Although glucocorticoids are widely used as antiinflammatory agents in clinical therapies, they may cause serious side effects that include insulin resistance and hyperinsulinemia. To study the potential functional adaptations of the islet of Langerhans to in vivo glucocorticoid treatment, adult Wistar rats received dexamethasone (DEX) for 5 consecutive days, whereas controls (CTL) received only saline. The analysis of insulin release in freshly isolated islets showed an enhanced secretion in response to glucose in DEX-treated rats. The study of Ca(2+) signals by fluorescence microscopy also demonstrated a higher response to glucose in islets from DEX-treated animals. However, no differences in Ca(2+) signals were found between both groups with tolbutamide or KCl, indicating that the alterations were probably related to metabolism. Thus, mitochondrial function was explored by monitoring oxidation of nicotinamide dinucleotide phosphate autofluorescence and mitochondrial membrane potential. Both parameters revealed a higher response to glucose in islets from DEX-treated rats. The mRNA and protein content of glucose transporter-2, glucokinase, and pyruvate kinase was similar in both groups, indicating that changes in these proteins were probably not involved in the increased mitochondrial function. Additionally, we explored the status of Ca(2+)-dependent signaling kinases. Unlike calmodulin kinase II, we found an augmented phosphorylation level of protein kinase C alpha as well as an increased response of the phospholipase C/inositol 1,4,5-triphosphate pathway in DEX-treated rats. Finally, an increased number of docked secretory granules were observed in the beta-cells of DEX animals using transmission electron microscopy. Thus, these results demonstrate that islets from glucocorticoid-treated rats develop several adaptations that lead to an enhanced stimulus-secretion coupling and secretory capacity. Royal palm tree peroxidase (RPTP) is a very stable enzyme in regards to acidity, temperature, H(2)O(2), and organic solvents. Thus, RPTP is a promising candidate for developing H(2)O(2)-sensitive biosensors for diverse applications in industry and analytical chemistry. RPTP belongs to the family of class III secretory plant peroxidases, which include horseradish peroxidase isozyme C, soybean and peanut peroxidases. Here we report the X-ray structure of native RPTP isolated from royal palm tree (Roystonea regia) refined to a resolution of 1.85 angstrom. RPTP has the same overall folding pattern of the plant peroxidase superfamily, and it contains one heme group and two calcium-binding sites in similar locations. The three-dimensional structure of RPTP was solved for a hydroperoxide complex state, and it revealed a bound 2-(N-morpholino) ethanesulfonic acid molecule (MES) positioned at a putative substrate-binding secondary site. Nine N-glycosylation sites are clearly defined in the RPTP electron-density maps, revealing for the first time conformations of the glycan chains of this highly glycosylated enzyme. Furthermore, statistical coupling analysis (SCA) of the plant peroxidase superfamily was performed. This sequence-based method identified a set of evolutionarily conserved sites that mapped to regions surrounding the heme prosthetic group. The SCA matrix also predicted a set of energetically coupled residues that are involved in the maintenance of the structural folding of plant peroxidases. The combination of crystallographic data and SCA analysis provides information about the key structural elements that could contribute to explaining the unique stability of RPTP. The purpose of this study was to evaluate the frequency of anti-Toxocara antibodies in sheep from Presidente Prudente, southeastern Brazil. Serum samples were obtained from 365 sheep of diverse breeds and different ages. Samples were collected at a slaughterhouse and at farms located in Presidente Prudente. Three groups of animal of different ages were evaluated according to age: Group I: between 1 and 6 months old; Group II: between 7 and 10 months old; and Group III: between 11 and 15 months old. An ELISA test was carried out to detect anti-Toxocara antibodies (IgG) using the excretory-secretory antigens of Toxocara canis (TES) larvae. In total, 183 out of 365 animals (50.1%) were positive for anti-Toxocara antibodies. The frequency of antibody detection was directly proportional to the age of the animals (p<0.0001). indicating a relationship between infection and aging. In Group III, there was a higher prevalence in females (p = 0.0041). The relevance of these animals to the epidemiology of toxocariasis in pets and human should be considered. (C) 2011 Elsevier B.V. The pentrophic membrane (PM) is an anatomical structure surrounding the food bolus in most insects. Rejecting the idea that PM has evolved from coating mucus to play the same protective role as it, novel functions were proposed and experimentally tested. The theoretical principles underlying the digestive enzyme recycling mechanism were described and used to develop an algorithm to calculate enzyme distributions along the midgut and to infer secretory and absorptive sites. The activity of a Spodoptera frugiperda microvillar aminopeptidase decreases by 50% if placed in the presence of midgut contents. S. frugiperda trypsin preparations placed into dialysis bags in stirred and unstirred media have activities of 210 and 160%, respectively, over the activities of samples in a test tube. The ectoperitrophic fluid (EF) present in the midgut caeca of Rhynchosciara americana may be collected. If the enzymes restricted to this fluid are assayed in the presence of PM contents (PMC) their activities decrease by at least 58%. The lack of PM caused by calcofluor feeding impairs growth due to an increase in the metabolic cost associated with the conversion of food into body mass. This probably results from an increase in digestive enzyme excretion and useless homeostatic attempt to reestablish destroyed midgut gradients. The experimental models support the view that PM enhances digestive efficiency by: (a) prevention of non-specific binding of undigested material onto cell Surface; (b) prevention of excretion by allowing enzyme recycling powered by an ectoperitrophic counterflux of fluid; (c) removal from inside PM of the oligomeric molecules that may inhibit the enzymes involved in initial digestion; (d) restriction of oligomer hydrolases to ectoperitrophic space (ECS) to avoid probable partial inhibition by non-dispersed undigested food. Spodoptera frugiperda beta-1,3-glucanase (SLam) was purified from larval midgut. It has a molecular mass of 37.5 kDa, an alkaline optimum pH of 9.0, is active against beta-1,3-glucan (laminarin), but cannot hydrolyze yeast beta-1,3-1,6-glucan or other polysaccharides. The enzyme is an endoglucanase with low processivity (0.4), and is not inhibited by high concentrations of substrate. In contrast to other digestive beta-1,3-glucanases from insects, SLam is unable to lyse Saccharomyces cerevisae cells. The cDNA encoding SLam was cloned and sequenced, showing that the protein belongs to glycosyl hydrolase family 16 as other insect glucanases and glucan-binding proteins. Multiple sequence alignment of beta-1,3-glucanases and beta-glucan-binding protein supports the assumption that the beta-1,3-glucanase gene duplicated in the ancestor of mollusks and arthropods. One copy originated the derived beta-1,3-glucanases by the loss of an extended N-terminal region and the beta-glucan-binding proteins by the loss of the catalytic residues. SLam homology modeling suggests that E228 may affect the ionization of the catalytic residues, thus displacing the enzyme pH optimum. SLam antiserum reacts with a single protein in the insect midgut. Immunocytolocalization shows that the enzyme is present in secretory vesicles and glycocalyx from columnar cells. (C) 2010 Elsevier Ltd. All rights reserved.

Both soluble (SfTre1) and membrane-bound (SfTre2) trehalases occur along the midgut of Spodoptera frugiperda larvae. Released SfTre2 was purified as a 67 kDa protein. Its K(m) (1.6 mM) and thermal stability (half life 10 min at 62 degrees C) are different from the previously isolated soluble trehalase (K(m) = 0.47 mM; 100% stable at 62 degrees C). Two cDNAs coding for S. frugiperda trehalases have been cloned using primers based on consensus sequences of trehalases and having as templates a cDNA library prepared from total polyA-containing RNA extracted from midguts. One cDNA codes for a trehalase that has a predicted transmembrane sequence and was defined as SfTre2. The other, after being cloned and expressed, results in a recombinant trehalase with a K(m) value and thermal stability like those of native soluble trehalase. This enzyme was defined as SfTre1 and, after it was used to generate antibodies, it was immunolocalized at the secretory vesicles and at the glycocalyx of columnar cells. Escherichia coli trehalase 3D structure and sequence alignment with SfTre1 support a proposal regarding the residue modulating the pKa value of the proton donor.

Este estudo apresenta os resultados de tratamento em 83 pacientes com Idade entre 2–10 anos, que apresentaram otite média secretora com efusão, bilateral ou unilateral, acompanhada ou não de hipertrofia de adenóides e cornetos, no Hospital Universitário São Francisco de Paula da Universidade Católica de Pelotas – UCPel, no período de julho de 1997 a julho de 1999. Os pacientes foram submetidos a timpanocentese com uso do Microcautério por Rádio-freqüência (modelo Lavinsky – HCPA), uni ou bilateralmente. Na avaliação periódica de 15 dias, um mês e três meses, observamos dois pontos principais: a) a cura dos pacientes (que apresentaram membrana e caixa timpânica normais); b) a relação com a literatura em trabalhos semelhantes com uso do Laser CO2. Relatamos também a incidência em percentuais, com identificação das alterações de imagem da membrana timpânica nestes três períodos cronológicos, e na distribuição conforme cor, sexo e idade, uni/bilateralidade e simultaneidade na presença das duas afecções relatadas. Na avaliação final, percebemos 80,5% de cura da patologia. Na comparação com a literatura existente sobre o uso de Laser CO2, os nossos resultados mostraram-se homogêneos. O tempo de oclusão da membrana timpânica foi de 2,73 meses (desvio – padrão = 1,39 meses), normalizando a secreção da caixa timpânica.

Uma significativa quantidade de proteínas vegetais apresenta-se compartimentalizada nas diversas estruturas celulares. A sua localização pode conduzir à elucidação do funcionamento dos processos biossintéticos e catabólicos e auxiliar na identificação de genes importantes. A fim de localizar produtos gênicos relacionados à resistência, foi utilizada a fusão de cDNAs de arroz (Oryza sativa L.) ao gene da proteína verde fluorescente (GFP). Os cDNAs foram obtidos a partir de uma biblioteca supressiva subtrativa de genes de arroz durante uma interação incompatível com o fungo Magnaporthe grisea. Estes cDNAs foram fusionados a uma versão intensificada de gfp e usados para transformar 500 plantas de Arabidopsis thaliana. Outras 50 plantas foram transformadas com o mesmo vetor, porém sem a fusão (vetor vazio). Foram obtidas aproximadamente 25.500 sementes oriundas das plantas transformadas com as fusões EGFP::cDNAs e 35.000 sementes das transformadas com o vetor vazio, produzindo, respectivamente, 750 e 800 plantas tolerantes ao herbicida glufosinato de amônio. Após a seleção, segmentos foliares das plantas foram analisados por microscopia de fluorescência, visando o estabelecimento do padrão de localização de EGFP. Foram observadas 18 plantas transformadas com a fusão EGFP::cDNAs e 16 plantas transformadas com o vetor vazio apresentando expressão detectável de GFP. Uma planta transformada com uma fusão EGFP::cDNA apresentou localização diferenciada da fluorescência, notadamente nas células guarda dos estômatos e nos tricomas. Após seqüenciamento do cDNA fusionado, foi verificado que esta planta apresentava uma inserção similar a uma seqüência codificante de uma quinase, uma classe de enzimas envolvidas na transdução de sinais em resposta à infecção por patógenos.

Peumus boldus Molina (Monimiaceae) é uma árvore comum e abundante no Chile, e suas folhas são amplamente empregadas pela medicina tradicional para o tratamento de uma variedade de afecções do sistema digestivo e hepatobiliar. Seus principais constituintes químicos são alcalóides do tipo aporfínicos, sendo a boldina seu maior e mais característico representante, cujas propriedades antioxidantes são extensivamente relatadas na literatura, servindo desta forma como marcador químico para a espécie. Com o objetivo de contribuir com parâmetros de controle de qualidade, um estudo botânico macro e microscópico das folhas e do pó da espécie foi realizado. Características morfológicas, identificadas pela análise macroscópica, como pilosidade, tipologia dos tricomas, proeminência ao redor da base dos tricomas e aparência do bordo, e características microscópicas como estômatos anomocíticos, folha hipoestomática e ocorrência de cristais de oxalato de cálcio denotam valor taxonômico. Parâmetros farmacopéicos como cinzas totais e perda por dessecação foram determinados, e um método de quantificação da boldina foi desenvolvido e validado utilizando a CLAE. O limite mínimo de boldina observado na análise de 10 amostras adquiridas foi de aproximadamente 0,016%. A fim de avaliarse o comportamento de seus constituintes químicos frente a variações de temperatura e umidade, realizou-se estudo de estabilidade acelerada (40ºC ± 2ºC / 75% ± 5% U.R.). A degradação máxima de boldina foi observada após 90 dias em câmara climática chegando a aproximadamente 52%. A reação de degradação sugerida foi de segunda ordem, e o tempo de vida útil e meia-vida calculados foram de aproximadamente 18 dias e 165 dias, respectivamente. Por fim, realizou-se a quantificação de alguns metais pesados nas folhas da espécie, que decresceram na ordem de Fe> Mn> Cu> Ni, sendo que Pb, Cr e Co tiveram níveis abaixo do limite de detecção de 5 μg/g.

Mothers with good vitamin A nutritional status during gestation and lactation are better able to nourish and protect their infant with maternal milk. Our hypothesis is that women with more serum retinol have more retinol and secretory immunoglobulin A in colostrum. 190 healthy puerperal women from a Brazilian public maternity were recruited and divided according to the cutoff point for serum retinol (30 μg/dL). A number of the women was supplemented with 200000 UI (60 mg) of retinyl palmitate in the immediate postpartum. Serum and colostrum were collected on the 1st day postpartum and colostrum again on the following day. Retinol (serum and colostrum) was analyzed by HPLC and SIgA (colostrum) by turbidimetry. The mothers presented with adequate biochemical indicators of nutritional status, according to serum retinol (44.6 μg/dL). There were significant differences (p= 0.0017 and p= 0.043, respectively) in retinol and SIgA levels in the colostrum of mothers with serum retinol > 30 μg/dL and < 30 μg/dL. The concentration of SIgA in the colostrum of non-supplemented mothers on the 1st day postpartum was 822.6 mg/dL, decreasing after 24 hours to 343.7 mg/dL. Supplemented mothers showed levels of SIgA in colostrum of 498.9 mg/dL on the 2nd day postpartum (p= 0.00006). The colostrum of women with good vitamin A nutritional status had more retinol and SIgA. Additionally, maternal supplementation increases the levels of SIgA in colostrum. The higher levels of SIgA on the 1st day postpartum showed the importance of early breastfeeding, given that it provides considerable immunological benefits to newborn infants

Ucides cordatus is the most commercially, important mangrove crab in Brazil. In spite of its economic importance, there are few studies of its reproduction, in particular the female reproductive system. The present study describes the histology and histochemistry of the spermathecae of U. cordatus. Adult females were can lit monthly from July 2004 through June 2005, at Iguape, State of São Paulo. The crabs were anaesthetized, and their spermathecae removed and fixed in Davidson's fluid, following the histological routine for paraffin. The slides were stained with HE, xylidine Ponceau, PAS, alcian blue (pH 1.0 and 2.5), Sudan black B and picrosirius-haematoxylin. Histologically, the spermathecae possesses a capsule of conjunctive tissue, rich in collalgen fibres, which surrounds the secretory columnar epithelium. In the lumen, individual sperm packets are not observed; the spermatophores are intermixed with the seminal fluid and secretions of the spermathecae itself. A large proportion of the free spermatozoids and spermatophores are arranged in homogeneous masses in the proximal part of the spermathecae. The secretion produced by the columnar epithelium appears to promote the movement of the gametes to the fertilization chamber, in a ventral position, allowing fertilization of the oocytes. Histochemically, the secretion produced by the columnar epithelium was strongly positive for neutral polysaccharides, positive for acid polysaccharides, and weakly positive for proteins and lipids. Tills secretion forms a glycoprotein matrix which is associated with maintenance of the spermatophores, which can remain stored for long periods.
