In this work, biocompatible and biodegradable poly(D-L-lactide-co-glycolide) (PLGA) microparticles with the potential for use as a controlled release system of vaccines and other drugs to the lung were manufactured using supercritical CO2, through the Supercritical Assisted Atomization (SAA) technique. After performing a controlled variance in production parameters (temperature, pressure, CO2/solution flow ratio) PLGA microparticles were characterized and later used to encapsulate active pharmaceutical ingredients (API). Bovine serum albumin (BSA) was chosen as model protein and vaccine, while sildenafil was the chosen drug to treat pulmonary artery hypertension and their effect on the particles characteristics was evaluated. All the produced formulations were characterized in relation to their morphology (Morphologi G3 and scanning electronic microscopy (SEM)), to their physical-chemical properties (X-ray diffraction (XRD, differential scanning calorimetry (DSC), Fourier transform infrared (FTIR)) and aerodynamic performance using an in vitro aerosolization study – Andersen cascade impactor (ACI) - to obtain data such as the fine particle fraction (FPF) and the mass median aerodynamic diameter (MMAD). Furthermore, pharmacokinetic, biodegradability and biocompatibility tests were performed in order to verify the particle suitability for inhalation. The resulting particles showed aerodynamic diameters between the 3 and 5 μm, yields up to 58% and FPF percentages rounding the 30%. Taken as a whole, the produced microparticles do present the necessary requests to make them appropriate for pulmonary delivery.

Bacillus subtilis tem a capacidade de utilizar arabinoligossacáridos presentes na parede das células vegetais, através de um consórcios de enzimas envolvidas na hidrólise enzimática dos mesmos. A captação destes carbohidratos por parte do Bacillus subtilis depende de dois transportadores membranares AraE e AraNPQ-MsmX. Após a captação de L-arabinose e arabinoligossacáridos, estes substratos serão metabolizados pelos produtos enzimáticos codificados por genes pertencentes ao operão responsável pela metabolização de arabinose araABDLMNPQabfA. Neste operão, para além dos genes responsáveis pelo transportador AraNPQ, o gene araL determina a síntese de uma enzima com atividade de fosfatase, AraL, que tem putativamente como principal função a destoxificação de intermediários metabólicos fosforilados, em situações particulares. O objectivo deste trabalho consistiu em descobrir quais os determinantes moleculares envolvidos em ambos os processos de reconhecimento molecular, ora o reconhecimento de arabinoligossacáridos por parte da AraN, do ponto de vista dos carbohidratos ora de carbohidratos fosforilados do ponto de vista da AraL. Para elucidar estes processos de reconhecimento molecular proteína-carbohidrato foi utilizada a técnica de RMN, modelação computacional e mutagénese dirigida. No primeiro capitulo são introduzidos conceitos fundamentais para a percepção da ação de ambas as proteínas (AraN e AraL), no organismo Bacillus subtilis. O segundo capitulo, refere-se ao estudo dos mecanismos envolvidos no reconhecimento proteína-carbohidrato através da técnica de STD-RMN para estudar interações do ponto de vista do carbohidrato, bem como abordagens bioinformáticas tais como alinhamentos de sequencia primária e dockings moleculares, para identificar resíduos do ponto de vista da proteína passiveis de se encontrarem envolvidos no reconhecimento molecular, que numa última instância são mutados para se confirmar a sua relevância no processo de reconhecimento molecular do ponto de vista da proteína. Neste capitulo é demonstrado que a AraN é responsável pelo reconhecimento de arabinoligossacáridos e celoligossacáridos. No que diz respeito aos primeiros, a AraN reconhece preferencialmente arabinoligossacáridos com três subunidades, verificando-se a perda de saturação na subunidade não redutora da arabinotetraose. O processo de reconhecimento molecular apresenta uma constante de dissociação com uma ordem de grandeza na ordem dos μM. Do ponto de vista da proteína são identificados resíduos (W253, Y254 e Y54) putativos de se encontrarem também eles envolvidos no reconhecimento molecular do ponto de vista da AraN através de interações CHstacking. Finalmente no terceiro capitulo, é apresentada a optimização da sobrexpressão da AraL, dado que numa primeira fase a sua produção ocorria sob a forma de corpos de inclusão. Para tal são descritas metodologias de solubilização e renaturação da mesmas, com recurso a agentes caotrópicos (ureia e GmdCl) e detergentes (SDS).

Amyotrophic lateral sclerosis (ALS) is a fatal neurodegenerative disease characterized by the pro-gressive loss of motoneurons (MN). Increasing evidence points glial cells as key players for ALS onset and progression. Indeed, MN-glia signalling pathways involving either neuroprotection or inflammation are likely to be altered in ALS. We aimed to study the molecules related with glial function and/or reactivity by evaluating glial markers and hemichannels, mainly present in astrocytes. We also studied molecules involved in mi-croglia-MN dialogue (CXCR3/CCL21; CX3CR1/CX3CL1; MFG-E8), as well as proliferation (Ki-67) and inflammatory-related molecules (TLR2/4, NLRP3; IL-18) and alarming/calming signals (HMGB1/autotaxin). We used lumbar spinal cord (SC) homogenates from mice expressing a mutant human-SOD1 protein (mSOD1) at presymptomatic and late-symptomatic ALS stages. SJL (WT) mice at same ages were used as controls. We observed decreased expression of genes associated with astrocytic (GFAP and S100B) and microglial (CD11b) markers in mSOD1 at the presymptomatic phase, as well as diminished levels of gap junction components pannexin1 and connexin43 and expression of Ki-67 and decreased autotax-in. In addition, microglial-MN communication was negatively affected in mSOD1 mice as well as in-flammatory response. Interestingly, we observed astrocytic (S100B) and microglial (CD11b) reactivity, increased proliferation (Ki-67) and increased autotaxin expression in symptomatic mSOD1 mice. In-creased MN-microglial dialogue (CXCR3/CCL21; CX3CR1/CX3CL1; MFG-E8) and hemichannel activ-ity, namely connexin43 and pannexin1, were also observed in mSOD1 at the symptomatic phase, along with an elevated inflammatory response as indicated by increased levels of HMGB1 and NLRP3. Our results suggest that decreased autotaxin expression is a feature of the presymptomatic stage, and precede the network of pro-inflammatory-related symptomatic determinants, including HMGB1, CCL21, CX3CL1, and NLRP3. The identification of the molecules and signaling pathways that are dif-ferentially activated along ALS progression will contribute for a better design of therapeutic strategies for disease onset and progression.

For the past decade, numerous imaging techniques gave rise to remarka-ble progresses in the understanding of brain’s structure and function. Amongst the wide variety of studies onto the field of neuroscience, neuropsychiatric re-searches with resource to neuroimaging have attracted increasing attention. The present study will focus on the identification of brain areas recruited while normative subjects read sentences related to past/present or future wor-ries. Our main aim was to accurately characterize these brain areas while providing them with a time-stamp that would hopefully help us understand the implications of past/present memories and future envisioning in worrying episodes. With that purpose, functional magnetic resonance imaging data was collected from ten healthy individuals. The obtained data was processed and statistically treated using the General Linear Model and both Fixed and Ran-dom Effects Analysis for group-level results. Thereafter, a Multi-Voxel Pattern Analysis with Searchlight Mapping was performed in order to find patterns of activation that allow differentiation between conditions. The obtained results indicate higher brain activation while reading sen-tences related to past/present worries when compared to future worry or neu-tral sentences. The main areas include frontal cortex, posterior parietal, occipital and temporal areas. Worrying, per se, was characterized by activation of the medial posterior parietal cortex, left posterior occipital lobe and left central temporal lobe. With the searchlight mapping approach we were able to further identify patterns of distinction between conditions, which were located in the parietal, limbic and frontal lobes.

As bactérias redutoras de sulfato (BRS) são um grupo diversificado de micro-organismos anaeróbios que obtêm energia a partir da redução dissimilativa do sulfato. Algumas espécies de BRS possuem versatilidade a nível respiratório, como é o caso de Desulfovibrio desulfuricans ATCC 27774, devido à utilização de aceitadores finais de eletrões alternativos. Neste contexto, o objetivo da primeira parte desta dissertação consistiu na identificação das ferramentas metabólicas (proteínas) envolvidas na flexibilidade respiratória desta bacteria induzidas por diferentes aceitadores de eletrões (nitrato vs sulfato). Assim, os extratos proteicos totais de células de D. desulfuricans crescidas em meios VMN com nitrato ou sulfato, foram analisados por eletroforese bidimensional (2DE), num gradiente de pH 4 - 7. Nas condições experimentais testadas, foram observados 604 e 519 spots de proteínas nos géis de células crescidas em meio contendo nitrato ou sulfato, respetivamente. Pela avaliação estatística foi possível observar aproximadamente 25 % de spots diferenciais. Os resultados obtidos sugerem que na presença de nitrato, a bactéria não só cresce mais rapidamente e com maior rendimento, como também produz uma maior quantidade de proteínas. Estes dados foram relacionados com a adaptação de D. desulfuricans ao substrato respiratório alternativo. Tal como esperado, nos ensaios das atividades enzimáticas das redutases do nitrito e do nitrato, foi possível observar maior atividade nos extratos das células crescidas em nitrato do que em sulfato. As actinobactérias marinhas do género Salinispora, têm vindo a ser exploradas como fontes de biofármacos naturais. Assim, na segunda parte deste trabalho, realizou-se um estudo preliminar, que pretendeu caracterizar as proteínas envolvidas na biossíntese destes compostos bioactivos em S. arenicola e de S. pacifica. Para tal, foi utilizada uma abordagem proteómica diferencial, baseada em 2DE. Surpreendentemente, os perfis proteicos das duas espécies mostraram-se bastante distintos, tendo-se identificado apenas 37 spots comuns entre os 650 observados no gel de S. arenicola e 510 no gel de S. pacifica.

The focus of this Thesis was the study of the sensor domains of two heme-containing methyl-accepting chemotaxis proteins (MCP) from Geobacter sulfurreducens: GSU0582 and GSU0935. These domains contain one c-type heme, form swapped dimers with a PAS-like fold and are the first examples of a new class of heme sensors. NMR spectroscopy was used to assign the heme and polypeptide signals in both sensors, as a first step to probe conformational changes in the vicinity of the hemes. However, the presence of two conformations in solution impaired the confident assignment of the polypeptide signals. To understand how conformational changes and swapped dimerization mechanism can effectively modulate the function of the two sensor domains and their signal transduction process, the sensor domains folding and stability were studied by circular dichroism and UV-visible spectroscopy. The results showed differences in the thermodynamic stability of the sensors, with GSU0582 displaying higher structural stability. These studies also demonstrated that the heme moiety undergoes conformational changes matching those occurring at the global protein structure and that the content of intrinsically disordered segments within these proteins (25% for GSU0935; 13% for GSU0582) correlates with the stability differences observed. The thermodynamic and kinetic properties of the sensor domains were determined at different pH and ionic strength by visible spectroscopy and stopped-flow techniques. Despite the remarkably similar spectroscopic and structural features of the two sensor domains, the results showed that their properties are quite distinct. Sensor domain GSU0935 displayed more negative reduction potentials and smaller reduction rate constants, which were more affected by pH and ionic strength. The available structures were used to rationalize these differences. Overall, the results described in this Thesis indicate that the two G. sulfurreducens MCP sensor domains are designed to function in different working potential ranges, allowing this bacterium to trigger an adequate cellular response in distinct anoxic subsurface environments.

Estamos na Primavera de 2012, de visita ao museu do Quai d’Orsay, em Paris. Enquanto nos deslocamos pelo espaço da exposição permanente, deparamos com um grupo de crianças, acompanhado por duas professoras. São meninos de uma escola primária de Paris e estão de visita ao museu. Uma das professoras pede-lhes que se sentem no chão, numa das partes da longa exposição permanente, enquanto se aproxima uma mulher vestida com um traje que se identifica de maneira difusa com a América Latina. Atriz ou antropóloga? A mulher fala com os meninos da dança dos Chunchus e da Morenada bolivianas, encena, dramatiza, chama-os a participar. Ensina e representa, enquanto o público infantil colabora. Alguns meninos menos timoratos entram no jogo, dão-lhe troco, agem e atuam, exibem-se e encenam. Escapam ao papel, reinventam-no e têm de ser reencaminhados para o contexto de partida. Play e ritual: a performatividade precipita a realidade, nesta transição entre o retorno da liberdade recreativa, do jogo, da brincadeira, até à gramática do ritual, repleta de regras, de interdições, de noções de correção, de um rito (Boissevain, 1992). Transforma as relações pela sua simples enunciação, no sentido que é dado ao termo «performativo»: dizer é fazer (Austin, 1962).

The development of devices based on heterostructured thin films of biomolecules conveys a huge contribution on biomedical field. However, to achieve high efficiency of these devices, the storage of water molecules into these heterostructures, in order to maintain the biological molecules hydrated, is mandatory. Such hydrated environment may be achieved with lipids molecules which have the ability to rearrange spontaneously into vesicles creating a stable barrier between two aqueous compartments. Yet it is necessary to find conditions that lead to the immobilization of whole vesicles on the heterostructures. In this work, the conditions that govern the deposition of open and closed liposomes of 1.2-dipalmitoyl-sn-Glycero-3-[Phospho-rac-(1-glycerol)] (sodium Salt) (DPPG) onto polyelectrolytes cushions prepared by the layer-by-layer (LbL) method were analyzed. Electronic transitions of DPPG molecules as well as absorption coefficients were obtained by vacuum ultraviolet spectroscopy, while the elemental composition of the heterostructures was characterized by x-ray photoelectron spectroscopy (XPS). The presence of water molecules in the films was inferred by XPS and infrared spectroscopy. Quartz crystal microbalance (QCM) data analysis allowed to conclude that, in certain cases, the DPPG adsorbed amount is dependent of the bilayers number already adsorbed. Moreover, the adsorption kinetics curves of both adsorbed amount and surface roughness allowed to determine the kinetics parameters that are related with adsorption processes namely, electrostatic forces, liposomes diffusion and lipids re-organization on surface. Scaling exponents attained from atomic force microscopy images statistical analysis demonstrate that DPPG vesicles adsorption mechanism is ruled by the diffusion Villain model confirming that adsorption is governed by electrostatic forces. The power spectral density treatment enabled a thorough description of the accessible surface of the samples as well as of its inner structural properties. These outcomes proved that surface roughness influences the adsorption of DPPG liposomes onto surfaces covered by a polyelectrolyte layer. Thus, low roughness was shown to induce liposome rupture creating a lipid bilayer while high roughness allows the adsorption of whole liposomes. In addition, the fraction of open liposomes calculated from the normalized maximum adsorbed amounts decreases with the cushion roughness increase, allowing us to conclude that the surface roughness is a crucial variable that governs the adsorption of open or whole liposomes. This conclusion is fundamental for the development of well-designed sensors based on functional biomolecules incorporated in liposomes. Indeed, LbL films composed of polyelectrolytes and liposomes with and without melanin encapsulated were successfully applied to sensors of olive oil.

A thermal Energy Storage Unit (ESU) could be used to attenuate inherent temperature fluctuations of a cold finger, either from a cryocooler working or due to sudden income heat bursts. An ESU directly coupled to the cold source acts as a thermal buffer temporarily increasing its cooling capacity and providing a better thermal stability of the cold finger (“Power Booster mode”). The energy storage units presented here use an enthalpy reservoir based on the high latent heat of the liquid-vapour transition of neon in the temperature range 38 - 44 K to store up to 900 J, and that uses a 6 liters expansion volume at RT in order to work as a closed system. Experimental results in the power booster mode will be described: in this case, the liquid neon cell was directly coupled to the cold finger of the working cryocooler, its volume (12 cm3) allowing it to store 450 J at around 40 K. 10 W heat bursts were applied, leading to liquid evaporation, with quite reduced temperature changes. The liquid neon reservoir can also work as a temporary cold source to be used after stopping the cryocooler, allowing for a vibration-free environment. In this case the enthalpy reservoir implemented (24 cm3) was linked to the cryocooler cold finger through a gas gap heat switch for thermal coupling/decoupling of the cold finger. We will show that, by controlling the enthalpy reservoir’s pressure, 900 J can be stored at a constant temperature of 40 K as in a triple-point ESU.

Cryocoolers have been progressively replacing the use of the stored cryogens in cryogenic chains used for detector cooling, thanks to their higher and higher reliability. However, the mechanical vibrations, the electromagnetic interferences and the temperature fluctuations inherent to their functioning could reduce the sensor’s sensitivity. In order to minimize this problem, compact thermal energy storage units (ESU) are studied, devices able to store thermal energy without significant temperature increase. These devices can be used as a temporary cold source making it possible to turn the cryocooler OFF providing a proper environment for the sensor. A heat switch is responsible for the thermal decoupling of the ESU from the cryocooler’s temperature that increases when turned OFF. In this work, several prototypes working around 40 K were designed, built and characterized. They consist in a low temperature cell that contains the liquid neon connected to an expansion volume at room temperature for gas storage during the liquid evaporation phase. To turn this system insensitive to the gravity direction, the liquid is retained in the low temperature cell by capillary effect in a porous material. Thanks to pressure regulation of the liquid neon bath, 900 J were stored at 40K. The higher latent heat of the liquid and the inexistence of triple point transitions at 40 K turn the pressure control during the evaporation a versatile and compact alternative to an ESU working at the triple point transitions. A quite compact second prototype ESU directly connected to the cryocooler cold finger was tested as a temperature stabilizer. This device was able to stabilize the cryocooler temperature ((≈ 40K ±1 K) despite sudden heat bursts corresponding to twice the cooling power of the cryocooler. This thesis describes the construction of these devices as well as the tests performed. It is also shown that the thermal model developed to predict the thermal behaviour of these devices, implemented as a software,describes quite well the experimental results. Solutions to improve these devices are also proposed.
