Lumbricus rubellus Hoffmeister, inhabiting soil at the 19th century Devon Great Consols mine at Tavistock, Devon, UK, show high tolerance to Cu- and As-toxicity and frequently have a striking yellow coloration. Specimens from this site (mature and immature) and from an uncontaminated site on Lancaster University campus (mature) were photographed, and the slide images digitized and analyzed. All L. rubellus showed reddish-purple pigmentation of the body wall that declined in intensity posteriorly. The metal- and metalloid-resistant earthworms, whether mature or immature, showed yellowing in the posterior half of the body. The source of the coloration was intense yellow pigmentation of the chloragogenous tissue surrounding the alimentary canal. The yellow pigmentation is masked by reddish-purple body wall pigmentation anteriorly. Total As concentrations in tissues were determined for the anterior, middle and posterior sections of resistant and non-resistant L. rubellus. Highest concentrations were in the middle sections of the mature and immature resistant L. rubellus (36.17 ± 19.77 and 27.77 ± 9.02 mg As kg-1, respectively). Resistant immature L. rubellus lost condition over 28 d in soil treated with 750 mg As kg-1, possibly due to a higher metabolism, whilst there was no loss in condition for resistant mature L. rubellus in the treated soil. As far as the authors are aware, this is the first report of yellow pigmentation of this kind in earthworms. The pigmentation may provide a useful indicator of exposure/resistance to soil contamination. © 2002 Elsevier Science Ltd. All rights reserved.

Two arsenic- and heavy metal-contaminated mine-spoil sites, at Carrock Fell, Cumbria and Devon Great Consols Mine, Devon, were found to support populations of the earthworms Lumbricus rubellus Hoffmeister and Dendrodrilus rubidus (Savigny). L. rubellus and D. rubidus collected from the Devon site and an uncontaminated site were kept for 28 days in uncontaminated soil and in soil containing sodium arsenate (494 mg As kg-1). The state of the specimens was recorded every 7 days using a semi-quantitative assessment of earthworm health (condition index, C. I.). The C. I. remained high for all specimens except those of L. rubellus and D. rubidus from uncontaminated sites, which displayed 60 and 10% mortality, respectively. L. rubellus collected from the Carrock Fell site, and L. rubellus and D. rubidus from an uncontaminated site, burrowed as rapidly into soil containing up to 1235 mg As kg-1 in the form of sodium arsenate as into uncontaminated soil when placed on the soil surface. When earthworms were allowed a choice between uncontaminated soil and soil contaminated with sodium arsenate in concentrations of up to 1235 mg As kg-1, the threshold concentration for avoidance of contaminated soil was lower for L. rubellus and D. rubidus from uncontaminated soil than for specimens from contaminated soil. There was no significant effect of pH on soil discrimination. The LC50 concentration of As for L. rubellus from Devon Great Consols was significantly higher (P < 0.001) than for L. rubellus from the uncontaminated site: 1510 and 96 mg As kg-1, respectively. © 2001 Elsevier Science Ltd. Mining and metallurgical industries are the most responsible for heavy metal contamination. These contaminants are often associated to poor management strategies and the lack of suitable containment areas. This is the case of one of the largest metal alloy producers in the São Francisco river basin, in Minas Gerais state, Brazil. During operation, the tailings of this activity have been accumulated in the surrounding area, leading to heavy metal contamination (Mn, Cu, Zn, Ni, Pb, Cd, Cr and As), much above the critical levels, which accumulate in the sediments of the nearby waterways, in the soils of the drainage area, and in the water column. In this work we are going to discuss both the contamination level and its extent in the Consciência river, a small tributary of the São Francisco river, the longest river entirely Brazilian, by analyzing the geochemistry and mineralogy of the accumulated sediments. It will be also discuss the reclamation strategies of this site and the impact on the environment.

Nas últimas décadas verificou-se um aumento da contaminação dos solos com metais pesados resultante de processos antropogénicos. As descargas de efluentes industriais, a actividade mineira e a aplicação de lamas residuais e de fertilizantes são as principais fontes de metais pesados. Em certas regiões, a acumulação destes elementos nos solos tem atingido níveis preocupantes para o equilíbrio dos ecossistemas. Vários estudos têm demonstrado que os metais influenciam os microrganismos afectando adversamente o seu crescimento, morfologia e actividades bioquímicas resultando num decréscimo da biomassa e diversidade. Entre os microrganismos do solo, as bactérias pertencentes ao género Rhizobium têm um elevado interesse científico, económico e ecológico devido à sua capacidade para fixar azoto. Deste modo, o trabalho desenvolvido ao longo desta tese incidiu sobre o efeito da toxicidade imposta pelos metais nas bactérias fixadoras de azoto, em particular em Rhizobium leguminosarum bv. trifolii e teve como principais objectivos: determinar o efeito dos metais pesados na sobrevivência e na capacidade de fixar azoto dos isolados de rizóbio; avaliar a influência dos metais na diversidade das populações de rizóbio isoladas de solos contaminados; determinar os níveis de tolerância do rizóbio a diferentes metais e analisar a resposta ao stresse oxidativo imposto pelo cádmio. A Mina do Braçal foi o local de estudo escolhido uma vez que os seus solos estão muito contaminados com metais em resultado da extracção de minério durante mais de 100 anos. Foram escolhidos 3 solos com diferentes graus de contaminação, o solo BC com concentrações reduzidas de metais, escolhido por estar numa zona já fora da mina e designado por solo controlo e os solos BD e BA considerados medianamente e muito contaminados, respectivamente. O Pb e o Cd foram os metais predominantes nestes solos, assim como o metalóide As, cujas concentrações ultrapassaram largamente os limites previstos na lei. Sendo as enzimas do solo boas indicadoras da qualidade do mesmo, foi determinada a actividade de algumas como a desidrogenase (DHA) e a catalase (CAT). Ambas as enzimas correlacionaramse negativamente com as concentrações de metais nos solos. A dimensão das populações indígenas de rizóbio nos solos contaminados (BD e BA) foi bastante baixa, 9,1 bactérias g-1 de solo e 7,3 bactérias g-1 de solo, respectivamente, quando em comparação com a população do solo BC (4,24x104 bactérias g-1 de solo). Estes resultados parecem estar relacionados com o elevado conteúdo em metais e com o pH ácido dos solos. A capacidade simbiótica também foi afectada pela presença de metais, uma vez que os isolados originários do solo BD mostraram menor capacidade em fixar azoto do que os isolados do solo controlo. A diversidade das populações de rizóbio foi determinada com recurso à análise dos perfis de plasmídeos, perfis de REP e ERIC-PCR de DNA genómico e perfis de proteínas e lipopolissacarídeos. No conjunto dos 35 isolados analisados foram identificados 11 plasmídeos com pesos moleculares entre 669 kb e 56 kb. Embora a incidência de plasmídeos tenha sido superior nos isolados do solo BC verificou-se maior diversidade plasmídica na população isolada do solo BD. Resultados similares foram obtidos com os perfis de REP e ERIC-PCR e perfis de proteínas, que indicaram maior diversidade nas populações dos solos contaminados (BD e BA), contrariamente ao verificado por outros autores. O grau de tolerância aos metais pesados e ao arsénio dos vários isolados testados dependeu do metal e do local de origem. No geral, os isolados do solo BD mostraram maior tolerância aos metais do que os isolados do solo controlo, o que está de acordo com o esperado uma vez que geralmente as populações dos locais contaminados são mais tolerantes. Contudo, os isolados do local mais contaminado (BA) foram muito tolerantes apenas ao chumbo mostrando-se sensíveis aos restantes metais. A inoculação dos solos BC, BD e BA após irradiação com estirpes seleccionadas de rizóbio permitiu avaliar a sua sobrevivência ao longo de 12 meses em condições mais realistas. Verificou-se que após um decréscimo inicial, os isolados inoculados no solo BC conseguiram recuperar a dimensão das suas populações para números similares aos inicialmente introduzidos, contrariamente ao verificado no solo BD onde o número de rizóbios decresceu ao longo dos 12 meses. As condições adversas do solo BA apenas permitiram a sobrevivência de 4 isolados até aos 3 meses e apenas dois deles conseguiram sobreviver após 12 meses, designadamente C 3-1 e A 17-3. Estes isolados possuem um plasmídeo de 669 kb que poderá estar na base da sobrevivência destas estirpes. Por outro lado, o último isolado é originário do solo contaminado e por isso estará também mais adaptado a sobreviver às elevadas concentrações de Pb existentes no solo BA. Por fim, constatou-se que o cádmio, um dos metais presente em concentrações mais elevadas nos solos em estudo, é um indutor de stresse oxidativo nos isolados de rizóbio menos tolerantes o que foi confirmado pelo aumento de ROS e danos celulares ao nível dos lípidos.

Os sistemas aquáticos naturais podem estar sujeitos frequentemente a entrada de tóxicos, quer seja através da lixiviação dos campos agrícolas ou da descarga por parte de unidades industriais. Avaliar o impacto potencial destes contaminantes nos sistemas aquáticos é muito importante, porque pode promover consequências sérias no balanço ecológico dos ecossistemas. Os efeitos de níveis sub-letais destes tóxicos nas populações aquáticas são detectados, em muitos casos, somente após diversas gerações, dependendo da espécie e do contaminante. O comportamento animal é considerado como sendo a primeira linha de defesa perante estímulos ambientais, e pode ser uma representação de alterações fisiológicas no organismo, sendo portanto um indicador excelente de alterações ambientais. O desenvolvimento dos sistemas de aviso prévio que integram parâmetros comportamentais pode ajudar a prever mais rapidamente possíveis alterações ao nível das populações naturais, do que a utilização de testes ecotoxicológicos padrão com a mesma finalidade. O conhecimento acerca de possíveis implicações devido a alterações comportamentais, em organismos bentónicos e em populações do campo sujeitas a tóxicos, é ainda escasso. Sabendo isto, neste estudo pretendeu-se investigar como o comportamento de Chironomus riparius – usando um biomonitor em tempo real – e outros parâmetros tais como crescimento, emergência de adultos, bioacumulação e biomarcadores, são afectados pela exposição a imidacloprid e ao mercúrio, que foram seleccionados como contaminantes. Os resultados demonstraram que a exposição às concentrações sub-letais de imidacloprid afecta o crescimento e o comportamento dos quironomídeos e que estes organismos podem recuperar de uma exposição curta ao insecticida. O comportamento que corresponde à ventilação de C. riparius revelou-se como um parâmetro mais sensível do que a locomoção e do que as respostas bioquímicas, quando as larvas foram sujeitas ao imidacloprid. Larvas de C. riparius expostas a concentrações sub-letais de mercúrio apresentaram uma tendência de diminuição de actividade comportamental, em testes com concentrações crescentes do tóxico; o crescimento das larvas foi também prejudicado, e as taxas de emergência de adultos e o tempo de desenvolvimento apresentaram retardamento. Estes organismos podem bioacumular rapidamente o mercúrio em condições de não alimentação e apresentam uma lenta depuração deste metal. Estes efeitos podem, em último caso, conduzir a prováveis repercussões ao nível da população e das comunidades. As reduções em actividades comportamentais, mesmo em concentrações baixas, podem diminuir a quantidade de tempo gasta na procura de alimento, produzindo efeitos aos níveis morfo-fisiológicos, e assim afectar severamente o desempenho dos quironomídeos no ambiente. O uso destes factores comportamentais como um parâmetro ecotoxicológico sub-letal relevante ao nível da toxicologia aumentará a versatilidade dos testes, permitindo uma resposta comportamental mensurável e quantitativa ao nível do organismo, utilizando uma avaliação não destrutiva, e assim certificando que esta aproximação pode ser usada em testes ecotoxicológicos futuros.

The present work describes the optimization of a short-term assay, based on the inhibition of the esterase activity of the alga Pseudokirchneriella subcapitata, in a microplate format. The optimization of the staining procedure showed that the incubation of the algal cells with 20 μmolL−1 fluorescein diacetate (FDA) for 40 min allowed discrimination between metabolic active and inactive cells. The shortterm assay was tested using Cu as toxicant. For this purpose, algal cells, in the exponential or stationary phase of growth, were exposed to the heavy metal in growing conditions. After 3 or 6 h, cells were subsequently stained with FDA, using the optimized procedure. For Cu, the 3- and 6-h EC50 values, based on the inhibition of the esterase activity of algal cells in the exponential phase of growth, were 209 and 130 μg L−1, respectively. P. subcapitata cells, in the stationary phase of growth, displayed higher effective concentration values than those observed in the exponential phase. The 3- and 6-h EC50 values for Cu, for cells in the stationary phase, were 443 and 268 μgL−1, respectively. This short-term microplate assay showed to be a rapid endpoint for testing toxicity using the alga P. subcapitata. The small volume required, the simplicity of the assay (no washing steps), and the automatic reading of the fluorescence make the assay particularly well suited for the evaluation of the toxicity of a high number of environmental samples.

The present work has as objective to contribute for the elucidation of the mechanism associated with Pb detoxification, using the yeast Saccharomyces cerevisiae as a model organism. The deletion of GTT1 or GTT2 genes, coding for functional glutathione transferases (GST) enzymes in S. cerevisiae, caused an increased susceptibility to high Pb concentrations (500-1000 μmol L(-1)). These results suggest that the formation of glutathione-Pb conjugate (GS-Pb), dependent of GSTs, is important in Pb detoxification. The involvement of ATP-binding cassette (ABC) vacuolar transporters, belonging to class C subfamily (ABCC) in vacuolar compartmentalization of Pb, was evaluated. For this purpose, mutant strains disrupted in YCF1, VMR1, YBT1 or BPT 1 genes were used. All mutants tested, without vacuolar ABCC transporters, presented an increased sensitivity to 500-1000 μmol L(-1) Pb comparative to wild-type strain. Taken together, the obtained results suggest that Pb detoxification, by vacuolar compartmentalization, can occur as a result of the concerted action of GSTs and vacuolar ABCC transporters. Pb is conjugated with glutathione, catalysed by glutathione transferases and followed to the transport of GS-Pb conjugate to the vacuole by ABCC transporters.

Although the knowledge on heavy metal hyperaccumulation mechanisms is increasing, the genetic basis of cadmium (Cd) hyperaccurnulation remains to be elucidated. Thlaspi caerulescens is an attractive model since Cd accumulation polymorphism observed in this species suggests genetic differences between populations with low versus high Cd hyperaccumulation capacities. In our study, a methodology is proposed to analyse at a regional scale the genetic differentiation of T. caerulescens natural populations in relation to Cd hyperaccumulation capacity while controlling for different environmental, soil, plant parameters and geographic origins of populations. Twenty-two populations were characterised with AFLP markers and cpDNA polymorphism. Over all loci, a partial Mantel test showed no significant genetic structure with regard to the Cd hyperaccumulation capacity. Nevertheless, when comparing the marker variation to a neutral model, seven AFLP fragments (9% of markers) were identified as presenting particularly high genetic differentiation between populations with low and high Cd hyperaccurnulation capacity. Using simulations, the number of outlier loci was showed to be significantly higher than expected at random. These loci presented a genetic structure linked to Cd hyperaccumulation capacity independently of the geography, environment, soil parameters and Zn, Pb, Fe and Cu concentrations in plants. Using a canonical correspondence analysis, we identified three of them as particularly related to the Cd hyperaccumutation capacity. This study demonstrates that populations with low and high hyperaccurnulation capacities can be significantly distinguished based on molecular data. Further investigations with candidate genes and mapped markers may allow identification and characterization of genomic regions linked to factors involved in Cd hyperaccumulation.

Nanoporous materials with large surface area and well-ordered pore structure have been synthesized. Thiol groups were grafted on the materials' surface to make heavy metal ion pre-concentration media. The adsorption properties ofthe materials were explored. Mercury, gold and silver can be strongly adsorbed by these materials, even in the presence of alkaline earth metal ion. Though the materials can adsorb other heavy metal ions such as lead and copper, they show differential adsorption ability when several ions are present in solution. The adsorption sequence is: mercury> == silver> copper » lead and cadmium. In the second part of this work, the memory effects of mercury, gold, silver and boron were investigated. The addition of 2% L-cysteine and 1% thiourea eliminates the problems of the three metal ions completely. The wash-out time for mercury dropped from more than 20 minutes to 18 seconds, and the wash-out time for gold decreased from more than 30 minutes to 49 seconds. The memory effect of boron can be reduced by the use of mannitol.

Although it is widely assumed that temperature affects pollutant toxicity, few studies have actually investigated this relationship. Moreover, such research as has been done has involved constant temperatures; circumstances which are rarely, if ever, actually experienced by north temperate, littoral zone cyprinid species. To investigate the effects of temperature regime on nickel toxicity in goldfish (Carassius auratus L.), 96- and 240-h LCSO values for the heavy metal pollutant, nickel (NiCI2.6H20), were initially determined at 2DoC (22.8 mg/L and 14.7 mg/L in artificially softened water). Constant temperature bioassays at 10°C, 20°C and 30°C were conducted at each of 0, 240-h and 96-h LCSO nickel concentrations for 240 hours. In order to determine the effects of temperature variation during nickel exposure it was imperative that the effects of a single temperature change be investigated before addressing more complex regimes. Single temperature changes of + 10°C or -10°C were imposed at rates of 2°C/h following exposures of between 24 hand 216 h. The effects of a single temperature change on mortality, and duration of toxicant exposure at high and low temperatures were evaluated. The effects of fluctuating temperatures during exposure were investigated through two regimes. The first set of bioassays imposed a sinewave diurnal cycle temperature (20.±.1DOC) throughout the 10 day exposure to 240-h LeSO Ni. The second set of investigations approximated cyprinid movement through the littoral zone by imposing directionally random temperature changes (±2°C at 2-h intervals), between extremes of 10° and 30°C, at 240-h LC50 Ni. Body size (i.e., total length, fork length, and weight) and exposure time were recorded for all fish mortalities. Cumulative mortality curves under constant temperature regimes indicated significantly higher mortality as temperature and nickel concentration were increased. At 1DOC no significant differences in mortality curves were evident in relation to low and high nickel test concentrations (Le., 16 mg/L and 20 mg/L). However at 20°C and 30°C significantly higher mortality was experienced in animals exposed to 20 mg/L Ni. Mortality at constant 10°C was significantly lower than at 30°C with 16 mg/L and was significantly loWer than each of 2DoC and 39°C tanks at 20 mg/L Ni exposure. A single temperature shift from 20°C to 1DoC resulted in a significant decrease in mortality rate and conversely, a single temperature shift from 20°C to 30°C resulted in a significant increase in mortality rate. Rates of mortality recorded during these single temperature shift assays were significantly different from mortality rates obtained under constant temperature assay conditions. Increased Ni exposure duration at higher temperatures resulted in highest mortality. Diurnally cycling temperature bioassays produced cumulative mortality curves approximating constant 20°C curves, with increased mortality evident after peaks in the temperature cycle. Randomly fluctuating temperature regime mortality curves also resembled constant 20°C tanks with mortalities after high temperature exposures (25°C - 30°C). Some test animals survived in all assays with the exception of the 30°C assays, with highest survival associated with low temperature and low Ni concentration. Post-exposure mortality occurred most frequently in individuals which had experienced high Ni concentrations and high temperatures during assays. Additional temperature stress imposed 2 - 12 weeks post exposure resulted in a single death out of 116 individuals suggesting that survivors are capable of surviving subsequent temperature stresses. These investigations suggest that temperature significantly and markedly affects acute nickel toxicity under both constant and fluctuating temperature regimes and plays a role in post exposure mortality and subsequent stress response.

Dans ses entretiens, George Steiner considère la musique rock et le heavy metal comme l’espéranto international des jeunes de notre époque (Steiner, Entretiens, p. 91). Or, cet espéranto porte une dimension religieuse manifeste, entre autres à travers l’usage fréquent d’images et de textes bibliques souvent passés au broyeur des guitares et des voix vociférantes. On peut se demander quelles figures construisent les fragments qui en résultent, souvent irrévérencieux voire scandaleux pour plusieurs. À travers les violences que le rock et surtout le métal infligent au texte biblique, faut-il voir des attaques contre la Bible et contre Dieu, comme le croient plusieurs chrétiens, ou, au contraire, un respect profond de la dynamique de ce texte et un usage légitime de celui-ci ? Davantage portée par la seconde hypothèse, ce texte vise à identifier les usages et les fonctions religieuses de la Bible dans la musique rock.

L’influence des contaminants environnementaux (CE) a été largement étudié en contexte d’exposition aigüe et d’intoxication. Qu’en est-il d’une exposition chronique à plus faibles doses sur le développement cognitif et sensoriel des enfants? Les études longitudinales de la présente thèse ont été réalisées au Nunavik. Bien que géographiquement éloignée des centres industriels, la communauté inuite est exposée aux CE via leur transport par les courants atmosphériques et océaniques ainsi que par leur bioaccumulation dans le poisson et les mammifères marins. Puisque l’alimentation autochtone traditionnelle, notamment la chair de béluga, est encore fréquemment consommée par la communauté, une proportion substantielle d’enfants inuits est exposée in utero aux CE. Ceux-ci sont également continuellement exposés à ces agents neurotoxiques durant leur développement postnatal. Or, la variation considérable dans l’adoption de l’alimentation traditionnelle au sein de la communauté représente une opportunité pour étudier la relation entre les niveaux d’exposition aux CE et le développement cognitif et sensoriel des enfants. Bien que certains déficits aient déjà été mis en lien avec l’exposition chronique aux CE dans la littérature, la présente thèse s’intéressa à cette relation plus spécifiquement chez les enfants inuits vivant dans le Grand Nord et plus exposés aux CE en raison de leur alimentation. Par ailleurs, les protocoles qui ont été développés pour cette thèse permettront d’évaluer des aspects qui ont été peu étudiés en lien avec les CE, soit l’attention visuospatiale et le traitement visuel cérébral précoce. Dans le premier volet de cette thèse, la relation entre trois CE et l’attention visuospatiale a été étudiée à l’aide d’une version adaptée de la tâche de Posner (M.I. Posner et al., 1980). Cette tâche psychophysique a été administrée à des enfants inuits (âge moyen = 11.2 ans) dont les niveaux d’exposition au mercure (Hg), au plomb (Pb) et aux biphényles polychlorés (BPCs) ont été documentés durant les périodes pré et postnatale. Les expositions in utero au Pb et aux BPCs ont été significativement associées à de l’impulsivité et de l’inattention, respectivement, alors que l’exposition postnatale au Pb a été associée à des temps de réaction plus longs. Bien qu’aucune relation spécifique avec l’attention visuospatiale n’ait été trouvée, les résultats de cette étude suggèrent que l’exposition aux CE est associée à une diminution des capacités attentionnelles générales chez les enfants résidant au Nunavik. Dans le second volet, le traitement cérébral précoce de l’information visuelle a été examiné à l’aide de potentiels évoqués visuels auprès d’enfants de la même communauté (âge moyen = 10.9 ans). La concentration de Hg dans le sang de cordon ombilical mesurée à la naissance a été associée à une réduction de l’amplitude et à une augmentation de la latence de la composante N75. L’exposition prénatale au Pb a quant à elle été associée à un délai dans la latence de la composante N150. Les résultats obtenus suggèrent ainsi que l’exposition aux métaux lourds, en particulier durant la période gestationnelle, serait associée à des altérations dans le développement visuel. Les résultats présentés dans cette thèse soutiennent l’hypothèse selon laquelle le cerveau en développement est vulnérable lors d’une exposition chronique aux CE, et cela même à des niveaux de concentration inférieurs aux limites recommandées par les organismes de santé publique. Les résultats permettent également d’apporter un éclairage nouveau sur les déficits cognitifs et sensoriels associés aux CE.
