We report, for the first time, extensive biologically-mediated phosphate removal from wastewater during high-rate anaerobic digestion (AD). A hybrid sludge bed/fixed-film (packed pumice stone) reactor was employed for low-temperature (12°C) anaerobic treatment of synthetic sewage wastewater. Successful phosphate removal from the wastewater (up to 78% of influent phosphate) was observed, mediated by biofilms in the reactor. Scanning electron microscopy and energy dispersive X-ray analysis revealed the accumulation of elemental phosphorus (~2%) within the sludge bed and fixed-film biofilms. 4’, 6-diamidino-2-phenylindole (DAPI) staining indicated phosphorus accumulation was biological in nature and mediated through the formation of intracellular inorganic polyphosphate (polyP) granules within these biofilms. DAPI staining further indicated that polyP accumulation was rarely associated with free cells. Efficient and consistent chemical oxygen demand (COD) removal was recorded, throughout the 732-day trial, at applied organic loading rates between 0.4-1.5 kg COD m-3 d-1 and hydraulic retention times of 8-24 hours, while phosphate removal efficiency ranged from 28-78% on average per phase. Analysis of protein hydrolysis kinetics and the methanogenic activity profiles of the biomass revealed the development, at 12˚C, of active hydrolytic and methanogenic populations. Temporal microbial changes were monitored using Illumina Miseq analysis of bacterial and archaeal 16S rRNA gene sequences. The dominant bacterial phyla present in the biomass at the conclusion of the trial were the Proteobacteria and Firmicutes and the dominant archaeal genus was Methanosaeta. Trichococcus and Flavobacterium populations, previously associated with low temperature protein degradation, developed in the reactor biomass. The presence of previously characterised polyphosphate accumulating organisms (PAOs) such as Rhodocyclus, Chromatiales, Actinobacter and Acinetobacter was recorded at low numbers. However, it is unknown as yet if these were responsible for the luxury polyP uptake observed in this system. The possibility of efficient phosphate removal and recovery from wastewater during AD would represent a major advance in the scope for widespread application of anaerobic wastewater treatment technologies.

A Czerny Mount double monochromator is used to measure Raman scattered radiation near 90" from a crystalline, Silicon sample. Incident light is provided by a mixed gas Kr-Ar laser, operating at 5145 A. The double monochromator is calibrated to true wavelength by comparison of Kr and Ar emission Une positions (A) to grating position (A) display [1]. The relationship was found to be hnear and can be described by, y = 1.219873a; - 1209.32, (1) where y is true wavelength (A) and xis grating position display (A). The Raman emission spectra are collected via C"*""*" encoded software, which displays a mV signal from a Photodetector and allows stepping control of the gratings via an A/D interface. [2] The software collection parameters, detector temperature and optics are optimised to yield the best quality spectra. The inclusion of a cryostat allows for temperatmre dependent capabihty ranging from 4 K to w 350 K. Silicon Stokes temperatm-e dependent Raman spectra, generally show agreement with Uterature results [3] in their frequency haxdening, FWHM reduction and intensity increase as temperature is reduced. Tests reveal that a re-alignment of the double monochromator is necessary before spectral resolution can approach literature standard. This has not yet been carried out due to time constraints.

Zinc salts of ethyl, isopropyl, and butyl xanthates were prepared in the laboratory. The effect of these xanthates in combination with zinc diethyldithiocarbamate (ZDC) on the vulcanization of silica-filled NBR compounds has been studied at different temperatures. The cure times of these compounds were compared with that of NBR compounds containing tetramethylthiuram disulphide/dibenzthiazyl disulphide. The rubber compounds with the xanthates and ZDC were cured at various temperatures from 60 to 150°C. The sheets were molded and properties such as tensile strength, tear strength, crosslink density, elongation at break, compression set, abrasion resistance, flex resistance, heat buildup, etc. were evaluated. The properties showed that zinc salt of xanthate/ZDC combination has a positive synergistic effect on the cure rate and mechanical properties of NBR compounds.

The properties show that zinc salt of the xanthate/ZDC accelerator system has a positive synergistic effect on the cure rate and mechanical properties of NBR compounds.

Microcellular (MC) soles based on polybutadiene (BR) and low-density polyethylene (LDPE) blends for low-temperature applications were developed. A part of BR in BR-LDPE blend was replaced by natural rubber (NR) for property improvement. The BR-NR-LDPE blend-based MC sole shows good technical properties. Sulphur curing and DCP curing were tried in BR-LDPE and NR-BR-LDPE blends. Study shows that sulphur-cured MC sheets possess better technical properties than DCPcured MC sheets. 90/10 BR-LDPE and 60/30/10 BR-NR-LDPE blend combinations are found to be suitable for low-temperature applications.

ABSTRACT: Zinc salts of ethyl, isopropyl, and butyl xanthates were prepared in the laboratory. They were purified by reprecipitation and were characterized by IR, NMR, and thermogravimetric analysis techniques. The melting points were also determined. The rubber compounds with different xanthate accelerators were cured at temperatures from 30 to 150°C. The sheets were molded and properties such as tensile strength, tear strength, crosslink density, elongation at break, and modulus at 300% elongation were evaluated. The properties showed that all three xanthate accelerators are effective for room temperature curing.

This thesis summarizes the results on the growth and characterisation of thin films of HA grown on TiAl6V4 (Ti) implant material at a lower substrate temperature by a combination of Pulsed laser deposition and a hydrothermal treatment to get sufficiently strong crystalline films suitable for orthopaedic applications. The comparison of the properties of the coated substrate has been made with other surface modification techniques like anodization and chemical etching. The in-vitro study has been conducted on the surface modified implants to assess its cell viability. A molecular level study has been conducted to analyze the adhesion mechanism of protein adhesion molecules on to HA coated implants.

The primary objective of this work is to develop an efficient accelerator system for low temperature vulcanization of rubbers. Although xanthates are known to act as accelerators for low temperature vulcanization, a systematic study on the mechanism of vulcanization, the mechanical properties of the vulcanizates at varying temperatures of vulcanization, cure characteristics etc are not reported. Further. xanthate based curing systems are not commonly used because of their chance for premature vulcanization during processing. The proposed study is to develop a novel accelerator system for the low temperature vulcanization of rubbers having enough processing safely. lt is also proposed to develop a method for the prevulcanisation of natural rubber latex at room temperature. As already mentioned the manufacture of rubber products at low temperature will improve its quality and appearance. Also, energy consumption can be reduced by low temperature vulcanization. in addition, low temperature vulcanization will be extremely useful in the area of repair of defective products, since subjecting finished products to high temperatures during the process of repair will adversely affect the quality of the product. Further. room temperature curing accelerator systems will find extensive applications in surface coating industries.

Nanocomposites with magnetic components possessing nanometric dimensions, lying in the range 1–10 nm, are found to be exhibiting superior physical properties with respect to their coarser sized counterparts. Magnetic nanocomposites based on gamma iron oxide embedded in a polymer matrix have been prepared and characterized. The behaviour of these samples at low temperatures have been studied using Mössbauer spectroscopy. Mössbauer studies indicate that the composites consist of very fine particles of g-Fe2O3 of which some amount exists in the superparamagnetic phase. The cycling of the preparative conditions were found to increase the amount of g-Fe2O3 in the matrix

Die Trocknung ist eines der am häufigsten verwendeten Verfahren, um die Haltbarkeit von landwirtschaftlichen Gütern zu verlängern. Von den vielen gebräuchlichen Trocknungsarten ist die Konvektionstrocknung, die Luft als Energie- und Feuchteträger verwendet, weiterhin die wichtigste. Trotz ihrer Nachteile und ihres hohen spezifischen Energieverbrauchs sind Satztrockner für die Getreide-, Kräuter- und Heutrocknung noch weit verbreitet. Ferner werden Konvektionstrockner gemeinhin zur künstlichen Trocknung eingesetzt. Diese Arbeit ist Teil eines Forschungsprojekts, welches eine sorptionsgestützte solare Trocknung von landwirtschaftlichen Gütern untersucht. Bei dieser wird kühle feuchte Luft in Kontakt mit einer konzentrierten hygroskopischen Salzlösung gebracht. Während dieses Prozesses wird die Luft entfeuchtet und mit Hilfe der freigesetzten Verdampfungsenthalpie erwärmt. Die Anwendung dieses Verfahrens zur Trocknung landwirtschaftlicher Güter ist besonders interessant für Produkte, die bei niedrigen Temperaturen verarbeitet werden, um ihre Qualität nicht negativ zu beeinflussen. Bei allen energieintensiven Prozessen und vor allem bei der Verwendung von Solarenergie ist ein effizienter Energieverbrauch äußerst wichtig für den technischen und ökonomischen Erfolg. Eine ungleichmäßige Trocknung ist oftmals bei Satztrocknern zu beobachten, was die Energieeffizienz negativ beeinflusst. Aus diesem Grund lag im agrartechnischen Teil des Projekts der Schwerpunkt auf der Trocknung bei niedrigen Temperaturen und geringer Luftfeuchte, die der Sorptionsprozess liefert, sowie auf der Verbesserung der Luftverteilung und der Gleichmäßigkeit der Trocknung in lockeren und verdichteten Schüttungen. Es wurden Dünnschicht-Trocknungsexperminente für Blattsellerie bei unterschiedlichen Lufttemperaturen und unterschiedlicher relativer Luftfeuchte durchgeführt. Die Ergebnisse zeigen eindeutig, dass nicht nur die Lufttemperatur einen großen Einfluss auf die Trocknungsgeschwindigkeit hat, sondern auch die relative Luftfeuchte. Dies ist vor allem bei niedrigen Temperaturen, wie sie vom Sorptionssystem geliefert werden, der Fall. Um die Luftverteilung und Gleichmäßigkeit der Trocknung lockerer Schüttungen zu untersuchen, wurde ein Kistentrockner experimentell und in Strömungssimulationen getestet. Die Leistung des ursprünglichen Trockners wurde signifikant durch einfache Veränderungen des Designs verbessert. Diese stellten einen gleichmäßigeren Luftstrom durch das Produkt sicher. Die Trocknung von Heu-Rundballen stellt eine Herausforderung dar, da diese sehr stark verdichtet sind und die Dichte innerhalb eines Ballens stark schwankt. Strömungs- und Trocknungssimulationen so wie experimentelle Messungen zeigten, dass das Trocknerdesign einen großen Einfluss auf die Gleichmäßigkeit der Trocknung hat. Bei den einfachsten Trocknervarianten war die Verteilung auch bei optimal gepressten Ballen unzureichend, während komplexere Trockner eine gleichmäßigere Trocknung erzielten. Jedoch werden auch hier die Ergebnisse stark von der Verteilung der Dichte im Ballen beeinflusst, welche in der Praxis weiterhin nicht ideal ist. Abschließend wurde, um den Trocknungsfortschritt zu überwachen, eine Methodik getestet, bei der zur Messung des durchschnittlichen aktuellen Feuchtegehalts des Produktes psychrometrische Messungen und Messungen des Luftstroms in kurzen Abständen durchgeführt wurden. Die Menge des tatsächlich entfernten Wassers stimmte in den meisten Fällen gut mit der geschätzten Menge überein. Jedoch hängt der Erfolg dieser Methode stark von der Genauigkeit der Messfühler ab.

Oxidation of amorphous silicon (a-Si) nanoparticles grown by plasma-enhanced chemical vapor deposition were investigated. Their hydrogen content has a great influence on the oxidation rate at low temperature. When the mass gain is recorded during a heating ramp in dry air, an oxidation process at low temperature is identified with an onset around 250°C. This temperature onset is similar to that of hydrogen desorption. It is shown that the oxygen uptake during this process almost equals the number of hydrogen atoms present in the nanoparticles. To explain this correlation, we propose that oxidation at low temperature is triggered by the process of hydrogen desorption

A set of free-drift experiments was undertaken to synthesize carbonates of mixed cation content (Fe, Ca, Mg) from solution at 25 and 70 degrees C to better understand the relationship between the mineralogy and composition of these phases and the solutions from which they precipitate. Metastable solid solutions formed at 25 degrees C which are not predicted from the extrapolation of higher temperature equilibrium assemblages; instead, solids formed that were intermediary in chemical composition to known magnesite-siderite and dolomite solid solutions. A calcite-siderite solid solution precipitated at 25 degrees C, with the percentage of CaCO3 in the solid being proportional to the aqueous Ca/Fe ratio of the solution, while Mg was excluded from the crystal structure except at relatively high aqueous Mg/Ca and Mg/Fe ratios and a low Ca content. Alternatively, at 70 degrees C Mg was the predominant cation of the solid solutions. These results are compatible with the hypothesis that the relative dehydration energies of Fe, Ca and Mg play an important role in the formation of mixed cation carbonates in nature. Soy isoflavones are thought to have a cardioprotective effect that is partly mediated by an inhibitory influence on the oxidation of low density lipoprotein (LDL). However, the aglycone forms investigated in many previous studies do not circulate in appreciable quantities because they are metabolised in the gut and liver. We investigated effects of various isoflavone metabolites, including for the first time the sulphated conjugates formed in the liver and the mucosa of the small intestine, on copper-induced LDL oxidation. The parent aglycones inhibited oxidation, although only 5% as well as quercetin. Metabolism increased or decreased their effectiveness. Equol inhibited 2.65-fold better than its parent compound daidzein and 8-hydroxydaidzein, not previously assessed, was 12.5-fold better than daidzein. However, monosulphated conjugates of genistein, daidzein and equol were much less effective and disulphates completely ineffective. Since almost all isoflavones circulate as conjugates, these data suggest that despite the increased potency produced by some metabolic changes, isoflavones may not be effective antioxidants in vivo unless they are deconjugated again.

Oxidised low density lipoprotein (LDL) may play a role in atherogenesis. We have investigated some of the mechanisms by which the thiol cysteine and the disulphide cystine can influence the oxidation of LDL by copper ions. Cysteine or cystine (100 PM) inhibited the oxidation of native LDL by copper in a simple phosphate buffer. One of the mechanisms by which cysteine (or more likely its oxidation products in the presence of copper) and cystine inhibited LDL oxidation was by decreasing the binding of copper to LDL (97% inhibition). Cysteine, but not cystine, rapidly reduced Cu2+ to Cu+. This may help to explain the antioxidant effect of cysteine as it may limit the amount of Cu2+ that is available to convert alpha-tocopherol in LDL into the prooxidant alpha-tocopherol radical. Cysteine (but not cystine) had a prooxidant effect, however, toward partially oxidised LDL in the presence of a low copper concentration, which may have been due to the rapid breakdown of lipid hydroperoxides in partially oxidised LDL by Cu+ generated by cysteine. To prove that cysteine can cause the rapid breakdown of lipid hydroperoxides in LDL, we enriched LDL with lipid hydroperoxides using an azo initiator in the absence of copper. Cysteine, but not cystine, increased the rate of lipid hydroperoxide decomposition to thiobarbituric acid-reactive substances (TBARS) in the presence of copper. (C) 2003 Elsevier Ireland Ltd.
