The cytogenetic analyses showed that Pseudopimelodns mangurus has a diploid number of 2n=54 chromosomes (6M, 26SM, 12ST and 10A), single Ag-NORs on the short arm of a middle-sized ST pair, identified as pair 19, and a very small amount of C-band positive segments in two chromosome pairs. The Ag-NORs are C-band positive. The staining of the chromosomes of P. mangurus with CMA 3 reveled the occurrence of bright signals corresponding to the Ag-NORs segments, to the C-band positive segments and also to some C-band negative segments. The occurrence of a diploid number of 2n=54 in all species of the family Pseudopimelodidae and its absence among representatives of Pimelodidae and Heptapteridae, two related families previously considered, reinforces the hypothesis that Pseudopimelodidae is a monophyletic group. © 2004 The Japan Mendel Society.

Cytogenetics studies in 12 specimens of Gymnothorax ocellatus reveled a diploid chromosome number of 2n=42 (16 metacentrics, 18 submetacentrics and 8 acrocentrics). The nucleolar organizer regions were located in a terminal position on the long arm of the chromosome pair number fifteen. Conspicuous blocks of constitutive heterochromatin were observed in the centromeric and pericentromeric regions of some chromosome pairs. The results obtained are similar to those previously described for others species of this family. However, the cytogenetic informations may be useful in the identification of a possible variety of this species in Brazilian coast and contribute to the understanding of relationships among the species and the process of diversification which occurred in this group. © 2005 The Japan Mendel Society.

There is high interest in the natural products properties due to their use in popular medicine. Agaricus blazei Murrill ss. Heinem. (Ab) is native to Brazil and has been widely disseminated because its medicinal properties. In the present study, the genotoxic and antigenotoxic potential of Ab extracts were investigated using the comet assay. The cells utilized were the non drug-metabolizing line CHO-k1 (Chinese hamster ovary) and the drug-metabolizing line HTC (rat hepatoma). Cells were treated for 3 h in the absence of fetal bovain serum (FBS) with methanolic, hexanic and n-butanolic extracts at 50 μg/ml and 0.75% aqueous extract to test for genotoxicity. Antigenotoxic effects of extracts were determined in cells exposed to the DNA damage inducing agent ethyl methanesulfonate under simultaneous or simultaneous with 1 h pre-incubation conditions. The extracts did not show genotoxicity in HTC, while they were genotoxic in CHO-k1. No antigenotoxic effect was observed with any extract under any condition. These results demonstrate that the metabolism in presence or in absence has a direct influence on the genotoxicity of these extracts. © 2006 The Japan Mendel Society.

The karyotype of a fish species belonging to the family Scoloplacidae, Scoloplax distolothrix, is described for the first time. Cytogenetic studies showed that this species has 2n=50 chromosomes, 18 metacentrics, 18 submetacentrics, 10 subtelocentrics and 4 acrocentrics. The Ag-NORs are restricted to a single large metacentric pair in the interstitial position on the long arm. © 2006 The Japan Mendel Society.

The clastogenic effect of the A. populnea leaves extract was tested in vivo on bone marrow cells of Wistar rats by evaluating the induction of chromosome aberrations and micronuclei induction on polychromatic erythrocytes. The extract was administered by gavage at doses of 300, 600 and 900mg/kg body weight. Experimental and control animals were submitted to euthanasia 24 h after the treatment. Under the conditions used, A. populnea leaves extract did not induce decrease in mitotic index and did not induce a statistically significant increase in the mean number of micronucleated polychromatic erythrocytes or chromosome aberrations in the bone marrow cells of Wistar rats. © 2007 The Japan Mendel Society.

Some cytogenetical aspects of spermatozoa formation were studied in 9 Coreidae Brazilian species: Anasa bellator, Athaumastus haematicus, Chariesterus armatus, Dallacoris obscura, Dallacoris pictus, Leptoglossus gonagra, Leptoglossus zonatus, Sphictyrtus fasciatus, and Zicca annulata. Similarly to the other species described to date, all the species studied herein showed cystic spermatogenesis, a reddish membrane covering the testes, a X0 sex determining system, a pair of m-chromosomes, intersticial chiasmata in most autosomes, and autosomes dividing reductionally at first meiotic division and equationally in the second 1 while sex chromosomes, divide equationally and reductionally at first and second meiotic division, respectively. In addition, it was observed that the sex chromosome is heteropycnotic at prophase and that heteropycnotic chromosomal material is found in the nuclei at spermiogenesis. In the species studied, the diploid chromosome number ranged from 19 to 25. It was 19 in S. fasciatus (16A+2m+X0); 21 in A. bellator, A. haematicus, D. obscura, D. pictus, L. gonagra, and L. zonatus (18A+2m+X0); 23 in Z. annulata (20A+2m+X0); and 25 in C. armatus (22A+2m+X0). © 2007 The Japan Mendel Society.

Three sympatric species of Gymnotus from the Fundo stream, a small tributary of the Sapucai river, Minas Gerais State, Brazil, were studied in relation to their karyology. Gymnotus sylvius presented 2n=40 chromosomes (36 m/sm+4 st/a), Gymnotus sp. presented 2n=50 (26 m/sm+ 24 st/a) and Gymnotus paraguensis had 2n=54 (50 m/sm+4 st/a). C-banding demonstrated positively stained heterochromatic blocks in the centromeric position of few chromosomes on G. sylvius and in the centromeric region of all chromosomes on G. paraguensis samples. The nucleolus organizer region (NOR) was located on the short arm of one st chromosome pair in G. sylvius and Gymnotus sp., and in the interstitial position on the short arm of the pair number one and below the centromere on a third chromosome on G. paraguensis. The cytogenetic data obtained indicate that Gymnotus sp. represent a new Gymnotus specie with a karyotypic constitution never observed on others species from this genus. Some aspects related to the chromosome diversification of these Gymnotus are discussed. © 2007 The Japan Mendel Society.

Cytogenetical data in 3 populations of characid fish assigned to the complex of Astyanax scabripinnis from São Francisco river basin and Grande river basin, Minas Gerais State, Brazil, are presented for the first time. The same diploid number, 2n=50, was detected in the 3 populations, which has conspicuous differences involving karyotype morphology: 8M, 20SM, 6ST and 16A (Cambeba stream), 6M, 28SM, 6ST and 10A (Machado headwater), 6M, 24SM, 8ST and 12A (Pedra Branca stream). Differences involving amount and/or locations of heterochromatin blocks, number and position of nucleolar organizer regions (NORs) and CMA3 positive signals were also observed. Some aspects related to the chromosome diversification of Astyanax scabripinnis are discussed. © 2007 The Japan Mendel Society.

Casearia sylvestris (Flacourtiaceae) is a plant which grows in wild and has been widely used in folk medicine. In this study, clastogenic/aneugenic properties of Casearia sylvestris crude ethanolic extract were evaluated using in vivo chromosomal aberrations (CAs) and micronucleus (MN) assays in rodents. The animals were treated by gavage with 3 concentrations of the extract: 150, 300 and 500 mg/kg body weight. Bone marrow cells from Wistar rats were collected 24 h after having been submitted to the MN and CAs test. Peripheral blood cells from Swiss mice were collected 48 and 72 h after having been submitted to the MN test. The results show that C. sylvestris extract does not induce a significant increase in mean values for micronucleated polychromatic erythrocytes (MNPCE) in Swiss mice and Wistar rats, or CAs in rat bone marrow cells, at the 3 tested doses, indicating that the extract showed no clastogenic/aneugenic effects on chromosomes of the rodent cells tested. © 2007 The Japan Mendel Society.

Ethnopharmacological relevance: In Brazil, a phytotherapeutic preparation produced from a standardized tincture of Cinchona calisaya Weddel such that each mL of product contains 400 mu g of quinine, known in Portuguese as Agua Inglesa (R) (English water), is indicated by the manufacturer as a tonic, appetite stimulant, and digestive. However, this preparation has long been used in folk medicine as a female fertility stimulant. Despite its widespread use in folk medicine to stimulate female fertility, no study has been undertaken to assess the potential teratogenic and genotoxic effects of this phytotherapeutic preparation. The aim of the present study was to investigate possible toxic reproductive effects in mice caused by exposure to Agua Inglesa (R), either before mating or during the pre- and post-embryo implantation periods. The genotoxic potential was evaluated using the micronucleus assay.Material, Methods, and Results: Virgin female mice, with at least one estrous cycle evidenced by vaginal cytology, were divided into five groups of 15 individuals each (Group I - control, Group II - treated with ethanol solution at 16%, Groups III, IV and V treated with phytotherapeutic preparation at 1.5 mL/kg/day, 3.0 mL/kg/day and 4.5 mL/kg/day, respectively). After the first 28 days of treatment, females were caged individually with adult fertile males. Pregnant females continued to receive treatment for seven days (preimplantation period). Body weight was recorded weekly during treatment. Signs of toxicity (weight loss, food intake, piloerection, apathy, prostration, diarrhea, seizures, behavioral changes, and locomotion) were also observed. The females were sacrificed on the 15th day of pregnancy, uterine horns were evaluated for implantation, and the placental index was recorded. In the micronucleus test, 2000 polychromatic erythrocytes (PCE) per animal, obtained from bone marrow, were scored. Results The results showed that exposure of the females during the pre- and post-implantation periods did not significantly alter the reproductive capacity (p < 0.05); however, in higher dose (three times human dose)reduction of fetal weight was observed. There was no difference between the control and phytotherapeutic preparation (p > 0.05) in terms of the average number of micronucleated polychromatic erythrocytes.Conclusions: Although folk medicine suggests that the Agua Inglesa (R) preparation is useful as a female fertility stimulant, no such effect was confirmed in mice. Small-scale farmers in the Brazilian Amazon collectively hold tenure over more than 12 million ha of permanent forest reserves, as required by the Forest Code. The trade-off between forest conservation and other land uses entails opportunity costs for them and for the country, which have not been sufficiently studied. We assessed the potential income generated by multiple use forest management for farmers and compared it to the income potentially derived from six other agricultural land uses. Income from the forest was from (i) logging, carried out by a logging company in partnership with farmers' associations; and (ii) harvesting the seeds of Carapa guianensis (local name andiroba) for the production of oil. We then compared the income generated by multiple-use forest management with the income from different types of agrarian systems. According to our calculations in this study, the mean annual economic benefits from multiple forest use are the same as the least productive agrarian system, but only 25% of the annual income generated by the most productive system. Although the income generated by logging may be considered low when calculated on an annual basis and compared to incomes generated by agriculture, the one-time payment after logging is significant (US$5,800 to US$33,508) and could be used to implement more intensive and productive cropping systems such as planting black pepper. The income from forest management could also be used to establish permanent fields in deforested areas for highly productive annual crops using conservation agriculture techniques. These techniques are alternatives to the traditional land use based on periodic clearing of the forest. Nevertheless, the shift in current practices towards adoption of more sustainable conservation agriculture techniques will also require the technical and legal support of the State to help small farmers apply these alternatives, which aim to integrate forest management in sustainable agricultural production systems.
