The effects of continuous sonication and presonication on the kinetics of oxidative dissolution of ruthenium dioxide hydrate by bromate ions under acidic conditions are reported. Compared with unsonicated and presonicated dispersions the overall rate of dissolution of continuously sonicated dispersions is significantly greater due to a reduction in the average particle size and, hence, an increase in the specific surface area. Powder dispersions subjected to continuous ultrasound and presonication exhibit an initial induction period in their corrosion kinetics; the length of this induction period increases with increasing presonication. This corrosion feature is retained in the dissolution kinetics of powder samples which have been subjected to pre-ultrasound, but which are then stirred during the dissolution process. It is believed that this apparent permanent change in the nature of the powder particles is due to the ultrasound induced formation of a very thin layer of a largely unreactive form of ruthenium dioxide (possibly due to partial dehydration) on the surface of the powder particles. A kinetic scheme, based on this model, is used to account for the observed kinetics of dissolution of RuO2 . xH2O which have been subjected to both continuous sonication and presonication.

Background
Neutrophil elastase (NE)-mediated inflammation contributes to lung damage in cystic fibrosis (CF). We investigated if DX-890, a small-protein NE inhibitor, could reduce neutrophil trans-epithelial migration and reduce activity released from neutrophils and NE-induced cytokine expression in airway epithelial cells.

Methods
Activated blood neutrophils (CF and healthy) treated ± DX-890 were assayed for NE activity. Transmigration of calcein-labeled neutrophils was studied using a 16HBE14o- epithelial monolayer. IL-8 release from primary nasal epithelial monolayers (CF and healthy) was measured after treatment ± DX-890 and NE or CF sputum.

Results
DX-890 reduced NE activity from neutrophils (CF and healthy) and reduced neutrophil transmigration. DX-890 pre-treatment reduced IL-8 release from epithelial cells of healthy or CF subjects after stimulation with NE and CF sputum sol. All improvements with DX-890 were statistically significant (p < 0.05).

Conclusions
DX-890 reduces NE-mediated transmigration and inflammation. NE inhibition could be useful in managing neutrophilic airway inflammation in CF.

An important difference between chemical agents that induce oxidative damage in DNA and ionizing radiation is that radiation-induced damage is clustered locally on the DNA, Both modelling and experimental studies have predicted the importance of clustering of lesions induced by ionizing radiation and its dependence on radiation quality. With increasing linear energy transfer, it is predicted that complex lesions will be formed within 1-20 bp regions of the DNA, As well as strand breaks, these sites may contain multiple damaged bases, We have compared the yields of single strand breaks (ssb) and double strand breaks (dsb) along with those produced by treatment of irradiated DNA with the enzyme endonuclease III, which recognizes a number of oxidized pyrimidines in DNA and converts them to strand breaks. Plasmid DNA was irradiated under two different scavenging conditions to test the involvement of OH radicals with either Co-60 gamma-rays or alpha-particles from a Pu-238 source. Under low scavenging conditions (10 mM Tris) gamma-irradiation induced 7.1x10(-7) ssb Gy/bp, which increased 3.7-fold to 2.6 x 10(-6) ssb Gy/bp with endo III treatment. In contrast the yields of dsb increased by 4.2-fold from 1.5 x 10(-8) to 6.3 x 10(-8) dsb Gy/bp, This equates to an additional 2.5% of the endo III-sensitive sites being converted to dsb on enzyme treatment. For alpha-particles this increased to 9%. Given that endo III sensitive sites may only constitute similar to 40% of the base lesions induced in DNA, this suggests that up to 6% of the ssb measured in X- and 22% in alpha-particle-irradiated DNA could have damaged bases associated with them contributing to lesion complexity.

Free radical-mediated damage to vascular cells may be involved in the pathogenesis of diabetic vasculopathy. The aim of this study was to compare the extent of glucose-induced oxidative stress in both vascular smooth muscle cells (VSMCs) and pericytes and the effect on antioxidant enzyme gene expression and activities. Porcine aortic VSMC and retinal pericytes were cultured in either 5 or 25 mmol/l glucose for 10 days. Intracellular malondialdehyde (MDA) was measured as a marker of peroxidative damage, and mRNA expression of CuZn-SOD, MnSOD, catalase, and glutathione peroxidase (GPX) were measured by Northern analysis. Glutathione (GSH) was also measured. Computer-assisted topology predictions are widely used to build low-resolution structural models of integral membrane proteins (IMPs). Experimental validation of these models by traditional methods is labor intensive and requires modifications that might alter the IMP native conformation. This work employs oxidative labeling coupled with mass spectrometry (MS) as a validation tool for computer-generated topology models. ·OH exposure introduces oxidative modifications in solvent-accessible regions, whereas buried segments (e.g., transmembrane helices) are non-oxidizable. The Escherichia coli protein WaaL (O-antigen ligase) is predicted to have 12 transmembrane helices and a large extramembrane domain (Pérez et al., Mol. Microbiol. 2008, 70, 1424). Tryptic digestion and LC-MS/MS were used to map the oxidative labeling behavior of WaaL. Met and Cys exhibit high intrinsic reactivities with ·OH, making them sensitive probes for solvent accessibility assays. Overall, the oxidation pattern of these residues is consistent with the originally proposed WaaL topology. One residue (M151), however, undergoes partial oxidation despite being predicted to reside within a transmembrane helix. Using an improved computer algorithm, a slightly modified topology model was generated that places M151 closer to the membrane interface. On the basis of the labeling data, it is concluded that the refined model more accurately reflects the actual topology of WaaL. We propose that the combination of oxidative labeling and MS represents a useful strategy for assessing the accuracy of IMP topology predictions, supplementing data obtained in traditional biochemical assays. In the future, it might be possible to incorporate oxidative labeling data directly as constraints in topology prediction algorithms.

The anionic speciation of chlorostannate(II) ionic liquids, prepared by mixing 1-alkyl-3-methylimidazolium chloride and tin(II) chloride in various molar ratios, chi(SnCl2), was investigated in both solid and liquid states. The room temperature ionic liquids were investigated by Sn-119 NMR spectroscopy, X-ray photoelectron spectroscopy, and viscometry. Crystalline samples were studied using Raman spectroscopy, single-crystal X-ray crystallography, and differential scanning calorimetry. Both liquid and solid systems (crystallized from the melt) contained [SnCl3](-) in equilibrium with Cl- when chi(SnCl2) < 0.50, [SnCl3](-) in equilibrium with [Sn2Cl5](-) when chi(SnCl2) > 0.50, and only [SnCl3](-) when chi(SnCl2) = 0.50. Tin(II) chloride was found to precipitate when chi(SnCl2) > 0.63. No evidence was detected for the existence of [SnCl4](-) across the entire range of chi(SnCl2) although such anions have been reported in the literature for chlorostannate(II) organic salts crystallized from organic solvents. Furthermore, the Lewis acidity of the chlorostannate(II)-based systems, expressed by their Gutmann acceptor number, has been determined as a function of the composition, chi(SnCl2), to reveal Lewis acidity for chi(SnCl2) > 0.50 samples comparable to the analogous systems based on zinc(II). A change of the Lewis basicity of the anion was estimated using H-1 NMR spectroscopy, by comparison of the measured chemical shifts of the C-2 hydrogen in the imidazolium ring. Finally, compositions containing free chloride anions (chi(SnCl2) < 0.50) were found to oxidize slowly in air to form a chlorostannate(IV) ionic liquid containing the [SnCl6](2-) anion.
