Individual planktonic microfossil species, or assemblage groups of different species, are often used to, qualitatively and/or quantitatively, reconstruct past (sub)surface-water conditions of the world's oceans and seas. Until now, little information has been available on the surface sediment distribution patterns and paleoenvironmental reconstruction potential of coccolith, calcareous dinoflagellate cyst and organic-walled dinoflagellate cyst assemblages of the South and equatorial Atlantic, especially at the species level. This paper (i) summarizes the distributions of these three phytoplanktonic microfossil groups in numerous Atlantic surface sediments from 20°N-50°S and 30°E-65°W and determines their relationship with the physicochemical and trophic conditions of the overlying (sub)surface-waters, and (ii) determines the synecology of the three phytoplankton groups by carrying out statistical analyses (i.e. detrended and canonical correspondence analyses) on all groups simultaneously. Ecological relationships are additionally strengthened by statistically comparing the distribution patterns of the phytoplankton groups with those of planktonic foraminifera (Pflaumann et al. 1996; Niebler et al. 1998), as the ecological preferences of the latter are much better known. Many of the analyzed phytoplanktonic microfossil species or groups of species in the surface sediments do show restricted distributions which primarily reflect the environmental conditions of the upper water masses above them (e.g. sea-surface temperature, productivity, stratification). The acquired 'reference' data sets are large and diverse enough to allow future development of transfer functions for the reconstruction of past surface-water conditions, and show that there is still an enormous paleoenvironmental reconstruction potential concealed in many fossil coccolith and dinoflagellate cyst assemblages.

Cores from the 11 sites drilled during Leg 180 showed radiolarian assemblages that appear only in the Quaternary sediments. The most diverse and well-preserved assemblages were found in hemipelagic sediments from Holes 1108A, 1110A, and 1115B.

Fingolimod is a Multiple Sclerosis treatment licensed in Europe since 2011. Its efficacy has been demonstrated in three large phase III trials, used in the regulatory submissions throughout the world. As usual, in these trials the inclusion and exclusion criteria were designed to obtain a homogeneous population, with interchangeable characteristics in the different treatment arms. Although this is the best strategy to achieve a robust answer to the investigation question, it does not guaranty the treatment efficacy in the clinical practice, since in the real world there are concomitant treatments, comorbidities, adherence and persistence challenges. But, to make informed treatment decision for a real life patient, we need to have evidence of the treatment efficacy, what has been called treatment effectiveness. This work aims to review fingolimod effectiveness, using as source of information abstracts, posters and manuscripts. This unorthodox strategy was developed because more than half of the published experience with fingolimod is still on abstracts and posters. Only a small part of the studies reviewed are already published in peer reviewed journals. Fingolimod seems to be, at least, as effective and safe as it was on clinical trials, and with its long term experience no new safety signals were observed. In the Portuguese hospital perspective, early treatment with fingolimod is expected to result in better clinical outcomes associated with a more efficient healthcare resources allocation.

Heat stress negatively affects wheat performance during its entire cycle, particularly during the reproductive stage. In view of the climate change and the prediction of a continued increase in temperature in the new future, it is urgent to concentrate efforts to discover novel genetic sources able to improve the resilience of wheat to heat stress. In this direction, this study addressed two different experiments in durum wheat to identify novel QTLs suitable to be applied in marker-assisted selection for heat tolerance. Chlorophyll fluorescence (ChlF) is a valuable indicator of plant response to environmental changes allowing a detailed assessment of PSII activity in view of its non-invasive measurement and high-throughput phenotyping. In the first study (Chapter 2), the Light-Induced Fluorescence Transient (LIFT) method was used to access ChlF data to map QTLs for ChlF-related traits during the vegetative growth stage in durum wheat under heat stress condition. Our results provide evidence that LIFT consistently measures ChlF at the level of high-throughput phenotyping combined with high accuracy which is required for Genome-Wide Association Study (GWAS) aimed at identifying genomic regions affecting PSII activity. The 50 QTLs identified for ChlF-related traits under heat stress mostly clustered into five chromosomes hotspots unrelated to phenology, a feature that makes these QTLs a valuable asset for marker-assisted breeding programs across different latitudes. In the second study (Chapter 3), a set of 183 accessions suitable for GWAS, was exposed to optimal and high temperature during two crop seasons under field conditions. Important agronomic traits were evaluated in order to identify valuable QTLs for GY and its components. The GWAS analysis identified several QTLs in the single years as well as in the joint analysis. From the total QTLs identified, 13 QTL clusters can be highlighted to be affecting heat tolerance across different years and/or different traits.

This thesis aimed to characterise two large tetraploid germplasm collections. The Global Durum Panel, involving modern cultivars and landrances and the Tetraploid Global Collection which comprises all the tetraploid wheat subgroups. Two distinct parallel studies were carried out. The first is focused on the characterisation of both collection for yield and quality related traits. The panel were phenotyped for two consecutive years each. In this phase the following traits were collected: the number of fertile spikelets per spike, the number of fertile florets of central spikelet for the spike-related traits. The following grain related traits were also phenotyped: the thousand kernel weight, the average grain area, average grain length, average grain width, grain brightness, grain redness, grain yellowness. GWAS analysis were performed for each collected trait and major QTLs were subjected to candidate gene analysis. Major QTLs emerging from GWA study were located on chromosome 2A with a strong bibliographic evidence for grain number-related traits such as the fertile spikelet number, the number of fertile florets per central spikelet. On the other hand two evident peaks were detected on chromosomes 6A and 7B for grain size and weight related traits. The second work was focused on the characterisation of the Global Durum Panel for root system architecture components, namely the root growth angle. GWAS analysis was perfomed and three major QTLs were detected on chromosome 2A, 6A and 7A. These three QTLs all have a bibliographic evidence.
