Previous studies carried out with Sm14 in experimental vaccination against Schistosoma mansoni or Fasciola hepatica infections were performed with recombinant Sm14 (rSm14) produced in Escherichia coli by the pGEMEX system (Promega). The rSm14 was expressed as a 40 kDa fusion protein with the major bacteriophage T7 capsid protein. Vaccination experiments with this rSm14 in animal models resulted in consistent high protective activity against S. mansoni cercariae challenge and enabled rSm14 to be included among the vaccine antigens endorsed by the World Health Organization for phase I/II clinical trials. Since the preparation of pGEMEX based rSm14 is time consuming and results in low yield for large scale production, we have tested other E. coli expression systems which would be more suitable for scale up and downstream processing. We expressed two different 6XHis-tagged Sm14 fusion proteins in a T7 promoter based plasmids. The 6XHis-tag fusions allowed rapid purification of the recombinant proteins through a Ni+2-charged resin. The resulted recombinant 18 and 16 kDa proteins were recognized by anti-Sm14 antibodies and also by antiserum against adult S. mansoni soluble secreted/excreted proteins in Western-Blot. Both proteins were also protective against S. mansoni cercariae infection to the same extent as the rSm14 expressed by the pGEMEX system.

Summary: Victims of crime are not only physical persons. Firms also experience some different forms of victimization either from outsiders or front employees. Crimes committed inside the firm are various: theft, embezzlement, fraud, fraudulent management, money laundering, vandalism etc. Assessed damage could be considerable. But in general, most of the criminological studies relating to victimization have been interested in individuals as victims. In Switzerland, to date, research on firms'victimization is simply rare and traditional research methods such as victimization survey, self-reported delinquency or controlled experiments are not very much utilized in Swiss white-collar crime studies. Thus, in order to improve the situation, we have decided to start a victimization study on white-collar crime committed in firms of the canton of Geneva. To narrow the field of research, our project seeks to apprehend white-collar crime as a series of offences committed by employees inside the firm, by fraud or deceit, aiming at an illegitimate enrichment. The phenomenon of white-collar crime perpetrated has been evaluated by examining the presence of risk factors and prevention strategies. Furthermore, the correlation between the victimization rate and some aspects of vulnerability has also been investigated. Using data collected from the victimization survey carried out in 2005, some of the key research questions are the following: - which are the types of offences committed? - Which types of measure of prevention are the most efficient to prevent such offences ? - Which type of business is more vulnerable? - Which are the risk factors? - Which is the profile of the authors? - Which are the motivations?, etc. Findings show that companies which valorize their employees and pursue a transparent management style suffer a lower victimization rate. Further, the retail sector seems to be more affected than the financial sector and big firms are more vulnerable.

The paper analyses the regional flows of domestic tourism that took place in Spain in year 2000, contributing to the state of knowledge on tourism required by authorities and private firms when faced with decision making, for example, for regional infrastructure planning. Although tourism is one of the main income-generating economic activities in Spain, domestic tourism has received little attention in the literature compared to inbound tourism. The paper uses among others, gravitational model tools and concentration indices, to analyse regional concentration of both domestic demand and supply; tourism flows among regions, and the causes that may explain the observed flows and attractiveness between regions. Among the most remarkable results are the high regional concentration of demand and supply, and the role of population and regional income as explanatory variables. Also remarkable are the attractiveness of own region and neighbour ones, and that domestic tourism may be acting as a regional income redistributing activity

Over recent years, both governments and international aid organizations have been devoting large amounts of resources to "simplifying" the procedures for setting up and formalizing firms. Many of these actions have focused on reducing the initial costs of setting up the firm, disregarding the more important role of business registers as a source of reliable information for judges, government departments and, above all, other firms. This reliable information is essential for reducing transaction costs in future dealings with all sorts of economic agents, both public and private. The priorities of reform policies should therefore be thoroughly reviewed, stressing the value of the legal institutions rather than trivializing them as is often the case.

Invasive aspergillosis is one of the most important infections in hematopoietic stem cell transplant recipients, with an incidence rate of 5-15% and an associated mortality of 30-60%. It remains unclear why certain patients develop invasive aspergillosis while others, undergoing identical transplant regimen and similar post transplant immunosuppression, do not. Over the last decade, pattern recognition receptors such as Toll-like receptors (TLRs) and the C-type lectin receptors (CLRs) have emerged as critical components of the innate immune system. By detecting specific molecular patterns from invading microbes and initiating inflammatory and subsequent adaptive immune responses, pattern recognition receptors are strategically located at the molecular interface of hosts and pathogens. Polymorphisms in pattern recognition receptors and downstream signaling molecules have been associated with increased or decreased susceptibility to infections, suggesting that their detection may have an increasing impact on the treatment and prevention of infectious diseases in the coming years. Infectious risk stratification may be particularly relevant for patients with hematologic malignancies, because of the high prevalence and severity of infections in this population. This review summarizes the innate immune mechanisms involved in Aspergillus fumigatus detection and the role of host genetic polymorphisms in susceptibility to invasive aspergillosis.

Wage inequality in the United States has grown substantially in thepast two decades. Standard supply-demand analysis in the empiricsof inequality (e.g.Katz and Murphy (1992)) indicates that we mayattribute some of this trend to an outward shift in the demand forhigh skilled labor. In this paper we examine a simple static channelin which the wage premium for skill may grow -increased firm entry.We consider a model of wage dispersion where there are two types ofworkers and homogeneous firms must set wages and preferences forwhat type of worker they would like to hire. We find that both thewage differential and the demand for high skill workers can increasewith the proportion of high skill workers -these high skill workerstherefore 'create' their own demand without exogenous factors. Inaddition, within group wage inequality can increase in step with thebetween group wage inequality. Simulations of the model are providedin order to compare the findings with empirical results.

We set up a dynamic model of firm investment in which liquidity constraintsenter explicity into the firm's maximization problem. The optimal policyrules are incorporated into a maximum likelihood procedure which estimatesthe structural parameters of the model. Investment is positively related tothe firm's internal financial position when the firm is relatively poor. This relationship disappears for wealthy firms, which can reach theirdesired level of investment. Borrowing is an increasing function of financial position for poor firms. This relationship is reversed as a firm's financial position improves, and large firms hold little debt.Liquidity constrained firms may be unused credits lines and the capacity toinvest further if they desire. However the fear that liquidity constraintswill become binding in the future induces them to invest only when internalresources increase.We estimate the structural parameters of the model and use them to quantifythe importance of liquidity constraints on firms' investment. We find thatliquidity constraints matter significantly for the investment decisions of firms. If firms can finance investment by issuing fresh equity, rather than with internal funds or debt, average capital stock is almost 35% higher overa period of 20 years. Transitory shocks to internal funds have a sustained effect on the capital stock. This effect lasts for several periods and ismore persistent for small firms than for large firms. A 10% negative shock to firm fundamentals reduces the capital stock of firms which face liquidityconstraints by almost 8% over a period as opposed to only 3.5% for firms which do not face these constraints.

Abstract The complexity of the current business world is making corporate disclosure more and more important for information users. These users, including investors, financial analysts, and government authorities rely on the disclosed information to make their investment decisions, analyze and recommend shares, and to draft regulation policies. Moreover, the globalization of capital markets has raised difficulties for information users in understanding the differences incorporate disclosure across countries and across firms. Using a sample of 797 firms from 34 countries, this thesis advances the literature on disclosure by illustrating comprehensively the disclosure determinants originating at firm systems and national systems based on the multilevel latent variable approach. Under this approach, the overall variation associated with the firm-specific variables is decomposed into two parts, the within-country and the between-country part. Accordingly, the model estimates the latent association between corporate disclosure and information demand at two levels, the within-country and the between-country level. The results indicate that the variables originating from corporate systems are hierarchically correlated with those from the country environment. The information demand factor indicated by the number of exchanges listed and the number of analyst recommendations can significantly explain the variation of corporate disclosure for both "within" and "between" countries. The exogenous influences of firm fundamentals-firm size and performance-are exerted indirectly through the information demand factor. Specifically, if the between-country variation in firm variables is taken into account, only the variables of legal systems and economic growth keep significance in explaining the disclosure differences across countries. These findings strongly support the hypothesis that disclosure is a response to both corporate systems and national systems, but the influence of the latter on disclosure reflected significantly through that of the former. In addition, the results based on ADR (American Depositary Receipt) firms suggest that the globalization of capital markets is harmonizing the disclosure behavior of cross-boundary listed firms, but it cannot entirely eliminate the national features in disclosure and other firm-specific characteristics.

Muscle stem cells and their progeny play a fundamental role in the regeneration of adult skeletal muscle. We have previously shown that activation of the canonical Wnt/beta-catenin signaling pathway in adult myogenic progenitors is required for their transition from rapidly dividing transient amplifying cells to more differentiated progenitors. Whereas Wnt signaling in Drosophila is dependent on the presence of the co-regulator Legless, previous studies of the mammalian ortholog of Legless, BCL9 (and its homolog, BCL9-2), have not revealed an essential role of these proteins in Wnt signaling in specific tissues during development. Using Cre-lox technology to delete BCL9 and BCL9-2 in the myogenic lineage in vivo and RNAi technology to knockdown the protein levels in vitro, we show that BCL9 is required for activation of the Wnt/beta-catenin cascade in adult mammalian myogenic progenitors. We observed that the nuclear localization of beta-catenin and downstream TCF/LEF-mediated transcription, which are normally observed in myogenic progenitors upon addition of exogenous Wnt and during muscle regeneration, were abrogated when BCL9/9-2 levels were reduced. Furthermore, reductions of BCL9/9-2 inhibited the promotion of myogenic differentiation by Wnt and the normal regenerative response of skeletal muscle. These results suggest a critical role of BCL9/9-2 in the Wnt-mediated regulation of adult, as opposed to embryonic, myogenic progenitors.

Diplomityön tavoitteena on selvittää projektituotteita valmistavan metallialan yrityksen toiminnan ohjaamisen ongelmat sekä hankkia yrityksen vaatimukset täyttävä integroitu toiminnanohjausjärjestelmä. Lisäksi työssä analysoidaan toiminnanohjausjärjestelmän tuomia etuja yritykselle. Työ tehtiin Hartolassa. Työn teoriaosassa käsitellään tietojärjestelmäprojektiin kuuluvia vaiheita sekä yleisesti liiketoimintaprosessien kehittämistä. Tämän jälkeen käsitellään tarkemmin tietojärjestelmän hankintaprojektia. Lopuksi kartoitetaan yleisesti tietojärjestelmiä ja toiminnanohjausjärjestelmiä. Työn empiirisessä osassa kerrotaan tietojärjestelmäprojektin toteuttamisesta metalliyrityksessä. Tällöin esitellään projektin alussa yrityksessä ollut tilanne ja kuvataan työntekijöiden haastatteluissa esiin tulleet ongelmat. Ongelmien analysoinnin jälkeen kuvataan toiminnanohjausjärjestelmän valintaprojektin kulkua. Toiminnanohjausjärjestelmän hankinnan lähtökohtana tulee olla yrityksen tuottavuuden ja toimintojen kehittäminen. Toiminnanohjausjärjestelmät tarjoavat tähän hyvän työkalun, mutta hyödyntääkseen toiminnanohjausjärjestelmää yritys joutuu käymään läpi suuren oppimisprosessin. Toiminnanohjausjärjestelmät parantavat yrityksen toimintaprosesseja vain, jos toiminnanohjausjärjestelmän implementointiprosessi onnistuu ja järjestelmästä näin saadaan kaikki hyöty irti. Integroidut toiminnanohjausjärjestelmät hyödyttävät yritystä reaaliaikaisen informaation ansiosta. Lisäksi järjestelmän avulla saadaan yrityksen prosesseja tehostettua ja yhdistettyä toisiinsa.

Globaalissa taloudessa yritysten rakenteet ovat kehittyneet yhteistyöverkostoiksi, joissa yritykset pyrkivät keskittymään ydinosaamiseensa ja tuottamaan muut toiminnot yhteistyökumppaneilla. Nykyisin yhä suurempi osa myös logistisista toiminnoista tuotetaan kolmansilla osapuolilla. Logistiikan piirissä saavutettavat synergiaedut ovat suuret ja monet yritykset niitä ovat myös jo saavuttaneet. Menestyksekkäät esimerkit ulkoistamisista ja toisaalta kehittyvän tietotekniikan tuomat uudet toimintamahdollisuudet johtavat ulkoistamisen, 3PL toiminnan, entistäkin suurempaan suosioon. Ongelmaksi on yrityksille muodostunut yhteistyökumppaneiden suuri määrä. Kun ydinosaamisalueen ulkopuolisia tehtäviä on jaettu useille eri yritykselle, tulee yhteistyöverkoston hallinnasta haasteellinen tehtävä, ja siihen vaaditaan kalliita tietojärjestelmiä. Kustannusten säästö saattoi kuitenkin olla ulkoistaneen yrityksen motiivi! Kaiken lisäksi toimitusketjun hallinnan periaatteiden mukaisesti tiukka toimitusketjun sisäinen koordinointi on välttämättömyys arvoprosessin tehokkuuudelle. Tutkimuksen tarkoituksena on selvittää 4PL toimintamallin logiikka, sekä kuinka sitä voidaan käyttää ratkaisemaan 3PL toiminnan ongelmakohtia. Case osuudessa esitellään kahden haastateluin selvitetyn logistiikkaintegraattorin toiminta ja tarkastellaan niiden toiminnan pohjalta teoreettista mallia. Tutkimuksen toisena päätavoitteena on selvittää toimitusketjun hallinnan periaatteet kehitettäessä toimitusketjua sekä tutkia 4PL toimintamallia näiden teorioiden valossa. Tutkielmassa esitetään toimitusketjun hallinnan integraatiofilosofian mukaiset periaatteet, jossa integroidaan toimitusketjun 8 avainprosessia. Tutkimuksen tulokset osoittavat, että joint venture yrityksenä perustettavan logistiikkaintegraattorin käyttö toimitusketjussa voi parantaa ennenkaikkea koordinointiongelmia, pienentää logistisia kustannuksia, sekä tehokkaasti vähentää niitä ongelmia, joita kolmannen osapuolen palveluntarjoajien käyttö yritykselle aiheuttaa. Tutkimuksessa esitetään myös kuinka logistiikkaintegraattorin toimintaperiaatteita voidaan laajentaa arvoketjuintegraattoriksi ja kuinka se voi auttaa toimitusketjun avainprosessien integroinnissa, synergiaetujen hyödyntämisessä ja koko toimitusketjun tehokkaassa hallinnassa.

The Internet has transformed the scope, boundaries and dynamics of social and economic interactions. It is argued to have broadened the notion of the community from physical, colocated groups towards collectives that are able to transcend time and space, i.e. virtual communities. Even if virtual communities have been on the academic agenda for a couple of decades, there is still surprisingly little research on knowledge sharing within them. In addition, prior research has largely neglected the complex dynamics between Internet-based communication channels and the surrounding communities in which they are embedded. This thesis aims at building a better understanding of knowledge sharing supported by conversational technologies in intra-organisational virtual communities and external virtual communities supporting relationships with customers. The focus is thus on knowledge sharing in types of virtual communities that seem to be of relevance to business organisations. The study consists of two parts. The first part introduces the research topic and discusses the overall results. The second part comprises seven research publications. Qualitative research methods are used throughout the study. The results of the study indicate that investigation of the processes of knowledge sharing in virtual communities requires a socio-technical perspective, combining the individual, social and technological levels, and understanding the interplay between them. It is claimed that collective knowledge in virtual communities creates the enabling structure for knowledge sharing, and forms the invisible structure of the community on the basis of which it operates. It consists of a shared context, social capital and a unique community culture. The Internet does not inevitably erode social interaction: it seems that supporting social relationships by means of communication technology is a matter of quantity rather than quality. In order to provide access to external knowledge and expertise, firms need to open themselves up to an array of Internet-based conversations, and to consider the relevance of virtual communities to their businesses.

The acidity and ester index are suggested, as simple and cheap methods, adequate to be employed in the study of the authenticity of copaíba oil by small drugstores and export firms. The acidity index permits both the detection and the evaluation of the content of adulteration of copaíba oil. The ester index permits to know if the adulterant either contain ester compounds,as the case of fatty oils, or contain only non saponifiable compounds as is the case of ethyl alchool or mineral oil. The results of the aplication of the methods to eight commercial samples, of copaiba oil, where three samples were adultered, are coherent with those obtained by more sophisticated instrumental methods.

The level of training provided by small firms to their employees is below that provided by their larger counterparts. The provision of firm-related training is believed to be associated to certain characteristics of the firm. In this paper we argue that small firms provide fewer training opportunities as they are less likely to be associated with these characteristics than large firms. The suitability of estimating training decisions as a double-decision process is examined here: first, a firm has to decide whether to provide training or not and, second, having decided to do so, the amount of training to provide. The differences in training provision between small and large firms are decomposed in order to analyse the individual contribution of these characteristics to explaining the gap. The results show that small firms face greater obstacles in accessing training and that the main reasons for that are related to their technological activity and the geographical scope of the market in which they operate.
