The humoral immune system plays a critical role in the clearance of numerous pathogens. In the setting of HIV-1 infection, the virus infects, integrates its genome into the host's cells, replicates, and establishes a reservoir of virus-infected cells. The initial antibody response to HIV-1 infection is targeted to non-neutralizing epitopes on HIV-1 Env gp41, and when a neutralizing response does develop months after transmission, it is specific for the autologous founder virus and the virus escapes rapidly. After continuous waves of antibody mediated neutralization and viral escape, a small subset of infected individuals eventually develop broad and potent heterologous neutralizing antibodies years after infection. In this dissertation, I have studied the ontogeny of mucosal and systemic antibody responses to HIV-1 infection by means of three distinct aims: 1. Determine the origin of the initial antibody response to HIV-1 infection. 2. Characterize the role of restricted VH and VL gene segment usage in shaping the antibody response to HIV-1 infection. 3. Determine the role of persistence of B cell clonal lineages in shaping the mutation frequencies of HIV-1 reactive antibodies.

After the introduction (Chapter 1) and methods (Chapter 2), Chapter 3 of this dissertation describes a study of the antibody response of terminal ileum B cells to HIV-1 envelope (Env) in early and chronic HIV-1 infection and provides evidence for the role of environmental antigens in shaping the repertoire of B cells that respond to HIV-1 infection. Previous work by Liao et al. demonstrated that the initial plasma cell response in the blood to acute HIV-1 infection is to gp41 and is derived from a polyreactive memory B cell pool. Many of these antibodies cross-reacted with commensal bacteria, Therefore, in Chapter 3, the relationship of intestinal B cell reactivity with commensal bacteria to HIV-1 infection-induced antibody response was probed using single B cell sorting, reverse transcription and nested polymerase chain reaction (RT- PCR) methods, and recombinant antibody technology. The dominant B cell response in the terminal ileum was to HIV-1 envelope (Env) gp41, and 82% of gp41- reactive antibodies cross-reacted with commensal bacteria whole cell lysates. Pyrosequencing of blood B cells revealed HIV-1 antibody clonal lineages shared between ileum and blood. Mutated IgG antibodies cross-reactive with both Env gp41 and commensal bacteria could also be isolated from the terminal ileum of HIV-1 uninfected individuals. Thus, the antibody response to HIV-1 can be shaped by intestinal B cells stimulated by commensal bacteria prior to HIV-1 infection to develop a pre-infection pool of memory B cells cross-reactive with HIV-1 gp41.

Chapter 4 details the study of restricted VH and VL gene segment usage for gp41 and gp120 antibody induction following acute HIV-1 infection; mutations in gp41 lead to virus enhanced neutralization sensitivity. The B cell repertoire of antibodies induced in a HIV-1 infected African individual, CAP206, who developed broadly neutralizing antibodies (bnAbs) directed to the HIV-1 envelope gp41 membrane proximal external region (MPER), is characterized. Understanding the selection of virus mutants by neutralizing antibodies is critical to understanding the role of antibodies in control of HIV-1 replication and prevention from HIV-1 infection. Previously, an MPER neutralizing antibody, CAP206-CH12, with the binding footprint identical to that of MPER broadly neutralizing antibody 4E10, that like 4E10 utilized the VH1-69 and VK3-20 variable gene segments was isolated from this individual (Morris et al., 2011). Using single B cell sorting, RT- PCR methods, and recombinant antibody technology, Chapter 4 describes the isolation of a VH1-69, Vk3-20 glycan-dependent clonal lineage from CAP206, targeted to gp120, that has the property of neutralizing a neutralization sensitive CAP206 transmitted/founder (T/F) and heterologous viruses with mutations at amino acids 680 or 681 in the MPER 4E10/CH12 binding site. These data demonstrate sites within the MPER bnAb epitope (aa 680-681) in which mutations can be selected that lead to viruses with enhanced sensitivity to autologous and heterologous neutralizing antibodies.

In Chapter 5, I have completed a comparison of evolution of B cell clonal lineages in two HIV-1 infected individuals who have a predominant VH1-69 response to HIV-1 infection--one who produces broadly neutralizing MPER-reactive mAbs and one who does not. Autologous neutralization in the plasma takes ~12 weeks to develop (Gray et al., 2007; Tomaras et al., 2008b). Only a small subset of HIV-1 infected individuals develops high plasma levels of broad and potent heterologous neutralization, and when it does occur, it typically takes 3-4 years to develop (Euler et al., 2010; Gray et al., 2007; 2011; Tomaras et al., 2011). The HIV-1 bnAbs that have been isolated to date have a number of unusual characteristics including, autoreactivity and high levels of somatic hypermutations, which are typically tightly regulated by immune control mechanisms (Haynes et al., 2005; 2012b; Kwong and Mascola, 2012; Scheid et al., 2009a). The VH mutation frequencies of bnAbs average ~15% but have been shown to be as high as 32% (reviewed in Mascola and Haynes, 2013; Kwong and Mascola, 2012). The high frequency of somatic hypermutations suggests that the B cell clonal lineages that eventually produce bnAbs undergo high-levels of affinity maturation, implying prolonged germinal center (GC) reactions and high levels of T cell help. To study the duration of HIV-1- reactive B cell clonal persistence, HIV-1 reactive and non HIV-1- reactive B cell clonal lineages were isolated from an HIV-1 infected individual that produces bnAbs, CAP206, and an HIV-1 infected individual who does not produce bnAbs, 004-0. Single B cell sorting, RT-PCR and recombinant antibody technology was used to isolate and produce monoclonal antibodies from multiple time points from each individual. B cell sequences clonally related to mAbs isolated by single cell PCR were identified within pyrosequences of longitudinal samples of these two individuals. Both individuals produced long-lived B cell clones that persisted from 0-232 weeks in CAP206, and 0-238 weeks in 004-0. The average length of persistence of clones containing members isolated from two separate time points was 91.5 weeks both individuals. Examples of the continued evolution of clonal lineages were observed in both the bnAb and non-bnAb individual. These data indicated that the ability to generate persistent and evolving B cell clonal lineages occurs in both bnAb and non-bnAb individuals, suggesting that some alternative host or viral factor is critical for the generation of highly mutated broadly neutralizing antibodies.

Together the studies described in Chapter 3-5 show that multiple factors influence the antibody response to HIV-1 infection. The initial antibody response to HIV-1 Env gp41 can be shaped by a B cell response to intestinal commensal bacteria prior to HIV-1 infection. VH and VL gene segment restriction can impact the B cell response to multiple HIV-1 antigens, and virus escape mutations in the MPER can confer enhanced neutralization sensitivity to autologous and heterologous antibodies. Finally, the ability to generate long-lived HIV-1 clonal lineages in and of itself does not confer on the host the ability to produce bnAbs.

The aim of this paper is to recognize the images that prospective elementary school teachers have about citizenship and citizenship education in school. The qualitative methodology with an ethnographic approach is used for that deep interview of ethnographic kind and analysis of institutional documents, for example, curricula, course syllabus and graduated profi le. The revision of documents helps to a better understanding of discourses. The analysis of the corpus obtained after the application of the paradigm of codifi cation proposed in the established theory. As the principal fi ndings we way point out that the concept of citizens that is common among prospective elementary school teachers is reduced to the school environment because they consider that the promotion of citizenship helps to the incorporation of children to society, thus reproducing a propaedeutic view of education.

Global amphibian declines are a major element of the current biodiversity crisis. Monitoring changes in the distribution and abundance of target species is a basic component in conservation decision making and requires robust and repeatable sampling. For EU member states, surveillance of designated species, including the common frog Rana temporaria, is a formal requirement of the 'EC Habitats & Species Directive'. We deployed established methods for estimating frog population density at local water bodies and extrapolated these to the national and ecoregion scale. Spawn occurred at 49.4% of water bodies and 70.1% of independent 500-m survey squares. Using spawn mat area, we estimated the number of adult breeding females and subsequently the total population assuming a sex ratio of 1:1. A negative binomial model suggested that mean frog density was 23.5 frogsha [95% confidence interval (CI) 14.9-44.0] equating to 196M frogs (95%CI 124M-367M) throughout Ireland. A total of 86% of frogs bred in drainage ditches, which were a notably common feature of the landscape. The recorded distribution of the species did not change significantly between the last Article 17 reporting period (1993-2006) and the current period (2007-2011) throughout the Republic of Ireland. Recording effort was markedly lower in Northern Ireland, which led to an apparent decline in the recorded distribution. We highlight the need to coordinate biological surveys between adjacent political jurisdictions that share a common ecoregion to avoid apparent disparities in the quality of distributional information. Power analysis suggested that a reduced sample of 40-50 survey squares is sufficient to detect a 30% decline (consistent with the International Union for Conservation of Nature Category of 'Vulnerable') at 80% power providing guidance for minimizing future survey effort. Our results provin assessments for R. temporaria and other clump-spawning amphibians. 2013 The Zoological Society of London.

BACKGROUND: A core outcome set (COS) can address problems of outcome heterogeneity and outcome reporting bias in trials and systematic reviews, including Cochrane reviews, helping to reduce waste. One of the aims of the international Core Outcome Measures in Effectiveness Trials (COMET) Initiative is to link the development and use of COS with the outcomes specified and reported in Cochrane reviews, including the outcomes listed in the summary of findings (SoF) tables. As part of this work, an earlier exploratory survey of the outcomes of newly published 2007 and 2011 Cochrane reviews was performed. This survey examined the use of COS, the variety of specified outcomes, and outcome reporting in Cochrane reviews by Cochrane Review Group (CRG). To examine changes over time and to explore outcomes that were repeatedly specified over time in Cochrane reviews by CRG, we conducted a follow-up survey of outcomes in 2013 Cochrane reviews.

METHODS: A descriptive survey of outcomes in Cochrane reviews that were first published in 2013. Outcomes specified in the methods sections and reported in the results section of the Cochrane reviews were examined by CRG. We also explored the uptake of SoF tables, the number of outcomes included in these, and the quality of the evidence for the outcomes.

RESULTS: Across the 50 CRGs, 375 Cochrane reviews that included at least one study specified a total of 3142 outcomes. Of these outcomes, 32 % (1008) were not reported in the results section of these reviews. For 23 % (233) of these non-reported outcomes, we did not find any reason in the text of the review for this non-report. Fifty-seven percent (216/375) of reviews included a SoF table.

CONCLUSIONS: The proportion of specified outcomes that were reported in Cochrane reviews had increased in 2013 (68 %) compared to 2007 (61 %) and 2011 (65 %). Importantly, 2013 Cochrane reviews that did not report specified outcomes were twice as likely to provide an explanation for why the outcome was not reported. There has been an increased uptake of SoF tables in Cochrane reviews. Outcomes that were repeatedly specified in Cochrane reviews by CRG in 2007, 2011, and 2013 may assist COS development.

Introdução: A profusão de informação na área médica cria problemas de gestão, sendo necessários métodos sistematizados para armazenamento e recuperação. Quando a informação se insere no contexto do processo clínico, os métodos devem integrar terminologias biomédicas controladas e igualmente devem integrar as características desejáveis dirigidas à estrutura, conteúdo e resultados clínicos. O objectivo deste artigo é testar a aplicabilidade e capacidade de recuperação, de um sistema multidimensional desenvolvido para classificação e gestão de informação em saúde. Métodos: A partir das questões recebidas em seis anos (Serviço de Informação de Medicamentos, Serviços Farmacêuticos, Hospitais da Universidade de Coimbra), seleccionaram-se 300 questões sobre informação clínica, por método aleatório informatizado. Caracterizou-se e avaliou-se a aplicabilidade pela quantidade classificada e pela necessidade de alterações ao sistema que é constituído por várias dimensões independentes e que englobam conceitos por vezes hierarquizados. A recuperação das questões foi testada pesquisando informação numa dimensão ou cruzamento de dimensões. Resultados: Todas as questões foram classificadas: 53% são casos clínicos com incidência nas doenças geniturinárias; doenças metabólicas, nutricionais e endócrinas; neoplasias; infecções e doenças do sistema nervoso. Em 81%, o objecto é o medicamento, sobretudo anti-infecciosos e anti-neoplásicos. As áreas de terapêutica e segurança foram as mais solicitadas, incidindo principalmente sobre os assuntos: utilização, reacções adversas, identificação de medicamentos e tecnologia farmacêutica. Na aplicabilidade, foi necessário adicionar alguns conceitos e modificar alguns grupos hierárquicos que não modificaram a estrutura base, nem colidiram com as características desejáveis. As limitações prenderam-se com os sistemas de classificação externos integrados. A pesquisa na dimensão assunto, do conceito administração de medicamentos, recuperou 19 questões. O cruzamento de duas dimensões: anti-infecciosos (externa) e teratogenicidade (assunto), recuperou três questões. Nos dois exemplos recupera-se informação a partir de qualquer um dos níveis da hierarquia, do mais geral ao mais específico e mesmo a partir de dimensões externas. Conclusões: A utilização do sistema nesta amostra demonstrou aplicabilidade na classificação e arquivo de informação clínica, capacidade de recuperação e flexibilidade, sofrendo alterações sem interferir com as características desejáveis. Esta ferramenta permite a recuperação da evidência que interessa orientada para o doente. Introduction: The large amount of information in the medical area creates management problems, being necessary systematic methods for filing and retrieval. With information on the context of clinical records, methods must integrate controlled biomedical terminologies and desirable characteristics oriented to the structure, content and clinical results. The objective is to test the applicability and capacity for retrieval of a multidimensional system developed for classification and management of health information. Methods: Three hundred questions were randomly selected, by computerized method, from the questions received in six years (Medicine Information Service, Pharmaceutical Department, Coimbra University Hospitals). They were characterized and applicability evaluated by classified amount and need to alter the system, which is composed of various independent dimensions, incorporating concepts sometimes hierarchical. Questions retrieval was tested searching information in a dimension or between dimensions. Results: All questions were classified: 53% are clinical cases with illnesses incidence in the genitourinary system; metabolic, nutritional and endocrine disease; cancer; infections and nervous system. In 81%, the object is a drug, mostly anti-infectious and anti-neoplastic agents. The therapeutic and safety areas had been the most requested, regarding the subjects: use, adverse reactions, drug identification and pharmaceutical technology. As to applicability, it was necessary to add some concepts and modify same hierarchical groups, that didn’t modify the basic structure, nor had collided with the desirable characteristics. The limitations were related with the incorporated external classification systems. The search in the subject dimension of the concept drug administration retrieved 19 questions. The search between two dimensions: antiinfectious (external) and teratogenicity (subject) retrieved three questions. In the two examples, it was possible to retrieve information from any one of the levels of the hierarchy, from the most general to the most specific and even from external dimensions. Conclusions: The use of the system in this sample showed its applicability in clinical information classification and filing, retrieval capacity and flexibility, supporting modifications without interfering with desirable characteristics. This tool allows retrieval of patient-oriented evidence that matters.

Managerial discretion is the focal theme bridging the clash between two schools of thoughts; whether executives have greater influence on their firms’ outcomes or other factors restrain their actions (Hambrick & Finkelstein, 1987). It is argued that constraints come from inertial, normative and environmental forces (e.g. DiMaggio & Powell, 1983). Of these restraints is the institutional environment in which a firm is headquartered. Our paper falls within this research stream and provides an extension for Crossland and Hambrick (2007, 2011) work. We investigate the national level of discretion in new cross-cultural contexts, provide deeper understanding of its concept, and shed the light on undiscovered discretion’s antecedents and consequences. We adopt a quantitative approach in which questionnaires represent our data collection instrument. We anticipate that in high discretion countries firms tend to follow what Miles & Snow (1978) labeled ‘Prospector’ strategy as opposed to low discretion countries in which firms incline to implement a ‘Defender’ strategy.

Ce mémoire s’inscrit dans une approche émergente en urbanisme qui cherche à mettre en dialogue les théories et les pratiques de l’urbanisme et la pensée du philosophe français Gilles Deleuze. Depuis quelques années, la pensée de Gilles Deleuze (surtout ses travaux coécrits avec Félix Guattari 1972; 1980) commence à s’immiscer dans les débats contemporains en urbanisme. Les travaux de Kim Dovey (2010; 2012), Jean Hillier (2005; 2007; 2011) et Colin McFarlane (2011a; 2011b) constituent les exemples les plus achevés d’une réflexion deleuzienne sur l’urbanisme. À degrés divers, ces auteurs mobilisent surtout cette pensée pour sa capacité à appréhender la complexité, le changement et l’instabilité (assemblage thinking). Pourtant, cette mobilisation de la pensée deleuzienne en urbanisme laisse largement intouchée le projet éthique et politique au coeur de la pensée de Gilles Deleuze. Le projet qui anime ce mémoire est d’explorer ce qu’une éthique deleuzienne peut apporter aux théories et pratiques de l’urbanisme. Cette éthique implique notamment un questionnement radical du cadre étatique dans lequel l’urbanisme s’insère, ce que ce mémoire appelle le «devenir- imperceptible » de l’urbanisme. Un travail empirique, constitué de 14 récits de ville prenant pour objet le territoire du pont Jacques-Cartier à Montréal, accompagne et poursuit cette réflexion théorique. Ces différents récits révèlent le pont Jacques-Cartier comme un lieu producteur de territoires, de mémoires et d’affects. Cette démarche terrain allie certains éléments de l’assemblage thinking et une éthique professionnelle deleuzienne. Elle explore la possibilité d’un rapport réellement sensible entre le territoire, l’urbaniste et les personnes concernées par une entreprise urbanistique.

Au Québec, le policier a l’autorité de faire respecter plusieurs lois et d’assurer le maintien de l’ordre. Puisque le policier peut être confronté à une multiplicité de problématiques, il est, dans certaines circonstances, contraint à employer une force pour se protéger lui-même ou pour protéger toute autre personne contre la mort ou contre des lésions corporelles graves. Cependant, bien que l’usage de la force par la police soit l’un des éléments les plus visibles et les plus controversés de l’intervention policière, les connaissances sur ce sujet spécifique demeurent limitées. Afin de pallier à certaines lacunes de la recherche et de la théorie, l’objectif général de ce mémoire vise à identifier les facteurs individuels, situationnels et contextuels liés au niveau de force utilisé par la police lors d’une intervention dans les postes de quartier et dans les secteurs de recensement. Les données dans le cadre de cette étude proviennent des rapports d’incidents déclarés d’usage de la force d’un service de police canadien pour une période couvrant les années 2007 à 2011, les données issues du recensement canadien de 2006 et les données criminelles issues du Programme de déclaration uniforme de la criminalité (DUC). La stratégie analytique privilégiée est la modélisation hiérarchique qui permettra de tester les liens entre les différents niveaux d’analyse. Au terme des analyses multiniveaux, les résultats indiquent que plusieurs facteurs ont un impact sur le niveau de force utilisé par le policier. Au niveau des déterminants individuels du sujet, c’est le fait d’être un homme, de résister physiquement à l’intervention et de posséder une arme qui est associé à des niveaux plus élevés de force. Au niveau des facteurs contextuels, on découvre que la proportion de minorité visible et le désavantage social ont un impact appréciable sur le niveau de force utilisé par le policier. Ces résultats nous amènent donc à conclure que les policiers tiennent surtout compte de la situation immédiate (facteurs individuels et situationnels) pour évaluer le degré de dangerosité ou de menace, mais qu’ils sont également influencés par l’endroit où ils se trouvent. D’autres études sont toutefois nécessaires pour vérifier la constance de nos résultats.

Cette recherche exploratoire a pour objet d’étude la mise en œuvre des interventions d’activation pour l’emploi auprès des prestataires de l’assistance sociale en situation de grande vulnérabilité au Québec. Elle cherche à décrire les pratiques mises en œuvre par des intervenants de la première ligne (« street-level ») au sein des organismes communautaires œuvrant en employabilité, dans le cadre des mesures et programmes de l’agence Emploi- Québec, dans un contexte de contractualisation et partenariat entre le gouvernement et le secteur à but non lucratif. Le mémoire cherche à identifier les processus qui peuvent influencer les pratiques de la première ligne de services lors de l’interaction avec les destinataires du système intégré d’aide financière de dernier recours et d’aide à l’emploi, de même qu’à comprendre les rapports de force en présence lors de la mise en œuvre des interventions. L’on veut répondre aux deux questions suivantes : Quel est le poids relatif du cadre institutionnel du système d’activation et du pouvoir discrétionnaire des intervenants de la première ligne travaillant directement avec les prestataires les plus vulnérables, au sein des organismes communautaires en employabilité? Plus précisément, comment ces deux types de processus interagissent-ils entre eux, pour produire la politique telle que mise en œuvre? On se base sur des approches théoriques sur la mise en œuvre des politiques inspirées des perspectives de « street level bureaucracy » (Lipsky, 1971, 1980; Smith et Lipsky, 1993 ; Hupe & Hill, 2007 ; Brodkin, 2007, 2011, 2012, 2015; Rice, 2012), du néo-institutionnalisme discursif (Schmidt, 2008, 2010) et de l’Ethnographie institutionnelle (Smith, 1999, 2006; Campbell & Gregor, 2008). On considère que la mise en œuvre de la politique d’activation entraîne non seulement une « appropriation » du cadre institutionnel et systémique par les intervenants de première ligne, mais aussi une co-construction de la politique dans un contexte de gouvernance spécifique. La stratégie de recherche est qualitative et s’inspire de l’approche de l’ethnographie institutionnelle en sociologie, qui propose une méthodologie pour retracer les effets des institutions sur les travailleurs dans leur vie quotidienne (Smith, 2005, 1999, 1987; Campbell and Gregor, 2008). Elle comprend : i) des entretiens avec des intervenants communautaires qui fournissent des services d’aide et d’accompagnement pour l’intégration en emploi portant sur certains dossiers de prestataires du système; ii) des entretiens avec des coordinatrices d’organismes communautaires œuvrant en employabilité, et iii) une collecte et une analyse de la documentation institutionnelle et du cadre de réglementation pour la mise en œuvre du système d’activation. L'analyse aborde le discours institutionnel d'une part et, d'autre part, le discours des répondants du secteur communautaire en employabilité concernant leurs interventions auprès des prestataires dans le cadre des programmes et mesures d’E-Q, et leurs « explications » des pratiques quotidiennes.

Si se considera que una inadecuada gestión financiera pone en riesgo la perdurabilidad de la empresa y los esfuerzos realizados por la organización en búsqueda constante del crecimiento, es de vital importancia que el profesional a cargo de funciones financieras cuente con una alta capacitación en herramientas de gestión. Este libro pretende acercar al estudiante a los conceptos básicos que soportan la gestión de las finanzas en el marco corporativo, familiarizándolo con las diferentes alternativas tradicionales con las que cuentan las empresas para financiarse en el largo plazo, los costos de estas fuentes de financiación, su relación con el riesgo empresarial y las expectativas de rentabilidad de los inversionistas. Al mismo tiempo, el texto evidencia la importancia de la administración del riesgo en las decisiones de inversión para la conservación de los activos y la perdurabilidad empresarial.
