresentou influência positiva na redução da força do gel, sendo que a aplicação de goma xantana entre 0,7 e 1,0% apresentou os menores valores de força do gel de amilose. Após 120h de armazenamento, a força do gel de amilose diminuiu com a adição de 0,5-1,0% de goma xantana e 0-0,15% de goma guar.

Este trabalho objetivou a otimização da imobilização das enzimas amilases extraídas do malte do milho, usando alginato de sódio. A concentração do malte no extrato, a porcentagem de alginato de sódio e o pH foram usados como fatores que influenciam na imobilização das enzimas. Os resultados mostraram que as melhores condições de imobilização foram obtidas quando se usou as soluções de malte de milho em duas faixas de concentrações, uma entre 3,75 a 5 g.L-1 e outra entre 15 a 16,25 g.L-1, em pH entre 4,83 a 6,6 e 4% (m/v) de alginato de sódio, condições nas quais se conseguiu imobilizar 100% das enzimas com baixa perda de atividade. Este trabalho mostrou como se obter amilases de malte de milho imobilizadas por oclusão em alginato de sódio e que podem ser usadas em processos industriais de hidrólise de amido.

Dentre os processos que precedem a conservação em longo prazo, a secagem tem papel fundamental, uma vez que o conteúdo de água das sementes afeta diretamente a sua longevidade. Os objetivos desse trabalho foram pesquisar o efeito da umidificação prévia das sementes após ultrassecagem em diferentes teores de água sobre a qualidade fisiológica de sementes de girassol. Sementes com conteúdo inicial de água de 4,7% foram embebidas previamente até conteúdo de água de 10,2%, e submetidas à secagem lenta, conduzida em câmara de secagem, e à secagem rápida, em sílica gel, até conteúdos de água de 7,4; 3,2; 2,9% e 5,3; 3,2; 2,1%, respectivamente. Após a secagem rápida ou lenta, as sementes foram submetidas ou não à umidificação em caixas tipo "gerbox" até o teor de água de 15,8 a 17% e então avaliadas quanto à germinação, peso da matéria seca de raiz, parte aérea e total das plântulas e teste de condutividade elétrica. O delineamento experimental foi o inteiramente casualizado em esquema fatorial 5 (teores de água) x 2 (com e sem umidificação). Quando utilizado a secagem rápida, a germinação das sementes não é prejudicada, e a secagem lenta, até conteúdo de água de 2,9%, proporciona uma redução na germinação das sementes. Sementes de girassol podem ser desidratadas tanto em sílica gel quanto em câmara de secagem até teores de água de 3,2%, sem perda de germinação e vigor. O tratamento de umidificação após secagem propicia um melhor desenvolvimento de raiz e menores valores de condutividade elétrica nas sementes desidratadas em sílica gel e em câmara de secagem.

A system comprised of a Martin-Puplett type polarizing interferometer and a Helium-3 cryostat was developed to study the transmission of materials in the very-far-infrared region of the spectrum. This region is of significant interest due to the low-energy excitations which many materials exhibit. The experimental transmission spectrum contains information concerning the optical properties of the material. The set-up of this system is described in detail along with the adaptations and improvements which have been made to the system to ensure the best results. Transmission experiments carried out with this new set-up for two different varieties of materials: superconducting thin films of lead and biological proteins, are discussed. Several thin films of lead deposited on fused silica quartz substrates were studied. From the ratio of the transmission in the superconducting state to that in the normal state the superconducting energy gap was determined to be approximately 25 cm-1 which corresponds to 2~/kBTc rv 5 in agreement with literature data. Furthermore, in agreement with theoretical predictions, the maximum in the transmission ratio was observed to increase as the film thickness was increased. These results provide verification of the system's ability to accurately measure the optical properties of thin low-Tc superconducting films. Transmission measurements were carried out on double deionized water, and a variety of different concentrations by weight of the globular protein, Bovine Serum Albumin, in the sol, gel and crystalline forms. The results of the water study agree well with literature values and thus further illustrate the reproducibility of the system. The results of the protein experiments, although preliminary, indicate that as the concentration increases the samples become more transparent. Some weak structure in the frequency dependent absorption coefficient, which is more prominent in crystalline samples, may be due to low frequency vibrations of the protein molecules.

La digestion enzymatique des protéines est une méthode de base pour les études protéomiques ainsi que pour le séquençage en mode « bottom-up ». Les enzymes sont ajoutées soit en solution (phase homogène), soit directement sur le gel polyacrylamide selon la méthode déjà utilisée pour l’isolation de la protéine. Les enzymes protéolytiques immobilisées, c’est-à-dire insolubles, offrent plusieurs avantages tels que la réutilisation de l’enzyme, un rapport élevé d’enzyme-sur-substrat, et une intégration facile avec les systèmes fluidiques. Dans cette étude, la chymotrypsine (CT) a été immobilisée par réticulation avec le glutaraldehyde (GA), ce qui crée des particules insolubles. L’efficacité d’immobilisation, déterminée par spectrophotométrie d’absorbance, était de 96% de la masse totale de la CT ajouté. Plusieurs différentes conditions d’immobilisation (i.e., réticulation) tels que la composition/pH du tampon et la masse de CT durant la réticulation ainsi que les différentes conditions d’entreposage tels que la température, durée et humidité pour les particules GA-CT ont été évaluées par comparaison des cartes peptidiques en électrophorèse capillaire (CE) des protéines standards digérées par les particules. Les particules de GA-CT ont été utilisés pour digérer la BSA comme exemple d’une protéine repliée large qui requit une dénaturation préalable à la digestion, et pour digérer la caséine marquée avec de l’isothiocyanate de fluorescéine (FITC) comme exemple d’un substrat dérivé afin de vérifier l’activité enzymatique du GA-CT dans la présence des groupements fluorescents liés au substrat. La cartographie peptidique des digestions par les particules GA-CT a été réalisée par CE avec la détection par absorbance ultraviolet (UV) ou fluorescence induite par laser. La caséine-FITC a été, en effet, digérée par GA-CT au même degré que par la CT libre (i.e., soluble). Un microréacteur enzymatique (IMER) a été fabriqué par immobilisation de la CT dans un capillaire de silice fondu du diamètre interne de 250 µm prétraité avec du 3-aminopropyltriéthoxysilane afin de fonctionnaliser la paroi interne avec les groupements amines. Le GA a été réagit avec les groupements amine puis la CT a été immobilisée par réticulation avec le GA. Les IMERs à base de GA-CT étaient préparé à l’aide d’un système CE automatisé puis utilisé pour digérer la BSA, la myoglobine, un peptide ayant 9 résidus et un dipeptide comme exemples des substrats ayant taille large, moyenne et petite, respectivement. La comparaison des cartes peptidiques des digestats obtenues par CE-UV ou CE-spectrométrie de masse nous permettent d’étudier les conditions d’immobilisation en fonction de la composition et le pH du tampon et le temps de réaction de la réticulation. Une étude par microscopie de fluorescence, un outil utilisé pour examiner l’étendue et les endroits d’immobilisation GA-CT dans l’IMER, ont montré que l’immobilisation a eu lieu majoritairement sur la paroi et que la réticulation ne s’est étendue pas si loin au centre du capillaire qu’anticipée.

Transition metal-loaded (3%) nanocrystalline sulfated titania (ST) powders are prepared using the sol–gel technique. Anatase is found as the active phase in all the samples. Sulfate ion impregnation decreases the crystallite size and stabilizes the anatase phase of TiO2. Acidity of the samples is found to increase by the incorporation of sulfate ion and also by the modification by transition metal ions. All the prepared catalysts are found stable up to 700 °C.

The fabrication and characterization of a fibre optic pH sensor based on evanescent wave absorption is presented. The unclad portion of a multi-mode optical fibre is coated with a pH sensitive dye, which is immobilized by the sol–gel route. The sensitivity of the device has been found to increase when multiple sol–gel coatings are used as the sensing region. The dynamic range and the temporal response of the sensor are investigated for two different dyes, namely bromocresol purple and bromocresol green. The performance of the device is evaluated in terms of the results obtained during actual measurements.

Introducción: En el tratamiento con Luz Pulsada Intensa (LPI) para el fotoenvejecimiento de las manos no se encuentran estudios que evidencien si existe alguna diferencia estadísticamente significativa en el grado de efectividad y seguridad al utilizar gel o aceite mineral como medios de acople. Objetivo: Determinar la efectividad y seguridad terapéutica en el uso de gel vs aceite mineral. Materiales y Métodos: Estudio observacional analítico de cohorte retrospectivo que involucró 29 pacientes. Realizado en tres fases; selección y recolección de las historias clínicas, evaluación fotográfica de registros pre tratamiento y pos tratamiento con determinación del grado de mejoría global en el fotoenvejecimiento de las manos por parte de tres evaluadores cegados, y análisis estadístico de los datos obtenidos por medio de las pruebas de Mann Whitney y Wilcoxon. Resultados: Se encontró mejoría dada por disminución en un grado del fotoenvejecimiento para los dos medios de acople con la misma significancia estadística. La percepción subjetiva mostró mejoría en todos los pacientes evaluados. La seguridad es similar en los dos grupos pero se evidenció mayor severidad en los efectos secundarios con el uso de aceite, con diferencias estadísticamente significativas en los efectos moderados y severos. Conclusión: La efectividad es la misma independiente del medio de acople que se use. La seguridad a pesar de evidenciar un perfil similar es mayor con el uso de gel en cuanto a la menor severidad de los efectos presentados. Se requieren más estudios de tipo ensayos clínicos controlados que permitan determinar una mayor evidencia.

En el presente trabajo de tesis se evalúa la posible contribución de otros compuestos al desarrollo del olor sexual mediante la aplicación de técnicas de Head Space dinámico y cromatografía de filtración de gel. El estudio se efectuó en canales de cerdos enteros previamente clasificadas con concentraciónes bajas de escatol (<0,10 µg /g) y androstenona (0,50 µg/ g) , aunque presentaban , según las respuestas de un panel sensorial, olor sexual. La selección de muestras de grasa clasificadas sin olor sexual permitió la comparación estadística de los resultados obtenidos. El análisis de los compuestos volátiles mostró una mayor abundancia de aldehidos (hexanal, heptanal), ácidos grasos de cadena corta (hexanoico, heptanico nonanoico), 1,4-diclorobenceno y estireno en las muestras de grasa con defecto organoléptico, ue puede, debido a los atributos sensoriales de estas sustancias y sus bajos umbrales de detección, favorecer el desarrollo de aromas desagradables asociados al olor sexual por los miembros de un papel sensorial. El análisis de las fracciones obtenidas por filtración de gel mediante cromatografía de gases acoplada a la espectometría de masas permitió la identificación de la 4-fenil-3-buten-2-ona en las muestras clasificadas con olor sexual y concentraciones bajas de escatol y androstenona, que junto a los resultados obtenidos en el estudio sensorial, indican una influencia de este compuesto en el olor sexual. Por una parte la 4-fenil-3-buten-2-ona presentaría una acción sinérgica con la androstenona al potenciar su detección por parte de los panelistas a concentraciones inferiores de 0,01µg/g. Además, el olor a naftalina que presenta la 4-fenil-3-buten-2-ona puede confundir a los miembros del panel debido a que es uno de los descriptores sensoriales que se asocia con la presencia de escatol en las muestras de grasa evaluadas. Así mismo, la baja transferencia a la fase vapor del escatol y, especialmente, de la androstenona determinada mediante técnicas de Head Space estático, indica que la concentración de cada uno de los compuestos en la fase vapor no se corresponde con la concentración real en las muestras de grasa . Según estos resultados, las respuestas del panel test pueden estar influenciadas por la poca volatilidad de los compuestos implicados en el olor sexual y por la presencia en la fase vapor de otros compuestos aromáticos de mayor volatilidad. La validación de métodos de análisis para la determinación de indol-escatol y androstenona-androstenoles en grasa dorsal porcina mediante cromatografía líquida en fase normal y cromatografía de gases acoplada a la espectrometría de masas, respectivamente, fue otro de los objetivos planteados del presente estudio. La simplificación del tratamiento de las muestras de grasa fue uno de los aspectos analíticos que se tuvo en mayor consideración con la finalidad de ofrecer una mayor capacidad de muestras en un intervalo de tiempo corto.

Whitish and whitish-light brown milky-like textural pedofeatures and impregnations were found in the voids and the matrix of buried paleosols older than 2.7 million years in a site in Sardinia, Italy. The pedofeatures were described and analysed using micromorphology, X-ray diffraction and microprobe techniques, and their spatial distribution correlated with field evidence. The suite of analyses showed that the main components of the pedofeatures were more or less ordered silica phases. As well as forming a matrix cement, these pedofeatures also occurred as coatings and infillings in pores. Significant amounts of alumina and, less, Mg, Ca and Fe were also present in the pedofeatures, possibly in the form of silicate coatings and inclusions/impurities, or alumino-silicates of the adjacent soil matrix. A number of hypotheses are drawn on the possible mechanisms of formation of these silica-rich pedofeatures, including the possibility of prolonged weathering of volcanic materials and the resulting formation of colloids and more or less ordered silica phases, with successive dehydration and progressive ordering of phases during the at least 2.5 million years. Silicon release from rice straw and amorphous silica when shaken in solution with five Sri Lankan soils was studied indirectly using sorption isotherms and changes in concentration and directly using straw in dialysis bags examined using electron microscopy. The aim was to further our understanding of the processes and factors affecting the release of straw-Si in soils and its availability to rice. The soils (alfisols and ultisols) shaken with 0.1 M NaCl (5 g per 125 mL for 250 days) produced concentrations of 1 - 4 mg L-1 of monosilicic acid-Si. Amorphous silica added to these suspensions (36.5 mg, containing 17 mg Si) raised the concentrations to 20 - 40 mg L-1, and added rice straw (0.5 g, containing 17 mg Si) gave 10 - 25 mg L-1. Sorption isotherms (7 days equilibrations) were used to calculate from the concentrations the amounts of Si released ( 24 - 38% and 8 - 21%, respectively). Both materials gave about 40 mg L-1 of monosilicic acid-Si plus 30 mg L-1 of disilicic acid-Si when shaken in solution alone (5 g per 125 mL). Straw in dialysis bags ( 0.5 g per 25 mL in 0.1 M NaCl) was shaken in soil suspension ( 5 g per 100 mL) for 60 days. Similar concentrations and releases were measured to those obtained above. About one fifth of the mass of straw was lost by decomposition in the first 15 days. A chloroform treatment prevented decomposition, but Si release was unaffected. Disintegration continued throughout the experiments, with phytoliths being exposed and dissolved. Compared to the rate of release from straw into solution without soil, the release of Si into soil suspensions was increased during the first 20 days by adsorption on the soil, but was then reduced probably through the effect of Fe and Al on the phytolith surfaces. The extent of this blocking effect varied between soils and was not simply related to soil pH.
