Bioprocesses use microorganisms or cells in order to produce and/or obtain some desired products. Nowadays these strategies appear as a fundamental alternative to the traditional chemical processes. Amongst the many advantages associated to their use in the chemical, oil or pharmaceutical industries, their low cost, easily scale-up and low environmental impact should be highlighted. This work reports two examples of bioprocesses as alternatives to traditional chemical processes used by the oil and pharmaceutical industries. In the first part of this work it was studied an example of a bioprocess based on the use of microorganisms in enhanced oil recovery. Currently, due to high costs of oil and its scarcity, the enhanced oil recovery techniques become very attractive. Between the available techniques the use of microbial enhanced oil recovery (MEOR) has been highlighted. This process is based on the stimulation of indigenous microorganisms or by the injection of microorganism consortia to produce specific metabolites and hence increase the amount of oil recovered. In the first chapters of this work the isolation of several microorganisms from samples of paraffinic Brazilian oils is described, and their tensioactive and biodegradability properties are presented. Furthermore, the chemical structures of the biosurfactants produced by those isolates were also characterized. In the final chapter of the first part, the capabilities of some isolated bacteria to enhance the oil recovery of paraffinic Brazilian oils entrapped in sand-pack columns were evaluated. In the second part of this work it was investigated aqueous two-phase systems or aqueous biphasic systems (ABS) as extractive strategies for antibiotics directly from the fermented broth in which they are produced. To this goal, several aqueous two-phase systems composed of ionic liquids (ILs) and polymers were studied for the first time and their phase diagrams were determined. The novel ATPS appear as effective and economic methods to extract different biomolecules or/and biological products. Thus, aiming the initial antibiotics extraction purpose it was studied the influence of a wide range of ILs and polymers in the aqueous two-phase formation ability, as well as their influence in the partitioning of several type-molecules, such as amino acids, alkaloids and dyes. As a final chapter it is presented the capacity of these novel systems to extract the antibiotic tetracycline directly from the fermented broth of Streptomyces aureofaciens.

Solubilities of red 153, (3-[[4-[[5,6(or 6,7)-dichloro-2-benzothiazolyl]azo]phenyl]ethylamino]propanenitrile), an azo compound, and disperse blue1 (1,4,5,8-tetraaminoantraquinone) in supercritical carbon dioxide (SC CO(2)) were measured at T = (333.2 to 393.2) K over the pressure range (12.0 to 40.0) MPa by a flow type apparatus. The solubility of red 153 (0.985. 10(-6) to 37.2. 10(-6)) in the overall region of measurements is found to be significantly higher than that of disperse blue 1 (1.12.10(-7) to 4.89.10(-7)). The solubility behavior of disperse red 153 follows the general solubility trend displayed by disperse dyes with a crossover pressure at about 20 MPa. On the other hand, blue 1, which is a disperse anthraquinone dye, exhibits unexpected behavior not recorded previously there is no crossover pressure at the temperature and pressure ranges studied, and the dye's solubility at T = 333.2 K practically does not increase with pressure. To the best of our knowledge, there are no previous measurements of blue 1 solubility in SC CO(2) reported in the literature. The experimental data were correlated by using the Soave Redlich Kwong equation of state (EoS) with the one-fluid van der Waals mixing rule, and an acceptable correlation of the solubility data for both dyes was obtained.

The main objective of the research work developed in the framework of this PhD thesis was the preparation and development of novel photorheological fluids. This was pursued following two distinct strategies. The first one focused on the synthesis of tripodal compounds functionalized with photodimerizable moieties of cinnamic acid, coumarin and anthracene. Two sets of compounds were prepared, varying the central unit as well as spacers resulting in molecules with different solubilities and molecular weight. All compounds were characterized towards their photochemical properties and all exhibited photoreactivity upon irradiation with ultra-violet light. In particular, both coumarin derivatives exhibited the greatest photopolymerization reactivity, resulting in the formation of dendrimeric nanoparticles or in the increase of viscosity of organic solutions. The second strategy was focused on the careful design of photosensitive ionic liquids, based on the results of several quantitative structure-property relationship studies. Thus, photosensitive ionic liquids were synthesized bearing cinnamic acid or coumarin moieties in the organic cation. Upon irradiation, all compounds exhibited reactivity, which resulted in changes in their physical properties, such as melting point or viscosity. In addition, novel coumarin chromophores with different photophysical and photochemical properties were developed. It is expected that these compounds may find application in the preparation of new photosensitive ionic liquids.

Nature has developed strategies to present us with a wide variety of colours, from the green of leaves to the bright colours seen in flowers. Anthocyanins are between these natural pigments that are responsible for the great diversity of colours seen in flowers and fruits. Anthocyanins have been used to sensitize titanium dioxide (TiO2) in Dye-Sensitized Solar Cells (DSSCs). DSSCs have become one of the most popular research topic in photovoltaic cells due to their low production costs when compared to other alternatives. DSSCs are inspired in what happens in nature during photosynthesis. A primary charge separation is achieved by means of a photoexcited dye capable of performing the electron injection into the conduction band of a wide band-gap semiconductor, usually TiO2. With this work we aimed to synthesize a novel mesoporous TiO2 structure as the semiconductor in order to increase the dye loading. We used natural occurring dyes such as anthocyanins and their synthetic flavylium relatives, as an alternative to the widely used metal complexes of Ru(II) which are expensive and are environmentally unsafe. This offers not only the chance to use safer dyes for DSSCs, but also to take profit of waste biological products, such as wine and olive oil production residues that are heavily loaded with anthocyanin dyes. We also performed a photodegradation study using TiO2 as the catalyst to degrade dye contaminants, such as those from the wine production waste, by photo-irradiation of the system in the visible region of the light spectrum. We were able to succeed in the synthesis of mesoporous TiO2 both powder and thin film, with a high capacity to load a large amount of dye. We proved the concept of photodegradation using TiO2 as catalyst. And finally, we show that wine production waste is a possible dye source to DSSCs application.

The advent of bioconjugation impacted deeply the world of sciences and technology. New biomolecules were found, biological processes were understood, and novel methodologies were formed due to the fast expansion of this area. The possibility of creating new effective therapies for diseases like cancer is one of big applications of this now big area of study. Off target toxicity was always the problem of potent small molecules with high activity towards specific tumour targets. However, chemotherapy is now selective due to powerful linkers that connect targeting molecules with affinity to interesting biological receptors and cytotoxic drugs. This linkers must have very specific properties, such as high stability in plasma, no toxicity, no interference with ligand affinity nor drug potency, and at the same time, be able to lyse once inside the target molecule to release the therapeutic warhead. Bipolar environments between tumour intracellular and extracellular medias are usually exploited by this linkers in order to complete this goal. The work done in this thesis explores a new model for that same task, specific cancer drug delivery. Iminoboronates were studied due to its remarkable selective stability towards a wide pH range and endogenous molecules. A fluorescence probe was design to validate this model by creating an Off/On system and determine the payload release location in situ. A process was optimized to synthetize the probe 8-(1-aminoethyl)-7-hydroxy-coumarin (1) through a reductive amination reaction in a microwave reactor with 61 % yield. A method to conjugate this probe to ABBA was also optimized, obtaining the iminoboronate in good yields in mild conditions. The iminoboronate model was studied regarding its stability in several simulated biological environments and each half-life time was determined, showing the conjugate is stable most of the cases except in tumour intracellular systems. The construction of folate-ABBA-coumarin bioconjugate have been made to complete this evaluation. The ability to be uptaken by a cancer cell through endocytosis process and the conjugation delivery of coumarin fluorescence payload are two features to hope for in this construct.

RESUME GRAND PUBLICLe cerveau est composé de différents types cellulaires, dont les neurones et les astrocytes. Faute de moyens pour les observer, les astrocytes sont très longtemps restés dans l'ombre alors que les neurones, bénéficiant des outils ad hoc pour être stimulés et étudiés, ont fait l'objet de toutes les attentions. Le développement de l'imagerie cellulaire et des outils fluorescents ont permis d'observer ces cellules non électriquement excitables et d'obtenir des informations qui laissent penser que ces cellules sont loin d'être passives et participent activement au fonctionnement cérébral. Cette participation au fonctionnement cérébral se fait en partie par le biais de la libération de substances neuro-actives (appellées gliotransmetteurs) que les astrocytes libèrent à proximité des synapses permettant ainsi de moduler le fonctionnement neuronal. Cette libération de gliotransmetteurs est principalement causée par l'activité neuronale que les astrocytes sont capables de sentir. Néanmoins, nous savons encore peu de chose sur les propriétés précises de la libération des gliotransmetteurs. Comprendre les propriétés spatio-temporelles de cette libération est essentiel pour comprendre le mode de communication de ces cellules et leur implication dans la transmission de l'information cérébrale. En utilisant des outils fluorescents récemment développés et en combinant différentes techniques d'imagerie cellulaire, nous avons pu obtenir des informations très précises sur la libération de ces gliotransmetteurs par les astrocytes. Nous avons ainsi confirmé que cette libération était un processus très rapide et qu'elle était contrôlée par des augmentations de calcium locales et rapides. Nous avons également décrit une organisation complexe de la machinerie supportant la libération des gliotransmetteurs. Cette organisation complexe semble être à la base de la libération extrêmement rapide des gliotransmetteurs. Cette rapidité de libération et cette complexité structurelle semblent indiquer que les astrocytes sont des cellules particulièrement adaptées à une communication rapide et qu'elles peuvent, au même titre que les neurones dont elles seraient les partenaires légitimes, participer à la transmission et à l'intégration de l'information cérébrale.RESUMEDe petites vésicules, les « SLMVs » ou « Synaptic Like MicroVesicles », exprimant des transporteurs vésiculaires du glutamate (VGluTs) et libérant du glutamate par exocytose régulée, ont récemment été décrites dans les astrocytes en culture et in situ. Néanmoins, nous savons peu de chose sur les propriétés précises de la sécrétion de ces SLMVs. Contrairement aux neurones, le couplage stimulussécrétion des astrocytes n'est pas basé sur l'ouverture des canaux calciques membranaires mais nécessite l'intervention de seconds messagers et la libération du calcium par le reticulum endoplasmique (RE). Comprendre les propriétés spatio-temporelles de la sécrétion astrocytaire est essentiel pour comprendre le mode de communication de ces cellules et leur implication dans la transmission de l'information cérébrale. Nous avons utilisé des outils fluorescents récemment développés pour étudier le recyclage des vésicules synaptiques glutamatergiques comme les colorants styryles et la pHluorin afin de pouvoir suivre la sécrétion des SLMVs à l'échelle de la cellule mais également à l'échelle des évènements. L'utilisation combinée de l'épifluorescence et de la fluorescence à onde évanescente nous a permis d'obtenir une résolution temporelle et spatiale sans précédent. Ainsi avons-nous confirmé que la sécrétion régulée des astrocytes était un processus très rapide (de l'ordre de quelques centaines de millisecondes). Nous avons découvert que cette sécrétion est contrôlée par des augmentations de calcium locales et rapides. Nous avons également décrit des compartiments cytosoliques délimités par le RE à proximité de la membrane plasmique et contenant les SLMVs. Cette organisation semble être à la base du couplage rapide entre l'activation des GPCRs et la sécrétion. L'existence de compartiments subcellulaires indépendants permettant de contenir les messagers intracellulaires et de limiter leur diffusion semble compenser de manière efficace la nonexcitabilité électrique des astrocytes. Par ailleurs, l'existence des différents pools de vésicules recrutés séquentiellement et fusionnant selon des modalités distinctes ainsi que l'existence de mécanismes permettant le renouvellement de ces pools lors de la stimulation suggèrent que les astrocytes peuvent faire face à une stimulation soutenue de leur sécrétion. Ces données suggèrent que la libération de gliotransmetteurs par exocytose régulée n'est pas seulement une propriété des astrocytes en culture mais bien le résultat d'une forte spécialisation de ces cellules pour la sécrétion. La rapidité de cette sécrétion donne aux astrocytes toutes les compétences pour pouvoir intervenir de manière active dans la transmission et l'intégration de l'information.ABSTRACTRecently, astrocytic synaptic like microvesicles (SLMVs), that express vesicular glutamate transporters (VGluTs) and are able to release glutamate by Ca2+-dependent regulated exocytosis, have been described both in tissue and in cultured astrocytes. Nevertheless, little is known about the specific properties of regulated secretion in astrocytes. Important differences may exist between astrocytic and neuronal exocytosis, starting from the fact that stimulus-secretion coupling in astrocytes is voltage independent, mediated by G-protein-coupled receptors and the release of Ca2+ from internal stores. Elucidating the spatiotemporal properties of astrocytic exo-endocytosis is, therefore, of primary importance for understanding the mode of communication of these cells and their role in brain signaling. We took advantage of fluorescent tools recently developed for studying recycling of glutamatergic vesicles at synapses like styryl dyes and pHluorin in order to follow exocytosis and endocytosis of SLMVs at the level of the entire cell or at the level of single event. We combined epifluorescence and total internal reflection fluorescence imaging to investigate, with unprecedented temporal and spatial resolution, the events underlying the stimulus-secretion in astrocytes. We confirmed that exo-endocytosis process in astrocytes proceeds with a time course on the millisecond time scale. We discovered that SLMVs exocytosis is controlled by local and fast Ca2+ elevations; indeed submicrometer cytosolic compartments delimited by endoplasmic reticulum (ER) tubuli reaching beneath the plasma membrane and containing SLMVs. Such complex organization seems to support the fast stimulus-secretion coupling reported here. Independent subcellular compartments formed by ER, SLMVs and plasma membrane containing intracellular messengers and limiting their diffusion seem to compensate efficiently the non-electrical excitability of astrocytes. Moreover, the existence of two pools of SLMVs which are sequentially recruited suggests a compensatory mechanisms allowing the refill of SLMVs and supporting exocytosis process over a wide range of multiple stimuli. These data suggest that regulated secretion is not only a feature of cultured astrocytes but results from a strong specialization of these cells. The rapidity of secretion demonstrates that astrocytes are able to actively participate in brain information transmission and processing.

Intercellular Ca(2+) wave propagation between vascular smooth muscle cells (SMCs) is associated with the propagation of contraction along the vessel. Here, we characterize the involvement of gap junctions (GJs) in Ca(2+) wave propagation between SMCs at the cellular level. Gap junctional communication was assessed by the propagation of intercellular Ca(2+) waves and the transfer of Lucifer Yellow in A7r5 cells, primary rat mesenteric SMCs (pSMCs), and 6B5N cells, a clone of A7r5 cells expressing higher connexin43 (Cx43) to Cx40 ratio. Mechanical stimulation induced an intracellular Ca(2+) wave in pSMC and 6B5N cells that propagated to neighboring cells, whereas Ca(2+) waves in A7r5 cells failed to progress to neighboring cells. We demonstrate that Cx43 forms the functional GJs that are involved in mediating intercellular Ca(2+) waves and that co-expression of Cx40 with Cx43, depending on their expression ratio, may interfere with Cx43 GJ formation, thus altering junctional communication.

The work described in this thesis has been divided into seven sections. The first section involves the preparation of N'-acyl-N'-arylN- benzothiohydrazides by the acylation of N'-aryl-N-benzothiohydrazides and is followed by a brief discussion of their possible conformation in solution. The second section deals with the preparation of 1,3,4-thiadiazolium salts by the action of perchloric acid/acetic anhydride on N'-acylN'- aryl-N-benzothiohydrazides and also by the reaction of N'-arylN- benzothiohydrazides with nitriles in an acidic medium. The preparation of 2-methylthio-I,3,4-thiadiazolium methosulfate by methylating the corresponding thione is also described. The third section deals with the reaction of 2-phenyl- and 2-methyl-I,3,4-thiadiazolium salts with alcohols in the presence of base. The stability and spectra of these compounds are discussed. Treatment of the 2-methyl-I,3,4-thiadiazolium salt with base was found to give rise to a dimeric anhydrobase and evidence supporting its structure is given. The anhydrobase could be trapped by a variety of acylating and thioacylating agents before dimerization occurred. In the fourth section, the reaction of N'-acyl-N'-aryl-N-benzothiohydrazides with a variety of acid anhydrides is described. These compounds were found to be identical with those obtained by acylating the anhydrobase. The mass spectral fragmentation of these compounds is described and the anomolous product obtained upon thiobenzoylation of 3-methyl-l-phenyl-pyrazal-5-one is also discussed. The fifth section deals with thioacyl derivatives of the anhydrobase which were prepared by the action of phosphorus pentasulfide upon the oxygen analogues and also obtained as the major product of the reaction of thioacetic acid with compounds related to N'-aryl-N-benzothiohydrazides. The mass spectra and p.m.r. spectra of these compounds are discussed. In the sixth section, the reaction of the 2-methylthio-l,3,4- thiadiazolium salt with active methylene compounds to give acyl and diacyl derivatives of the anhydrobase is described. Some aspects of these compounds are discussed. The seventh section describes the synthesis of ncyanine~' type dyes incorporating the l,3,4-thiadiazole ring and their spectra are briefly discussed.

A study was undertaken' to determine the applicability of gas liquid chromatography to the simultaneous analysis of sugars and sugar phosphates from biological samples. A new method of silylation involving dimethylsulfoxide, hexamethyldisilazane, trimethylchlorosilane and cyclohexane (1:0.2:0.1:1) which rapidly silylated sugars and sugar phosphates was developed. Subsequent chromatography on a 5% SE-52 column gave good resolution of the sugar and sugar phosphate samples. Sugar phosphates decomposed during chromatography and were lost at the 7 x 10-3 ~mole level. Acidic ethanol extraction of yeast samples revealed background contamination from the yeast sample, the culture medium and the silylation reagents which would further limit the level of detection obtainable with the glc for sugars in biological samples to the 3 x 10-4 ~mole level.

The Ontario Tender Fruit Marketing Board operates under the Farm Producers Marketing Act. It covers all tender fruit farmers who produce either fresh or canned products. Today the board has over 500 grower-members. Tender fruit in the Niagara region includes: peaches, pears, plums, grapes and cherries. The fruits are used in a number of different ways, from jams and jellies to desserts, sauces and wine. Peaches were first harvested along the Niagara river in 1779. Peter Secord (Laura Secord’s uncle) is thought to be the first farmer to plant fruit trees when he took a land grant near Niagara in the mid 1780s. Since the beginnings of Secord’s farm, peaches, pears and plums have been grown in the Niagara region ever since. However, none of the original varities of peach trees remain today. Peaches were often used for more than eating by early settlers. The leaves and bark of the tree was used to make teas for conditions such as chronic bronchitis, coughs and gastritis. Cherries have been known to have anti-inflammatory and pain relieving properties. Like peaches and cherries, pears had many uses for the early pioneers. The wood was used to make furniture. The juice made excellent ciders and the leaves provided yellow dyes. Plums have been around for centuries, not only in the Niagara region, but throughout the world. They have appeared in pre-historic writings and were present for the first Thanksgiving in 1621. The grape industry in Ontario has also been around for centuries. It began in 1798 when land was granted to Major David Secord (brother-in-law to Laura Secord) slightly east of St. David’s, on what is Highway No. 8 today. Major Secord’s son James was given a part of the land in 1818 and in 1857 passed it onto Porter Adams. Adams is known to be the first person to plant grapes in Ontario1. Tender fruits are best grown in warm temperate climates. The Niagara fruit belt, stretching 65km from Hamilton to Niagara on the Lake, provides the climate necessary for this fruit production. This belt produces 90% of Ontario’s annual tender fruit crop. It is one of the largest fruit producing regions in all of Canada.

This thesis describes a method involving the preparation of an L-proline-derived imidazolone protected with an N-triethylsilyl group that undergoes diastereoselective lithiation followed by electrophile quench to give C5-substituted products with syn stereochemistry. The N-silylated derivatives may be more easily N-deprotected as compared to previous N-t-Bu analogues to give secondary ureas. These may serve as precursors to N-phenyl chiral bicyclic guanidines or as NHC precursors for synthesis of corresponding complexes.
