Semi-aquatic animals represent a transitional locomotor condition characterised by the possession of morphological features that allow locomotion both in water and on land. Most ecologically important behaviours of crocodilians occur in the water, raising the question of whether their 'terrestrial construction' constrains aquatic locomotion. Moreover, the demands for aquatic locomotion change with life-history stage. It was the aim of this research to determine the kinematic characteristics and efficiency of aquatic locomotion in different-sized crocodiles (Crocodylus porosus). Aquatic propulsion was achieved primarily by tail undulations, and the use of limbs during swimming was observed only in very small animals or at low swimming velocities in larger animals. Over the range of swimming speeds we examined, tail beat amplitude did not change with increasing velocity, but amplitude increased significantly with body length. However, amplitude expressed relative to body length decreased with increasing body length. Tail beat frequency increased with swimming velocity but there were no differences in frequency between different-sized animals. Mechanical power generated during swimming and thrust increased non-linearly with swimming velocity, but disproportionally so that kinematic efficiency decreased with increasing swimming velocity. The importance of unsteady forces, expressed as the reduced frequency, increased with increasing swimming velocity. Amplitude is the main determinant of body-size-related increases in swimming velocity but, compared with aquatic mammals and fish, crocodiles are slow swimmers probably because of constraints imposed by muscle performance and unsteady forces opposing forward movement. Nonetheless, the kinematic efficiency of aquatic locomotion in crocodiles is comparable to that of fully aquatic mammals, and it is considerably greater than that of semi-aquatic mammals.

Objective: This study compares myoelectric manifestations of fatigue of the sternocleidomastoid (SCM) and anterior scalene (AS) muscles between 10 chronic neck pain subjects and 10 normal matched controls. Methods: Surface electromyography (sEMG) signals were recorded from the sternal bead of SCM and AS muscles bilaterally during submaximal isometric cervical flexion contractions at 25 and 50% of the maximum voluntary contraction (MVC). The mean frequency, average rectified value and conduction velocity of the sEMG signal were calculated to quantify myoelectric manifestations of muscle fatigue. Results: For both the SCM and AS muscles, the Mann-Whitney U test indicated that the initial value and slope of the mean frequency in neck pain patients were greater than in healthy subjects (P < 0.05). This was significant both at 25 and 50% of MVC. Conclusions: These results suggest: (a) a predominance of type-II fibres in the neck pain patients and/or (b) greater fatigability of the superficial cervical flexors in neck pain patients. These results are in agreement with previous muscle biopsy studies in subjects with neck pain, which identified transformation of slow-twitch type-I fibres to fast-twitch type-IIB fibres, as well as the clinical observation of reduced endurance in the cervical flexors in neck pain patients. No information is available on the decomposition and nutrient release pattern of Piper aduncum and Imperata cylindrica despite their importance in shifting cultivation systems of Papua New Guinea and other tropical regions. We conducted a litter bag study (24 weeks) on a Typic Eutropepts in the humid lowlands to assess the rate of decomposition of Piper aduncum, Imperata cylindrica and Gliricidia sepium leaves under sweet potato (Ipomoea batatas). Decomposition rates of piper leaf litter were fastest followed closely by gliricidia, and both lost 50% of the leaf biomass within 10 weeks. Imperata leaf litter decomposed much slower and half-life values exceeded the period of observation. The decomposition patterns were best explained by the lignin plus polyphenol over N ratio which was lowest for piper (4.3) and highest for imperata (24.7). Gliricidia leaf litter released 79 kg N ha(-1), whereas 18 kg N ha(-1) was immobilised in the imperata litter. The mineralization of P was similar for the three species, but piper litter released large amounts of K. The decomposition and nutrient release patterns had significant effects on the soil. The soil contained significantly more water in the previous imperata plots at 13 weeks due to the relative slow decomposition of the leaves. Soil N levels were significantly reduced in the previous imperata plots due to immobilisation of N. Levels of exchangeable K were significantly increased in the previous piper plots due to the large addition of K. It can be concluded that piper leaf litter is a significant and easily decomposable source of K which is an important nutrient for sweet potato. Gliricidia leaf litter contained much N, whereas imperata leaf litter releases relatively little nutrients and keeps the soil more moist. Gliricidia fallow is more attractive than an imperata fallow for it improves the soil fertility and produces fuelwood as additional saleable products.

The consumption of excess alcohol in patients with liver iron storage diseases, in particular the iron-overload disease hereditary haemochromatosis (HH), has important clinical consequences. HH, a common genetic disorder amongst people of European descent, results in a slow, progressive accumulation of excess hepatic iron. If left untreated, the condition may lead to fibrosis, cirrhosis and primary hepatocellular carcinoma. The consumption of excess alcohol remains an important cause of hepatic cirrhosis and alcohol consumption itself may lead to altered iron homeostasis. Both alcohol and iron independently have been shown to result in increased oxidative stress causing lipid peroxidation and tissue damage. Therefore, the added effects of both toxins may exacerbate the pathogenesis of disease and impose an increased risk of cirrhosis. This review discusses the concomitant effects of alcohol and iron on the pathogenesis of liver disease. We also discuss the implications of co-existent alcohol and iron in end-stage liver disease.

Disposition kinetics of [H-3] palmitate and its low-molecular-weight metabolites in perfused rat livers were studied using the multiple-indicator dilution technique, a selective assay for [H-3] palmitate and its low-molecular-weight metabolites, and several physiologically based pharmacokinetic models. The level of liver fatty acid binding protein (L-FABP), other intrahepatic binding proteins (microsomal protein, albumin, and glutathione S-transferase) and the outflow profiles of [H-3] palmitate and metabolites were measured in four experimentalgroups of rats: 1) males; 2) clofibrate-treated males; 3) females; and 4) pregnant females. A slow-diffusion/bound model was found to better describe the hepatic disposition of unchanged [H-3] palmitate than other pharmacokinetic models. The L-FABP levels followed the order: pregnant female > clofibrate-treated male > female > male. Levels of other intrahepatic proteins did not differ significantly. The hepatic extraction ratio and mean transit time for unchanged palmitate, as well as the production of low-molecular-weight metabolites of palmitate and their retention in the liver, increased with increasing L-FABP levels. Palmitate metabolic clearance, permeability-surface area product, retention of palmitate by the liver, and cytoplasmic diffusion constant for unchanged [H-3] palmitate also increased with increasing L-FABP levels. It is concluded that the variability in hepatic pharmacokinetics of unchanged [H-3] palmitate and its low-molecular-weight metabolites in perfused rat livers is related to levels of L-FABP and not those of other intrahepatic proteins.

delta-Atracotoxin-Ar1a (delta-ACTX-Ar1a) is the major polypeptide neurotoxin isolated from the venom of the male Sydney funnel-web spider, Atrax robustus. This neurotoxin targets both insect and mammalian voltage-gated sodium channels, where it competes with scorpion alpha-toxins for neurotoxin receptor site-3 to slow sodium-channel inactivation. Progress in characterizing the structure and mechanism of action of this toxin has been hampered by the limited supply of pure toxin from natural sources. In this paper, we describe the first successful chemical synthesis and oxidative refolding of the four-disulfide bond containing delta-ACTX-Ar1a. This synthesis involved solid-phase Boc chemistry using double coupling, followed by oxidative folding of purified peptide using a buffer of 2 M GdnHCl and glutathione/glutathiol in a 1:1 mixture of 2-propanol (pH 8.5). Successful oxidation and refolding was confirmed using both chemical and pharmacological characterization. Ion spray mass spectrometry was employed to confirm the molecular weight. H-1 NMR analysis showed identical chemical shifts for native and synthetic toxins, indicating that the synthetic toxin adopts the native fold. Pharmacological studies employing whole-cell patch clamp recordings from rat dorsal root ganglion neurons confirmed that synthetic delta-ACTX-Ar1a produced a slowing of the sodium current inactivation and hyperpolarizing shifts in the voltage-dependence of activation and inactivation similar to native toxin. Under current clamp conditions, we show for the first time that delta-ACTX-Ar1a produces spontaneous repetitive plateau potentials underlying the clinical symptoms seen during envenomation. This successful oxidative refolding of synthetic delta-ACTX-Ar1a paves the way for future structure-activity studies to determine the toxin pharmacophore.

The HERG K+ channel has very unusual kinetic behavior that includes slow activation but rapid inactivation. These features are critical for normal cardiac repolarization as well as in preventing lethal ventricular arrhythmias. Mutagenesis studies have shown that the extracellular peptide linker joining the fifth transmembrane domain to the pore helix is critical for rapid inactivation of the HERG K+ channel. This peptide linker is also considerably longer in HERG K+ channels, 40 amino acids, than in most other voltage-gated K+ channels. In this study we show that a synthetic 42-residue peptide corresponding to this linker region of the HERG K+ channel does not have defined structural elements in aqueous solution; however, it displays two well defined helical regions when in the presence of SDS micelles. The helices correspond to Trp(585)-Ile(593) and Gly(604)-Tyr(611) of the channel. The Trp(585)-Ile(593) helix has distinct hydrophilic and hydrophobic surfaces. The Gly(604)-Tyr(611) helix corresponds to an N-terminal extension of the pore helix. Electrophysiological studies of HERG currents following application of exogenous S5P peptides show that the amphipathic helix in the S5P linker interacts with the pore region of the channel in a voltage-dependent manner.

The origin of intracellular Ca2+ concentration ([Ca2+](i)) transients stimulated by nicotinic ( nAChR) and muscarinic ( mAChR) receptor activation was investigated in fura-2-loaded neonatal rat intracardiac neurons. ACh evoked [Ca2+](i) increases that were reduced to similar to 60% of control in the presence of either atropine ( 1 muM) or mecamylamine ( 3 muM) and to < 20% in the presence of both antagonists. Removal of external Ca2+ reduced ACh-induced responses to 58% of control, which was unchanged in the presence of mecamylamine but reduced to 5% of control by atropine. The nAChR-induced [Ca2+](i) response was reduced to 50% by 10 μM ryanodine, whereas the mAChR-induced response was unaffected by ryanodine, suggesting that Ca2+ release from ryanodine-sensitive Ca2+ stores may only contribute to the nAChR-induced [Ca2+](i) responses. Perforated-patch whole cell recording at - 60 mV shows that the rise in [Ca2+](i) is concomitant with slow outward currents on mAChR activation and with rapid inward currents after nAChR activation. In conclusion, different signaling pathways mediate the rise in [Ca2+](i) and membrane currents evoked by ACh binding to nicotinic and muscarinic receptors in rat intracardiac neurons.

A fines de los ???80, el Estado venezolano inici?? una descentralizaci??n pol??tico-territorial produciendo cambios en las gobernaciones, instancias receptoras de competencias en este proceso, las cuales realizan reformas a trav??s de la estrategia selectiva, incorporando algunas de la propuestas de la nueva gesti??n p??blica. Este trabajo explora estas reformas. Se observan: diferencias entre los servicios sociales y los relacionados directamente con la econom??a; privatizaci??n de ??stos; traspaso a la sociedad civil de algunos servicios sociales, creaci??n de entes descentralizados; automatizaci??n de procesos; flexibilizaci??n laboral; iniciativas con lento avance en rendici??n de cuentas por resultados y capacitaci??n para una nueva cultura administrativa. En conclusi??n: la tendencia es a la conformaci??n de una gesti??n p??blica eficiente sobre la base de la reducci??n del aparato estatal, de costos sociales, as?? como de costos y conflictos laborales y escasa incorporaci??n de las propuestas de participaci??n que permiten profundizar la democracia y promover el bienestar social.

A idade e o crescimento da cioba da costa central do Brasil foram estudados a partir de leituras de otólitos obtidos em amostragens mensais de desembarques comerciais, realizados nas cidades de Vitória e Vila Velha, entre os anos de 1998 e 1999. Através da análise do incremento marginal, foi verificada a formação anual de uma zona translúcida a partir do final do outono e durante o inverno. Foram observadas ciobas de 2 a 19 anos. Os comprimentos furcais (CF) médios retrocalculados variaram de 108 mm para idade 1 a 524 mm para idade 19. A equação de Von Bertalanffy ajustada aos CF médios retrocalculados foi CFt = 567,1 (1 - e -0.130 (t + 0.773)). A relação comprimento peso foi P = 2,68x10-5CF2.914, onde P = peso total em gramas. O crescimento é lento, com os incrementos anuais em peso aumentando gradativamente até atingir o máximo de 164 g entre as idades 7 e 8 anos. Os resultados deste trabalho indicam que a cioba tem longa expectativa de vida e baixas taxas de crescimento somático, características das espécies mais sensíveis à exploração pesqueira.

Animais híbridos representam um desafio à taxonomia e sistemática, pois correspondem a unidades evolutivas geralmente sem clara delimitação morfológica, comportamental e genética. Híbridos podem ser morfologicamente intermediários aos parentais ou, devido à introgressão e retrocruzamentos, suas características podem se misturar tornando difícil sua identificação. Uma das formas de identificação de híbridos é por meio de ferramentas de biologia molecular, que ao utilizarem marcadores de DNA mitocondrial (herança exclusiva materna) e DNA nuclear (herança materna e paterna), permitem a comparação entre informações genéticas. Além da hibridização existem outras fontes de conflito entre dados moleculares provenientes do DNA mitocondrial e DNA nuclear, como por exemplo a retenção de polimorfismos ancentrais. Em localidades do Espírito Santo, Brasil, foram coletados indivíduos de morfologia distinta de Trachycephalus mesophaeus e T. nigromaculatus, que são as únicas espécies do gênero conhecidas nesse estado. Porém, estudos piloto usando o gene mitocondrial Citocromo Oxidase subunidade I (COI) agruparam esses espécimes com amostras de T. typhonius. Devido a estas incongruências, foram sequenciados fragmentos de dois genes mitocondriais - COI e Nicotinamida Desidrogenase subunidade 2 (ND2) e um exon nuclear (tirosinase) de 173 indivíduos de Trachycephalus, de forma a esclarecer as identificações taxonômicas e investigar a correspondência entre caracteres morfológicos e genéticos nesta linhagem, na sua área de ocorrência As filogenias moleculares, divergências genéticas, redes de haplótipos e polimorfismos de nucleotídeos únicos (SNPs) confirmaram as três espécies acima mencionadas como linhagens evolutivas distintas e revelaram mais sete indivíduos potencialmente híbridos, mas morfologicamente assinalados a T. mesophaeus, T. nigromaculatus ou T. typhonius.. Devido à taxa de evolução lenta da tirosinase, as espécies mais recentes T. typhonius e T. nigromaculatus parecem não terem sido sorteadas completamente nesse gene. Já T. mesophaeus, que é a espécie mais antiga das três, foi recuperada inequivocamente em todas as análises. De forma inédita, as análises moleculares evidenciaram a ocorrência de introgressão bidirecional entre T. nigromaculatus e T. typhonius e entre T. nigromaculatus e T. mesophaeus, sendo que há indícios de indivíduos F1 (cruzamentos entre espécies parentais puras gerando híbridos). A utilização do gene ND2 mostrou-se mais eficiente do que o gene COI nas filogenias e, apesar da tirosinase ser um gene nuclear de evolução lenta, contribuiu para a identificação de incongruências citonucleares. Nossos resultados mostram que a história filogenética de Trachycephalus é complexa e que o uso de marcadores nucleares de evolução mais rápida e ampliação dessas análises para outras espécies do gênero podem revelar mais eventos de hibridização.

Nosso país é um dos principais produtores agrícolas do mundo e altamente dependente da importação de fertilizantes, principalmente o potássio. Visando reduzir essa dependência, materiais provenientes de rochas para fornecimento desse elemento estão sendo testados. Este estudo foi divido em dois experimentos: um experimento em laboratório e um em casa de vegetação. O experimento em laboratório teve por objetivo avaliar os diferentes tipos de tratamentos térmicos e extratores, associados a fontes alternativas de potássio, no aumento da disponibilidade de K. O ensaio foi realizado num delineamento experimental inteiramente casualizado num esquema fatorial 4 x 5 x 3, com quatro repetições. Os fatores estudados foram: quatro fontes alternativas de potássio (verdete, fonolito, gnaisse e granito); cinco tratamentos térmicos (radiação em forno micro-ondas; autoclavagem; aquecimento em forno mufla com resfriamento rápido; aquecimento em forno mufla com resfriamento lento; e o material in natura); e três extratores: água, ácido cítrico 2 % e solução Mehlich-1. As extrações foram realizadas utilizando 4 g das fontes alternativas, adicionando-se 40 mL de cada extrator, as amostras foram agitadas, centrifugadas, filtradas e avolumadas. Foram realizadas seis extrações sucessivas e determinada a concentração de potássio nos extratos. Os dados obtidos foram submetidos à análise de variância, sendo seus efeitos desdobrados em contrastes ortogonais. O experimento em casa de vegetação teve por objetivo avaliar o efeito da utilização de fontes alternativas de potássio, submetidas a tratamentos térmicos, no fornecimento de potássio para plantas de milho (Zea mays L.). O delineamento experimental adotado foi em blocos casualizados, em um esquema fatorial (3 x 5) + 1, sendo três fontes alternativas de K (verdete, fonolito e granito), submetidas à cinco diferentes tratamentos (aquecimento em forno mufla com resfriamento lento, aquecimento em forno mufla com resfriamento rápido, radiação em forno micro-ondas, autoclavagem, e o material in natura). As fontes alternativas foram comparadas com o cloreto KCl. Amostras de solo contendo 2 dm³ de TFSA, foram incubadas durante 20 dias. Após a adubação, em cada vaso foram aplicadas as fontes alternativas de potássio. Realizou-se o plantio com cinco sementes de milho e posterior desbaste, deixando três plantas por vaso. Decorridos 40 dias da semeadura, foi realizado o corte da parte aérea das plantas, a coleta de amostras de solo para análise dos teores de potássio e separação das raízes. Os dados obtidos foram submetidos à análise de variância e desdobrados em contrastes ortogonais. Observando os resultados obtidos nos experimentos de laboratório e casa de vegetação, podemos concluir que a fonte alternativa e proporcionou melhores resultados e que possui potencial agronômico para ser utilizada como fonte alternativa de potássio foi o fonolito. Utilizando o fonolito como fonte alternativa de nutrientes, o tratamento térmico que proporcionou melhores resultados foi o com aquecimento em forno mufla, principalmente quando o resfriamento foi realizado lentamente.

A N-acetiltransferase 2 é a principal enzima responsável pelo metabolismo e inativação da isoniazida no organismo humano. Mutações no gene NAT2 levam a 3 perfis genotípicos de acetilação que alteram os níveis séricos do fármaco: acetiladores lentos, intermediários e rápidos, o que pode alterar o desfecho terapêutico. O objetivo do estudo foi investigar se os diferentes perfis podem influenciar no tempo de negativação da cultura de escarro, e se existe correlação entre carga bacilar e gravidade da doença com tempo de conversão da cultura. A população de estudo foi composta por 62 pacientes, que tiveram seus DNAs sequenciados para identificação de mutações no gene NAT2 e seus perfis de acetilação determinados. A análise genotípica detectou 10 SNPs, sendo as mutações 341 T>C (39,65%) e 481 C>T (38,71%) as mais frequentes. A determinação das variantes alélicas identificou NAT2*5B (29,03%), NAT2*6A (23,39%) e NAT2*4 (24,19%) como os alelos mais frequentes e NAT2*5B/*5B como o genótipo mais frequente (20,4%). Dentre os 62 pacientes, foi possível correlacionar tempo de negativação da cultura e perfil de acetilação entre 43 deles, os quais 58,3% e 55,6% tiveram o genótipo lento com maior frequência no mês 1 e mês 3, respectivamente. Por meio de dados microbiológicos, a carga bacilar e a gravidade da doença também foram comparadas com o tempo de negativação, indicando que os pacientes com doença moderada ou avançada (76,7%) e aqueles com carga bacilar alta (60,4%), não tiveram associação estatística com o tempo de conversão da cultura. Por último, curvas de crescimento de isolados de M. tuberculosis de pacientes foram construídas para verificar possíveis diferenças na duração da fase lag entre os isolados, porém não foi observada diferença estatística entre elas. Com base nos resultados encontrados, verifica-se que não existe associação entre o perfil de acetilação do paciente, a carga bacilar, a gravidade da doença e o tempo de negativação da cultura de escarro

Laparoscopic surgery (LS) has revolutionized traditional surgical techniques introducing minimally invasive procedures for diagnosis and local therapies. LSs have undeniable advantages, such as small patient incisions, reduced postoperative pain and faster recovery. On the other hand, restricted vision of the anatomical target, difficult handling of the surgical instruments, restricted mobility inside the human body, need of dexterity to hand-eye coordination and inadequate and non-ergonomic surgical instruments may restrict LS only to more specialized surgeons. To overcome the referred limitations, this work presents a new robotic surgical handheld system – the EndoRobot. The EndoRobot was designed to be used in clinical practice or even as a surgical simulator. It integrates an electromechanical system with 3 degrees of freedom. Each degree can be manipulated independently and combined with different levels of sensitivity allowing fast and slow movements. As other features, the EndoRobot has battery power or external power supply, enables the use of bipolar radiofrequency to prevent bleeding while cutting and allows plug-and-play of the laparoscopic forceps for rapid exchange. As a surgical simulator, the system was also instrumented to measure and transmit, in real time, its position and orientation for a training software able to monitor and assist the trainee’s surgical movements.

ABSTRACT Sorghum arundinaceum (Desv.) Stapf is a weed that belongs to the Poaceae family and is widespread throughout Brazil. Despite the frequent occurrence, infesting cultivated areas, there is little research concerning the biology and physiology of this species. The objective of this research was to evaluate the growth, carbon partitioning and physiological characteristics of the weed Sorghum arundinaceum in greenhouse. Plants were collected at regular intervals of seven days, from 22 to 113 days after transplanting (DAT). In each sample, we determined plant height, root volume, leaf area and dry matter, and subsequently we perfomed the growth analysis, we have determined the dry matter partitioning among organs, the accumulation of dry matter, the specific leaf area, the relative growth rate and leaf weight ratio. At 36, 78 and 113 DAT, the photosynthetic and transpiration rates, stomatal conductance, CO2 concentration and chlorophyll fluorescence were evaluated. The Sorghum arundinaceum reached 1.91 in height, with slow initial growth and allocated much of the biomass in the roots. The photosynthetic rate and the maximum quantum yield of FSII are similar throughout the growth cycle. At maturity the Sorghum arundinaceum presents higher values of transpiration rate, stomatal conductance and non-photochemical quenching coefficient (NPQ).
