Background: Anhedonia, the loss of pleasure in usually enjoyable activities, is a central feature of major depressive disorder (MDD). The aim of the present study was to examine whether young people at a familial risk of depression display signs of anticipatory, motivational or consummatory anhedonia, which would indicate that these deficits may be trait markers for MDD. Methods: The study was completed by 22 participants with a family history of depression (FH+) and 21 controls (HC). Anticipatory anhedonia was assessed by asking participants to rate their anticipated liking of pleasant and unpleasant foods which they imagined tasting when cued with images of the foods. Motivational anhedonia was measured by requiring participants to perform key presses to obtain pleasant chocolate taste rewards or to avoid unpleasant apple tastes. Additionally, physical consummatory anhedonia was examined by instructing participants to rate the pleasantness of the acquired tastes. Moreover, social consummatory anhedonia was investigated by asking participants to make preference-based choices between neutral facial expressions, genuine smiles, and polite smiles. Results: It was found that the FH+ group’s anticipated liking of unpleasant foods was significantly lower than that of the control group. By contrast, no group differences in the pleasantness ratings of the actually experienced tastes or in the amount of performed key presses were observed. However, controls preferred genuine smiles over neutral expressions more often than they preferred polite smiles over neutral expressions, while this pattern was not seen in the FH+ group. Conclusion: These findings suggest that FH+ individuals demonstrate an altered anticipatory response to negative stimuli and show signs of social consummatory anhedonia, which may be trait markers for depression.

Recent concern over global pollinator declines has led to considerable research on the effects of pesticides on bees1, 2, 3, 4, 5. Although pesticides are typically not encountered at lethal levels in the field, there is growing evidence indicating that exposure to field-realistic levels can have sublethal effects on bees, affecting their foraging behaviour1, 6, 7, homing ability8, 9 and reproductive success2, 5. Bees are essential for the pollination of a wide variety of crops and the majority of wild flowering plants10, 11, 12, but until now research on pesticide effects has been limited to direct effects on bees themselves and not on the pollination services they provide. Here we show the first evidence to our knowledge that pesticide exposure can reduce the pollination services bumblebees deliver to apples, a crop of global economic importance. Bumblebee colonies exposed to a neonicotinoid pesticide provided lower visitation rates to apple trees and collected pollen less often. Most importantly, these pesticide-exposed colonies produced apples containing fewer seeds, demonstrating a reduced delivery of pollination services. Our results also indicate that reduced pollination service delivery is not due to pesticide-induced changes in individual bee behaviour, but most likely due to effects at the colony level. These findings show that pesticide exposure can impair the ability of bees to provide pollination services, with important implications for both the sustained delivery of stable crop yields and the functioning of natural ecosystems.

P>Pineapple pulp was homogenised at different pressures, and its stability investigated by way of flow curves, particle size distribution, morphology, cloudiness and sedimentation. The particle size of the homogenised pulp ranged from 400 to 100 mu m for homogenisation pressures of between 0 and 700 bar. The pineapple pulp showed shear thinning behaviour with increasing flow index (n) after processing at higher pressures. In addition, the pulps with smaller particles showed less serum cloudiness, even though the sedimentation tests showed the highest stability for pulp homogenised between 200 and 300 bar. Above 400 bar, the pulp showed phase separation and higher sedimentation indexes, similar to that observed for the untreated samples, which was attributed to the formation of aggregates because of interparticle attraction.

This work evaluated the effect of pressure and temperature on yield and characteristic flavour intensity of Brazilian cherry (Eugenia uniflora L) extracts obtained by supercritical CO(2) using response surface analysis, which is a simple and efficient method for first inquiries. A complete central composite 2(2) factorial experimental design was applied using temperature (ranging from 40 to 60 degrees C) and pressure (from 150 to 250 bar) as independent variables. A second order model proved to be predictive (p <= 0.05) for the extract yield as affected by pressure and temperature, with better results being achieved at the central point (200 bar and 50 degrees C). For the flavour intensity, a first order model proved to be predictive (p <= 0.05) showing the influence of temperature. Greater characteristic flavour intensity in extracts was obtained for relatively high temperature (> 50 degrees C), Therefore, as far as Brazilian cherry is concerned, optimum conditions for achieving higher extract yield do not necessarily coincide to those for obtaining richer flavour intensity. Industrial relevance: Supercritical fluid extraction (SFE) is an emerging clean technology through which one may obtain extracts free from organic solvents. Extract yields from natural products for applications in food, pharmaceutical and cosmetic industries have been widely disseminated in the literature. Accordingly, two lines of research have industrial relevance, namely, (i) operational optimization studies for high SFE yields and (ii) investigation on important properties extracts are expected to present (so as to define their prospective industrial application). Specifically, this work studied the optimization of SFE process to obtain extracts from a tropical fruit showing high intensity of its characteristic flavour, aiming at promoting its application in natural aroma enrichment of processed foods. Purpose: The interference of electric fields (EF) with biological processes is an issue of considerable interest. No studies have as yet been reported on the combined effect of EF plus ionising radiation. Here we report studies on this combined effect using the prokaryote Microcystis panniformis, the eukaryote Candida albicans and human cells. Materials and methods: Cultures of Microcystis panniformis (Cyanobacteria) in glass tubes were irradiated with doses in the interval 0.5-5kGy, using a 60Co gamma source facility. Samples irradiated with 3kGy were exposed for 2h to a 20Vcm-1 static electric field and viable cells were enumerated. Cultures of Candida albicans were incubated at 36C for 20h, gamma-irradiated with doses from 1-4kGy, and submitted to an electric field of 180Vcm-1. Samples were examined under a fluorescence microscope and the number of unviable (red) and viable (apple green fluorescence) cells was determined. For crossing-check purposes, MRC5 strain of lung cells were irradiated with 2 Gy, exposed to an electric field of 1250 V/cm, incubated overnight with the anti-body anti-phospho-histone H2AX and examined under a fluorescence microscope to quantify nuclei with -H2AX foci. Results: In cells exposed to EF, death increased substantially compared to irradiation alone. In C. albicans we observed suppression of the DNA repair shoulder. The effect of EF in growth of M. panniformis was substantial; the number of surviving cells on day-2 after irradiation was 12 times greater than when an EF was applied. By the action of a static electric field on the irradiated MRC5 cells the number of nuclei with -H2AX foci increased 40%, approximately. Conclusions: Application of an EF following irradiation greatly increases cell death. The observation that the DNA repair shoulder in the survival curve of C. albicans is suppressed when cells are exposed to irradiation+EF suggests that EF likely inactivate cellular recovering processes. The result for the number of nuclei with -H2AX foci in MRC5 cells indicates that an EF interferes mostly in the DNA repair mechanisms. A molecular ad-hoc model is proposed.

The inactivation kinetics of enzymes polyphenol oxidase (PPO) and peroxidase (POD) was studied for the batch (discontinuous) microwave treatment of green coconut water. Inactivation of commercial PPO and POD added to sterile coconut water was also investigated. The complete time-temperature profiles of the experimental runs were used for determination of the kinetic parameters D-value and z-value: PPO (D(92.20 degrees C) = 52 s and z = 17.6 degrees C); POD (D(92.92 degrees C) = 16 s and z = 11.5 degrees C); PPO/sterile coconut water: (D(84.45 degrees C) = 43 s and z = 39.5 degrees C) and POD/sterile coconut water: (D(86.54 degrees C) = 20 s and z = 19.3 degrees C). All data were well fitted by a first order kinetic model. The enzymes naturally present in coconut water showed a higher resistance when compared to those added to the sterilized medium or other simulated solutions reported in the literature. The thermal inactivation of PPO and POD during microwave processing of green coconut water was significantly faster in comparison with conventional processes reported in the literature. The influence of the sample matrix in the CC-electron-capture detection analysis of the pesticides dimethoate, diazinon, chlorothalonil.. parathion methyl and fenitrothion in fruits samples has been studied. Experiments have been carried out where the pesticide responses in standard solutions prepared in selected solvent were compared with their response when present in apple, mango, papaya, banana, pineapple and melon extracts. The presence of matrix effects (MEs) and their extent were shown to be simultaneously influenced by several factors (matrix concentration, matrix type, pesticide concentration, analytical range). Pronounced MEs were observed particularly for dimethoate and diazinon in all matrices tested; in lower concentrations, all pesticides presented significant ME. The other pesticides presented variable ME. Higher ME enhancement was detected at lower pesticide concentration levels of and/or at higher matrix concentration solutions. The ME detected for fenitrothion, in the analytical range evaluated, were dependent on matrix type. For each pesticide, solvent and matrix-matched calibrations were compared for all fruit samples, and it could be concluded that quantitation based on standard solutions prepared in blank matrix extract (matrix-matched calibration) should be used to compensate the MEs and to obtain more accurate results for the pesticides studied.

Behovet av att analysera volatilt minne från Macintosh-datorer med OS X har blivit allt mer betydelsefull på grund av att deras datorer blivit allt populärare och att volatil minnesanalysering blivit en allt viktigare del i en IT-forensikers arbete. Anledningen till att volatil minnesanalysering blivit allt viktigare är för att det går att finna viktig information som inte finns lagrad permanent på datorns interna hårddisk. Problemet som låg till grunden för det här examensarbetet var att det uppenbart fanns brist på undersökningsmetoder av det volatila minnet för Mac-datorer med OS X.Syftet med detta arbete var därför att undersöka möjligheten att utvinna information från ett volatilt minne från en Mac-dator med OS X genom att kartlägga och bedöma olika undersökningsmetoder. För att göra denna undersökning har litteraturstudier, informella intervjuer, egna kunskaper och praktiska försök genomförts.Slutsatsen blev att möjligheten att utvinna information från det volatila minnet från en Mac-dator med OS X är relativt begränsad. Det största problemet är själva dumpningen av minnet. Många av dumpningsmetoderna som finns att tillgå kräver administrativa rättigheter. Vid analysering av en minnesdump bör man aldrig förlita sig på en analysmetod då olika analysmetoder ger olika resultat som kan vara till nytta för en vidare undersökning av en Mac-dator.

Neste trabalho é avaliada a aplicação da técnica de ICP OES (Espectrometria de Emissão com Plasma Indutivamente Acoplado) para a determinação de elementos traço em amostras ambientais. Foram investigados os comportamentos de As, Ba, Cd, Co, Cu, Cr, Mn, Ni, Pb, V e Zn, utilizando-se espectrômetro com vista de observação axial/radial do plasma e sistema de detecção baseado em dispositivos de carga acoplada (CCD). No presente estudo, foi avaliado o desempenho dos nebulizadores pneumáticos do tipo concêntrico (Meinhard), “cross flow” e GemCone® acoplados às câmaras de nebulização de duplo passo (Scott) e ciclônica, bem como do nebulizador ultra-sônico para a introdução de amostras no plasma. Investigou-se a robustez do plasma, potência de radiofreqüência (RF), vazão dos gases de nebulização e auxiliar, bem como a altura de observação, para ambas as vistas de observação do plasma. Sob condições otimizadas os limites de detecção (LD), em ng mL-1, para os elementos As, Ba, Cd, Co, Cr, Cu, Mn, Ni, Pb, V e Zn, em solução aquosa de HNO3 5% (v/v), utilizando-se a configuração axial do plasma, foram: 1,1 - 16; 0,002 - 0,32; 0,03 - 1,2; 0,02 - 0,72; 0,03 - 0,82; 0,04 - 3,0; 0,003 - 0,76; 0,08 - 3,8; 0,22 - 8,9; 0,04 - 2,6; e 0,02 - 1,2 respectivamente. Utilizando-se a configuração radial, os LDs (ng mL-1) dos mesmos elementos foram: 10 - 87; 0,01 - 0,91; 0,07 - 3,8; 0,16 - 4,3; 0,13 - 8,1; 0,16 - 4,3; 0,01 - 0,81; 0,43 - 7,6; 1,4 - 37; 0,28 - 6,0 e 0,77 - 9,5 respectivamente. Com relação à nebulização pneumática, os LDs são relativamente mais baixos quando é utilizado o nebulizador concêntrico acoplado à câmara de nebulização ciclônica. LDs ainda melhores são obtidos mediante o uso de nebulização ultra-sônica mas, neste caso, foi observado que o plasma é menos robusto. As metodologias foram desenvolvidas mediante o uso de materiais de referência certificados sendo analisados os seguintes materiais: sedimento marinho (PACS-2/NRCC), folhas de maçã (apple leaves – 1515/NIST) e água (natural water – 1640/NIST), obtendo-se concentrações concordantes com as certificadas, com exceção do Ni em folha de maçã, que não foi detectado utilizando-se a nebulização pneumática e também não pôde ser determinado com exatidão mediante o uso de nebulizador ultra-sônico. Interferências não espectrais observadas na análise de sedimento marinho foram contornadas através da diluição da amostra, ou através da lavagem da câmara de nebulização com solução de HNO3 5% (v/v), por 60 s entre cada ciclo de leitura. Estas interferências não puderam ser contornadas com o uso de padrão interno (PI). A interferência espectral do As sobre o Cd não foi observada fazendo-se a medição do sinal em área de pico, demarcado com somente 3 pontos/pico. Após serem estabelecidas as metodologias de análise, foram analisadas as seguintes amostras não certificadas: água de rio, água subterrânea (poço artesiano), água tratada e de chuva, folhas de eucalipto e bambu, acículas de pinus e infusão de chá preto, sendo possível quantificar baixas concentrações dos elementos investigados, utilizando-se a calibração externa.

The association in a form of cooperative can become a guarantee factor to the activities of the small and medium producers of the apple culture in the region of Vacaria-RS, Brazil. The study shows the problems the small and medium producers that grow apples in the city have. After an overview of the literature, the thesis presents a project of a hypothetical structure with the objective of evaluating the financial and economic viability of a cooperative. In this stage the utilized methodology was the collected data available between representatives of the sector and local companies that operate on this activity. The available data was used to construct a cash flow ¿ projecting costs and expenses to an alleged project; utilizing on the sequence to the evaluation the Net Present Value Method ( NPV / Valor Presente Líquido ), the Internal Rate of Return ( IRR / Taxa Interna de Retorno ) and Pay Back ( Período de Recuperação do Investimento ¿ PRI ). The results of the economic and financial evaluation obtained reveal the acceptance of the proposed project. However, on the exemplification phase of the cooperatives that work in other regions, the social, institutional and organizational aspects showed as important as the financial aspects of the project.

Esta dissertação aborda a implementação de um protótipo de aplicação multimídia referente à Ilha Rei George (61º 50’ S, 57º 30’ W e 62º 15’ S, 59º 00' W), Antártica. Nessa ilha o Brasil mantém sua única estação polar, onde realiza parte das pesquisas do Programa Antártico Brasileiro (PROANTAR). O presente trabalho visa reduzir a carência por materiais didáticos em língua portuguesa sobre as regiões polares e aborda aspectos gerais da biota, geografia física (geologia, climatologia e oceanografia) e geografia humana (história de ocupação, turismo, pesquisa e impactos gerados pela presença humana). O público alvo são pessoas com nível de instrução de ensino médio incompleto ou níveis mais avançados. O conteúdo é apresentado de forma a cativar a atenção do usuário, sem exageros estéticos. A fim de obter material para subsidiar o projeto, foram realizados registros por meio de fotografias, gravações em vídeo e entrevistas durante os trabalhos de campo na ilha, realizados no verão de 2004. Além dos registros em campo, são apresentados materiais didáticos tais como textos, ilustrações e animações em 3D. Parte do material gerado está disponível para utilização posterior em outros projetos O ambiente de desenvolvimento utilizado para a aplicação foi o Macromedia Director, para Apple Macintosh. O produto do projeto é um protótipo que pode ser distribuído em mídia removível, CD-ROM (Compact Disk Read Only Memory) ou DVD-ROM (Digital Versatile Disc Read Only Memory) e é encontrado como um encarte no final desta dissertação. A informação compilada e reunida na aplicação permite ao público acesso a dados atualizados sobre o ambiente antártico de forma simples, didática e interativa. O trabalho esclarece as relações entre a Antártica e o mundo, principalmente no que faz referência à ilha Rei George, buscando a valorização das regiões polares.

Image stitching is the process of joining several images to obtain a bigger view of a scene. It is used, for example, in tourism to transmit to the viewer the sensation of being in another place. I am presenting an inexpensive solution for automatic real time video and image stitching with two web cameras as the video/image sources. The proposed solution relies on the usage of several markers in the scene as reference points for the stitching algorithm. The implemented algorithm is divided in four main steps, the marker detection, camera pose determination (in reference to the markers), video/image size and 3d transformation, and image translation. Wii remote controllers are used to support several steps in the process. The built‐in IR camera provides clean marker detection, which facilitates the camera pose determination. The only restriction in the algorithm is that markers have to be in the field of view when capturing the scene. Several tests where made to evaluate the final algorithm. The algorithm is able to perform video stitching with a frame rate between 8 and 13 fps. The joining of the two videos/images is good with minor misalignments in objects at the same depth of the marker,misalignments in the background and foreground are bigger. The capture process is simple enough so anyone can perform a stitching with a very short explanation. Although real‐time video stitching can be achieved by this affordable approach, there are few shortcomings in current version. For example, contrast inconsistency along the stitching line could be reduced by applying a color correction algorithm to every source videos. In addition, the misalignments in stitched images due to camera lens distortion could be eased by optical correction algorithm. The work was developed in Apple’s Quartz Composer, a visual programming environment. A library of extended functions was developed using Xcode tools also from Apple.

A presente dissertação de Mestrado é apresentada para a obtenção do Grau de Mestre em Gestão Cultural pela Universidade da Madeira. Esta investigação pretende efectuar uma breve revisão da bibliografia teórica acerca do livro em suporte impresso e do livro em suporte digital. Naturalmente que foram convocados conceitos como hipertexto, hipermédia e multimédia. A pergunta a formular no decurso desta investigação poderá parecer simples: como é que a linguagem (nas suas dimensões oral e escrita) no ecrã poderá funcionar de diferente forma no papel? Para tentar responder a esta questão foi imprescindível uma investigação em contexto prático para se poder obter uma resposta fundamentada e rigorosa ao problema colocado. O trabalho de campo (em contexto prático, portanto) realizou-se a partir da criação de um texto inédito, escrito pela autora da presente dissertação, através do qual se pretendeu aferir e analisar os efeitos da transição da escrita em suporte papel para meios digitais e/ou a coexistência de ambos os suportes, de forma a tentar compreender a forma como a substituição e/ou a co-presença de meios poderá afectar (e de que modo) a compreensão textual de crianças em contexto educativo, tendo em grande medida por objectivo analisar a influência da escrita e dos áudio-livros no processo de aprendizagem de crianças do 4º ano do 1º Ciclo do Ensino Básico. Para este efeito, elaborou-se um livro em papel e um e-book para ser visualizado no ecrã de um computador e, por comparação, aferir a reacção das crianças perante os dois artefactos. Após as conclusões obtidas neste trabalho de campo, estas foram motivo de reflexão e ponto de partida para a realização de uma aplicação interactiva que neste momento está disponibilizada na store da Apple apenas para iPads.

The volatile composition of different apple varieties of Malus domestica Borkh. species from different geographic regions at Madeira Islands, namely Ponta do Pargo (PP), Porto Santo (PS), and Santo da Serra (SS) was established by headspace solid-phase microextraction (HS-SPME) procedure followed by GC-MS (GC-qMS) analysis. Significant parameters affecting sorption process such as fiber coating, extraction temperature,extractiontime,sampleamount,dilutionfactor,ionicstrength,anddesorption time,wereoptimizedanddiscussed.TheSPMEfibercoatedwith50/30 lmdivinylbenzene/carboxen/PDMS (DVB/CAR/PDMS) afforded highest extraction efficiency of volatile compounds, providing the best sensitivity for the target volatiles, particularly whenthesampleswereextractedat508Cfor30 minwithconstantmagneticstirring. A qualitative and semi-quantitative analysis between the investigated apple species has been established. It was possible to identify about 100 of volatile compounds amongpulp(46,45,and39),peel(64,60,and64),andentirefruit(65,43,and50)inPP, PS,andSSapples,respectively.Ethylesters,terpenes,andhigheralcoholswerefound tobethemostrepresentativevolatiles. a-Farnesene,hexan-1-olandhexyl2-methylbutyratewerethecompoundsfoundinthevolatileprofileofstudiedappleswiththelargestGCarea,representing,onaverage,24.71,14.06,and10.80%ofthetotalvolatilefractionfromPP,PS,andSSapples.InPPentireapple,themostabundantcompoundsidentified were a-farnesene (30.49%), the unknown compound m/z (69, 101, 157) (21.82%) andhexylacetate(6.57%).RegardingPSentireapplethemajorcompoundswere a-farnesene(16.87%),estragole(15.43%),hexan-1-ol(10.94),andE-2-hexenal(10.67).a-Farnesene(30.3%),hexan-1-ol(18.90%),2-methylbutanoicacid(4.7%),andpentan-1-ol(4.6%) werealsofoundasSSentireapplevolatilespresentinahigherrelativecontent.Principal component analysis (PCA) of the results clustered the apples into three groups according to geographic origin. Linear discriminant analysis (LDA) was performed in order to detect the volatile compounds able to differentiate the three kinds of apples investigated. The most important contributions to the differentiation of the PP, PS, and SS apples were ethyl hexanoate, hexyl 2-methylbutyrate, E,E-2,4-heptadienal, pethylstyrene,andE-2-hexenal.

O conhecimento da profundidade ideal de germinação de sementes de plantas daninhas é importante para o desenvolvimento de estratégias de manejo eficientes, seguras e econômicas. Com o objetivo de estudar a emergência de plântulas de Sida rhombifolia L. e Solanum viarum Dunal, em resposta à época (setembro de 2008 e janeiro de 2009) e às profundidades de semeadura (0, 1, 2, 3, 4 e 5 cm), foram realizados dois experimentos em casa de vegetação. Sida rhombifolia mostrou-se sensível às variações de temperatura, em decorrência das épocas de semeadura, e os maiores percentuais de emergência ocorreram nas profundidades entre 1 e 4 cm. Para S. viarum, observou-se forte influência da temperatura sobre a sua emergência, sendo, observado o máximo de emergência, nas profundidades de 1 a 5 cm e sua redução para as sementes locadas na superfície do solo.
