Asymptomatic influenza virus infections in pigs are frequent and the lack of measures for controlling viral spread facilitates the circulation of different virus strains between pigs. The goal of this study was to demonstrate the circulation of influenza A virus strains among asymptomatic piglets in an abattoir in Brazil and discuss the potential public health impacts. Tracheal samples (n = 330) were collected from asymptomatic animals by a veterinarian that also performed visual lung tissue examinations. No slaughtered animals presented with any noticeable macroscopic signs of influenza infection following examination of lung tissues. Samples were then analysed by reverse transcription-polymerase chain reaction that resulted in the identification of 30 (9%) influenza A positive samples. The presence of asymptomatic pig infections suggested that these animals could facilitate virus dissemination and act as a source of infection for the herd, thereby enabling the emergence of influenza outbreaks associated with significant economic losses. Furthermore, the continuous exposure of the farm and abattoir workers to the virus increases the risk for interspecies transmission. Monitoring measures of swine influenza virus infections and vaccination and monitoring of employees for influenza infection should also be considered. In addition regulatory agencies should consider the public health ramifications regarding the potential zoonotic viral transmission between humans and pigs.

Estudo de diagnóstico de situação, que buscou reconhecer e comparar as condições de inspeção sanitária de Programas de Controle de Infecção Hospitalar (PCIH), junto a agentes de dois Grupos Técnicos de Vigilância Sanitária, em 2002, por meio de questionários. Os resultados mostraram que os agentes possuíam algum conhecimento sobre IH, mas a maioria não inspecionava PCIH, não realizou treinamento, não utilizava roteiro específico e outros recursos. A estrutura física foi o tipo de inspeção mais citado, seguindo-se atas e estatísticas do PCIH. Concluiu-se que as principais dificuldades para a inspeção de PCIH concentram-se na insuficiência de pessoal, recursos, motivação e capacitação técnica. Observou-se, também, heterogeneidade, nesses resultados, entre os Grupos.

Dermatophytes cause the majority of superficial mycoses in humans and animals. However, little is known about the pathogenicity of this specialized group of filamentous fungi, for which molecular research has been limited thus far. During experimental infection of guinea pigs by the human pathogenic dermatophyte Arthroderma benhamiae, we recently detected the activation of the fungal gene encoding malate synthase AcuE, a key enzyme of the glyoxylate cycle. By the establishment of the first genetic system for A. benhamiae, specific ΔacuE mutants were constructed in a wild-type strain and, in addition, in a derivative in which we inactivated the nonhomologous end-joining pathway by deletion of the A. benhamiae KU70 gene. The absence of AbenKU70 resulted in an increased frequency of the targeted insertion of linear DNA by homologous recombination, without notably altering the monitored in vitro growth abilities of the fungus or its virulence in a guinea pig infection model. Phenotypic analyses of ΔacuE mutants and complemented strains depicted that malate synthase is required for the growth of A. benhamiae on lipids, major constituents of the skin. However, mutant analysis did not reveal a pathogenic role of the A. benhamiae enzyme in guinea pig dermatophytosis or during epidermal invasion of the fungus in an in vitro model of reconstituted human epidermis. The presented efficient system for targeted genetic manipulation in A. benhamiae, paired with the analyzed infection models, will advance the functional characterization of putative virulence determinants in medically important dermatophytes.

Differences in physico-chemical characteristics of bone grafts to fill bone defects have been demonstrated to influence in vitro bacterial biofilm formation. Aim of the study was to investigate in vivo staphylococcal biofilm formation on different calcium phosphate bone substitutes. A foreign-body guinea-pig infection model was used. Teflon cages prefilled with β-tricalcium phosphate, calcium-deficient hydroxyapatite, or dicalcium phosphate (DCP) scaffold were implanted subcutaneously. Scaffolds were infected with 2 × 10(3) colony-forming unit of Staphylococcus aureus (two strains) or S. epidermidis and explanted after 3, 24 or 72 h of biofilm formation. Quantitative and qualitative biofilm analysis was performed by sonication followed by viable counts, and microcalorimetry, respectively. Independently of the material, S. aureus formed increasing amounts of biofilm on the surface of all scaffolds over time as determined by both methods. For S. epidermidis, the biofilm amount decreased over time, and no biofilm was detected by microcalorimetry on the DCP scaffolds after 72 h of infection. However, when using a higher S. epidermidis inoculum, increasing amounts of biofilm were formed on all scaffolds as determined by microcalorimetry. No significant variation in staphylococcal in vivo biofilm formation was observed between the different materials tested. This study highlights the importance of in vivo studies, in addition to in vitro studies, when investigating biofilm formation of bone grafts.

Este trabalho tem como objetivo efetuar um estudo da orgânica da direção-geral dos impostos, missão, competências e meios territoriais instalados para conseguir atingir a missão que lhe está atribuída, e pretende igualmente conhecer e dar a conhecer uma das mais importantes instituições públicas do país. A escolha do tema “AUTORIDADE TRIBUTÁRIA E ADUANEIRA (AT) – INSPEÇÃO TRIBUTÁRIA E AS AÇÕES CONJUNTAS COM OUTRAS ENTIDADES” para este relatório deve-se aos seguintes fatores: o facto de estar a realizar um estágio de seis meses nas direções de finanças do porto, pelo que, existe uma maior identificação na abordagem dos temas, e por ser o sector da administração pública do estado que mais gera receitas públicas e também aquela que mais sofreu transformações ao longo do tempo e modificações estruturais significativas de curto e médio prazo.

A indústria dos transportes marítimos é uma das atividades mais importantes de qualquer país, não apenas para o seu desenvolvimento económico, mas também como garante da sua soberania e independência constituindo a segurança marítima uma componente fundamental desta atividade. Tendo como princípios orientadores a segurança e o ambiente marinho, a comunidade marítima internacional adotou, ao longo dos anos, vários tratados tendo como principais objetivos fornecer instrumentos para uma melhor cooperação entre governos no campo da regulamentação e das práticas relacionadas com assuntos técnicos de qualquer género aplicáveis aos navios operando no tráfego internacional. Outro dos objetivos desses instrumentos, foi o de estimular e facilitar a adoção de padrões elevados no que respeita à segurança, eficiência da navegação e à prevenção e controlo da poluição pelos navios. Apesar do esforço das diversas entidades envolvidas, as normas, regulamentos e diretrizes aplicáveis à atividade marítima não se mostram totalmente eficazes quando o assunto é acidente de trabalho a bordo, pois não preveem, salvo em alguns casos, a recolha e o tratamento sistemático dos incidentes e acidentes a bordo, o que poderia servir como base de estudo para aperfeiçoamento das regras para segurança dos navios e do trabalho a bordo.

Forensic Entomology research has been concentrated in only a few localities of the "Cerrado" vegetation, the Brazilian Savannah. The present study had, as its objective, an examination of the diversity of arthropod fauna associated with the carcasses of Sus scrofa (Linnaeus) in this biome. The study was conducted during the dry and humid periods in two Cerrado vegetation profiles of the State of Minas Gerais. The decaying process was slower and greater quantities of arthropods were collected during the dry period. Insects represented 99% of 161,116 arthropods collected. The majority of these were Diptera (80.2%) and Coleoptera (8.8%). The entomofauna belong to 85 families and at least 212 species. Diptera were represented by 31 families and at least 132 species. Sarcophagidae (Diptera) and Scarabaeidae (Coleoptera) were the richest groups. Oxysarcodexia (Sarcophagidae) presented the largest number of attracted species, however none of these species bred in the carcasses. The Coleoptera collected belong to at least 50 species of 21 families. Among these species, Dermestes maculatus and Necrobia rufipes were observed breeding in the carcasses. This study showed species with potential importance for estimating the postmortem interval (PMI), indicative of seasonal and environmental type located.

Sarchophagid flies (Insecta, Diptera) from pig carcasses in Minas Gerais, Brazil, with nine new records from the Cerrado, a threatened Neotropical biome. The diversity of the Sarcophagidae fauna of the Cerrado biome, also know as the Brazilian Savanna, is still underestimated. In this research we collected flies in the state of Minas Gerais, Brazil, during a Forensic Entomology experiment. Samples were collected throughout the decomposition process of domestic pig (Sus scrofa Linnaeus) carcasses, and the experiments were conducted in areas of pasture and semideciduous forest. A total of 85,694 adult flesh flies belonging to 57 species were collected from all carcasses. New records for nine species of Sarcophaginae are provided, including the first record of Blaesoxipha (Acridiophaga) caridei (Brèthes, 1906) to Brazil, and new occurrences of the following species for the Cerrado and/or for the state of Minas Gerais: Blaesoxipha (Acanthodotheca) acridiophagoides (Lopes & Downs, 1951), Malacophagomyia filamenta (Dodge, 1964), Nephochaetopteryx orbitalis (Curran & Walley, 1934), Nephochaetopteryx cyaneiventris Lopes, 1936, Nephochaetopteryx pallidiventris Townsend, 1934, Oxysarcodexia occulta Lopes, 1946, Ravinia effrenata (Walker, 1861) and Sarcophaga (Neobellieria) polistensis (Hall, 1933).

A degradação de estruturas de betão armado em larga escala, seja nos grandes centros urbanos, no meio rural, industrial, nas proximidades ou não dos ambientes marítimos, atualmente, deixa observar problemas patológicos relacionados com a oxidação e a degradação do betão. Muitas vezes esses problemas aparecem de uma forma precoce, cuja recuperação envolve custos elevados, outras vezes é de tal forma que se torna difícil a sua recuperação. Nesta óptica, a aplicação de técnicas de intervenção inovadoras e destinadas a solucionar estes problemas tem cada vez mais uma importância primordial. Baseando na Norma Europeia EN 1504 (2004), por ainda não existir uma Norma Caboverdiana, pretende-se fazer um estudo da aplicação das técnicas, bem como dos métodos de intervenção a elas associadas. A alternativa de intervenção para o edifício em estudo é apresentada como proposta, por forma a melhorar o entendimento sobre a aplicabilidade das técnicas de proteção e de reparação. As escolhas dessas técnicas baseiam-se na visita de inspeção visual, levantamento fotográfico e realização de ensaios para o diagnóstico. O LEC (Laboratório de Engenharia Civil), foi o colaborador para a realização dos ensaios.

ABSTRACTPostmortem injuries are a source of misinterpretations in forensic analysis and therefore are subject matter of taphonomic interest. Many types of injuries can cause different artifacts, which deserve attention of the forensic pathologists when evaluating corpses, either at the crime scene or during an autopsy. Insects can be important biotaphonomic agents and their activity may result in artifacts that resemble antemortem injuries. Here, we describe postmortem injuries caused by the Neotropical wasp Agelaia fulvofasciata (Degeer, 1773) on domestic pig carcasses weighting 15 kg. The specimens showed extensive injuries to the lower lip, similar to lacerations, and some minor lesions on the snout and anus. In addition, we observed the same wasp species preying on larvae of Sarcophagidae (Peckia sp.). Besides causing postmortem injuries, the ability of this species to detect carcasses in the early and fresh decomposition stages should be noted. Thus, future applications aiming criminal, any biotaphonomic events caused by carrion insects need to be disclosed.

Limited treatment options are available for implant-associated infections caused by methicillin (meticillin)-resistant Staphylococcus aureus (MRSA). We compared the activity of daptomycin (alone and with rifampin [rifampicin]) with the activities of other antimicrobial regimens against MRSA ATCC 43300 in the guinea pig foreign-body infection model. The daptomycin MIC and the minimum bactericidal concentration in logarithmic phase and stationary growth phase of MRSA were 0.625, 0.625, and 20 microg/ml, respectively. In time-kill studies, daptomycin showed rapid and concentration-dependent killing of MRSA in stationary growth phase. At concentrations above 20 microg/ml, daptomycin reduced the counts by >3 log(10) CFU/ml in 2 to 4 h. In sterile cage fluid, daptomycin peak concentrations of 23.1, 46.3, and 53.7 microg/ml were reached 4 to 6 h after the administration of single intraperitoneal doses of 20, 30, and 40 mg/kg of body weight, respectively. In treatment studies, daptomycin alone reduced the planktonic MRSA counts by 0.3 log(10) CFU/ml, whereas in combination with rifampin, a reduction in the counts of >6 log(10) CFU/ml was observed. Vancomycin and daptomycin (at both doses) were unable to cure any cage-associated infection when they were given as monotherapy, whereas rifampin alone cured the infections in 33% of the cages. In combination with rifampin, daptomycin showed cure rates of 25% (at 20 mg/kg) and 67% (at 30 mg/kg), vancomycin showed a cure rate of 8%, linezolid showed a cure rate of 0%, and levofloxacin showed a cure rate of 58%. In addition, daptomycin at a high dose (30 mg/kg) completely prevented the emergence of rifampin resistance in planktonic and adherent MRSA cells. Daptomycin at a high dose, corresponding to 6 mg/kg in humans, in combination with rifampin showed the highest activity against planktonic and adherent MRSA. Daptomycin plus rifampin is a promising treatment option for implant-associated MRSA infections.
