During early vertebrate development, the correct establishment of the body axes is critical. The anterior pole of the mouse embryo is established when Distal Visceral Endoderm (DVE) cells migrate to form the Anterior Visceral Endoderm (AVE). Symmetrical expression of Lefty1, Cer1 and Dkk1 determines the direction of DVE migration and the future anterior side. In addition to the establishment of the Anterior-Posterior axis, the AVE has also been implicated in anterior neural specification. To better understand the role of the AVE in these processes, we have performed a differential screening using Affymetrix GeneChip technology with AVE cells isolated from cer1P-EGFP transgenic mouse embryos. We found 175 genes which were upregulated in the AVE and 36 genes in the Proximal-posterior sample. Using DAVID software, we characterized the AVE cell population regarding cellular component, molecular function and biological processes. Among the genes that were found to be upregulated in the AVE, several novel genes were identified. Four of these transcripts displaying high-fold change in the AVE were further characterized by in situ hybridization in early stages of development in order to validate the screening. From those four selected genes, one, denominated Adtk1, was chosen to be functionally characterized by targeted inactivation in ES cells. Adtk1 encodes for a serine/threonine kinase. Adtk1 null mutants are smaller and present short limbs due to decreased mineralization, suggesting a potential role in chondrogenesis during limb development. Taken together, these data point to the importance of reporting novel genes present in the AVE.

Anti-signal recognition particle (SRP) myopathy is a rare idiopathic inflammatory myositis that usually affects middle-age women, and is characterized by rapidly progressive proximal and symmetrical muscle weakness, elevated creatine kinase levels, severe necrotizing immune-mediated myopathy, presence of anti-SRP autoantibodies and poor response to steroid therapy. We report a geriatric case of a previously independent patient, presenting with slow onset of proximal paraparesis, myalgia and severe gait impairment. The patient was treated with steroid and azathioprine, with laboratory and pain response but modest muscle strength improvement. The clinical presentation of this unusual patient was atypical, which hampered the correct diagnosis.

Boloceroides spp. are looking like an untidy mop, this anemone is sometimes seen in sea grass areas on many of our shores. It is possibly seasonal. Sometimes, large numbers are seen (up to 10-20 animals in a trip) and then none at all.  Tiny swimming anemones may sometimes be confused with Sea grass anemones which have translucent tentacles with tiny spots. The swimming anemone harbors symbiotic single-celled algae (zooxanthellae). The algae undergo photosynthesis to produce food from sunlight. The food produced is shared with the sea anemone, which in return provides the algae with shelter and minerals. The oral disk and tentacle muscles are used to obtain, retain, and ingest prey; in Boloceroides spp. tentacles can autotomize if it is needed to evade a predator. Tentacles can control body form by use of their endodermal muscles. Retractors are longitudinal muscles that will aid in withdrawing tentacles and the oral disk if they are exposed to the open air.  This hypothesis is furthered because in comparison to other sea anemones, Boloceroides is loosely attached to its respective substrate, thus allowing the pedal disk to detach quickly resulting in a rapid swimming response. Boloceroides can reproduce both sexually and asexually. As Anthozoans, Boloceroides produce sexually by bypassing the medusa life cycle stage; this allows Boloceroides (and all Anthozoans) to release their egg and sperm creating planula a bilaterally symmetrical, flattened, ciliated, motile larva.

The present report refers to research developed in the context of Supervised Teaching Practice in Pre-school Education and in Primary School, integrated in Master’s degree in Pre-school Education and Teaching Primary School at University of Évora. The research was conducted in context of a pre-school room and in context of 1st cycle of basic education, in a 2nd class year. The two contexts are part of the Manuel Ferreira Patrício School, headquarters of cluster of Évora schools. The respective research has aimed understand, analyze and reflect about reflection symmetry exploration, in Pre-school and in Primary School. Thus, was intented to answer the questions: “How childrens deal with the identification and exploration of reflection symmetry?" And "What practices should be developed to contribute to the understanding and use of reflection symmetry by childrens?". During the research it was developed a didactic intervention, in each context, consisting of sequences of symmetry exploration tasks that featured by resorting to diverse materials and the computer with the SIMIS program in the Primary School, with open tasks exploratory. Data collection builds on the training books on STP, the flat patterns, in productions of children, in response tasks and excerpts from the additions accruing dialogues. This research has shown that it is possible to explore the symmetry in Preschool and Primary School, since children recognized figures with symmetry, they were able to identify lines of symmetry in figures and built symmetrical figures. For such practices have developed that have contributed to the understanding and use of symmetry, through exploratory teaching and learning strategies of mathematics, using manipulatives and specific software, which proved to be essential tools in the exploration tasks and identification symmetry.

The evaluation of the mesh opening stiffness of fishing nets is an important issue in assessing the selectivity of trawls. It appeared that a larger bending rigidity of twines decreases the mesh opening and could reduce the escapement of fish. Nevertheless, netting structure is complex. A netting is made up of braided twines made of polyethylene or polyamide. These twines are tied with non-symmetrical knots. Thus, these assemblies develop contact-friction interactions. Moreover, the netting can be subject to large deformation. In this study, we investigate the responses of netting samples to different types of solicitations. Samples are loaded and unloaded with creep and relaxation stages, with different boundary conditions. Then, two models have been developed: an analytical model and a finite element model. The last one was used to assess, with an inverse identification algorithm, the bending stiffness of twines. In this paper, experimental results and a model for netting structures made up of braided twines are presented. During dry forming of a composite, for example, the matrix is not present or not active, and relative sliding can occur between constitutive fibres. So an accurate modelling of the mechanical behaviour of fibrous material is necessary. This study offers experimental data which could permit to improve current models of contact-friction interactions [4], to validate models for large deformation analysis of fibrous materials [1] on a new experimental case, then to improve the evaluation of the mesh opening stiffness of a fishing net

This thesis describes a study of various methods to produce bioactive peptides. Initially, the generation of anti-Cronobacter spp. peptides by fermentation of milk protein is described. Lactobacillus johnsonii DPC6026 was used to generate two previously described antimicrobial peptides. Phenotypic analysis indicated unsatisfactory casein hydrolysis. The genome of the strain was sequenced and annotated. Results showed a number of unique features present, most notably a large symmetrical inversion of approximately 750kb in comparison with the human isolate L. johnsonii NCC 533. The data suggest significant genetic diversity and intra-species genomic rearrangements within the L. johnsonii spp.. Cronobacter spp. have emerged as pathogens of concern to the powdered infant formula industry. Chapters 3 and 4 of this thesis describe novel methods to generate two antimicrobial peptides, Caseicin A and B. In Chapter 3 a bank of Bacillus strains was generated and investigated for caseicin production. Following casein hydrolysis by specific B. cereus and B. thuringiensis strains the peptides of interest were generated. Chapter 4 describes a sterile enzymatic method to generate peptides from casein. Bioinformatic tools were used to predict enzymes capable of liberating caseicin peptides from casein. Hydrolysates were generated using suitable enzymes, examined and some were found to produce peptides with activity against Cronobacter spp.. This study establishes a potential industrial-grade method to generate antimicrobial peptides. Administration of GLP-1 leads to improved glycaemic control in diabetes patients. Generation of a recombinant lactic acid bacteria capable of producing a GLP-1 analogue is described in Chapter 5. In-vivo analysis confirmed insulinotropic activity. The results illustrate a method using bacteriocin producing cellular machinery to generate bioactive peptides. This thesis describes the generation of bioactive peptides by bacterial fermentation, tailored enzymatic hydrolysis and recombinant bacterial methods. The techniques described contribute to bioactive peptide research with regards novel methods of production and industrial scale-up.

Several definitions exist that offer to identify the boundaries between languages and dialects, yet these distinctions are inconsistent and are often as political as they are linguistic (Chambers & Trudgill, 1998). A different perspective is offered in this thesis, by investigating how closely related linguistic varieties are represented in the brain and whether they engender similar cognitive effects as is often reported for bilingual speakers of recognised independent languages, based on the principles of Green’s (1998) model of bilingual language control. Study 1 investigated whether bidialectal speakers exhibit similar benefits in non-linguistic inhibitory control as a result of the maintenance and use of two dialects, as has been proposed for bilinguals who regularly employ inhibitory control mechanisms, in order to suppress one language while speaking the other. The results revealed virtually identical performance across all monolingual, bidialectal and bilingual participant groups, thereby not just failing to find a cognitive control advantage in bidialectal speakers over monodialectals/monolinguals, but also in bilinguals; adding to a growing body of evidence which challenges this bilingual advantage in non-linguistic inhibitory control. Study 2 investigated the cognitive representation of dialects using an adaptation of a Language Switching Paradigm to determine if the effort required to switch between dialects is similar to the effort required to switch between languages. The results closely replicated what is typically shown for bilinguals: Bidialectal speakers exhibited a symmetrical switch cost like balanced bilinguals while monodialectal speakers, who were taught to use the dialect words before the experiment, showed the asymmetrical switch cost typically displayed by second language learners. These findings augment Green’s (1998) model by suggesting that words from different dialects are also tagged in the mental lexicon, just like words from different languages, and as a consequence, it takes cognitive effort to switch between these mental settings. Study 3 explored an additional explanation for language switching costs by investigating whether changes in articulatory settings when switching between different linguistic varieties could - at least in part – be responsible for these previously reported switching costs. Using a paradigm which required participants to switch between using different articulatory settings, e.g. glottal stops/aspirated /t/ and whispers/normal phonation, the results also demonstrated the presence of switch costs, suggesting that switching between linguistic varieties has a motor task-switching component which is independent of representations in the mental lexicon. Finally, Study 4 investigated how much exposure is needed to be able to distinguish between different varieties using two novel language categorisation tasks which compared German vs Russian cognates, and Standard Scottish English vs Dundonian Scots cognates. The results showed that even a small amount of exposure (i.e. a couple of days’ worth) is required to enable listeners to distinguish between different languages, dialects or accents based on general phonetic and phonological characteristics, suggesting that the general sound template of a language variety can be represented before exact lexical representations have been formed. Overall, these results show that bidialectal use of typologically closely related linguistic varieties employs similar cognitive mechanisms as bilingual language use. This thesis is the first to explore the cognitive representations and mechanisms that underpin the use of typologically closely related varieties. It offers a few novel insights and serves as the starting point for a research agenda that can yield a more fine-grained understanding of the cognitive mechanisms that may operate when speakers use closely related varieties. In doing so, it urges caution when making assumptions about differences in the mechanisms used by individuals commonly categorised as monolinguals, to avoid potentially confounding any comparisons made with bilinguals.
