Os estuários são ambientes costeiros semifechados, que atuam como um sítio de troca de água e de matéria. De acordo com suas características, estuários podem exportar ou importar matéria orgânica (MO). A MO é um complexo bioquímico formado por diversos compostos orgânicos, e sua quantificação é normalmente traduzida pela quantidade de carbono orgânico total (COT), seu elemento mais abundante, tanto na forma de carbono orgânico dissolvido (COD), quanto carbono orgânico particulado (COP). Determinar a origem do carbono orgânico pode auxiliar na compreensão da dinâmica da MO, podendo ser feita com auxílio de isótopos estáveis de carbono (13C) e de nitrogênio (15N). Vila Dois Rios, é um vilarejo localizado ao sul da Ilha Grande, onde se localiza o rio Barra Grande, que possui pouca intervenção antrópica. Esse é um rio de porte pequeno, com uma mata ciliar bem preservada, representada por espécies de Mata Atlântica, com presença de manguezal, ao lado está localizado o Centro de Estudos Ambientais e Desenvolvimento Sustentável (CEADS). Foram realizadas amostragens sazonais, todas em maré de sizígia, ao longo do ano de 2012, totalizando quatro coletas trimestrais. Todas as campanhas ocorreram em uma estação fixa, na foz do rio Barra Grande. A amostragem de água foi realizada em duas profundidades, superfície e fundo, coletadas em intervalos de 30 minutos, ao longo de 25 horas, para as seguintes determinações: carbono orgânico particulado (COP), carbono orgânico dissolvido (COD) e isótopos estáveis de carbono (13C) e de nitrogênio (15N). Concomitantemente, foram medidos in situ: pH, salinidade, temperatura, oxigênio dissolvido (OD), profundidade do Disco de Secchi e medidas de fluxo. Ao fim da amostragem na Ilha Grande, o material coletado foi transportado para o Laboratório de Geoquímica Orgânica Marinha (LaGOM), na UERJ, onde foi analisado posteriormente. Durante todas as campanhas, foi observada uma maré mista, predominantemente semidiurna com desigualdade. Os resultados de temperatura e salinidade evidenciaram uma estratificação vertical em todas as campanhas. Os valores medianos de MPS foram sempre maiores no fundo em relação às amostras coletadas na superfície. O carbono orgânico apresentou maiores concentrações sob a forma dissolvida do que na particulada, variando entre as profundidades e as campanhas, sendo observadas maiores concentrações medianas, tanto de COD quanto de COP, em superfície e no fundo, na campanha de verão. As concentrações medianas de Nt observadas ao longo das três campanhas foram baixas, estando relacionadas às baixas concentrações de COP. A razão molar C/N e as isotópicas indicaram que o manguezal demonstrou ser a fonte mais abundante de COP nesse estuário. Os cálculos de fluxo indicaram que o estuário tanto importou, quanto exportou material, e apenas o COD foi importado em todas as campanhas.

O presente trabalho tem o objetivo geral de estudar a permanência do positivismo criminológico no Brasil e sua parcela de contribuição para a naturalização da desigualdade característica da seletividade de nosso sistema de controle social. Tendo como pano de fundo a hegemônica ideologia da democracia racial, a pesquisa pretende afastar ocultações de harmonia racial e demonstrar como, a despeito delas, a incorporação da criminologia positivista carregou a reificação da distinção racializada no olhar para a questão criminal, se enraizando no sistema penal. Partindo de um problema presente o componente racista da seleção preferencial do sistema penal brasileiro a pesquisa busca na recuperação histórica a leitura das traduções realizadas pelos intelectuais brasileiros que, problematizando a nacionalidade e a cidadania no momento de transição representado pela Primeira República, construíram ideias sobre o crime, o criminoso e a defesa social a partir de critérios de distinção ancorados em visões racializadas, gerais e individualizantes. Para tanto, coloca-se em questão a inserção do positivismo na polícia, dentro do contexto das reformas policiais do início do século XX. Chama-se a atenção para o papel de determinados tradutores traidores do positivismo chamados de intelectuais de polícia no campo policial, assim como para a forma de introdução dessa criminologia, usando-se a ideia de luta simbólica pela mudança da prática policial. Pretendendo, desse modo, contribuir com o estudo da questão criminal e com a compreensão do sistema penal brasileiro a partir do diálogo entre criminologia, história e sociologia, as conclusões do trabalho apontam para a oportunidade da abordagem da polícia como um campo social dotado de um habitus específico. Essa visão possibilita, por um lado, a interpretação das pretensões modernizantes dos intelectuais de polícia alinhadas com o pensamento positivista como pressões sobre as estruturas desse campo, passíveis de rejeição e de retradução no interior desse mundo social específico. De outro, admitindo-se a possibilidade de tensionamento e reestruturação do habitus policial como resultado dessas demandas externas, pode-se encontrar nessas disposições duráveis elementos da permanência do positivismo que imprimiram na prática policial as desigualdades ocultadas pelo mito da democracia racial.

One of the avenues through which the Government objective of poverty eradication in Uganda can be achieved is Fisheries development and management. Up to 20% of Uganda’s surface area is covered by aquatic systems i.e. lakes, rivers, streams and swamps and to a large extent, all these are interconnected. The large lakes: Victoria, Albert, Kyoga, George and Edward are sites of the more important commercial fisheries, but even the smaller water bodies, rivers (e.g. the Rivers Nile and Kagera) and the surrounding swamps provide sources of livelihood to rural areas. Fish is an important source of high quality food, employment revenue and is currently the second most important export commodity next to coffee generating approximately US $ 80 million annually. Fish exports to regional markets are worth at least US $ 20 million annually. Fish flesh is rich in proteins, which are superior to those of beef and poultry. Fish flesh contains an anticholesterol which assists in reducing heart diseases. Some fishes are of medicinal value e.g. haplochromines (Nkejje) are used to treat measles. Most of the fish in Uganda is got from lakes Victoria, Kyoga, Albert and Albert Nile, Edward and George production systems as well as from the 160 minor lakes and rivers and the associated wetland systems. Capture fisheries based in these systems contribute up to 99% of the fish production in Uganda but aquaculture is also picking up. The fishing industry employs up to one million Ugandans

About 18% of Uganda’s surface area is covered with water from which 300,000 metric tonnes of fish are produced. Fish are currently the second most important export commodity generating approximately US$100 million. Fish provides 50% of protein diet for the 20 million people translating into per capita consumption of 12 kg. Close to the production system, this figure rises to 50 – 100 kg. It is estimated that fishery-related activities employ at least one million people countrywide (i.e. 5% of the population). Fish is an important source of high quality food, employment, and revenue and it is currently the second most important export commodity next to coffee generating approximately US $ 80 million annually. Fish exports to regional markets are worth at least US $ 20 million annually. Fish flesh is rich in proteins, which are superior to those of beef and poultry. Fish flesh contains an anticholesterol which assists in reducing heart diseases. Some fishes are of medicinal value e.g. haplochromines (Nkejje) are used to treat measles. The specific plasminogen activator from Trimeresurus stejnegeri venom (TSV-PA) is a serine proteinase presenting 23% sequence identity with the proteinase domain of tissue type plasminogen activator, and 63% with batroxobin, a fibrinogen clotting enzyme from Bothrops atrox venom that does not activate plasminogen. TSV-PA contains six disulfide bonds and has been successfully overexpressed in Escherichia coli (Zhang, Y., Wisner, A., Xiong, Y. L,, and Bon, C, (1995) J. Biol. Chem. 270, 10246-10255), To identify the functional domains of TSV-PA, we focused on three short peptide fragments of TSV-PA showing important sequence differences with batroxobin and other venom serine proteinases. Molecular modeling shows that these sequences are located in surface loop regions, one of which is next to the catalytic site, When these sequences were replaced in TSV-PA by the equivalent batroxobin residues none generated either fibrinogen-clotting or direct fibrinogenolytic activity, Two of the replacements had little effect in general and are not critical to the specificity of TSV-PA for plasminogen. Nevertheless, the third replacement, produced by the conversion of the sequence DDE 96a-98 to NVI, significantly increased the K-m for some tripeptide chromogenic substrates and resulted in undetectable plasminogen activation, indicating the key role that the sequence plays in substrate recognition by the enzyme.

Fish diversity in Nira River in Pune District has been studied. The study revealed the presence of 24 species of fish belonging to eight orders (Cypriniformes, Siluriformes, Perciformes, Osteoglossiformes, Synbranchiformes, Clupeiformes, Mugiliformes and Aulopiformes). The predominant orders of fishes in this area (Sangavi) are Cypriniformes, Siluriformes and Perciformes. The highest number of ten species was recorded in the order Cypriniformes. The fishes recorded were found to be widely distributed and were present in good numbers in the river.

Sea cucumbers belong to phylum Echinodermata, order Holothuroidea are an abundant and diverse group of Invertebrates, with over 1400 species occuring from the intertidal to the deepest oceanic trenches. Sea cucumbers are important components of the food chain in temperate and coral reef ecosystems and they play an important role as deposite feeders and suspension feeders. Rapid decline in populations may have serious consequences for the survival of other species that are part of the same complex food web,as the eggs, larve and juveniles constitute an important food source for the other marine species including crustaceans, fish and mollusks. In addition sea cucumbers are often called the earthworms of the sea, because they are responsible for the extensive shifting and mixing of the substrate, and recycling of detrital matter. Sea cucumbers consume and grind sediment and organic material into finer particles , turning over the top layers of sediment in lagoons , reefs and other habitats and allowing the penetration of oxygen. While the taxonomy of the holothurian families is generally well known , the distinction of similar species is difficult. There are relatively few holothurian taxonomist.Most sea cucumber species can be identified by Holothurin taxonomists by using the calcareous skeletal ossicles found in the body wall. In this study , at first a sea cucumber from Kish island in Persian gulf has recognized. Individuals collected from west and east extend far away into north and south of coral reefs by diving. I have checked them morphologically and anatomically.Then with key to the orders of the Holothuroidea, They belong to the Aspidochirotida with key to the families of Aspidochirotida, they were in Stichopodidae families and with key to the genus of Stchopodidae, they were Stichopus. Then ossicles were extracted at National Museum of Natural History, by Dr David Pawson. The ossicles were measured on a transect across a slide prepared from the mid-dorsal region of each specimen.The one we have in the shallow waters of Kish island, is Stichopus hermanni, a massive holothurian, body broad, considerably flattened ventraly ,the dorsal side slightly arched and the lateral sides almost vertical; body wall fairy thick and soft ; mouth subterminal; anus central; tentacles usually 20 in number of length and leaf shaped. Numerous ossicles consisting of table with large discs having usually 7 to 15 peripheral holes, but often irregular or incomplete and spire of moderate height ending in a group of spinelets, rosettes of variable development, and c-shaped rods. Color (exept papillae)partly remained after preservation in alcohol which is found at the depth of 4 to 8 meters, on coral reef. Furthermore, the sexual reproductive cycle was described using standard methods. Gonads were removed and transferred to Bouin's fixative for four weeks and then processed according to standard embedding technique. To prevent the loss of tubule contents during embedding, the tubule sections, were cut well beyond the segment selected for sectioning. For each individual, six sections, each section with 5µm diameter by microtome were cut from tubules. These sections were first placed on gelatin coated slides (the gelatin was heated to 42°c) and then transferred to the oven at 37°c for one hour. This technique usually prevents the fragil tubules from breaking and the loss of gametes. The slides were stained with Eosin and Hematoxylin, and good resolution of the various cell types achieved.A second series of slides was stained with the Periodic Acid Schiff(PAS) to identify polysaccharides(glycogen). Monthly sampling was occurred.The sexual reproductive cycle was defined through the combined use of these criteria: Monthly percentages of the gonad stages for each sex, the monthly gonad index (GI) , given as the ratio of the wet gonad weight (G) to the dray weight (DW)and the monthly percentage of individuals that undetermined sex. The gonad consists of two tufts of tubules on which saccules develop. Gonadal development was classified into five stages: post spawning, recovery, growth, advanced growth, and mature stage that were adapted from the earlier studies of holothurians. Histological preparations showed that the sex of larger individuals could be identified by the presence of oogonia and young oocytes in females, and spermatogonic stages in males.The mean diameter of the tubules and gonadal mass follow annual cycles, increasing from late winter through spring, and dropping abruptly after spawning in the summer. Gametogenesis is generally a prolongate process and begins in March. By summer the ovarian tubules contain oocytes with diameter of 120-240 pm and the testicular tubules contain an abundance of spermatozoa (diameter 5-6 gm ).Following spawning the predominant activity within the spent tubules is phagocytosis of the residual gamets.The active phase of gametogenesis (March to July), coincides with an increasing photoperiod regim, and an accelerated gametogenesis occurs in July when temperature is high. Throughout the year, the gonad of Stichopus hermanni is larger in males than in females, and this is due to the number of tubules in the testis rather than to tubules length or diameter.

The formation energies of the oxygen vacancy and titanium interstitial in rutile TiO 2 were calculated by the screened-exchange (sX) hybrid density functional method, which gives a band gap of 3.1 eV, close to the experimental value. The oxygen vacancy gives rise to a gap state lying 0.7 eV below the conduction band edge, whose charge density is localized around the two of three Ti atoms next to the vacancy. The Ti interstitial (Ti int) generates four defect states in the gap, whose unpaired electrons lie on the interstitial and the adjacent Ti 3d orbitals. The formation energy for the neutral oxygen vacancy is 1.9 eV for the O-poor chemical potential. The neutral Ti interstitial has a lower formation energy than the O vacancy under O-poor conditions. This indicates that both the O vacancy and Ti int are relevant for oxygen deficiency in rutile TiO 2 but the O vacancy will dominate under O-rich conditions. This resolves questions about defect localization and defect predominance in the literature. © 2012 American Physical Society.

Jacked piles are becoming a valuable installation method due to the low noise and vibration involved in the installation procedure. Cyclic jacking may be used in an attempt to decrease the required installation force. Small scale models of jacked piles were tested in sand and silt in a 10 m beam centrifuge. Two different piles were tested: smooth and rough. Piles were driven in two ways with monotonic and cyclically jacked installations. The cyclically jacked installation involves displacement reversal at certain depth for a fixed number of cycles. The depth of reversal and amplitude of the cycle vary for different tests. Data show that the base resistance increases during cyclic jacking due to soil compaction at the pile toe. On the other hand, shaft load decreases with the number of cycles applied due to densification of soil next to the pile shaft. Cyclic jacking may be used in unplugged tubular piles to decrease the required installation load. © 2013 Taylor & Francis Group, London.

Nanoindentation is ideal for the characterization of inhomogeneous biological materials. However, the use of nanoindentation techniques in biological systems is associated with some distinctively different techniques and challenges. For example, engineering materials used in the microelectronics industry (e.g. ceramics and metals) for which the technique was developed, are relatively stiff and exhibit time-independent mechanical responses. Biological materials, on the other hand, exhibit time-dependent behavior, and can span a range of stiffness regimes from moduli of Pa to GPa - eight to nine orders of magnitude. As such, there are differences in the selection of instrumentation, tip geometry, and data analysis in comparison with the "black box" nanoindentation techniques as sold by commercial manufacturers. The use of scanning probe equipment (atomic force miscroscopy) is also common for small-scale indentation of soft materials in biology. The book is broadly divided into two parts. The first part presents the "basic science" of nanoindentation including the background of contact mechanics underlying indentation technique, and the instrumentation used to gather mechanical data. Both the mechanics background and the instrumentation overview provide perspectives that are optimized for biological applications, including discussions on hydrated materials and adaptations for low-stiffness materials. The second part of the book covers the applications of nanoindentation technique in biological materials. Included in the coverage are mineralized and nonmineralized tissues, wood and plant tissues, tissue-engineering substitute materials, cells and membranes, and cutting-edge applications at molecular level including the use of functionalized tips to probe specific molecular interactions (e.g. the ligand-receptor binding). The determination of lacunar-canalicular permeability is essential for understanding local fluid flow in bone, which may indicate how bone senses changes in the mechanical environment to regulate mechano-adaptation. The estimates of lacunar-canalicular permeability found in the literature vary by up to eight orders of magnitude, and age-related permeability changes have not been measured in non-osteonal mouse bone. The objective of this study is to use a poroelastic approach based on nanoindentation data to characterize lacunar-canalicular permeability in murine bone as a function of age. Nine wild type C57BL/6 mice of different ages (2, 7 and 12 months) were used. Three tibiae from each age group were embedded in epoxy resin, cut in half and indented in the longitudinal direction in the mid-cortex using two spherical fluid indenter tips (R=238 μm and 500 μm). Results suggest that the lacunar-canalicular intrinsic permeability of mouse bone decreases from 2 to 7 months, with no significant changes from 7 to 12 months. The large indenter tip imposed larger contact sizes and sampled larger ranges of permeabilities, particularly for the old bone. This age-related difference in the distribution was not seen for indents with the smaller radius tip. We conclude that the small tip effectively measured lacunar-canalicular permeability, while larger tip indents were influenced by vascular permeability. Exploring the age-related changes in permeability of bone measured by nanoindentation will lead to a better understanding of the role of fluid flow in mechano-transduction. This understanding may help indicate alterations in bone adaptation and remodeling.

The paper describes the rapid and label-free detection of the white spot syndrome virus (WSSV) using a surface plasmon resonance (SPR) device based on gold films prepared by electroless plating. The plating condition for obtaining films suitable for SPR measurements was optimized. Gold nanoparticles adsorbed on glass slides were characterized by transmission electron microscopy (TEM). Detection of the WSSV was performed through the binding between WSSV in solution and the anti-WSSV single chain variable fragment (scFv antibody) preimmobilized onto the sensor surface. Morphologies of the as-prepared gold films, gold films modified with self-assembled alkanethiol monolayers, and films covered with antibody were examined using an atomic force microscope (AFM). To demonstrate the viability of the method for real sample analysis, WSSV of different concentrations present in a shrimp hemolymph matrix was determined upon optimizing the surface density of the antibody molecules. The SPR device based on the electroless-plated gold films is capable of detecting concentration of WSSV as low as 2.5 ng/mL in 2% shrimp hemolymph, which is one to two orders of magnitude lower than the level measurable by enzyme-linked immunosorbant assays. Ultrasonic solvent extraction combined with solid-phase microextraction (SPME) with calix[4]arene/hydroxy-terminated silicone (C[4]/OHTSO) oil coated fiber was used to extract phthalate acid esters (PAEs) plasticizers in plastic, such as blood bags, transfusion tubing, food packaging bag, and mineral water bottle for analysis by gas chromatography (GC). Both extraction parameters (i.e. extraction time, extraction temperature, ionic strength) and conditions of the thermal desorption in a GC injector were optimized by analysis of eight phthalates. The fiber shows wonderful sensitivity and selectivity to the tested compounds. Owing to its high thermal stability (380 degreesC), the carryover effect that often encountered when using conventional fibers can be reduced by appropriately enhancing the injector temperature. The method showed linear response over two to four orders of magnitude with correlation coefficients (r) better than 0.996, and limits of detection (LOD) ranged between 0.006 and 0.084 mug l(-1). The relative standard deviation values obtained were less than or equal to 10%. bis-2-Ethylhexyl phthalate (DEHP) was the sole analyte detected in these plastics and recoveries were in the ranges 95.5-101.4% in all the samples. Nematoda is a metazoan group with extremely high diversity only next to Insecta. Caenorhabditis elegans is now a favorable experimental model animal in modern developmental biology, genetics and genomics; studies. However, the phylogeny of Nematoda and the phylogenetic position of the phylum within animal kingdom have long been in debate. Recent molecular phylogenetic studies gave great challenges to the traditional nematode classification. The new phylogenies not only placed the Nematoda in the Ecdysozoan and divided the phylum into five clades, but also provided new insights into animal molecular identification and phylogenetic biodiversity studies. The present paper reviews major progress and remaining problems in the current molecular phylogenetic studies of Nematoda, and prospects the developmental tendencies of this field.
