In Escherichia coli and Salmonella typhimurium it has been shown that selenophosphate serves as the selenium donor for the conversion of seryl-tRNA to selenocysteyl-tRNA and for the synthesis of 2-selenouridine, a modified nucleoside present in tRNAs. Although selenocysteyl-tRNA also is formed in eukaryotes and is used for the specific insertion of selenocysteine into proteins, the precise mechanism of its biosynthesis from seryl-tRNA in these systems is not known. Because selenophosphate is extremely oxygen labile and difficult to identify in biological systems, we used an immunological approach to detect the possible presence of selenophosphate synthetase in mammalian tissues. With antibodies elicited to E. coli selenophosphate synthetase the enzyme was detected in extracts of rat brain, liver, kidney, and lung by immunoblotting. Especially high levels were detected in Methanococcus vannielii, a member of the domain Archaea, and the enzyme was partially purified from this source. It seems likely that the use of selenophosphate as a selenium donor is widespread in biological systems.

The level and structure of yeast iso-1-cytochrome c and iso-2-cytochrome c, encoded by the nuclear genes CYC1 and CYC7, respectively, are normally not altered in rho- mutants, which completely lack the cytochromes a.a3 subunits and cytochrome b that are encoded by mitochondrial DNA. In contrast, iso-cytochromes c containing the amino acid change Thr-78-->Ile (T78I) were observed at the normal or near-normal wild-type level in rho+ strains but were completely absent in rho- mutants. We have demonstrated with the "global" suppressor mutation Asn-52-->Ile and by pulse-chase labeling that the T78I iso-1-cytochrome c undergoes rapid cellular degradation in rho- mutants. Furthermore, specific mutations revealed that the deficiency of T78I iso-1 cytochrome c can be caused by the lack of cytochrome a.a3 or cytochrome c1, but not by the lack of cytochrome b. Thus, this and certain other, but not all, labile forms of cytochrome c are protected from degradation by the interaction with its physiological partners.

Plasmid-encoded addiction genes augment the apparent stability of various low copy number bacterial plasmids by selectively killing plasmid-free (cured) segregants or their progeny. The addiction module of plasmid prophage P1 consists of a pair of genes called phd and doc. Phd serves to prevent host death when the prophage is retained and, should retention mechanisms fail, Doc causes death on curing. Doc acts as a cell toxin to which Phd is an antidote. In this study we show that host mutants with defects in either subunit of the ClpXP protease survive the loss of a plasmid that contains a P1 addiction module. The small antidote protein Phd is fully stable in these two mutant hosts, whereas it is labile in a wild-type host. We conclude that the role of ClpXP in the addiction mechanism of P1 is to degrade the Phd protein. This conclusion situates P1 among plasmids that elicit severe withdrawal symptoms and are able to do so because they encode both a cell toxin and an actively degraded macromolecule that blocks the synthesis or function of the toxin.

In the facultative anaerobe Escherichia coli, the transcription factor FNR (fumarate nitrate reduction) regulates gene expression in response to oxygen deprivation. To investigate how the activity of FNR is regulated by oxygen availability, two mutant proteins, DA154 and LH28-DA154, which have enhanced in vivo activity in the presence of oxygen, were purified and compared. Unlike other previously examined FNR preparations, the absorption spectrum of LH28-DA154 had two maxima at 324 nm and 419 nm, typical of iron-sulfur (Fe-S)-containing proteins. Consistent with these data, metal analysis showed that only the LH28-DA154 protein contained a significant amount of iron and acid-labile sulfide, and, by low temperature EPR spectroscopy, a signal typical of a [3Fe-4S]+ cluster was detected. The LH28-DA154 protein that contained the Fe-S cluster also contained a higher proportion of dimers and had a 3- to 4-fold higher apparent affinity for the target DNA than the DA154 protein. In agreement with this, we found that when the LH28-DA154 protein was treated with an iron chelator (alpha,alpha'-dipyridyl), it lost its characteristic absorption and the apparent affinity for DNA was reduced 6-fold. However, increased DNA binding and the characteristic absorption spectrum could be restored by in vitro reconstitution of the Fe-S center. DNA binding of the LH28-DA154 protein was also affected by the redox state of the Fe-S center, since protein exposed to oxygen bound 1/10th as much DNA as the protein reduced anaerobically with dithionite. The observation that DNA binding is enhanced when the Fe-S center is reduced indicates that the redox state of the Fe-S center affects the DNA-binding activity of this protein and suggests a possible mechanism for regulation of the wild-type protein.

Neste trabalho foram investigados os mecanismos e o perfil cinético de processos nitrosativos do ponto de vista da nitrosação do indicador 4,5-diamino fluoresceina (DAF2) em células do tipo RAW 264.7. Também foi investigado o papel que ferro lábil (LIP) exerce em tais processos. O estudo cinético mostrou que a nitrosação do DAF2 é dependente de superóxido intracelular e se processa por dois mecanismos distintos denominados nitrosilação oxidativa e nitrosação. Observou-se que o perfil cinético da nitrosaçao do DAF2 sofre uma transição passando de dependente para independente com relação à concentração de NO, quando a concentração de NO se aproxima de 100-110nM. Este perfil está relacionado com a dinâmica de recombinação entre NO e O2¯ que dispara todo o processo de nitrosação do DAF2. No trabalho fica claro que processos nitrosativos que ocorrem pelos mesmos mecanismos podem apresentar perfis cinéticos completamente diferentes dependendo da localização onde ocorre a recombinação entre NO e O2¯. O ponto mais interessante foi a constatação de que quelantes permeáveis à membranas biológicas estimulam a nitrosação do DAF2 intracelular. Este efeito é decorrente da remoção de LIP intracelular que, surpreendementemente, apresenta papel antinitrosativo nas condições experimentais estudadas. O papel incomum antinitrosativo apresentado por LIP é analizado do ponto de vista da reação entre LIP e ONOO¯ que tem como produto nitrito, uma espécie não nitrosante. Estes resultados podem alterar a forma como LIP é visto em processos oxidativos e nitrosativos.

With increased warming in the Arctic, permafrost thaw may induce localized physical disturbance of slopes. These disturbances, referred to as active layer detachments (ALDs), redistribute soil across the landscape, potentially releasing previously unavailable carbon (C). In 2007–2008, widespread ALD activity was reported at the Cape Bounty Arctic Watershed Observatory in Nunavut, Canada. Our study investigated organic matter (OM) composition in soil profiles from ALD-impacted and undisturbed areas. Solid-state 13C nuclear magnetic resonance (NMR) and solvent-extractable biomarkers were used to characterize soil OM. Throughout the disturbed upslope profile, where surface soils and vegetation had been removed, NMR revealed low O-alkyl C content and biomarker analysis revealed low concentrations of solvent-extractable compounds suggesting enhanced erosion of labile-rich OM by the ALD. In the disturbed downslope region, vegetation remained intact but displaced material from upslope produced lateral compression ridges at the surface. High O-alkyl content in the surface horizon was consistent with enrichment of carbohydrates and peptides, but low concentrations of labile biomarkers (i.e., sugars) suggested the presence of relatively unaltered labile-rich OM. Decreased O-alkyl content and biomarker concentrations below the surface contrasted with the undisturbed profile and may indicate the loss of well-established pre-ALD surface drainage with compression ridge formation. However, pre-ALD profile composition remains unknown and the observed decreases may result from nominal pre-ALD OM inputs. These results are the first to establish OM composition in ALD-impacted soil profiles, suggesting reallocation of permafrost-derived soil C to areas where degradation or erosion may contribute to increased C losses from disturbed Arctic soils.

Increased temperature and precipitation in Arctic regions have led to deeper thawing and structural instability in permafrost soil. The resulting localized disturbances, referred to as active layer detachments (ALDs), may transport organic matter (OM) to more biogeochemically active zones. To examine this further, solid state cross polarization magic angle spinning 13C nuclear magnetic resonance (CPMAS NMR) and biomarker analysis were used to evaluate potential shifts in riverine sediment OM composition due to nearby ALDs within the Cape Bounty Arctic Watershed Observatory, Nunavut, Canada. In sedimentary OM near ALDs, NMR analysis revealed signals indicative of unaltered plant-derived material, likely derived from permafrost. Long chain acyclic aliphatic lipids, steroids, cutin, suberin and lignin occurred in the sediments, consistent with a dominance of plant-derived compounds, some of which may have originated from permafrost-derived OM released by ALDs. OM degradation proxies for sediments near ALDs revealed less alteration in acyclic aliphatic lipids, while constituents such as steroids, cutin, suberin and lignin were found at a relatively advanced stage of degradation. Phospholipid fatty acid analysis indicated that microbial activity was higher near ALDs than downstream but microbial substrate limitation was prevalent within disturbed regions. Our study suggests that, as these systems recover from disturbance, ALDs likely provide permafrost-derived OM to sedimentary environments. This source of OM, which is enriched in labile OM, may alter biogeochemical patterns and enhance microbial respiration within these ecosystems.

El lenguado de California Paralichthys californicus es una especie con alto valor comercial debido a su gran tamaño y calidad de su carne. Esta especie presenta dimorfismo sexual en el crecimiento donde las hembras crecen más rápido que los machos, por lo tanto el cultivo monosexual de hembras resulta favorable para la producción. En el presente estudio se registró el proceso de diferenciación sexual mediante cortes histológicos de las larvas y gónadas del lenguado de California asimismo se probó el efecto de diferentes concentraciones de 17β-estradiol (E2) (2.5, 5 y 10 mg/kg) a través de la dieta para incrementar la proporción de hembras en el cultivo. A los 25 días después de la eclosión (DDE) se observó el primordio gonadal en ejemplares con una longitud total promedio de 6.96 ± 0.92 mm, hasta el día 75 DDE (37.58 ± 6.58 mm) se observó una gónada indiferenciada evidenciada por la presencia de células germinales primordiales. La primera evidencia de diferenciación se registró a los 115 DDE (55.93 ± 14.67 mm) donde se observó la cavidad ovárica y posteriormente a los 180 DDE (115.70 ± 17.02 mm) se evidencian ovarios con ovocitos en crecimiento y testículos con espermátidas. Por lo tanto, el periodo lábil para la diferenciación sexual se encuentra entre los días 75 y 115 después de la eclosión. Por otro lado, el suministro de E2 a través de la dieta a concentraciones de 2.5, 5 y 10 mg/kg incrementó el porcentaje de hembras de 26.67% (control, no adición de E2) a un 100% en todos los tratamientos. Se encontraron diferencias en la proporción de los tipos celulares (ovogonias, ovocitos primarios en fase I y II) entre el control y los tratamientos mientras que no se registraron alteraciones histológicas como atrofia gonadal en ninguno de los casos.

La diversidad en los estadios de desarrollo de los poliquetos bentónicos fue estudiada en un perfil batimétrico de la plataforma continental central frente a Callao (Perú, 12°S) del verano al invierno del 2015, con el objetivo de determinar el asentamiento de larvas y reclutamiento de poliquetos bentónicos, bajo la influencia de los factores abióticos del agua y sedimento: temperatura, salinidad, oxígeno disuelto de fondo, sulfuro de hidrógeno en agua intersticial, fitopigmentos totales, materia orgánica total y biopolímeros lábiles. Se encontró un total de 25 especies, pertenecientes a 15 familias. La familia Spionidae presentó la mayor diversidad de larvas (06 especies), seguida de la familia Pilargidae (03 especies). Las larvas de Magelonidae fueron dominantes en verano y otoño. La disponibilidad larval en la capa de fondo estuvo asociada al régimen de oxigenación en el gradiente batimétrico. El número de larvas de poliquetos disminuyó desde el ambiente somero hacia la plataforma externa. Asimismo, la abundancia total de larvas de poliquetos y la abundancia de larvas de Magelona phyllisae se redujeron con el aumento de la temperatura y de la profundidad de la oxiclina durante el período de estudio, caracterizado por la influencia del evento El Niño 2015 - 2016. Por otro lado, se encontró que el número de especies de poliquetos juveniles y adultos fue mayor en ambientes someros, con mayor grado de oxigenación pero también condiciones más reductoras en el sedimento. No obstante, el éxito del reclutamiento, inferido a partir de la abundancia de individuos adultos y la proporción entre el número de especies en estadio adulto y el estadio juvenil fueron mayores en la plataforma externa deficiente en oxígeno, lo cual fue explicado principalmente por la contribución de Paraprionospio pinnata, especie dominante y típica de sedimentos de la plataforma continental.

The distribution and microhabitat of living benthic fora- minifera (15 calcareous and 6 agglutinated) have been studied in two box cores from the Tagus Prodelta. Stable oxygen and carbon isotopes were analysed for eight different species from six surface samples from the Tagus Prodelta and Estuary. At the two box core stations, most of the living foraminifera were restricted to the oxygenated top cm of the sediment and generally show a shallow infaunal behavior. Those taxa are e.g. Rectuvigerina phlegeri, Stainforthia fusiformis and species of the genus Bolivina, which is the most abundant genus in the Tagus Prodelta. Infaunal species are found down to 10 cm depth, and some infaunal taxa, e.g. Bulimina marginata, Globobulimina auriculata and Nonionella turgida, inhabit the low oxic or anoxic sediments. The deep infaunal species are suggested to feed selectively, on refractory organic matter or on the bacterial stocks, while the opportunistic shallow infaunal species are believed to feed on fresh phytodetritus or labile organic matter. Our data show that there is a close connection between the concentration of foraminifera and the distribution of organic matter in the area. The highest abundance of living benthic foraminifera was found in sediments close to the Tagus river plume, where the sediments have relatively high organic carbon contents. The spatial distribution of the stable isotope values of different benthic foraminifera reflects the distribution of the low salinity and relatively high temperature water with high organic carbon fluxes within the Tagus Estuary.
