The recent description of the respiratory pathogen human metapneumovirus (hMPV) has highlighted a deficiency in current diagnostic techniques for viral agents associated with acute lower respiratory tract infections. We describe two novel approaches to the detection of viral RNA by use of reverse transcriptase PCR (RT-PCR). The PCR products were identified after capture onto a solid-phase medium by hybridization with a sequence-specific, biotinylated oligonucleotide probe. The assay was applied to the screening of 329 nasopharyngeal aspirates sampled from patients suffering from respiratory tract disease. These samples were negative for other common microbial causes of respiratory tract disease. We were able to detect hMPV sequences in 32 (9.7%) samples collected from Australian patients during 2001. To further reduce result turnaround times we designed a fluorogenic TaqMan oligoprobe and combined it with the existing primers for use on the LightCycler platform. The real-time RT-PCR proved to be highly reproducible and detected hMPV in an additional 6 out of 62 samples (9.6%) tested during the comparison of the two diagnostic approaches. We found the real-time RT-PCR to be the test of choice for future investigation of samples for hMPV due to its speed, reproducibility, specificity, and sensitivity.

We assayed mtDNA haplotype [300 base pairs (bp) control region] geography and genealogy in the Indo-Pacific tasselfish, Polynemus sheridani from its contiguous estuarine distribution across northern Australia (n = 169). Eight estuaries were sampled from three oceanographic regions (Timor Sea, Gulf of Carpentaria and the Coral Sea) to assess the impact of Pleistocene sea level changes on the historical connectivity among P. sheridani populations. Specifically, we investigated the genetic consequences of disruption to Indian-Pacific Ocean connectivity brought about by the closure of the Torres Strait. Overall there was significant population subdivision among estuaries (F-ST = 0.161, (Phi(ST) = 0.187). Despite a linear distribution, P. sheridani did not show isolation by distance over the entire sampled range because of genetic similarity of estuaries greater than 3000 km apart. However, significant isolation by distance was detected between estuaries separated by less than 3000 km of coastline. Unlike many genetic studies of Indo-Pacific marine species, there was no evidence for an historical division between eastern and western populations. Instead, phylogeographical patterns were dominated by a starlike intraspecific phylogeny coupled with evidence for population expansion in both the Gulf of Carpentaria and the Coral Sea but not the Timor Sea. This was interpreted as evidence for recent west to east recolonization across of northern Australia following the last postglacial marine advance. We argue that although sufficient time has elapsed postcolonization for populations to approach gene flow-drift equilibrium over smaller spatial scales (< 3000 km), the signal of historical colonization persists to obscure the expected equilibrium pattern of isolation by distance over large spatial scales (> 3000 km).

Epstein-Barr virus (EBV)-encoded oncogene latent membrane protein (LMP) 1, which is consistently expressed in multiple EBV-associated malignancies, has been proposed as a potential target antigen for any future vaccine designed to control these malignancies. However, the high degree of genetic variation in the LMP1 sequence has been considered a major impediment for its use as a potential immunotherapeutic target for the treatment of EBV-associated malignancies. In the present study, we have employed a highly efficient strategy, based on ex vivo functional assays, to conduct an extensive sequence-wide analysis of LMP1-specific T-cell responses in a large panel of healthy virus carriers of diverse ethnic origin and nasopharyngeal carcinoma patients. By comparing the frequencies of T cells specific for overlapping peptides spanning LMP1, we mapped a number of novel HLA class I- and class II-restricted LMP1 T-cell epitopes, including an epitope with dual HLA class I restriction. More importantly, extensive sequence analysis of LMP1 revealed that the majority of the T-cell epitopes were highly conserved in EBV isolates from Caucasian, Papua New Guinean, African, and Southeast Asian populations, while unique geographically constrained genetic variation was observed within one HLA A2 supertype-restricted epitope. These findings indicate that conserved LMP1 epitopes should be considered in designing epitope-based immunotherapeutic strategies against EBV-associated malignancies in different ethnic populations.

The Epstein-Barr virus latent membrane protein (LMP 1) functions as a constitutively active signalling molecule and associates in lipid rafts clustered with other signalling molecules. Using immunofluorescent confocal microscopy, LMP 1 was shown to have an heterogeneous distribution among individual cells which was not related to the cell cycle stage. LMP 1 was shown to localize to intracellular compartments in cells other than the plasma membrane, Co-labelling of cells with both an LIMP 1 antibody and an antibody to the Golgi protein GS15 revealed that the intracellular LMP 1 partly co-localized with the Golgi apparatus. Further confirmation of intracellular LMP 1 localization was obtained by immunoelectron microscopy with rabbit polyclonal LIMP 1 antibodies and cryosectioning. As well as being present in intracellular foci, LMP 1 co-localized in part with MHC-II and was present on exosomes derived from a lymphoblastoid cell line. Preparations of LMP 1 containing exosomes were shown to inhibit the proliferation of peripheral blood mononuclear cells, suggesting that LIMP 1 could be involved in immune regulation. This may be of particular relevance in EBV-associated tumours such as nasopharyngeal carcinoma and Hodgkin's disease, as LMP 1-containing exosomes may be taken up by infiltrating T-lymphocytes, where LMP 1 could exert an anti-proliferative effect, allowing the tumour cells to evade the immune system.

This study reports on a block clinical trial of two types of central venous catheters (CVCs): antiseptic-impregnated catheters (AIC) and non-impregnated catheters (non-AIC), on catheter tip colonization and bacteraemia. In total, 500 catheters were inserted in 390 patients over the 18 month study period, 260 (52.0%) AIC and 240 (48.0%) non-AIC. Of these, 460 (92.0%) tips (237 AIC and 223 non-AIC) were collected. While significantly fewer AIC, 14 (5.9%), than non-AIC, 30 (13.5%), catheters were colonized (P < 0.01), there was no difference in the rates of bacteraemias in the two groups (0.8% vs. 2.7%, respectively, P = 0.16). There were 6.87 (95% CI 3.38-14.26) and 16.92 (95% CI 10.61-27.12) colonized AIC and non-AIC catheters, respectively, per 1000 catheter days, a difference that was significant (P < 0.01). However, no difference emerged between bacteraemias in AIC and non-AIC catheters per 1000 catheter days measured at 0.98 (95% CI 0.24-5.54) and 3.38 (95% CI 1.29-9.34), respectively (P = 0.10). Of the 444 CVCs that were sited in the subclavian or jugular veins and had tips collected, significantly more catheters were colonized in the jugular group, 19 (20%), compared with the subclavian group, 24 (6.9%; P less than or equal to 0.01). Overall, the low rates of colonization and bacteraemia may be explained by the population studied, the policies used and the employment of a clinical nurse dedicated to CVC management. A hipótese defendida nesta pesquisa se baseia na possibilidade de a arquitetura jesuítica implantada em terras brasileiras (século XVI) dialogar e agenciar, num mesmo corpo edificado, e de modo inter-relacionado, aspectos relativos à morfologia urbana, tipologia e paisagem. Lama explica que, como disciplina, a morfologia urbana agrega para si não somente o ambiente construído, mas os meios pelos quais este foi construído em sua interação com a forma urbana, ou seja, os “fenômenos sociais, econômicos e outros motores da urbanização” (LAMAS, 1992). Entender a forma urbana é entender seus elementos constituintes, “quer em ordem à leitura ou análise do espaço, quer em ordem à sua concepção ou produção” (LAMAS, 1992). Estudar a forma urbana significa compreender o lugar onde se insere a cidade e seus elementos constituintes, seus espaços e a inter-relação entre eles e seu contexto, em um espectro abrangente do que se denomina cidade, e urbano. A tipologia arquitetônica e a morfologia urbana estão interligadas no cerne de suas análises, considerando que ambas, segundo Pereira, estudam “duas ordens de fatos homogêneos” (PEREIRA, 2012); estudam elementos constituintes da cidade – arquitetônicos e espaciais – que se sobrepõem ou se complementam de acordo com a escala de análise utilizada. A arquitetura jesuítica do Brasil colonial modela de modo determinante a construção de distintos núcleos urbanos originários na costa brasileira no século XVI. Isso, por meio da implantação de tipologia edilícia que acompanha a doutrina jesuítica de localização e escolha do sítio para suas construções, preconizando segurança, visibilidade do entorno e facilidade de acesso por rios ou pelo mar. Essas construções, realizadas em áreas elevadas, marcaram, por conseguinte, no tempo e no espaço, a paisagem dos primeiros núcleos urbanos brasileiros. A pesquisa analisou um dos exemplares históricos da arquitetura jesuítica no Estado do Espírito Santo, especificamente na cidade de Vitória, capital e núcleo urbano original da colonização portuguesa neste Estado. A instalação dos jesuítas na antiga Vila da Vitória, no séc. XVI, através de sua igreja dedicada a São Tiago e de seu colégio anexo, marca a presença tipológica de uma arquitetura religiosa que influencia a própria morfologia da cidade – caracterizando esta arquitetura como um tipomorfológico - e, por reflexo, participa da construção de sua paisagem urbana secular. Entende-se que o antigo complexo jesuítico de São Tiago e atual Palácio Anchieta, sede governamental e prédio cultural capixaba, é uma arquitetura que permeia estas três grandes narrativas arquitetônicas e urbanas: a tipologia, a morfologia e a paisagem.

O estudo teve como objetivo compreender de forma crítica como se caracteriza a gestão participativa no campus Serra do Ifes, a partir do entendimento e do processo interativo dos servidores Técnico-administrativos (TAEs) das classes C, D e E, e das chefias imediatas. Para tanto, a Teoria da Ação Comunicativa (TAC) de Jürgen Habermas, bem como sua proposta de democracia deliberativa foram empreendidas como marco teórico para compreensão desse processo e os conceitos norteadores da TAC (atos de fala, mundo da vida, sistema, colonização do mundo da vida, esfera pública, ação comunicativa e instrumental ou estratégica) foram empregados como elementos de análise das estruturas relevantes identificadas na pesquisa. Esse trabalho caracteriza-se como uma pesquisa de abordagem qualitativa e possui um enfoque crítico com visão dialética da realidade social. A análise foi realizada considerando o duplo efeito que a prática da gestão participativa pode significar: como comprometimento com o desempenho e reforço do sistema capitalista; e como resistência dos trabalhadores organizados às formas de dominação e controle. Os dados empíricos foram produzidos por meio de pesquisa documental, observação participante e entrevista semiestruturada. Foram entrevistados oito TAEs subordinados e 13 chefias imediatas, selecionados conforme o critério da bola de neve. Utilizou-se a análise de conteúdo para o tratamento das informações obtidas nas entrevistas. Os resultados apontam para a construção de ações participativas de cunho instrumental, estabelecidas a partir dos interesses da gestão, de grupos específicos e individuais. Sendo assim, a prática da gestão participativa caracteriza-se como um espaço estratégico para alcance do êxito e não do entendimento, onde os TAEs são corresponsáveis no processo de manutenção e construção dos fenômenos que emperram o desenvolvimento de uma participação democrática.

A história de Cabo Verde teve início com o seu achamento em 1460 pelos navegadores portugueses que, apesar dos condicionalismos da região, deram arranque ao povoamento, começando uma vida baseada no comércio com a costa africana e, depois, com a Europa e a América por causa do tráfico negreiro. Para Portugal, Cabo Verde era uma fonte de receitas por causa da posição geoestratégica. O rei nomeava representantes que administravam as ilhas para recolherem os lucros do comércio e os enviarem para a metrópole. Cabo Verde demorou cinco séculos até ser independente e no arquipélago nunca houve luta armada contra a presença portuguesa. A 5 de Julho de 1975, tornou-se independente optou pelo monopartidarismo que vigorou quinze anos. Em 1990, fruto da alteração da conjuntura internacional e da saturação a nível interno, deu-se a abertura ao multipartidarismo, com a criação do MpD, que venceu as primeiras eleições multipartidárias. Depois o PAICV regressaria ao Poder pela força dos votos e esta alternância foi bem aceite por todos porque provou a existência de democracia no arquipélago. Cabo Verde tem aproveitado a cooperação para o desenvolvimento, como se verifica pela evolução do IDH. Actualmente, a parceria com a UE constitui um desafio e uma oportunidade.

Formaldehyde (FA), also known as formalin, formal and methyl aldehydes, is a colorless, flammable, strong-smelling gas. It has an important application in embalming tissues and that result in exposures for workers in the pathology anatomy laboratories and mortuaries. Occupational exposure to FA has been shown to induce nasopharyngeal cancer and has been classified as carcinogenic to humans (group 1) on the basis of sufficient evidence in humans and sufficient evidence in experimental animals. Manifold in vitro studies clearly indicated that FA is genotoxic. FA induced various genotoxic effects in proliferating cultured mammalian cells. The cytokinesis-block micronucleus (CBMN) assay was originally developped as an ideal system form easuring micronucleus (MN), however it can also be used to measure nucleoplasmic bridges (NBP) and nuclear buds (NBUD). Over the past decade another unique mechanism of micronucleus formation, known as nuclear budding has emerged. NBUDS is considered as a marker of gene amplification and/or altered gene dosage because the nuclear budding process is the mechanism by which cells removed amplified and/excess DNA.

Formaldehyde (FA) the most simple and reactive of all aldehydes, is a colorless, reactive and readily polymerizing gas at normal temperature. It has a pungent, suffocating odour that is recognized by most human subjects at concentrations below 1ppm. According to the Report on Carcinogens, FA ranks 25th in the overall U.S. chemical production with more than 11 billion pounds (5 million tons) produced each year. Is an important industrial compound that is used in the manufacture of synthetic resins and chemical compounds such as lubricants and adhesives. It has also applications as a disinfectant, preservative and is used in cosmetics. Estimates of the number of persons who are occupationally exposed to FA indicate that, at least at low levels, may occur in a wide variety of industries. The occupational settings with most extensive use of formaldehyde is in the production of resins and in anatomy and pathology laboratories. Several studies reported a carcinogenic effect in humans after inhalation of FA, in particular an increased risk for nasopharyngeal cancer. Nowadays, the International Agency for Research on Cancer (IARC) classifies FA as carcinogenic to humans (group 1), on the basis of sufficient evidence in humans and sufficient evidence in experimental animals. Manifold in vitro studies clearly indicated that FA is genotoxic. FA induced various genotoxic effects in proliferatin cultured mammalian cells. A variety of evidence suggests that the primary DNA alterations after FA exposure are DNA-protein crosslinks. Incomplete repair of DPX can lead to the formation of mutations.

Formaldehyde (FA) is ubiquitous in the environment and is a chemical agent that possesses high reactivity. Occupational exposure to FA has been shown to induce nasopharyngeal cancer and has been classified as carcinogenic to humans (group 1) on the basis of sufficient evidence in humans and sufficient evidence in experimental animals. The exposure to this substance is epidemiologically linked to cancer and nuclear changes detected by the cytokinesis-block micronucleus test (CBMN). This method is extensively used in molecular epidemiology, since it determines several biomarkers of genotoxicity, such as micronucleus (biomarkers of chromosomes breakage or loss), nucleoplasmic bridges (biomarker of chromosome rearrangement, poor repair and / or telomeres fusion) and nuclear buds (biomarker of elimination of amplified DNA). The gene X-ray repair cross-complementing group 3 (XRCC3) is involved in homologous recombination repair of cross-links and chromosomal double-strand breaks and at least one polymorphism has been reported in codon 241, a substitution of a methionine for a threonine.

Formaldehyde is classified by IARC as carcinogenic to humans (nasopharyngeal cancer). Tobacco smoke has been epidemiologically associated to a higher risk of development of cancer, especially in the oral cavity, larynx and lungs, as these are places of direct contact with many carcinogenic tobacco’s compounds. XRCC3 is involved in homologous recombination repair of cross-links and chromosomal double-strand breaks (Thr241Met polymorphism). The aim of the study is to determine whether there is an in vivo association between genetic polymorphism of the gene XRCC3 and the frequency of genotoxicity biomarkers in subjects exposed or not to formaldehyde and with or without tobacco consumption.

Occupational exposure to formaldehyde (FA) has been shown to induce nasopharyngeal cancer and has been classified as carcinogenic to humans (group 1) on the basis of sufficient evidence in humans. Tobacco smoke has been associated to a higher risk of development of cancer, especially in the oral cavity, larynx and lungs, as these are places of direct contact with many carcinogenic tobacco’s compounds. Alcohol is a recognized agent that influence cells in a genotoxic form, been citied as a strong agent with potential in the development of carcinogenic lesions. Epidemiological evidence points to a strong synergistic effect between cigarette smoking and alcohol consumption in the induction of cancers in the oral cavity. Approximately 90% of human cancers originate from epithelial cells. Therefore, it could be argued that oral epithelial cells represent a preferred target site for early genotoxic events induced by carcinogenic agents entering the body via inhalation and ingestion. The MN assay in buccal cells was also used to study cancerous and precancerous lesions and to monitor the effects of a number of chemopreventive agents.

The International Agency for Research on Cancer classified formaldehyde as carcinogenic to humans because there is “sufficient epidemiological evidence that it causes nasopharyngeal cancer in humans”. Genes involved in DNA repair and maintenance of genome integrity are critically involved in protecting against mutations that lead to cancer and/or inherited genetic disease. Association studies have recently provided evidence for a link between DNA repair polymorphisms and micronucleus (MN) induction. We used the cytokinesis-block micronucleus (CBMN assay) in peripheral lymphocytes and MN test in buccal cells to investigate the effects of XRCC3 Thr241Met, ADH5 Val309Ile, and Asp353Glu polymorphisms on the frequency of genotoxicity biomarkers in individuals occupationally exposed to formaldehyde (n = 54) and unexposed workers (n = 82). XRCC3 participates in DNA double-strand break/recombination repair, while ADH5 is an important component of cellular metabolism for the elimination of formaldehyde. Exposed workers had significantly higher frequencies (P < 0.01) than controls for all genotoxicity biomarkers evaluated in this study. Moreover, there were significant associations between XRCC3 genotypes and nuclear buds, namely XRCC3 Met/Met (OR = 3.975, CI 1.053–14.998, P = 0.042) and XRCC3 Thr/Met (OR = 5.632, CI 1.673–18.961, P = 0.005) in comparison with XRCC3 Thr/Thr. ADH5 polymorphisms did not show significant effects. This study highlights the importance of integrating genotoxicity biomarkers and genetic polymorphisms in human biomonitoring studies.
