Myostatin, ein Mitglied der TGF-β Familie von Wachstumsfaktoren, ist ein negativer Regulator des Skelettmuskelwachstums. Obwohl Myostatin nach einer Vielzahl pathologischer Zustände im Herzen massiv hochreguliert wird, ist die physiologische und pathophysiologische Funktion von Myostatin im Herzen noch kaum erforscht. Deshalb wurde im Rahmen dieser Dissertation die Funktion von Myostatin im adulten Herzen untersucht. Dazu wurden Mausmodelle, in denen Myostatin in Kardiomyozyten deletiert und überexprimiert wird, verwendet. Ich konnte zeigen, dass die akute Deletion von Myostatin in Kardiomyozyten zu einer erhöhten Lethalität, Herzinsuffizienz und Hypertrophie führt. Dabei konnte ich eine Aktivierung der AMP-aktivierten Kinase (AMPK) als Ursache der Hypertrophie identifizieren und mit Hilfe eines AMPK Inhibitors die Entstehung der Hypertrophie in vivo verhindern. Des Weiteren konnte ich in vivo und in vitro zeigen, dass Myostatin AMPK über die TGF-β-aktivierte Kinase 1 (TAK1) und seinen kanonischen Rezeptor inhibiert. Die akute Deletion von Myostatin hemmte auch die Expression von Rgs2, einem Inhibitor der Gq Signalkaskade, und führte dadurch zu einer Aktivierung dieses für Herzinsuffizienz elementaren Signalweges. Außerdem verbesserte die akute adulte Überexpression von Myostatin die Herzkontraktilität leicht, während eine langfristige Überexpression eine interstitielle Fibrose, die über TAK1 und p38 vermittelt wird, induzierte. Hiermit konnte ich Myostatin als neuen Regulator der Hypertrophie und Herzinsuffizienz etablieren.rn

The dissertation is devoted to the investigation of the process of triplet-triplet annihilation assisted upconversion (TTA – UC). The current state of the art of the used nowadays emitters and sensitizers was overviewed. The new emitters, synthesized mainly on the base of perylene, were created and analyzed for the applicability for TTA – UC in the combination with the sensitizers from tetrabenzo- and tetranaphtoporphyrin families. A strong influence of the excitation beam parameters on UC efficiency was shown. The new strategy of an effective UC emitter’s creation by comprising the functions of two chromophors in a dyad, where the function of the triplet-triplet transfer (TTT) and the triplet-triplet annihilation (TTA) are attributed to different parts of the molecule, was represented. The successful transfer of an UC medium into the water environment by the encapsulation of hydrophobic UC dyes into the micelles, formed by the PTS surfactant, was done. Resulting quantum efficiency is comparable with that in an organic solvent. An extremely strong dependence of the efficiency of the TTA – UC process on temperature in such micellar structures was obtained

Das Ziel dieser Arbeit ist die Konstruktion eines Homomorphismus von partiell definierten, graduiert-kommutativen Algebren, der nach Ubergang zu rationalen Kohomologiegruppen mit der Regulatorabbildung reg zwischen motivischer und Deligne-Beilinson Kohomologie übereinstimmt.rnZu Beginn der Arbeit werden verschiedene Komplexe beschrieben, mit denen sich die motivische und die Deligne-Beilinson Kohomologie berechnen lassen.rnIm ersten Kapitel wird der Komplex der höheren Chow Ketten und der Unterkomplex der "alternierenden" Ketten "in guter Lage" eingeführt, die beide die motivische Kohomologie berechnen (letzterer mit rationalen Koeffizienten).rnIn den folgenden beiden Kapiteln werden Komplexe C_D und P_D beschrieben, mit denen sich die (rationale) Deligne-Beilinson Kohomologie berechnen lässt. Diese sind aufgebaut aus sogenannten Strömen, die im zweiten Kapitel eingeführt werden. Verknüpft sind die beiden Komplexe durch eine Auswertungsabbildung ev, die für rationale Koeffizienten zu einem Quasi-Isomorphismus wird. Auf beiden Komplexen lassen sich (Schnitt-)Produkte definieren, von denen jedoch nur das Produkt auf P_D gleichzeitig assoziativ und graduiert-kommutativ ist.rnIm vierten Kapitel wird ganz allgemein für eine Familie von Komplexen, die einer Reihe an Anforderungen genügt, ein (partiell definierter) Homomorphismus (der Regulator) von dem Komplex der höheren Chow Ketten in eben diese Komplexe konstruiert. Die beiden oben genannten Komplexe erfüllen diese Anforderungen und liefern daher Regulatoren reg_C und reg_P

Among the many cell types that may prove useful to regenerative medicine, mounting evidence suggests that human term placenta-derived cells will join the list of significant contributors. In making new cell therapy-based strategies a clinical reality, it is fundamental that no a priori claims are made regarding which cell source is preferable for a particular therapeutic application. Rather, ongoing comparisons of the potentiality and characteristics of cells from different sources should be made to promote constant improvement in cell therapies, and such comparisons will likely show that individually tailored cells can address disease-specific clinical needs. The principle underlying such an approach is resistance to the notion that comprehensive characterization of any cell type has been achieved, neither in terms of phenotype nor risks-to-benefits ratio. Tailoring cell therapy approaches to specific conditions also requires an understanding of basic disease mechanisms and close collaboration between translational researchers and clinicians, to identify current needs and shortcomings in existing treatments. To this end, the international workshop entitled "Placenta-derived stem cells for treatment of inflammatory diseases: moving toward clinical application" was held in Brescia, Italy, in March 2009, and aimed to harness an understanding of basic inflammatory mechanisms inherent in human diseases with updated findings regarding biological and therapeutic properties of human placenta-derived cells, with particular emphasis on their potential for treating inflammatory diseases. Finally, steps required to allow their future clinical application according to regulatory aspects including good manufacturing practice (GMP) were also considered. In September 2009, the International Placenta Stem Cell Society (IPLASS) was founded to help strengthen the research network in this field.

Background During production and processing of multi-walled carbon nanotubes (MWCNTs), they may be inhaled and may enter the pulmonary circulation. It is essential that interactions with involved body fluids like the pulmonary surfactant, the blood and others are investigated, particularly as these interactions could lead to coating of the tubes and may affect their chemical and physical characteristics. The aim of this study was to characterize the possible coatings of different functionalized MWCNTs in a cell free environment. Results To simulate the first contact in the lung, the tubes were coated with pulmonary surfactant and subsequently bound lipids were characterized. The further coating in the blood circulation was simulated by incubating the tubes in blood plasma. MWCNTs were amino (NH2)- and carboxyl (-COOH)-modified, in order to investigate the influence on the bound lipid and protein patterns. It was shown that surfactant lipids bind unspecifically to different functionalized MWCNTs, in contrast to the blood plasma proteins which showed characteristic binding patterns. Patterns of bound surfactant lipids were altered after a subsequent incubation in blood plasma. In addition, it was found that bound plasma protein patterns were altered when MWCNTs were previously coated with pulmonary surfactant. Conclusions A pulmonary surfactant coating and the functionalization of MWCNTs have both the potential to alter the MWCNTs blood plasma protein coating and to determine their properties and behaviour in biological systems.

Background Since late 2003, the highly pathogenic influenza A H5N1 had initiated several outbreak waves that swept across the Eurasia and Africa continents. Getting prepared for reassortment or mutation of H5N1 viruses has become a global priority. Although the spreading mechanism of H5N1 has been studied from different perspectives, its main transmission agents and spread route problems remain unsolved. Methodology/Principal Findings Based on a compilation of the time and location of global H5N1 outbreaks from November 2003 to December 2006, we report an interdisciplinary effort that combines the geospatial informatics approach with a bioinformatics approach to form an improved understanding on the transmission mechanisms of H5N1 virus. Through a spherical coordinate based analysis, which is not conventionally done in geographical analyses, we reveal obvious spatial and temporal clusters of global H5N1 cases on different scales, which we consider to be associated with two different transmission modes of H5N1 viruses. Then through an interdisciplinary study of both geographic and phylogenetic analysis, we obtain a H5N1 spreading route map. Our results provide insight on competing hypotheses as to which avian hosts are responsible for the spread of H5N1. Conclusions/Significance We found that although South China and Southeast Asia may be the virus pool of avian flu, East Siberia may be the source of the H5N1 epidemic. The concentration of migratory birds from different places increases the possibility of gene mutation. Special attention should be paid to East Siberia, Middle Siberia and South China for improved surveillance of H5N1 viruses and monitoring of migratory birds.

The authors examine children's access to and caregiver's satisfaction with organizations that provide leisure time activities for children on Saturdays. The authors argue that access and satisfaction are a function of familie's financial, cultural and social capital. Using data on 1,036 households in the Phoenix metropolitan area in 2003-04, the authors found that families' financial and cultural capital affected whether or not children participate din activities organized by organizations, but family ties to the organization directly (e.g., either worked there, volunteered, donated) resulted in caregivers being more satisfied with the services. The authors also found that the benefits of network closure (caregivers knowing the parents of other children on site) were greater the riskier the activities of the child (e.g., sports or cheerleading). Contrary to the authors expectations, having family or friends in the area did not affect caregiver's satisfaction with the child's provider.

Background Increasing concern has been expressed regarding the potential adverse health effects that may be associated with human exposure to inhaled multi-walled carbon nanotubes (MWCNTs). Thus it is imperative that an understanding as to the underlying mechanisms and the identification of the key factors involved in adverse effects are gained. In the alveoli, MWCNTs first interact with the pulmonary surfactant. At this interface, proteins and lipids of the pulmonary surfactant bind to MWCNTs, affecting their surface characteristics. Aim of the present study was to investigate if the pre-coating of MWCNTs with pulmonary surfactant has an influence on potential adverse effects, upon both (i) human monocyte derived macrophages (MDM) monocultures, and (ii) a sophisticated in vitro model of the human epithelial airway barrier. Both in vitro systems were exposed to MWCNTs either pre-coated with a porcine pulmonary surfactant (Curosurf) or not. The effect of MWCNTs surface charge was also investigated in terms of amino (−NH2) and carboxyl (−COOH) surface modifications. Results Pre-coating of MWCNTs with Curosurf affects their oxidative potential by increasing the reactive oxygen species levels and decreasing intracellular glutathione depletion in MDM as well as decreases the release of Tumour necrosis factor alpha (TNF-α). In addition, an induction of apoptosis was observed after exposure to Curosurf pre-coated MWCNTs. In triple cell-co cultures the release of Interleukin-8 (IL-8) was increased after exposure to Curosurf pre-coated MWCNTs. Effects of the MWCNTs functionalizations were minor in both MDM and triple cell co-cultures. Conclusions The present study clearly indicates that the pre-coating of MWCNTs with pulmonary surfactant more than the functionalization of the tubes is a key factor in determining their ability to cause oxidative stress, cytokine/chemokine release and apoptosis. Thus the coating of nano-objects with pulmonary surfactant should be considered for future lung in vitro risk assessment studies. The role of platelets in hemostasis is to produce a plug to arrest bleeding. During thrombocytopenia, spontaneous bleeding is seen in some patients but not in others; the reason for this is unknown. Here, we subjected thrombocytopenic mice to models of dermatitis, stroke, and lung inflammation. The mice showed massive hemorrhage that was limited to the area of inflammation and was not observed in uninflamed thrombocytopenic mice. Endotoxin-induced lung inflammation during thrombocytopenia triggered substantial intra-alveolar hemorrhage leading to profound anemia and respiratory distress. By imaging the cutaneous Arthus reaction through a skin window, we observed in real time the loss of vascular integrity and the kinetics of skin hemorrhage in thrombocytopenic mice. Bleeding-observed mostly from venules-occurred as early as 20 minutes after challenge, pointing to a continuous need for platelets to maintain vascular integrity in inflamed microcirculation. Inflammatory hemorrhage was not seen in genetically engineered mice lacking major platelet adhesion receptors or their activators (alphaIIbbeta3, glycoprotein Ibalpha [GPIbalpha], GPVI, and calcium and diacylglycerol-regulated guanine nucleotide exchange factor I [CalDAG-GEFI]), thus indicating that firm platelet adhesion was not necessary for their supporting role. While platelets were previously shown to promote endothelial activation and recruitment of inflammatory cells, they also appear indispensable to maintain vascular integrity in inflamed tissue. Based on our observations, we propose that inflammation may cause life-threatening hemorrhage during thrombocytopenia.

Die nachhaltige Veränderung der Gesellschaft stellt die Schule vor neue Herausforderungen und verlangt eine Neuausrichtung ihres Zusammenspiels mit der Familie. Die steigende Nachfrage nach Institutionen der Familien ergänzenden Kinderbetreuung befördert die Einrichtung von schulischen Tagesstrukturen und den Ausbau des Angebots an Tagesschulen. Dabei stehen jedoch oft sozial-, wirtschafts- und gleichstellungspolitische Anliegen im Vordergrund, während pädagogische Argumente, die das Wohl des Kindes und die kindliche Entwicklung im Auge haben, leicht vergessen gehen. Wie aber soll eine Tagesschule gestaltet sein, damit auch den Ansprüchen auf optimale Bildung und förderliche Betreuung der Kinder Rechnung getragen wird? Diese Frage steht im Zentrum des vorliegenden Bandes. Er versammelt namhafte Beiträge aus Theorie und Praxis, die der Frage nach der pädagogischen Qualität von Tagesschulen und ganztägigen Bildungseinrichtungen nachgehen.

Die Biografie eines Einzelnen ist immer zugleich eine Familiengeschichte. Die Familienverhältnisse werden als prägende Faktoren in die (auto-)biografischen Darstellungen einbezogen. Diesem Zusammenhang zwischen Familie und Biografie gehen hier Autorinnen und Autoren aus Literaturwissenschaft, Historischer Pädagogik und Geschichtswissenschaft nach.
