PV Off-Grid systems have demonstrated to be a good solution for the electrification of remote areas [1]. A hybrid system is one kind of these systems. The principal characteristic is that it uses PV as the main generator and has a backup power supply, like a diesel generator, for instance, that is used when the CPV generation is not enough to meet demand. To study the use of CPV in these systems, ISFOC has installed a demonstration hybrid system at its headquarters. This hybrid system uses CPV technology as main generator and the utility grid as the backup generator. A group of batteries have been mounted as well to store the remaining energy from the CPV generator when nedeed. The energy flows are managed by a SMA system based on Sunny Island inverters and a Multicluster-Box (figure 1). The Load is the air-conditioning system of the building, as it has a consumption profile higher than the CPV generator and can be controlled by software [2]. The first results of this system, as well as the first chances of improvement, as the need of a bigger CPV generator and a better management of the energy stored in the batteries, are presented in this paper.

En este Proyecto Fin de Carrera trata de dar a conocer de forma detallada el comportamiento real en frecuencia de una red de distribución SMATV y establecer un procedimiento que pueda permitir una predicción más o menos precisa de su funcionamiento. Se realizará una reflexión sobre los procedimientos de cálculo que se emplean en entornos profesionales para el diseño de estas redes y sus carencias. En concreto nos centraremos en el uso de conectores y kits de conexión que se utilizan frecuentemente en redes SMATV. A la hora de realizar un proyecto de una red de este tipo solo se tienen en cuenta las perdidas por inserción de los conectores, y en muchas ocasiones ni siquiera eso, se realiza una estimación al alza y con eso basta. Este proyecto pretende demostrar la relevancia de los conectores dentro de una red, y tratar de arrojar luz sobre los aspectos más importantes a tener en cuenta a la hora de elegir e instalar un conector. Abstract In this Thesis is to present in detail the actual behavior frequency SMATV distribution network and establish a procedure to allow more or less accurate prediction of its operation. There will be a reflection on the calculation procedures used in professional environments for the design of these networks and their shortcomings. In particular we will focus on the use of connectors and connection kits that are frequently used in SMATV networks. When a project of this type network only takes into account the insertion loss of the connectors, and often not even that, an estimate is made up and that's enough. This project aims to demonstrate the relevance of the connectors within a network, and try to shed light on the most important aspects to consider when choosing and installing a connector.

ML 1.4 is widely accepted as the standard for representing the various software artifacts generated by a development process. For this reason, there have been attempts to use this language to represent the software architec- ture of systems as well. Unfortunately, these attempts have ended in representa- tions (boxes and lines) already criticized by the software architecture commu- nity. Recently, OMG has published a draft that will constitute the future UML 2.0 specification. In this paper we compare the capacities of UML 1.4 and UML 2.0 to describe software architectures. In particular, we study extensions of both UML versions to describe the static view of the C3 architectural style (a simplification of the C2 style). One of the results of this study is the difficulties found when using the UML 2.0 metamodel to describe the concept of connector in a software architecture.

Connectors are essential in many circuits to communicate with exterior world. When working in millimeter wave frequencies, connectors require very tight tolerances and being mounted exactly in the correct way. Main problems which use to appear when working with millimeter wave connectors have been collected in this article. Besides that, some basic guidelines to get optimal connector performance are given. To illustrate how these advices can improve connector operation, they have been used in a real case.

PAMELA (Phased Array Monitoring for Enhanced Life Assessment) SHMTM System is an integrated embedded ultrasonic guided waves based system consisting of several electronic devices and one system manager controller. The data collected by all PAMELA devices in the system must be transmitted to the controller, who will be responsible for carrying out the advanced signal processing to obtain SHM maps. PAMELA devices consist of hardware based on a Virtex 5 FPGA with a PowerPC 440 running an embedded Linux distribution. Therefore, PAMELA devices, in addition to the capability of performing tests and transmitting the collected data to the controller, have the capability of perform local data processing or pre-processing (reduction, normalization, pattern recognition, feature extraction, etc.). Local data processing decreases the data traffic over the network and allows CPU load of the external computer to be reduced. Even it is possible that PAMELA devices are running autonomously performing scheduled tests, and only communicates with the controller in case of detection of structural damages or when programmed. Each PAMELA device integrates a software management application (SMA) that allows to the developer downloading his own algorithm code and adding the new data processing algorithm to the device. The development of the SMA is done in a virtual machine with an Ubuntu Linux distribution including all necessary software tools to perform the entire cycle of development. Eclipse IDE (Integrated Development Environment) is used to develop the SMA project and to write the code of each data processing algorithm. This paper presents the developed software architecture and describes the necessary steps to add new data processing algorithms to SMA in order to increase the processing capabilities of PAMELA devices.An example of basic damage index estimation using delay and sum algorithm is provided.

La determinación de la línea histórica de deforestación como parte del establecimiento de la línea de referencia de emisiones, en el marco del programa REDD (Reducing Emissions from Deforestation and Forest Degradation), permite medir la evolución de la pérdida de bosque en un periodo definido de tiempo. El objetivo fue calcular la línea histórica de deforestación mediante estudio multitemporal para el periodo 1998-2011, en la región de San Martín (Perú), utilizando la metodología de Análisis de Mezclas Espectrales (Spectral Mixtures Analysis) con imágenes Landsat 5-TM. Las transiciones en guía de onda son las estructuras utilizadas en microondas para transferir y adaptar la señal que viaja en un determinado sistema de transmisión (por ejemplo, un cable coaxial) a otro sistema de transmisión o a un sistema radiante (por ejemplo, una antena de bocina). Los dos sistemas de transmisión entre los que la transición adapta la señal pueden ser distintos o del mismo tipo pero con alguna de sus dimensiones diferente. Existen diferentes transiciones de guía de onda que dependiendo de su utilidad son diseñadas y construidas con diferentes secciones: circular, rectangular, elíptica o incluso combinaciones de éstas. No es necesario que la sección de la guía presente una forma geométrica conocida pero los estándares que se van a seguir hacen referencia en concreto a secciones rectangulares y circulares. En el trabajo que aquí se desarrolla se pretende optimizar por medio de simulaciones paramétricas una transición entre cable coaxial con conector tipo K y una guía de onda de sección circular que sigue el estándar presentado por Flann, Millitech y Antarfs para la banda WR34. La transición que va a ser objeto de este estudio se denomina transición tapered o transición conformada. Este tipo de transiciones se caracterizan por variar una de sus dimensiones progresivamente hasta llegar al tamaño definido en el estándar correspondiente. La manera de realizar la optimización de la transición se basará en el estudio del parámetro S11 que presente la estructura a lo largo de la banda de trabajo. Ya que se sigue el estándar WR34 la banda de trabajo que éste comprende va de 21,7 a 33 GHz. Se pretende conseguir que la respuesta del parámetro S11 se encuentre por debajo de -20 dB en la banda de WR34 como resultado del diseño para poder contar de esta manera con una buena adaptación. Finalmente se propondrá un criterio a seguir para optimizar este tipo de transiciones siguiendo el objetivo de mejor adaptación teniendo en cuenta el impacto de cada tramo sobre el rango de frecuencias en el que influye y se presentarán las características finales que presenta la transición bajo estudio. En este documento se introduce de manera breve la utilidad de los transformadores de impedancias lambda cuartos en líneas de transmisión, el estado del arte de las diferentes técnicas para su diseño, y la propuesta de diseño y caracterización objeto del presente trabajo. Posteriormente, se presenta el caso de estudio para el diseño de la transición para ser integrada a una bocina de choke. Luego, se introduce el marco teórico del trabajo presentando algunos ejemplos ilustrativos de tramos de guía de onda rectangular y guía de onda circular, introduciendo adaptadores de λ/4 en simulaciones. A continuación, se explica la implementación del modelo bajo estudio en CST (Computer Simulation Technology) Studio Suite. Finalmente se presenta la discusión de los resultados obtenidos, las conclusiones y líneas futuras de trabajo. ABSTRACT. Waveguide transitions are structures used in microwave in order to transfer and adapt the signal that travels from a certain transmission system (e.g. coaxial cable) to other transmission system or to a radiant system (e.g. horn antenna). Both transmission systems between which the transition adapts the signal can be different or from the same type, but with differences in some of their dimensions. There are different waveguide transitions that, depending on their utility, are designed and constructed in different sections: circular, rectangular, elliptic or combinations of the former. The section of the guide does not have to have a known geometric shape, although the standards to be followed in this thesis apply to rectangular and circular sections. In the work here presented, we aim to optimize by means of parametric simulations, a transition between a coaxial cable with a K-type connector and a waveguide with circular section following the standard presented by Flann, Millitech y Antarfs for the band WR34. The transition studied is called tapered transition, which is characterized by the progressive variation of its dimensions, until reaching the defined size of the corresponding standard. The way of optimizing the transition will be based in the study of the parameter S11 presented by the structure along the bandwidth. Since the standard used is the WR34, the bandwidth can be defined from 21.7 up to 33 GHz. It is aimed that the response of the S11 parameter be lower equal than -20dB in the frequency band under study according to the design in order to be well matched. Finally, a criterion to follow is proposed in order to optimize these transitions type, following the better-match objective. That will be done taking into account the impact of each section on the frequency range in which influences and the final characteristics of the studied transition will be presented. In this document, it is briefly introduced the utility of quarter-wave impedance transformers in transmission lines, the state-of-art of the different techniques for their design, and the proposal of design and characterization aimed with this work. Afterwards, the study case for the design of the transition to be integrated with a choke horn antenna will be presented. Later, the theoretical frame work is introduced, giving some illustrative examples of rectangular and circular waveguide sections, and also introducing λ/4 adaptors in the simulations. Subsequently, the implementation of the model under study in CST (Computer Simulation Technology) Studio Suite will be explained. Finally, the discussion of the results, conclusion and future lines of the work are presented.

La aparición y avance de la enfermedad del marchitamiento del pino (Pine Wilt Desease, PWD), causada por Bursaphelenchus xylophilus (Nematoda; Aphelenchoididae), el nematodo de la madera del pino (NMP), en el suroeste de Europa, ha puesto de manifiesto la necesidad de estudiar la fenología y la dispersión de su único vector conocido en Europa, Monochamus galloprovincialis (Col., Cerambycidae). El análisis de 12 series de emergencias entre 2010 y 2014, registradas en Palencia, València y Teruel, con material procedente de diversos puntos de la península ibérica, demostró una alta variabilidad en la fenología de M. galloprovincialis y la divergencia térmica respecto de las poblaciones portuguesas. Para éstas, el establecimiento de los umbrales térmicos de desarrollo de las larvas post-dormantes del vector (12,2 y 33,5ºC) permitió la predicción de la emergencia mediana para la fecha en la que se acumulaban de 822 grados-día. Ninguna de las series analizadas en este trabajo necesitó de dichos grados-día estimados para la emergencia mediana. Asimismo, la emergencia se adelantó en las regiones más calurosas, mientras que se retrasó en las zonas más templadas. Más allá de la posible variabilidad entre poblaciones locales peninsulares, se detectaron indicios de que la diferencia en la acumulación de calor durante el otoño puede afectar el grado de maduración de las larvas invernantes, y su posterior patrón temporal de emergencia. Por último, también fueron observados comportamientos de protandria en las emergencias. Respecto a la fenología de su vuelo, entre los años 2010 y 2015, fueron ejecutados un total de 8 experimentos de captura de M. galloprovincialis mediante trampas cebadas con atrayentes en diferentes regiones (Castellón, Teruel, Segovia y Alicante) permitiendo el seguimiento del periodo de vuelo. Su análisis permitió constatar la disminución de las capturas y el acortamiento del periodo de vuelo con la altitud, el inicio del vuelo en el mes de mayo/junio a partir de los 14ºC de temperatura media diaria, la influencia de las altas temperaturas en la disminución de las capturas estivales (potencial causante de perfiles bimodales en las curvas de vuelo en las zonas menos frías), la evolución de la proporción de sexos a lo largo del periodo de vuelo (que muestra una mayor captura de hembras al inicio y de machos al final) y el comportamiento diurno y ligado a las altas temperaturas del vuelo circadiano del insecto. Dos redes de muestreo sistemático de insectos saproxílicos instaladas en la Comunitat Valencia (Red MUFFET, 15 parcelas, año 2013) y en Murcia (Red ESFP, 20 parcelas, años 2008-2010) permitieron el estudio de la comunidad de insectos relacionada con M. galloprovincialis. Cada una de las parcelas contaba con una trampa cebada con atrayentes y una estación meteorológica. El registro de más de 250 especies de coleópteros saproxílicos demostró el potencial que tiene el empleo de redes de trampas vigía para la detección temprana de organismos exóticos, además de permitir la caracterización y evaluación de las comunidades de entomofauna útil, representando una de las mejores herramientas de la gestión integrada de plagas. En este caso, la comunidad de saproxílicos estudiada mostró ser muy homogénea respecto a la variación ambiental de las zonas de muestreo, y que pese a las pequeñas variaciones entre las comunidades de los diferentes ecosistemas, el rol que M. galloprovincialis desempeña en ellas a lo largo de todo el gradiente estudiado es el mismo. Con todo, el análisis mediante redes de interacción mostró su relevancia ecológica al actuar de conector entre los diferentes niveles tróficos. Por último, un total de 12 experimentos de marcaje-liberación-recaptura desarrollados entre 2009 y 2012 en Castellón, Teruel, Valencia y Murcia permitieron evaluar el comportamiento dispersivo de M. galloprovincialis. Las detecciones mediante trampas cebadas de los insectos liberados se dieron por lo menos 8 días después de la emergencia. La abundancia de población pareció relacionada con la continuidad, la naturalización de la masa, y con la afección previa de incendios. La dispersión no estuvo influida por la dirección ni la intensidad de los vientos dominantes. La abundancia de material hospedante (en lo referente a las variables de masa y a los índices de competencia) influyó en la captura del insecto en paisajes fragmentados, aunque la ubicación de las trampas optimizó el número de capturas cuando se ubicaron en el límite de la masa y en zonas visibles. Por último también se constató que M. galloprovincialis posee suficiente capacidad de dispersión como para recorrer hasta 1500 m/día, llegando a alcanzar distancias máximas de 13600m o de 22100 m. ABSTRACT The detection and expansion of the Pine Wilt Desease (PWD), caused by Bursaphelenchus xylophilus (Nematoda; Aphelenchoididae), Pine Wood Nematode (PWN), in southwestern Europe since 1999, has triggered off the study of the phenology and the dispersion of its unique vector in the continent, Monochamus galloprovincialis (Coleoptera, Cerambycidae). The analysis of 12 emergence series between 2010 and 2014 registered in Palencia, Teruel and Valencia (Spain), registered from field colonized material collected at several locations of the Iberian Peninsula, showed a high variability in the emergence phenology of M. galloprovincialis. In addition, these patterns showed a very acute thermal divergence regarding a development model fitted earlier in Portugal. Such model forecasted the emergence of 50% of M. galloprovincialis individuals in the Setúbal Peninsula (Portugal) when an average of 822 degree-days (DD) were reached, based on the accumulation of heat from the 1st of March until emergence and lower and upper thresholds of 12.2 ºC and 33,5 °C respectively. In our results, all analyzed series needed less than 822 DD to complete the 50% of the emergence. Also, emergency occurred earlier in the hottest regions, while it was delayed in more temperate areas. Beyond the possible variability between local populations, the difference in the heat accumulation during the fall season may have affected the degree of maturation of overwintering larvae, and subsequently, the temporal pattern of M. galloprovincialis emergences. Therefore these results suggest the need to differentiate local management strategies for the PWN vector, depending on the location, and the climatic variables of each region. Finally, protandrous emergence patterns were observed for M. galloprovincialis in most of the studied data-sets. Regarding the flight phenology of M. galloprovincialis, a total of 8 trapping experiments were carried out in different regions of the Iberian Peninsula (Castellón, Teruel, Segovia and Alicante) between 2010 and 2015. The use of commercial lures and traps allowed monitoring of the flight period of M. galloprovincialis. The analyses of such curves, helped confirming different aspects. First, a decline in the number of catches and a shortening of the flight period was observed as the altitude increased. Flight period was recorded to start in May / June when the daily average temperature went over 14 ° C. A significant influence of high temperatures on the decrease of catches in the summer was found in many occasions, which frequently lead to a bimodal profile of the flight curves in warm areas. The evolution of sex ratio along the flight period shows a greater capture of females at the beginning of the period, and of males at the end. In addition, the circadian response of M. galloprovincialis to lured traps was described for the first time, concluding that the insect is diurnal and that such response is linked to high temperatures. Two networks of systematic sampling of saproxylic insects were installed in the Region of Valencia (Red MUFFET, 15 plots, 2013) and Murcia (Red ICPF, 20 plots, 2008-2010). These networks, intended to serve the double purpose of early-detection and long term monitoring of the saproxylic beetle assemblies, allowed the study of insect communities related to M. galloprovincialis. Each of the plots had a trap baited with attractants and a weather station. The registration of almost 300 species of saproxylic beetles demonstrated the potential use of such trapping networks for the early detection of exotic organisms, while at the same time allows the characterization and evaluation of useful entomological fauna communities, representing one of the best tools for the integrated pest management. In this particular case, the studied community of saproxylic beetles was very homogeneous with respect to environmental variation of the sampling areas, and despite small variations between communities of different ecosystems, the role that M. galloprovincialis apparently plays in them across the studied gradient seems to be the same. However, the analysis through food-webs showed the ecological significance of M. galloprovincialis as a connector between different trophic levels. Finally, 12 mark-release-recapture experiments were carried out between 2009 and 2012 in Castellón, Teruel, Valencia and Murcia (Spain) with the aim to describe the dispersive behavior of M. galloprovincialis as well as the stand and landscape characteristics that could influence its abundance and dispersal. No insects younger than 8 days were caught in lured traps. Population abundance estimates from mark-release-recapture data, seemed related to forest continuity, naturalization, and to prior presence of forest fires. On the other hand, M. galloprovincialis dispersal was not found to be significantly influenced by the direction and intensity of prevailing winds. The abundance of host material, very related to stand characteristics and spacing indexes, influenced the insect abundance in fragmented landscapes. In addition, the location of the traps optimized the number of catches when they were placed in the edge of the forest stands and in visible positions. Finally it was also found that M. galloprovincialis is able to fly up to 1500 m / day, reaching maximum distances of up to 13600 m or 22100 m.

En el estado de Veracruz, al sur de México, se ubican empresas dedicadas a la obtención de etanol a partir de melaza de azúcar de caña. Las más pequeñas, tienen una producción promedio de 20,000 L de alcohol/día. Los efluentes de la producción de etanol incluyen agua de enfriamiento de condensadores, agua del lavado de tanques de fermentación y vinazas, estas últimas son los efluentes más contaminantes en las destilerías, por su concentración de material orgánico biodegradable y no biodegradable. Las vinazas se generan en grandes volúmenes, produciéndose de 12 a 15 litros de vinazas por cada litro de alcohol destilado. Estos efluentes se caracterizan por tener altas temperaturas, pH ácido y una elevada concentración de DQO así como de sólidos totales. La determinación de la biodegradabilidad anaerobia de un agua residual, permite estimar la fracción de DQO que puede ser transformada potencialmente en metano y la DQO recalcitrante que queda en el efluente. Para el desarrollo de una prueba de biodegradabilidad, es importante considerar diversos factores relacionados con la composición del agua a tratar, composición de los lodos y las condiciones bajo las cuales se lleva a cabo la prueba. La digestión anaerobia de aguas residuales industriales es comúnmente usada en todo el mundo, ofrece significativas ventajas para el tratamiento de efluentes altamente cargados. Los sistemas anaerobios de tratamiento de aguas residuales industriales incluyen tecnologías con biopelículas, estos sistemas de tratamiento anaerobio con biopelícula son una tecnología bien establecida para el tratamiento de efluentes industriales. El Reactor de Lecho Fluidizado Inverso Anaerobio (LFI) ha sido diseñado para el tratamiento de aguas residuales de alta carga, teniendo como ventajas el empleo de un soporte que proporciona una gran superficie y un bajo requerimiento de energía para la fluidización del lecho. En el presente trabajo, se lleva a cabo el análisis de un proceso de producción de etanol, identificando a los efluentes que se generan en el mismo. Se encuentra que el efluente final está compuesto principalmente por las vinazas provenientes del proceso de destilación. En la caracterización de las vinazas provenientes del proceso de producción de etanol a partir de melaza de azúcar de caña, se encontraron valores promedio de DQO de 193.35 gDQO/L, para los sólidos totales 109.78 gST/L y pH de 4.64. Así mismo, en esta investigación se llevó a cabo una prueba de biodegradabilidad anaerobia, aplicada a la vinaza proveniente de la producción de etanol. En la caracterización de los lodos empleados en el ensayo se obtiene una Actividad Metanogénica Especifica de 0.14 g DQO/gSSV.d. El porcentaje de remoción de DQO de la vinaza fue de 62.7%, obteniéndose una k igual a 0.031 h-1 y una taza de consumo de sustrato de 1.26 gDQO/d. El rendimiento de metano fue de 0.19 LCH4/g DQOremovida y el porcentaje de biodegradabilidad de 54.1%. El presente trabajo también evalúa el desempeño de un LFI, empleando Extendospher® como soporte y tratando efluentes provenientes de la producción de etanol. El reactor se arrancó en batch y posteriormente se operó en continuo a diferentes Cargas Orgánicas Volumétricas de 0.5, 1.0, 3.3, 6.8 y 10.4 g DQO/L.d. Además, se evaluaron diferentes Tiempos de Residencia Hidráulica de 10, 5 y 1 días. El sistema alcanzó las siguientes eficiencias promedio de remoción de DQO: 81% para la operación en batch; 58, 67, 59 y 50 % para las cargas de 0.5, 1.0, 3.3, 6.8 g DQO/L.d respectivamente. Para la carga de 10.4 g DQO/L.d, la eficiencia promedio de remoción de DQO fue 38%, en esta condición el reactor presentó inestabilidad y disminución del rendimiento de metano. La generación de metano inició hasta los 110 días de operación del reactor a una carga de 1.0 g DQO/L.d. El sistema alcanzó un rendimiento de metano desde 0.15 hasta 0.34 LCH4/g DQO. Durante la operación del reactor a una carga constante de 6.4 g DQO/L.d, y un TRH de 1 día, se alcanzó una eficiencia promedio de remoción de DQO de 52%. In the state of Veracruz, to the south of Mexico, there are located companies dedicated to the production of ethanol from molasses of cane sugar. The smallest, have a average production of 20,000 L ethanol/day. The effluent of production of ethanol include water of condensers, water originated from the cleanliness of tanks of fermentation and vinasses, the above mentioned are more effluent pollutants in the distilleries, for the poor organic matter degradability. The vinasses are generated in high volumes, producing from 12 to 15 L of vinasses per every liter of distilled ethanol. These effluent are characterized by its high temperature, pH acid and a high concentration of DQO as well as high concentration of TS. The determination of the anaerobic degradability of a waste water, it allows to estimate the fraction of DQO that can be transformed potentially into methane and the recalcitrant DQO that stays in the effluent. For the development of degradability test, it is important to consider factors related to the composition of the water to be treated, composition of the sludge and the conditions under which the test is carried out. The anaerobic digestion of industrial wastes water is used commonly in the whole world, it offers significant advantages for the treatment of effluent highly loaded. The anaerobic treatment of industrial wastes water include technologies with biofilms, this anaerobic treatment whit biofilms systems, is a well-established technology for treatment of industrial effluents. The Anaerobic Inverse Fluidized Bed Reactor (IFBR) has been developed to provide biological treatment of high strength organic wastewater for their large specific surface and their low energy requirements for fluidization. In this work, there is carried out the analysis of a process of production of ethanol, identifying the effluent ones that are generated in the process. One determined that the effluent end is composed principally by the vinasses originated from the process of distillation. In the characterization of the vinasses originated from the process of production of ethanol from cane sugar molasses, there were average values of DQO of 193.35 gDQO/L, average values of solid of 109.78 gST/L and pH of 4.64. In this investigation there was carried out a anaerobic degradability test of the vinasses generated in the production of ethanol. In the characterization of the sludge used in the essay, the specific methanogenic activity (SMA) was 0.14 gDQO/gSSV.d. The average removal of DQO of the vinasses was 62.7 %, k equal to 0.031 h-1 was obtained one and a rate of removal substrate of 1.26 gDQO/d. The methane yield was 0.19 LCH4/gDQO removed and the anaerobic biodegradability was a 54.1 %. This study describes the performance of IFBR with Extendospher®, for the treatment of vinasses. The start-up was made in batch, increasing gradually the Organic Load Rate (OLR): 0.5, 1.0, 3.3, 6.8 and 10.4 g COD/L.d. Different Hydraulic Retention Times (HRT) were evaluated: 10, 5 and 1 days. During the operation in batch, the COD removal obtained was of 81 %, and for OLR of 0.5, 1.0, 3.3, 6.8 g COD/L.d the removal obtained was 58, 67, 59 and 50 % respectively. For a maximum OLR of 10.4 g COD/L.d, the COD removal was 38 %, and the system presented instability and decrease of the yield methane. The methane production initiated after 110 days of the start-up of the IFBR, to organic load rate of 1.0 g COD/L.d. The system reached values in the methane yield from 0.15 up to 0.34 LCH4/g CODremoved, for the different organic load rates. During the operation to a constant OLR of 6.4 g COD/L.d, and a HRT of 1 day, the Anaerobic Inverse Fluidized Bed Reactor reached a maximum efficiency of removal of 52 %.

Aggregation of Ig light chains to form amyloid fibrils is a characteristic feature of light-chain amyloidosis, a light-chain deposition disease. A recombinant variable domain of the light chain SMA was used to form amyloid fibrils in vitro. Fibril formation was monitored by atomic force microscopy imaging. Single filaments 2.4 nm in diameter were predominant at early times; protofibrils 4.0 nm in diameter were predominant at intermediate times; type I and type II fibrils 8.0 nm and 6.0 nm in diameter, respectively, were predominant at the endpoints. The increase in number of fibrils correlated with increased binding of the fluorescent dye thioflavin T. The fibrils and protofibrils showed a braided structure, suggesting that their formation involves the winding of protofibrils and filaments, respectively. These observations support a model in which two filaments combine to form a protofibril, two protofibrils intertwine to form a type I fibril, and three filaments form a type II fibril.

Connector enhancer of KSR (CNK) is a multidomain protein required for RAS signaling. Its C-terminal portion (CNKC-term) directly binds to RAF. Herein, we show that the N-terminal portion of CNK (CNKN-term) strongly cooperates with RAS, whereas CNKC-term efficiently blocks RAS- and RAF-dependent signaling when overexpressed in the Drosophila eye. Two effector loop mutants of RASV12, S35 and C40, which selectively activate the mitogen-activated protein kinase (MAPK) and phosphatidylinositol-3-kinase pathways, respectively, do not cooperate with CNK. However, a strong cooperation is observed between CNK and RASV12G37, an effector loop mutant known in mammals to activate specifically the RAL pathway. We have identified two domains in CNKN-term that are critical for cooperation with RAS. Our results suggest that CNK functions in more than one pathway downstream of RAS. CNKc-term seems to regulate RAF, a component of the MAPK pathway, whereas CNKN-term seems to be involved in a MAPK-independent pathway.

Members of the transforming growth factor-β family play critical roles in body patterning, in both vertebrates and invertebrates. One transforming growth factor-β-related gene, dbl-1, has been shown to regulate body length and male ray patterning in Caenorhabditis elegans. We screened arrayed cDNAs to identify downstream target genes for the DBL-1 signaling by using differential hybridization. C. elegans cDNAs representing 7,584 independent genes were arrayed on a nylon membrane at high density and hybridized with 33P-labeled DNA probes synthesized from the mRNAs of wild-type, dbl-1, sma-2, and lon-2 worms. Signals for all the spots representing hybridized DNA were quantified and compared among strains. The screening identified 22 and 2 clones, which were positively and negatively regulated, respectively, by the DBL-1 signal. Northern hybridization confirmed the expression profiles of most of the clones, indicating good reliability of the differential hybridization using arrayed cDNAs. In situ hybridization analysis revealed the spatial and temporal expression patterns of each clone and showed that at least four genes, including the gene for the type I receptor for DBL-1, sma-6, were transcriptionally regulated by the DBL-1 signal.

This study was undertaken to determine the modulation of uterine function by chorionic gonadotrophin (CG) in a nonhuman primate. Infusion of recombinant human CG (hCG) between days 6 and 10 post ovulation initiated the endoreplication of the uterine surface epithelium to form distinct epithelial plaques. These plaque cells stained intensely for cytokeratin and the proliferating cell nuclear antigen. The stromal fibroblasts below the epithelial plaques stained positively for α-smooth muscle actin (αSMA). Expression of αSMA is associated with the initiation of decidualization in the baboon endometrium. Synthesis of the glandular secretory protein glycodelin, as assessed by Western blot analysis, was markedly up-regulated by hCG, and this increase was confirmed by immunocytochemistry, Northern blot analysis, and reverse transcriptase-PCR. To determine whether hCG directly modulated these uterine responses, we treated ovariectomized baboons sequentially with estradiol and progesterone to mimic the hormonal profile of the normal menstrual cycle. Infusion of hCG into the oviduct of steroid-hormone-treated ovariectomized baboons induced the expression of αSMA in the stromal cells and glycodelin in the glandular epithelium. The epithelial plaque reaction, however, was not readily evident. These studies demonstrate a physiological effect of CG on the uterine endometrium in vivo and suggest that the primate blastocyst signal, like the blastocyst signals of other species, modulates the uterine environment prior to implantation.

SMN1 and SMN2 (survival motor neuron) encode identical proteins. A critical question is why only the homozygous loss of SMN1, and not SMN2, results in spinal muscular atrophy (SMA). Analysis of transcripts from SMN1/SMN2 hybrid genes and a new SMN1 mutation showed a direct relationship between presence of disease and exon 7 skipping. We have reported previously that the exon-skipped product SMNΔ7 is partially defective for self-association and SMN self-oligomerization correlated with clinical severity. To evaluate systematically which of the five nucleotides that differ between SMN1 and SMN2 effect alternative splicing of exon 7, a series of SMN minigenes was engineered and transfected into cultured cells, and their transcripts were characterized. Of these nucleotide differences, the exon 7 C-to-T transition at codon 280, a translationally silent variance, was necessary and sufficient to dictate exon 7 alternative splicing. Thus, the failure of SMN2 to fully compensate for SMN1 and protect from SMA is due to a nucleotide exchange (C/T) that attenuates activity of an exonic enhancer. These findings demonstrate the molecular genetic basis for the nature and pathogenesis of SMA and illustrate a novel disease mechanism. Because individuals with SMA retain the SMN2 allele, therapy targeted at preventing exon 7 skipping could modify clinical outcome.
