This paper describes new advances in the exploitation of oxygen A-band measurements from POLDER3 sensor onboard PARASOL, satellite platform within the A-Train. These developments result from not only an account of the dependence of POLDER oxygen parameters to cloud optical thickness τ and to the scene's geometrical conditions but also, and more importantly, from the finer understanding of the sensitivity of these parameters to cloud vertical extent. This sensitivity is made possible thanks to the multidirectional character of POLDER measurements. In the case of monolayer clouds that represent most of cloudy conditions, new oxygen parameters are obtained and calibrated from POLDER3 data colocalized with the measurements of the two active sensors of the A-Train: CALIOP/CALIPSO and CPR/CloudSat. From a parameterization that is (μs, τ) dependent, with μs the cosine of the solar zenith angle, a cloud top oxygen pressure (CTOP) and a cloud middle oxygen pressure (CMOP) are obtained, which are estimates of actual cloud top and middle pressures (CTP and CMP). Performances of CTOP and CMOP are presented by class of clouds following the ISCCP classification. In 2008, the coefficient of the correlation between CMOP and CMP is 0.81 for cirrostratus, 0.79 for stratocumulus, 0.75 for deep convective clouds. The coefficient of the correlation between CTOP and CTP is 0.75, 0.73, and 0.79 for the same cloud types. The score obtained by CTOP, defined as the confidence in the retrieval for a particular range of inferred value and for a given error, is higher than the one of MODIS CTP estimate. Scores of CTOP are the highest for bin value of CTP superior in numbers. For liquid (ice) clouds and an error of 30 hPa (50 hPa), the score of CTOP reaches 50% (70%). From the difference between CTOP and CMOP, a first estimate of the cloud vertical extent h is possible. A second estimate of h comes from the correlation between the angular standard deviation of POLDER oxygen pressure σPO2 and the cloud vertical extent. This correlation is studied in detail in the case of liquid clouds. It is shown to be spatially and temporally robust, except for clouds above land during winter months. The analysis of the correlation's dependence on the scene's characteristics leads to a parameterization providing h from σPO2. For liquid water clouds above ocean in 2008, the mean difference between the actual cloud vertical extent and the one retrieved from σPO2 (from the pressure difference) is 5 m (−12 m). The standard deviation of the mean difference is close to 1000 m for the two methods. POLDER estimates of the cloud geometrical thickness obtain a global score of 50% confidence for a relative error of 20% (40%) of the estimate for ice (liquid) clouds over ocean. These results need to be validated outside of the CALIPSO/CloudSat track.

A quality assessment of the CFC-11 (CCl3F), CFC-12 (CCl2F2), HF, and SF6 products from limb-viewing satellite instruments is provided by means of a detailed intercomparison. The climatologies in the form of monthly zonal mean time series are obtained from HALOE, MIPAS, ACE-FTS, and HIRDLS within the time period 1991–2010. The intercomparisons focus on the mean biases of the monthly and annual zonal mean fields and aim to identify their vertical, latitudinal and temporal structure. The CFC evaluations (based on MIPAS, ACE-FTS and HIRDLS) reveal that the uncertainty in our knowledge of the atmospheric CFC-11 and CFC-12 mean state, as given by satellite data sets, is smallest in the tropics and mid-latitudes at altitudes below 50 and 20 hPa, respectively, with a 1σ multi-instrument spread of up to ±5 %. For HF, the situation is reversed. The two available data sets (HALOE and ACE-FTS) agree well above 100 hPa, with a spread in this region of ±5 to ±10 %, while at altitudes below 100 hPa the HF annual mean state is less well known, with a spread ±30 % and larger. The atmospheric SF6 annual mean states derived from two satellite data sets (MIPAS and ACE-FTS) show only very small differences with a spread of less than ±5 % and often below ±2.5 %. While the overall agreement among the climatological data sets is very good for large parts of the upper troposphere and lower stratosphere (CFCs, SF6) or middle stratosphere (HF), individual discrepancies have been identified. Pronounced deviations between the instrument climatologies exist for particular atmospheric regions which differ from gas to gas. Notable features are differently shaped isopleths in the subtropics, deviations in the vertical gradients in the lower stratosphere and in the meridional gradients in the upper troposphere, and inconsistencies in the seasonal cycle. Additionally, long-term drifts between the instruments have been identified for the CFC-11 and CFC-12 time series. The evaluations as a whole provide guidance on what data sets are the most reliable for applications such as studies of atmospheric transport and variability, model–measurement comparisons and detection of long-term trends. Cutoff lows (COLs) pressure systems climatology for the Southern Hemisphere (SH), between 10 degrees S and 50 degrees S, using the National Center for Environmental Prediction-National Center for Atmospheric Research (NCEP-NCAR) and the ERA-40 European Centre for Medium Range Weather Forecast (ECMWF) reanalyses are analyzed for the period 1979-1999. COLs were identified at three pressure levels (200, 300, and 500 hPa) using an objective method that considers the main physical characteristics of the conceptual model of COLs. Independently of the pressure level analyzed, the climatology from the ERA-40 reanalysis has more COLs systems than the NCEP-NCAR. However, both reanalyses present a large frequency of COLs at 300 hPa, followed by 500 and 200 hPa. The seasonality of COLs differs at each pressure level, but it is similar between the reanalyses. COLs are more frequent during summer, autumn, and winter at 200, 300, and 500 hPa, respectively. At these levels, they tend to occur around the continents, preferentially from southeastern Australia to New Zealand, the south of South America, and the south of Africa. To study the COLs at 200 and 300 hPa from a regional perspective, the SH was divided in three regions: Australia-New Zealand (60 E-130 W), South America (130 degrees W-20 degrees W), and southern Africa (20 degrees W-60 degrees E). The common COLs features in these sectors for both reanalyses are a short lifetime (similar to 80.0% and similar to 70.0% of COLs at 200 and 300 hPa, respectively, persisting for up to 3 days), mobility (similar to 70.0% and similar to 50% of COLs at 200 and 300 hPa, respectively, traveling distances of up to 1200 km), and an eastward propagation.

The impact of the inter-El Nio (EN) variability on the moisture availability over Southeastern South America (SESA) is investigated. Also, an automatic tracking scheme was used to analyze the extratropical cyclones properties (system density - SD and central pressure - CP) in this region. During the austral summer period from 1977-2000, the differences for the upper-level wave train anomaly composites seem to determine the rainfall composite differences. In fact, the positive rainfall anomalies over most of the SESA domain during the strong EN events are explained by an upper-level cyclonic center over the tropics and an anticyclonic center over the eastern subtropical area. This pattern seems to contribute to upward vertical motion at 500 hPa and reinforcement of the meridional moisture transport from the equatorial Atlantic Ocean and western Amazon basin to the SESA region. These features may contribute to the positive SD and negative CP anomalies explaining part of the positive rainfall anomalies found there. On the other hand, negative rainfall anomalies are located in the northern part of SESA for the weak EN years when compared to those for the strong events. Also, positive anomalies are found in the southern part, albeit less intense. It was associated with the weakening of the meridional moisture transport from the tropics to the SESA that seems have to contributed with smaller SD and CP anomalies over the most part of subtropics, when compared to the strong EN years.

This work presents an analysis of a lowermost stratospheric air intrusion event over the coast of Brazil, which may have been responsible for a secondary surface cyclogenesis over the southwestern Atlantic Ocean. The surface cyclone initiated at 0600 UTC 17 April 1999 in a cold air mass in the rear of a cold front after a primary cyclone developed over the same region. The analysis of the secondary cyclone revealed the presence of lowermost stratospheric air intrusion characterized by anomalous potential vorticity (PV), dry air, and high concentration of ozone in atmospheric column. The system developed on the eastern side of an upper level core of PV anomaly, which induced a cyclonic wind circulation at lower levels and favored the onset of the secondary cyclone. In midlevels (500 hPa), the cutoff low development contributed to reduce the propagation speed of the wave pattern. This feature seemed to (1) allow the low-level cold/dry air to heat/moisten associated with sensible and latent fluxes transferred from the ocean to the atmosphere, which intensified a baroclinic zone parallel to the coast, and (2) contribute to the long duration of the system. The present analysis indicates that this secondary cyclone development could be the result of the coupling between the PV anomaly in the upper levels and low-level air-sea interaction.

Intraseasonal and interannual variability of extreme wet and dry anomalies over southeastern Brazil and the western subtropical South Atlantic Ocean are investigated. Precipitation data are obtained from the Global Precipitation Climatology Project (GPCP) in pentads during 23 austral summers (December-February 1979/80-2001/02). Extreme wet (dry) events are defined according to 75th (25th) percentiles of precipitation anomaly distributions observed in two time scales: intraseasonal and interannual. The agreement between the 25th and 75th percentiles of the GPCP precipitation and gridded precipitation obtained from stations in Brazil is also examined. Variations of extreme wet and dry anomalies on interannual time scales are investigated along with variations of sea surface temperature (SST) and circulation anomalies. The South Atlantic SST dipole seems related to interannual variations of extreme precipitation events over southeastern Brazil. It is shown that extreme wet and dry events in the continental portion of the South Atlantic convergence zone (SACZ) are decoupled from extremes over the oceanic portion of the SACZ and there is no coherent dipole of extreme precipitation regimes between tropics and subtropics on interannual time scales. On intraseasonal time scales, the occurrence of extreme dry and wet events depends on the propagation phase of extratropical wave trains and consequent intensification (weakening) of 200-hPa zonal winds. Extreme wet and dry events over southeastern Brazil and subtropical Atlantic are in phase on intraseasonal time scales. Extreme wet events over southeastern Brazil and subtropical Atlantic are observed in association with low-level northerly winds above the 75th percentile of the seasonal climatology over central-eastern South America. Extreme wet events on intraseasonal time scales over southeastern Brazil are more frequent during seasons not classified as extreme wet or dry on interannual time scales.

This paper examines moisture transport on intraseasonal timescales over the continent and over the South Atlantic convergence zone (SACZ) during the South America (SA) summer monsoon. Combined Empirical Orthogonal Function analysis (EOFc) of Global Precipitation Climatology Project pentad precipitation, specific humidity, air temperature, zonal and meridional winds at 850 hPa (NCEP/NCAR reanalysis) are performed to identify the large-scale variability of the South America monsoon system and the SACZ. The first EOFc was used as a large-scale index for the South American monsoon (LISAM), whereas the second EOFc characterized the SACZ. LISAM (SACZ) index showed spectral variance on 30-90 (15-20) days and were both band filtered (10-100 days). Intraseasonal wet anomalies were defined when LISAM and SACZ anomalies were above the 75th percentile of their respective distribution. LISAM and SACZ wet events were examined independently of each other and when they occur simultaneously. LISAM wet events were observed with the amplification of wave activity in the Northern Hemisphere and the enhancement of northwesterly cross-equatorial moisture transport over tropical continental SA. Enhanced SACZ was observed with moisture transport from the extratropics of the Southern Hemisphere. Simultaneous LISAM and SACZ wet events are associated with cross-equatorial moisture transport along with moisture transport from Subtropical Southwestern Atlantic.

The temporal organization of mammals presents a daily adjustment to the environmental light/dark cycle. The environmental light detected by the retina adjusts the central clock in the suprachiasmatic nuclei, which innervate the pineal gland through a polysynaptic pathway. During the night, this gland produces and releases the nocturnal hormone melatonin, which circulates throughout the whole body and adjusts several bodily functions according to the existence and duration of darkness. We have previously shown that during the time frame of an inflammatory response, pro-inflammatory cytokines, such as tumor necrosis factor-a, inhibit while anti-inflammatory mediators, such as glucocorticoids, enhance the synthesis of melatonin, interfering in the daily adjustment of the light/dark cycle. Therefore, injury disconnects the organism from environmental cycling, while recovery restores the light/dark information to the whole organism. Here, we extend these observations by evaluating the effect of a mild restraint stress, which did not induce macroscopic gastric lesions. After 2 h of restraint, there was an increase in circulating corticosterone, indicating activation of the hypothalamus-pituitary-adrenal (HPA) axis. In parallel, an increase in melatonin production was observed. Taking into account the data obtained with models of inflammation and stress, we reinforce the hypothesis that the activity of the pineal gland is modulated by the state of the immune system and the HPA axis, implicating the darkness hormone melatonin as a modulator of defense responses.

The effects of single or repeated amphetamine (AMPH) treatment and those of AMPH withdrawals on immune-mediated lung inflammatory response were studied in rats. Two experiments were done. In the first, rats egg-albumin (OVA) sensitized were singularly or repeatedly (21 days, once daily) treated with AMPH (1.0 mg/kg) or with a similar number and volume of 0.9% NaCl. The OVA aerosol challenge was performed 12 h after the single or last repeated AMPH treatment and also 72 and 120 h after AMPH withdrawal. In the second experiment, the effects of reserpine (1.0 mg/kg/day for 5 consecutive days) on single AMPH actions on lung allergic response of rats were analyzed. Single and repeated AMPH treatment induced opposite actions on Bronchoalveolar lavage fluid (BAL) cellularity of allergic rats: single treatment decreased and repeated treatment increased the total number of cells as well as those of macrophages, neutrophils and eosinophils. Our data also showed that single but not repeated AMPH treatment decreased the number of neutrophils, monocytes and lymphocytes in the peripheral blood, and increased the total number of bone marrow cells in rats sensitized and challenged with OVA. Furthermore, it was shown that reserpine treatment precluded the effects of single AMPH treatment on cellular migration to the lung of OVA-sensitized and challenged rats. It was concluded that AMPH effects on lung inflammatory response and cell recruitment to the lung in allergic rats rely at least partially on corticosterone serum levels. The possible involvement of vesicular monoamine transporter type 2 (VMAT2) with these observed effects was discussed. (c) 2008 Elsevier B.V. All rights reserved.

Let M -> B, N -> B be fibrations and f(1), f(2): M -> N be a pair of fibre-preserving maps. Using normal bordism techniques we define an invariant which is an obstruction to deforming the pair f(1), f(2) over B to a coincidence free pair of maps. In the special case where the two fibrations axe the same and one of the maps is the identity, a weak version of our omega-invariant turns out to equal Dold`s fixed point index of fibre-preserving maps. The concepts of Reidemeister classes and Nielsen coincidence classes over B are developed. As an illustration we compute e.g. the minimal number of coincidence components for all homotopy classes of maps between S(1)-bundles over S(1) as well as their Nielsen and Reidemeister numbers.

Depression is associated with glucocorticoid hypersecretion, due to dysfunction of the hypothalamo-pituitary-adrenocorticol axis (HPA-axis). Because excess glucocorticoids are associated with depressive-like features in humans, glucocorticoid receptor antagonists are currently being tested for antidepressant efficacy in clinical trials. In the current study the hypothesis that mifepristone (RU486), a glucocorticoid receptor antagonist, would decrease the neuroendocrine and central HPA-axis responses to an acute stressor and attentuate depressive like behavior in an animal model of behavioral helplessness (forced swim test) was tested. Adult male rats were treated with 10 mglkg RU486 (subcutaneous) for five days and then exposed to a IO-minute forced swim test (FST), conducted in Plexiglas cylinders. FST sessions were videotaped for later analysis of behavioral immobility. Plasma ACTH and corticosterone CORT were measured at 15min and 90min after FST cessation. Animals were perfused and brains were collected for immunocytochemical assessment of c-Fos expression in the medial prefrontal cortex (mPFC), a brain region implicated in both depression and central control of the HPA axis. RU486 significantly decreased peak ACTH and CORT concentrations following FST exposure. In addition, glucocorticoid negative feedback was at1enuated in RU486-treated animals exposed to the FST. Exposure to FST alone induced c-FOS expression in the mPFC, as measured by the number of c-Fos positive neurons. Treatment with RU486 significantly increased the number of rnPFC c-Fos positive cell following FST exposure. The behavioral data obtained from FST paradigm, demonstrated that RU486 decreased immobility in the FST illustrating the potential efficacy of this drug as an antidepressant. Collectively these data suggest that RU486 dampens HPA-axis responses to stress, possibly by enhancing the excitability of stress-inhibitory neurons in the mPFC. This is particularly exciting, given the fact that this neural region is associated with decreased neural activity during depression in humans.

A fenilcetonúria (PKU) é um erro inato do metabolismo de aminoácidos causada pela deficiência da enzima fenilalanina hidroxilase hepática (PAH) que converte fenilalanina (Phe) em tirosina. Caracteriza-se clinicamente por retardo mental severo e, em alguns pacientes, por convulsões e eczema cutâneo. Bioquimicamente os pacientes afetados por esta doença apresentam acúmulo de Phe e seus metabólitos no sangue e nos tecidos. Phe é considerada o principal agente neurotóxico nesta doença, cujos mecanismos de neurotoxicidade são pouco conhecidos. O metabolismo energético cerebral é caracterizado por níveis altos e variáveis de síntese e de utilização de ATP. O cérebro contém altos níveis de creatinaquinase (CK), uma enzima que transferere reversivelmente um grupo fosforil entre ATP e creatina e entre ADP e fosfocreatina (PCr). Considerando que a CK parece estar envolvida em certas condições patológicas relacionadas com deficiência de energia cerebral e sabendo que a PKU está associada à redução de produção e de utilização de energia pelo cérebro, no presente trabalho verificamos a atividade da CK em homogeneizado total de córtex cerebral, cerebelo e cérebro médio de ratos Wistar submetidos aos modelos experimentais agudo e crônico de hiperfenilalaninemia (HPA) quimicamente induzida. Também investigamos o efeito in vitro da Phe e da α-metil-DL-fenilalanina (MePhe), um inibidor da PAH, nas mesmas estruturas de ratos Wistar de 22 dias de idade não tratados. Nossos resultados mostraram uma redução significativa na atividade da CK nas estruturas cerebrais estudadas de ratos sujeitos a HPA. Também verificamos que Phe e MePhe inibiram in vitro a atividade da CK nas mesmas estruturas. O estudo da interação cinética entre Phe e MePhe, sugere a existência de um único sítio de ligação na CK para os dois compostos. Considerando a importância da CK para a manutenção do metabolismo energético cerebral, nossos resultados sugerem que a alteração da homeostasia energética pode contribuir para a neurotoxicidade da Phe na PKU.

A fenilcetonúria (PKU) é caracterizada bioquimicamente pelo acúmulo de fenilalanina (Phe) e seus metabólitos nos tecidos dos pacientes afetados. O dano neurológico é a marca da PKU e a Phe é considerada o principal agente neurotóxico nesta doença, cujos mecanismos de neurotoxicidade são pouco conhecidos. A alanina (Ala) é nutricionalmente um aminoácido não essencial. Ela é o principal aminoácido gliconeogênico porque pode originar piruvato e glicose, tendo sido, por este motivo, usada como um suplemento dietético em combinação com o hormônio de crescimento no tratamento de crianças subnutridas afetadas por algumas doenças metabólicas herdadas, para induzir o anabolismo. O principal objetivo do presente trabalho foi medir as atividades dos complexos da cadeia respiratória mitocondrial (CCR) e succinato desidrogenase (SDH) no córtex cerebral de ratos Wistar sujeitos à hiperfenilalaninemia (HPA) quimicamente induzida e à administração crônica de Ala, desde o 6± até o 21± dia de vida pós-natal. Também investigamos o efeito in vitro da Phe e Ala nas atividades da SDH e CCR em córtex cerebral de ratos de 22 dias de idade. Os resultados mostraram uma redução nas atividades da SDH e complexos I + III no córtex cerebral de ratos sujeitos à HPA e também no córtex cerebral de ratos sujeitos à administração de Ala. Também verificamos que ambos: Phe e Ala inibiram in vitro a atividade dos complexos I + III por competição com NADH. Considerando a importância da SDH e CCR para a sustentação do suprimento energético para o cérebro, nossos resultados sugerem que o déficit energético possa contribuir para a neurotoxicidade da Phe em PKU. Em relação à Ala, ficou evidenciado que mais investigações serão necessárias antes de considerar a suplementação com Ala como uma terapia adjuvante válida para crianças com estas doenças.

O objetivo geral deste trabalho é desenvolver estudos relacionados à dispersão de poluentes, considerando a queima de carvão para geração de energia nas Usinas Termoelétricas de Charqueadas e São Jerônimo. O período de estudo foi do dia 17 a 23 de junho de 2003. Neste período houve a passagem de um sistema frontal. Sendo possível avaliar a dispersão dos poluentes em condições pré-frontal, frontal e pós-frontal. Para simular o comportamento dos poluentes neste período, foi utilizada uma subrotina de dispersão acoplada ao modelo RAMS (Regional Atmospheric Modeling System). Os resultados mostraram que nos dias classificados como pré-frontal e pós-frontal as concentrações do material particulado, dióxido de enxofre e do óxido de nitrogênio, atingiram seus valores máximos, pelo fato da umidade relativa do ar estar bastante baixa em torno de 60%, pressão atmosférica da ordem de 1021 hPa e a intensidade dos ventos fraca. No dia classificado como frontal, as concentrações estavam praticamente nulas, devido à passagem do sistema frontal que causou a queda na pressão atmosférica, aumento da umidade relativa e também pelo fato da ocorrência de precipitação atmosférica neste dia. As comparações dos resultados simulados, com os dados observados na estação de qualidade do ar mostraram-se satisfatórios. Com exceção do dia 22 de junho, que apresentou uma diferença da ordem de 90%.

A manipulação neonatal é um modelo experimental utilizado para avaliar o modo pelo qual interferências precoces na vida do animal podem alterar funções neuroendócrinas e comportamentos na vida adulta. O procedimento de manipulação neonatal, além de alterar a atividade do eixo hipotálamo-hipófise-adrenal (eixo HPA) em ratos machos e fêmeas, pode causar profundas mudanças na função reprodutiva de ratas adultas. De fato, há evidências de que a manipulação neonatal diminui a atividade do eixo HPA através da redução da síntese e secreção de hormônios que são liberados quando os animais são expostos ao estresse na vida adulta e, além disso, induz à presença de ciclos anovulatórios e diminui a receptividade sexual de ratas. Considerando essas alterações induzidas pela manipulação neonatal que são relacionadas à função reprodutiva de ratas, essa tese teve por objetivo estudar as possíveis causas da alteração no comportamento sexual e ovulação induzidas pela manipulação neonatal. Para isto, além de estudar o perfil hormonal desses animais, o que incluiu os esteróides gonadais, as gonadotrofinas e a prolactina (PRL) em diferentes fases e horários do ciclo estral, o conteúdo do hormônio liberador de gonadotrofinas (LHRH) em algumas regiões do sistema nervoso central (SNC) e na eminência mediana (EM); foi avaliada a possível participação do sistema angiotensinérgico central, através da análise da densidade dos receptores de angiotensina II (Ang II) pela técnica de auto-radiografia, na mediação dos efeitos da manipulação neonatal sobre a função do eixo hipotálamo-hipófise-gônada (eixo HPG) e do eixo HPA. O presente estudo confirmou dados obtidos em nosso laboratório sobre a redução do comportamento sexual e da ovulação em ratas manipuladas no período neonatal. Na tarde do proestro, período no qual ocorrem os eventos necessários para a ovulação na próxima fase do ciclo estral, os resultados mostram que os animais do grupo manipulado têm redução significativa da concentração plasmática de estradiol, de gonadotrofinas e de PRL, assim como um aumento no conteúdo de LHRH na área pré-óptica medial (APOM). A manipulação neonatal também reduziu a concentração plasmática de progesterona analisada após o coito que pode ser decorrente da reduzida estimulação vaginocervical recebida por essas ratas, já que houve uma redução significativa da freqüência de intromissão realizada pelo macho sobre as ratas do grupo manipulado. A densidade de receptores de Ang II na APOM e no núcleo paraventricular do hipotálamo (PVN) também foi alterada pela manipulação neonatal, pois houve redução significativa na densidade desses receptores nessas duas regiões. Em conclusão, a manipulação neonatal reduz profundamente a atividade do eixo HPG e essa alteração é causada por modificações nas concentrações de estradiol no plasma que, por sua vez, pode alterar a secreção de LHRH, de gonadotrofinas e de PRL, comprometendo dessa maneira a ovulação e a receptividade sexual. Contribuindo para os efeitos deletérios da manipulação neonatal sobre a função reprodutiva em ratas, este procedimento pode alterar a implantação do blastocisto devido à redução da secreção de progesterona observada após o coito no grupo manipulado. Alguns dos efeitos da manipulação neonatal parecem ser mediados pelo sistema angiotensinérgico central, como o aumento do conteúdo de LHRH na APOM e a diminuída resposta do eixo HPA observada em animais manipulados submetidos a situações estressantes na vida adulta.
