Recombinant tau protein is widely used to study the biochemical, cellular and pathological aspects of tauopathies, including Alzheimer's disease and frontotemporal dementia with Parkinsonism linked to chromosome 17 (FTPD-17). Pure tau in high yield is a requirement for invitro evaluation of the protein's physiological and toxic functions. However, the preparation of recombinant tau is complicated by the protein's propensity to aggregate and form truncation products, necessitating the use of multiple, time-consuming purification methods. In this study, we investigated parameters that influence the expression of wild type and FTPD-17 pathogenic tau, in an attempt to identify ways to maximise expression yield. Here, we report on the influence of the choice of host strain, induction temperature, duration of induction, and media supplementation with glucose on tau expression in Escherichia coli. We also describe a straightforward process to purify the expressed tau proteins using immobilised metal affinity chromatography, with favourable yields over previous reports. An advantage of the described method is that it enables high yield production of functional oligomeric and monomeric tau, both of which can be used to study the biochemical, physiological and toxic properties of the protein.

A doena de Wilson uma doena hereditria autossmica recessiva que afecta o gene ATP7B, localizado no cromossoma 13. Caracteriza-se por uma alterao do transporte transmembranar do cobre, acumulando-se este no fgado e noutros rgos, principalmente crebro, rins e crnea. A prevalncia mundial de aproximadamente 30 casos por milho de habitantes. Cerca de 60 a 70% dos casos so diagnosticados entre os 8 e os 20 anos de idade. Clinicamente, apresenta-se como doena heptica, neurolgica ou psiquitrica. O seu diagnstico baseado num alto ndice de suspeio e na combinao de achados clnicos e laboratoriais. O exame gold standard para o diagnstico a bipsia heptica com doseamento do cobre. O tratamento consiste na teraputica farmacolgica permanente ou no transplante heptico. Existem actualmente trs frmacos disponveis: a penicilamina e a trientina (agentes quelantes) e o zinco. O tetratiomolibdato um agente quelante ainda em investigao. A capacidade reprodutiva das mulheres com doena de Wilson tem melhorado com o aumento da eccia destas teraputicas e, como tal, a gravidez torna-se mais frequente. Esta parece no afectar o curso da doena e o principal problema que se coloca a escolha da teraputica mais adequada durante a gestao, dado que a segurana destes frmacos na gravidez no est garantida. Os autores descrevem 3 casos de grvidas com doena de Wilson, vigiadas na sua instituio, e discutem a controvrsia em torno do uso destes frmacos no tratamento da doena durante a gravidez e amamentao.

Frente a la poltica de apertura comercial que se viene desarrollando desde 1990 en Colombia, el manejo econmico ha tenido como propsito fundamental lograr integrar la economa nacional al exterior. Una de las herramientas de poltica econmica ms importantes es la TASA DE CAMBIO; frente a sta, es preciso adoptar una posicin y conocer el manejo que se le ha dado a esta variable durante los ltimos veinte aos. A partir de 1967, la poltica cambiara colombiana se bas en un sistema de devaluacin gota a gota (Crawling Peg). Esta poltica continu hasta 1991, cuando la Junta Monetaria cambi la devaluacin gota a gota, por el sistema de Certificado de Cambio con descuento. En enero de 1994, la nueva Junta Directiva del Banco de la Repblica elimin el sistema de certificado de cambio con descuento e introdujo el sistema de Bandas Cambiaras

Introduccin Los planificadores de la poltica exterior de Estados Unidos siempre le han presentado gran inters a la estratgica regin centroamericana. Los europeos haban invertido intensamente en la regin y Estados Unidos comprenda que, a menos que asumiera la responsabilidad por la estabilidad econmica y poltica de los estados centroamericanos, los gobiernos de Europa ignoraran la Doctrina Monroe cuando se tratase e proteger los intereses del exterior, Estados Unidos deber mantener el orden, per la abierta intervencin militar era costosa y era generadora de mala voluntad en todo el hemisferio. En consecuencia, el gobierno de Wilson busc otros medios para promover la estabilidad y la hegemona estadunidense. La Primera Guerra Mundial produjo una coyuntura de especiales oportunidades, as como de particulares riesgos.

This paper examines empirically the relative influence of the degree of endangerment of wildlife species and their stated likeability on individuals' allocation of funds for their conservation. To do this, it utilises data obtained from the IUCN Red List, and likeability and fund allocation data obtained from two serial surveys of a sample of the Australian public who were requested to assess 24 Australian wildlife species from three animal classes: mammals, birds and reptiles. Between the first and second survey, respondents were provided with extra information about the focal species. This information resulted in the dominance of endangerment as the major influence on the allocation of funding of respondents for the conservation of the focal wildlife species. Our results throw doubts on the proposition in the literature that the likeability of species is the dominant influence on willingness to pay for conservation of wildlife species. Furthermore, because the public's allocation of fund for conserving wildlife species seems to be more sensitive to information about the conservation status of species than to factors influencing their likeability, greater attention to providing accurate information about the former than the latter seems justified.
