American visceral leishmaniasis (AVL), caused by Leishmania infantum chagasi (L.i.chagasi), stands as a public health problem in Brazil, with human and canine cases related in all states..Lipid metabolism can be modified in several status of infection. For example, experimental studies show that the cholesterol is necessary to internalization and replication of L.i.chagasi in macrophages through caveolar domains. Patients with AVL present low levels of cholesterol and a visible triglycerides increase. This work aimed to evaluate the lipid metabolism in several post-infection status by L.i.chagasi, including individuals with symptomatic infection (AVL), and asymptomatic. The levels of cholesterol, triglycerides, HDL and reactive C protein, were measured. Individuals with AVL were compared with individuals with assymptomatic infection and presented low levels of total cholesterol (128 ± 6.180 mg/dL vs. 158 ±5.733 mg/dL, p=0.0001), HDL (29 ± 1.746 mg/dL vs. 37 ± 1.647 mg/dL, p=0.0001), increased levels of triglycerides (149.5 mg/dL ± 12.72 vs. 78.00 ± 10.43 mg/dL, p=0.0095) and higher levels of reactive C protein (1.750± 0.4939 mg/dL vs. 0.40 ± 0.1707 mg/dL; p=0.0001). The expression of genes related to lipid metabolism, such as LXR-a, LXR-b, PPAR-a, PPAR-d, PPAR-g and APOE was evaluated by real time PCR. A reduction in the expression of those genes was found in the group of AVL patients corroborating the serum levels of the metabolites earlier quantified. Our findings suggest a modulation of metabolism of lipids, in the chronic phase of AVL, this could facilitate the survival of leishmania, due to the known reduction on the ability of macrophages in presenting antigens efficiently to the T cells due to the reduction in the cholesterol available, it results in a subversion of the host immunity.

La levadura metilotrófica Pichia pastoris es de gran importancia industrial principalmente en la producción de proteínas heterólogas. En un estudio reciente se emplearon cinco factores ambientales para definir condiciones de cultivo a nivel de bioreactor que condujeron a altos (CM) y bajos (CP) niveles de la producción extracelular de una fitasa recombinante en una cepa Muts de P. pastoris. Los resultados de este estudio mostraron que bajo las condiciones CM, la demanda y consumo de O2 y de metanol fueron más altos y condujeron a valores más altos en la velocidad específica de crecimiento (μ), biomasa (2.7 veces), niveles de producción de fitasa extracelular (5.5 veces) y rendimientos (Yp/x) que en CP. Con el fin de comprender los mecanismos de regulación transcripcional que afectan a la fisiología de P. pastoris por la sobre-producción de la proteína recombinante y las condiciones de cultivo, en este trabajo se realizó un análisis de expresión diferencial de genes (DGE) empleando la tecnología de secuenciación masiva de mRNA (RNAseq) de la cepa Muts de P. pastoris crecida bajo las condiciones CM y CP reportadas previamente. Además se validaron los resultados del estudio de DGE mediante RT-qPCR. Resultados: La expresión de 4,950 genes, el 93% de los genes totales anotados, fueron detectados. Se sub- y sobre-expresaron 350 y 413 genes respectivamente en CM respecto a CP. En CM vs CP se sobre-expresaron significativamente términos relacionados con la biosíntesis de aminoácidos, biosíntesis de nucleósidos de purina, regulación de la traducción, glicosilación de proteínas y mitosis, indicando una mayor actividad anabólica en CM. La transcripción del gen heterólogo y de los genes de la ruta de desasimilación del metanol no mostraron diferencias entre ambas condiciones de cultivo y fue inducida en metanol. Sin embargo las enzimas claves (DAS1 y DAS2) de la ruta de asimilación del metanol se sobre-expresaron significativamente en CM vs CP, indicando que CM está favorecida la producción de biomasa y la generación de energía a través de esta vía, explicando los valores más altos para la μ y biomasa obtenidos en CM respecto a CP. De 110 genes analizados involucrados en la vía de secreción, 20 se sobre-expresaron en CM vs CP, la sobre-expresión de estos genes indicaron que bajo las condiciones de CM, se presenta una mayor actividad transcipcional de los genes implicados en el transporte y translocación hacia el RE (15%), genes implicados en el plegamiento de proteínas en RE (25%), así como genes relacionados en el procesamieto de las proteínas a través del RE (30%) y Golgi (35%) que permitieron un estado fisiológico favorable para la secreción de la proteína heteróloga. De los 44 genes relacionados con el estrés en RE durante la secreción, en CM vs CP se sobre-expresaron genes UPR indicando, que bajo condiciones de CM, se promueve la expresión de genes relacionados con el plegamiento de proteínas y probablemente se evita el acumulamiento de proteínas mal plegadas. La sub-expresión de todos los genes relacionados con autofagia, es uno de los factores que podría explicar la menor actividad proteolítica observada en CM. Finalmente se observó una correlación entre los métodos de RNA-seq y RTqPCR (r2=0.7). Conclusiones: El análisis de la DGE señala que los factores ambientales en CM condujeron a la regulación de la expresión de genes del proceso de secreción y genes relacionados al estrés en RE durante la secreción que condujeron a valores de Yp/x, más altos en CM que en CP y no se atribuyen a una expresión diferencial del gen heterólogo. La regulación de la ruta del metanol hacia la asimilación y una mejor respuesta de adaptación al estrés en CM condujeron a un mayor crecimiento y producción de biomasa en CM que en CP.

La diabetes es una enfermedad que ha ido en aumento en los últimos años, partiendo de esta problemática el objetivo principal de esta tesis es el de desarrollar una interfaz visual basada en modelos de metabolismo de glucosa, de manera que esta sea una herramienta que sirva de apoyo para observar el comportamiento de dicho metabolismo en un paciente sano, en un paciente con diabetes tipo I o en un paciente con diabetes tipo II, siendo que en un futuro esta se pueda modificar de forma que, con ciertos parámetros establecidos, se pueda llegar a predecir el comportamiento del metabolismo de glucosa en un paciente en específico y con ello saber cómo es que se tiene que actuar frente a las condiciones actuales para mejorar la calidad de vida de la persona en cuestión, aportando de esta manera en el desarrollo de la tecnología de generación de pacientes virtuales. El desarrollo de la interfaz se realizó en MATLAB© y permite el manejo de los tres tipos de pacientes virtuales (sano, diabetes tipo I, y diabetes tipo II) y reproduce el comportamiento dinámico de la concentración de glucosa e insulina en sangre. También permite manejar señales de entrada (dosificación de insulina prescrita por el medico) y perturbaciones (ingesta de alimentos).

The natural raw materials acquired special importance beside the mineral raw materials with the need for using alternative sources to oil, because they can be used to produce biopolymers. Gelatin, produced from the denaturation of collagen, and starch, an abundant polysaccharide in various plants, are examples of biopolymers which have several technological applications, especially in films. The objective of this work is to produce polymeric bioblends with gelatin and corn starch using two types of gelatin: commercial bovine gelatin and gelatin produced from mechanically separated flesh of tilapia (Oreochromis niloticus). For the extraction of tilapia gelatin 3 distinct pretreatments, followed by extraction in distilled water under heating were performed. The properties of gelatin extracted were similar to bovine gelatin, and the differences can be explained by the difference in extraction processes and sources. Blends of commercial gelatin and starch were produced in an internal mixer from a Haake torque rheometer, to study the behavior of the gelatin mixture with starch, thus, the same compositions were processed by twin screw extrusion, to define the mixing parameters. Subsequently, the extrusion of blends of tilapia gelatin and corn starch was carried out in the same twin screw extruder. The physico-chemical, rheological and morphological properties of the blends with thermoplastic starch and gelatin were studied. It was found that various properties vary linearly with increasing concentration of the components. The blends produced are immiscible, and among the two gelatins, tilapia gelatin showed a better interfacial adhesion with the corn starch. Regarding the morphology, gelatins formed the dispersed phase in all compositions studied, even in compositions rich in starch. Can be concluded that the procedure for tilapia gelatin extraction is feasible and advantageous, and the increasing in its scale to a reactor of 30 liters is possible, with a satisfactory yield. The bioblends of bovine gelatin/corn starch and tilapia gelatin/corn starch were successfully produced, and the processing conditions were appropriate

O experimento foi realizado com o objetivo de avaliar a inclusão de amido em dietas para larvas de tilápia-do-nilo (Oreochromis niloticus). Foram utilizadas 375 larvas com 8,02 ± 2,11 mg de peso inicial, distribuídas em 25 aquários com capacidade unitária de 30 L, em um delineamento experimental inteiramente casualizado, com cinco tratamentos e cinco repetições. As rações foram formuladas para conter 0; 2,5; 5; 7,5 e 10% de amido, com 3.500 kcal ED/kg e 38,6% proteína digestível. Os peixes foram alimentados, à vontade, cinco vezes ao dia. O peso final, o ganho de peso, o fator de condição e a sobrevivência dos peixes alimentados com rações acrescidas de amido não diferiram em relação ao daqueles alimentados com a ração controle. O amido pode ser incluído como fonte energética em níveis de até 10% em rações para larvas de tilápia-do-nilo, pois não prejudica o desempenho dos peixes.

No Brasil, estima-se que sejam registrados cerca de 3.000 novos casos de Erros Inatos de Metabolismo (EIM) a cada ano. Os EIM são geralmente multissistêmicos. Muitos evoluem com comprometimento neurológico e óbito precoce. O diagnóstico dos EIM é complexo e compreende várias etapas de investigação. Há possibilidade de intervenção terapêutica em boa parte dos casos. O tratamento específico envolve dietoterapia, uso de fármacos, reposição enzimática e até transplante de órgãos e tecidos. Pacientes com EIM necessitam de acompanhamento especializado cuidadoso. Neste quarto módulo do curso de Formação de Teleconsultores sobre a Política Nacional de Atenção Integral às Pessoas com Doenças Raras no SUS são abordados os seguintes tópicos: classificação dos EIM; roteiro de anamnese; sinais e sintomas que indiquem a presença de EIM; fluxograma atendimento recomendado para EIM.

Annatto seeds do not germinate during early stages of their development because of insufficient reserve substances. In situ analysis showed that the principal reserves are proteins and starch, deposited in endosperm cells. During the early stages of development, the starch grains were elliptic, because amylose was the minor component. During development, these grains became more spherical due to an increase in amylose relative to amylopectin. Endosperm cells do not contain protein bodies, but they accumulate proteins dispersed in the cytoplasm. At the final stage of development the proteins became compacted due to the dehydration of the seeds wich is part of the global process of orthodox seeds maturation. Natural fluorescence revealed aromatic amino acids, principally tryptophan and tyrosine in the proteins. The seeds reached their maximum dry weight after moisture contents had declined to around 60%. At this point the seeds presented maximum germination capacity.

Kohleria eriantha (Benth.) Hanst is a plant belonging to the family Gesneriaceae, with an underground organ, which is associated with vegetative reproduction. This organ is a rhizome, whose stem bears buds covered with modified leaves that store up starch. In small sections of this rhizome, containing six buds (1.5 to 2.0cm long), only one bud sprouted. The sprouted bud could be differentiated into two morphological pattern: aerial part or rhizome. Sprouting of the rhizome pattern occurred in sections kept on substrate with low water content (1mL of water), or lacking water, whereas sprouting of the aerial part pattern occurred in sections on substrate with high water content (12mL of water). Temperature at 20ºC also stimulated sprouting of the rhizome pattern, regardless of the water volume in the substrate. Sprouting of the rhizome pattern occurred still in sections on substrate to which polyethylene glycol 6000 (PEG) solution was added at the concentrations of 161.2, 235.2 and 340.0g/L, resulting in potentials of -3, -6 and -12 MPa, respectively. Sections kept on substrate with low water content (1 ml of water) showed a reduction in the dry matter content and high osmotic concentration in comparison with those on substrate with high water content. The results obtained revealed that forming of the rhizome pattern was influenced by water content and temperature. It is suggested that sprouting of the rhizome pattern was induced by the low water potential in the sections, when kept on substrate with low water content. Moreover, it was observed that the rhizome buds of Kohleria eriantha showed a high degree of plasticity.

INTRODUCTION: Like in humans, lower amounts of glycogen are present in tissues of diabetic rats. However, training or drugs that lower glycemia can improve the metabolic control. Metformin increased glycogen while decreased glycemia in normal rats stressed by exercise. OBJECTIVE: In this work we investigated if regular exercise and metformin effects improve the metabolism of diabetic rats. METHODS: Alloxan diabetic Wistar rats treated with metformin (DTM) or not (DT) were trained. Training consisted of 20 sessions of 30 min, 5 days a week. Sedentary diabetic rats served as control (SD and SDM). Metformin (5.6 µg/g) was given in the drinking water. After 48 h resting, glucose (mg/dl) and insulin (ng/mL) was measured in plasma and glycogen (mg/100 mg of wet tissue) in liver, soleus and gastrocnemius. RESULTS: Glycemia decreased in DM group from 435±15 to 230±20, in DT group to 143±8.1 and in DTM group to 138±19 mg/dl. DM group had proportional increase in the hepatic glycogen from 1.69±0.22 to 3.53±0.24, and the training increased to 3.36 ± 0.16 mg/100 mg. Metformin induced the same proportional increase in the muscles (soleus from 0.21±0.008 to 0.42±0.03 and gastrocnemius from 0.33±0.02 to 0.46±0.03), while the training promoted increase on gastrocnemius to 0,53 ± 0,03, only. A high interaction was observed in liver (glycogen increased to 6.48±0.34). CONCLUSION: Very small oral doses of metformin and/or, partially restored glycemia in diabetic rats and decreased glycogen in tissues. Its association with an exercise program was beneficial, helping lower glycemia further and increase glycogen stores on liver of diabetic rats.
