La synthèse d’un ARNm eucaryotique dépend d’une suite d’étapes qui inclut notamment l’ajout d’une queue poly(A) à son extrémité 3’. Au noyau, la queue poly(A) des ARNms est liée par PABPN1 (poly(A)-binding protein nuclear 1). PABPN1 fut notamment caractérisée, d’après des études in vitro, pour stimuler la réaction de polyadénylation en plus de contrôler la taille ultime des queues poly(A). Cela dit, la ou les fonction(s) biologique(s) de PABPN1 est/sont cependant largement méconnue(s). Chez Schizosaccharomyces pombe (S. pombe), Pab2 est l’orthologue présumé de PABPN1. Or, mes travaux indiquent que Pab2 est fonctionnellement différente de PABPN1 à l’égard de son rôle sur le processus général de polyadénylation. Ainsi, in vivo, l’absence de Pab2 entraîne l’expression et l’accumulation d’un groupe limité d’ARNs hyperadénylés parmi lesquels se trouvent de nombreux petits ARNs nucléolaires non-codants (snoRNAs) lesquels constituent normalement un groupe abondant d’ARN poly(A)-. Mes résultats supportent ainsi un mécanisme par lequel des snoRNAs immatures poly(A)+, sont convertis en une forme mature poly(A)- par le biais de Pab2 et de l’activité 3’-->5’ exoribonucléase de l’exosome à ARN. Ces observations sont inusitées dans la mesure où elles associent une fonction pour une PABP dans la maturation d'ARNs non-codants, contrairement à la notion que les PABPs travaillent exclusivement au niveau des ARNms, en plus de procurer une nouvelle perspective face au mécanisme de recrutement de l'exosome à ARN à des substrats poly(A)+. La formation de l’extrémité 3’ d’un ARN est un processus étroitement lié à la terminaison de sa transcription. Pour les gènes codants, la terminaison transcriptionnelle est initiée par le clivage endonucléolytique du pré-ARNm. Ce clivage génère une extrémité d’ARN 5’ libre laquelle sera ciblée par une exoribonucléase 5'-->3’ afin de mener à bien l’éviction de l’ARNPII de la matrice d’ADN (terminaison transcriptionnelle de type torpedo). Au contraire, chez Saccharomyces cerevisiae (S. cerevisiae), la majorité des gènes non-codants, incluant les snoRNAs, dépendent plutôt du complexe NNS (Nrd1/Nab3/Sen1) pour la terminaison de leur transcription. Cela dit, il est incertain si le complexe NNS est conservé chez d’autres espèces. À cet égard, mes travaux indiquent que S. pombe est dépourvu d’un mécanisme de terminaison de la transcription de type NNS. Seb1, l’orthologue présumé de Nrd1 chez S. pombe, s’associe plutôt à la machinerie de clivage et de polyadénylation et influence la sélection de site de polyadénylation à l’échelle du génome. Mes résultats supportent ainsi l’utilisation de la machinerie de maturation 3’ des ARNms comme principal vecteur de terminaison transcriptionnelle chez S. pombe et identifient Seb1 comme un facteur clé de ce processus. L’évènement transcriptionnel étant hautement complexe, des erreurs peuvent arriver de manière stochastique menant à l’accumulation d’ARNs aberrants potentiellement néfastes pour la cellule. Or, mes travaux ont mis en lumière un mécanisme de surveillance co-transcriptionnel des ARNs impliquant l’exosome à ARN et lié à la terminaison de la transcription. Pour ce faire, l’exosome à ARN promeut la terminaison transcriptionnelle via la dégradation d’une extrémité 3’ libre d’ARN devenue émergente suite au recul de l’ARNPII le long de la matrice d’ADN (phénomène de backtracking). Mes résultats supportent ainsi une terminaison de la transcription de type torpedo inversé (3'-->5’) réévaluant par la même occasion le concept voulant que la terminaison de la transcription s’effectue uniquement selon une orientation 5’-->3’. Somme toute, mes travaux de doctorat auront permis d’identifier et de caractériser plus en détail les facteurs et mécanismes impliqués dans la maturation 3’ et la terminaison de la transcription des gènes codants et non-codants chez l’organisme modèle S. pombe.

Este artículo presenta los resultados de una investigación realizada al interior de dos contextos. Por un lado, el teórico, en el marco de uno de los discursos más relevantes en los campos de la estrategia organizacional, de la managerial and organizational cognition (MOC) y, en general, de los estudios organizacionales (organization studies): la construcción de sentido (sensemaking). Por el otro, el empírico, en una de las grandes compañías multinacionales del sector automotriz con presencia global. Esta corporación enfrenta una permanente tensión entre lo que dicta la casa matriz, en relación con el cumplimiento de metas y estándares específicos, considerando el mundo entero, y los retos que, teniendo en cuenta lo regional y lo local, experimentan los altos directivos encargados de hacer prosperar la empresa en estos lugares. La aproximación implementada fue cualitativa. Esto en atención a la naturaleza de la problemática abordada y la tradición del campo. Los resultados permiten ampliar el actual nivel de comprensión acerca de los procesos de sensemaking de los altos directivos al enfrentar un entorno estratégico turbulento.

Off-site Manufacture (OSM) has long been recognised, both in Australia and internationally, as offering numerous benefits to all parties in the construction process. More importantly, it is recognised as a key vehicle for driving improvement within the construction industry. The uptake of OSM in construction is however limited, despite well documented benefits. The research aims to determine the ‘state-of-the-art’ of OSM in Australia. It confirms the benefits and identifies the real and perceived barriers to the widespread adoption of OSM. Further the project identifies opportunities for future investment and research. Although numerous reports have been produced in the UK on the state of OSM adoption within that region, no prominent studies exist for the Australian context. This scoping study is an essential component upon which to build any initiatives that can take advantage of the benefits of OSM in construction. The Construction 2020 report predicted that OSM is set to increase in use over the next 5-15 years, further justifying the need for such a study. The long-term goal of this study is to contribute to the improvement of the Australian construction industry through a realisation of the potential benefits of OSM.

Off-site manufacture (OSM) offers numerous benefits to all parties in the construction process. The uptake of OSM in Australia has, however, been limited. This limited uptake corresponds to similar trends in the UK and US, although the level of OSM there appears to be increasing. This project undertook three workshops — one each in Victoria, Queensland and Western Australia — and 18 interviews with key stakeholders to assist in identifying the general benefits and barriers to OSM uptake in the Australian construction industry. Seven case studies were also undertaken, involving construction projects that used OSM, ranging from civil projects through to residential. Each of these case studies has been analysed to identify what worked and what didn’t, and suggest the lessons to be learned from each project.

Much has been written on Off-site Manufacture (OSM) in construction, particularly regarding the perceived benefits and barriers to implementation. However, very little understanding of the state of OSM in the Australian construction industry exists. A ‘scoping study' has recently been undertaken to determine the ‘state-of-the-art’ of OSM in Australia. This involved several industry workshops, interviews and case studies across four major states of Australia. The study surveyed a range of suppliers across the construction supply-chain, incorporating the civil, commercial and housing segments of the market. This revealed that skills shortages and lack of adequate OSM knowledge are generally the greatest issues facing OSM in Australia. The drivers and constraints that emerged from the research were, in large measure, consistent with those found in the US and UK, although some Australian anomalies are evident, such as the geographical disparity of markets. A comparative analysis with similar studies in the UK and US is reported, illustrating both the drivers and constraints confronting the industry in Australia. OSM uptake into the future is however dependent on many factors, not least of which is a better understanding of the construction process and its associated costs.

In 2004, with the increasing overloading restriction requirements of society in Anhui, a provincial comprehensive overloading transportation survey has been developed to take evaluations on overloading actuality and enforcement efficiency with the support of the World Bank. A total of six site surveys were conducted at Hefei, Fuyang, Luan, Wuhu, Huainan and Huangshan Areas with four main contents respectively: traffic volume, axle load, freight information and registration information. Via statistical analysis on the survey data, conclusions were gained that: vehicle overloading are very universal and serious problems at arterial highways in Anhui now. The traffic loads have far exceeded the designed endure capacity of highways and have caused prevalent premature pavement damage, especially for rigid pavement. The overloading trucks are unimpeded engaged in highway freight transportation actually due to the disordered overloading enforcement strategies and the deficient inspecting technologies.

The Internet theoretically enables marketers to personalize a Website to an individual consumer. This article examines optimal Website design from the perspective of personality trait theory and resource-matching theory. The influence of two traits relevant to Internet Web-site processing—sensation seeking and need for cognition— were studied in the context of resource matching and different levels of Web-site complexity. Data were collected at two points of time: personality-trait data and a laboratory experiment using constructed Web sites. Results reveal that (a) subjects prefer Web sites of a medium level of complexity, rather than high or low complexity; (b)high sensation seekers prefer complex visual designs, and low sensation seekers simple visual designs, both in Web sites of medium complexity; and (c) high need-for-cognition subjects evaluated Web sites with high verbal and low visual complexity more favourably.

The interaction of quercetin, which is a bioflavonoid, with bovine serum albumin (BSA) was investigated under pseudo-physiological conditions by the application of UV–vis spectrometry, spectrofluorimetry and cyclic voltammetry (CV). These studies indicated a cooperative interaction between the quercetin–BSA complex and warfarin, which produced a ternary complex, quercetin–BSA–warfarin. It was found that both quercetin and warfarin were located in site I. However, the spectra of these three components overlapped and the chemometrics method – multivariate curve resolution-alternating least squares (MCR-ALS) was applied to resolve the spectra. The resolved spectra of quercetin–BSA and warfarin agreed well with their measured spectra, and importantly, the spectrum of the quercetin–BSA–warfarin complex was extracted. These results allowed the rationalization of the behaviour of the overlapping spectra. At lower concentrations ([warfarin] < 1 × 10−5 mol L−1), most of the site marker reacted with the quercetin–BSA, but free warfarin was present at higher concentrations. Interestingly, the ratio between quercetin–BSA and warfarin was found to be 1:2, suggesting a quercetin–BSA–(warfarin)2 complex, and the estimated equilibrium constant was 1.4 × 1011 M−2. The results suggest that at low concentrations, warfarin binds at the high-affinity sites (HAS), while low-affinity binding sites (LAS) are occupied at higher concentrations.

Embryogenic callus was initiated by culturing in vitro taro corm slices on agar-solidified half-strength MS medium containing 2.0 mg/L 2,4-dichlorophenoxyacetic acid (2,4-D) for 20 days followed by transfer to 1.0 mg/L thidiazuron (TDZ). Callus was subsequently proliferated on solid medium containing 1.0 mg/L TDZ, 0.5 mg/L 2,4- D and 800 mg/L glutamine before transfer to liquid medium containing the same components but with reduced glutamine (100 mg/L). After 3 months in liquid culture on an orbital shaker, cytoplasmically dense cell aggregates began to form. Somatic embryogenesis was induced by plating suspension cells onto solid media containing reduced levels of hormones (0.1 mg/L TDZ, 0.05 mg/L 2,4-D), high concentrations of sucrose (40–50 g/L) and biotin (1.0 mg/L). Embryo maturation and germination was then induced on media containing 0.05 mg/L benzyladenine (BA) and 0.1 mg/L indole-3-acetic acid (IAA). Histological studies of the developing embryos revealed the presence of typical shoot and root poles suggesting that these structures were true somatic embryos. The rate of somatic embryos formation was 500–3,000 per mL settledcell volume while approximately 60% of the embryos regenerated into plants.
