Asian soybean rust (ASR), caused by the fungus Phakopsora pachyrhizi, is one of the most economically important crop diseases, but is only treatable with fungicides, which are becoming less effective owing to the emergence of fungicide resistance. There are no commercial soybean cultivars with durable resistance to P. pachyrhizi, and although soybean resistance loci have been mapped, no resistance genes have been cloned. We report the cloning of a P. pachyrhizi resistance gene CcRpp1 (Cajanus cajan Resistance against Phakopsora pachyrhizi 1) from pigeonpea (Cajanus cajan) and show that CcRpp1 confers full resistance to P. pachyrhizi in soybean. Our findings show that legume species related to soybean such as pigeonpea, cowpea, common bean and others could provide a valuable and diverse pool of resistance traits for crop improvement.

TEM-1 is the dominant β-lactamase of Haemophilus influenzae and can be located on small plasmids. Three distinct plasmids with sizes from 4,304 to 5,646 nucleotides (nt) were characterized: pA1606, pA1209, and pPN223. In addition to TEM-1 and a replication enzyme of the Rep 3 superfamily, pA1606 carries a Tn3 resolvase gene and pA1606 and pA1209 carry an open reading frame (ORF) similar to a plasmid recombination enzyme gene described in Gram-positive bacteria. The plasmids transformed strain Rd to the ampicillin-resistant phenotype.

Haemophilus parasuis, the causative agent of Glässer's disease, is one of the early colonizers of the nasal mucosa of piglets. It is prevalent in swine herds, and lesions associated with disease are fibrinous polyserositis and bronchopneumonia. Antibiotics are commonly used in disease control, and resistance to several antibiotics has been described in H. parasuis. Prediction of H. parasuis virulence is currently limited by our scarce understanding of its pathogenicity. Some genes have been associated with H. parasuis virulence, such as lsgB and group 1 vtaA, while biofilm growth has been associated with nonvirulent strains. In this study, 86 H. parasuis nasal isolates from farms that had not had a case of disease for more than 10 years were obtained by sampling piglets at weaning. Isolates were studied by enterobacterial repetitive intergenic consensus PCR and determination of the presence of lsgB and group 1 vtaA, biofilm formation, inflammatory cell response, and resistance to antibiotics. As part of the diversity encountered, a novel 2,661-bp plasmid, named pJMA-1, bearing the blaROB-1 β-lactamase was detected in eight colonizing strains. pJMA-1 was shown to share a backbone with other small plasmids described in the Pasteurellaceae, to be 100% stable, and to have a lower biological cost than the previously described plasmid pB1000. pJMA-1 was also found in nine H. parasuis nasal strains from a separate collection, but it was not detected in isolates from the lesions of animals with Glässer's disease or in nontypeable Haemophilus influenzae isolates. Altogether, we show that commensal H. parasuis isolates represent a reservoir of β-lactam resistance genes which can be transferred to pathogens or other bacteria.

The objective of this work was to determine the genotypic profile specific to scrapie in codons 136, 154, and 171 of the PRNP gene of the Pantanal creole sheep. Genomic DNA was extracted from blood samples collected from 66 sheep, and the regions of interest on the DNA strand were amplified by PCR. Five haplotypes were identified: ARR, alanine, arginine, arginine; ARQ, alanine, arginine, glutamine; AHQ, alanine, histidine, glutamine; ARH, alanine, arginine, histidine; and VRQ, valine, arginine, glutamine. The most common genotypes were ARQ/ARQ (27%) and ARR/ARQ (24%). The genotypic profile of the Pantanal creole sheep shows low to moderate susceptibility.

The continuous soybean-maize crop succession in the tropical region of Brazil has led to significant increases in the population size of root-knot (Meloidogyne incognita and M. javanica ) and root-lesion nematodes (Pratylenchus brachyurus), which make soils unsuitable for soybean cropping. A greenhouse study was conducted to identify sunflower genotypes adapted to the tropical region of Brazil and that are resistant to M. incognita, M. javanica and/or P. brachyurus . Two experiments for each nematode were conducted in a completely randomized design with six replicates. Gall index was calculated from visual scores (0?5) of gall intensity on roots for the root-knot nematode. Initial and final population density and reproduction factor were also measured for each nematode. Sunflower genotypes varied in resistance to the nematodes. Sunflower hybrids BRS 321 and BRS 323 were resistant to M. javanica and P. brachyurus and exhibited low gall index for M. incognita . The cultivars are good alternatives to using in the succession of soybean in nematode-infested areas of the tropical regions of Brazil. No sunflower genotype was identified as resistant to M. incognita and thus sunflower cropping is not indicated in areas infested with this nematode.

O uso de cultivares resistentes é a principal medida para o manejo da antracnose do colmo em milho. Neste trabalho, objetivou-se identificar linhagens com níveis de resistência à antracnose do colmo, similar ao híbrido 2B710, considerado resistente. Dois experimentos foram conduzidos na Embrapa Milho e Sorgo. No primeiro experimento, foram avaliados 234 linhagens e os híbridos BRS1010 (suscetível) e 2B710 (resistente). Foi realizada inoculação artificial com um isolado de C. graminicola, na fase de pré-pendoamento e, após 30 dias, foi realizada a avaliação da severidade da antracnose no colmo. O segundo experimento foi conduzido com 48 linhagens e os híbridos inoculados com dois isolados de C. graminicola. No primeiro experimento, os genótipos formaram oito grupos com base na severidade da doença e as linhagens do último grupo foram consideradas as mais resistentes, incluindo o híbrido 2B710, em que os genótipos apresentaram valores de severidade entre 11,50 a 23%. No segundo experimento, houve interação entre os fatores linhagens e isolados e, de modo geral, as linhagens apresentaram a mesma tendência de reação obtida no primeiro experimento, no entanto, a severidade da doença foi maior para a maioria das linhagens, mesmo quando utilizado o outro isolado. Com isso, foi possível realizar a seleção de linhagens com bons níveis de resistência, as quais podem ser utilizadas em programas de melhoramento, em estudos de herança, desenvolvimento de híbridos e identificação de marcadores moleculares, associados com resistência à antracnose do colmo.

From the 70's, Africa was a massive wave of independence process. However, such processes are not meant processes of decolonization. According with Fanon (1961) also decolonization is based not only on the territorial and political, but mostly mental. While some former colonies resist colonial hegemony still visible in the local culture, others are colluding with the colonizing process. Thus, it will analyze the subservience and resistance to colonization in Lusophone African poetry, especially with the Angolan Viriato da Cruz, with the poem MaKézú (1961) and Cape Verdian Jorge Barbosa with Prelude (1956).

The development of resistance to anthelmintics has prompted research into alternative methods of controlling intestinal nematodes in ruminants. This study aimed to assess the activity of Ananas comosus on Haemonchus contortus in Santa Inês sheep. The aqueous extract of pineapple skin (AEPS), bromelain from pineapple stems (B4882) and residue from pineapple processing was evaluated in in vitro and in vivo tests. The enzymatic activity of substances was analyzed by the azocasein method. The egg hatch test (EHT) and larval development test (LDT) were performed using the Embrapa2010 isolate of H. contortus. In the in vivo test, 36 sheep artificially infected with H. contortus were divided into six groups: G1: 2g/kg BW of the aqueous extract administered for three days; G2: 2g/kg BW of the industrial pineapple residue for 60 days; G3: 180mg/animal of bromelain in a single dose; G4: negative control I; G5: positive control (levamisole phosphate); and G6: negative control II. The eggs per gram (EPG) in the feces were counted till 28 days after treatment. LC50 and LC90 were obtained by the probit procedure, while the in vivo test results were analyzed by GLM. The aqueous extract in the in vitro and in vivo test, the bromelain and industrial residue presented 0.102, 0.157, 1.864 and 0.048 enzyme units/mL, respectively. In the egg hatch test, the LC50 and LC90 were respectively 31 and 81mg/mL for the aqueous extract and 0.50 and 2mg/mL for bromelain. In the larval development test, the LC50 and LC90 were respectively 1.7 and 7.3mg/mL for the aqueous extract and 0.019 and 0.086mg/mL for bromelain. In the in vivo test, the general efficacies of the treatments in relation to the negative control were 22.6%, 42.2%, 3.65% and 89% for the aqueous extract, industrial pineapple residue, bromelain and positive control respectively. The transformed EPG values were 3.19±0.59, 3.32±0.25, 2.85±0.66, 3.44±0.50, 2.28±0.93 and 2.75±0.94 for the aqueous extract, industrial residue, bromelain, negative control I, positive control and negative control II respectively. The results for all the treated groups differed significantly (p<0.05) from the positive control, and although the residue presented efficacy of 42.2%, there was no statistical difference (p>0.05) in relation to the negative control. Therefore, both the aqueous extract and bromelain were effective in vitro, but showed reduced anthelmintic efficacy in vivo. For the pineapple residue, the 42.2% in vivo efficacy in reducing the EPG and the possibility of reducing environmental contamination through reuse of industrial residue indicate it can also be useful for control of this parasite. © 2013 Elsevier B.V.
