Os experimentos foram conduzidos para avaliar a digestibilidade ileal (CDILEAL) e total (CDTOTAL) do amido e a atividade das enzimas amilase (ATAM) e maltase (ATMAL) em coelhos de 35 e 45 dias alimentados com amido proveniente do milho ou da raspa de mandioca. em cada ensaio foram utilizados 20 coelhos da raça Nova Zelândia Branco, sendo dez machos e dez fêmeas, alojados individualmente em gaiolas de arame e distribuídos em um delineamento inteiramente casualizado. Ao final do período experimental, os animais foram abatidos para retirada de uma amostra do conteúdo digestivo de uma porção do jejuno e do íleo. Os CDILEAL e CDTOTAL do amido da raspa de mandioca (59,25±1,38% e 99,89±0,04%, respectivamente) foram superiores ao do milho (54,21±1,51% e 99,21±0,05%, respectivamente). O CDTOTAL dos animais com 45 dias de idade foi superior ao dos com 35 dias (99,75±0,04% vs 99,35±0,05%). A ATAM, no conteúdo do jejuno e do íleo, foi maior nos animais com 45 dias (0,952±0,118 mmol de glicose/mg/min e 0,647±0,093 mmol de glicose/mg/min, respectivamente) do que nos de 35 (0,469±0,111 mmol de glicose/mg/min e 0,375±0,088 mmol de glicose/mg/min, respectivamente). A ATMAL foi maior nos animais alimentados com raspa de mandioca do que nos que receberam milho (0,061±0,007 mmol de glicose/mg/min vs 0,032±0,007 mmol de glicose/mg/min). Conclui-se que o amido da raspa de mandioca é mais digestível que o do milho, e o sistema enzimático dos coelhos com 35 dias de idade ainda não está suficientemente desenvolvido para que ocorra otimização da digestão do amido.

Eukaryotic translation initiation factor 5A (eIF5A) is the only cellular protein that contains the polyamine-modified lysine, hypusine [N(epsilon)-(4-amino-2-hydroxybutyl)lysine]. Hypusine occurs only in eukaryotes and certain archaea, but not in eubacteria. It is formed post-translationally by two consecutive enzymatic reactions catalyzed by deoxyhypusine synthase (DHS) and deoxyhypusine hydroxylase (DOHH). Hypusine modification is essential for the activity of eIF5A and for eukaryotic cell proliferation. eIF5A binds to the ribosome and stimulates translation in a hypusine-dependent manner, but its mode of action in translation is not well understood. Since quantities of highly pure hypusine-modified eIF5A is desired for structural studies as well as for determination of its binding sites on the ribosome, we have used a polycistronic vector, pST39, to express eIF5A alone, or to co-express human eIF5A-1 with DHS or with both DHS and DOHH in Escherichia coli cells, to engineer recombinant proteins, unmodified eIF5A, deoxyhypusine- or hypusine-modified eIF5A. We have accomplished production of three different forms of recombinant eIF5A in high quantity and purity. The recombinant hypusine-modified eIF5A was as active in methionyl-puromycin synthesis as the native, eIF5A (hypusine form) purified from mammalian tissue. The recombinant eIF5A proteins will be useful tools in future structure/function and the mechanism studies in translation.

In this work we investigate the possible toxicity of vanadyl sulfate (VOSO4), a compound capable of reducing hyperglycemia, on the following serum enzymes of diabetic young rats: alanine aminotransferase (ALT), aspartate aminotransferase (AST), lactate dehydrogenase (LD) and creatine kinase (CK), as well as its effects on serum lipids. We find that at a concentration of 1 mg/mL VOSO4 has no toxic effect on the liver and muscles of diabetics young rats. These findings suggest that VOSO4 may be an alternative to insulin in the near future, due to its low cost, low toxicity and ready availability.

The digestive tract of insects is an important natural, physical, and chemical defense barrier against pathogen invasion. Certain. lepidopteran caterpillars are serious pests of agricultural crops and their biology has received much attention, but little is known about the larval noctuid gut. The morphological analysis of the digestive tract in Anticarsia gemmatalis under scanning electron microscopy (SEM) is a good model for studies about its defense mechanism. The material was fixed (2,5% glutaraldehyde solution; 0.1 M-phosphate buffer, pH 7.3), post-fixed (1% osmium tetroxide in the same buffer), dried at critical point, gold coated and analyzed in a SEM 515-Philips. A. gemmatalis digestive tract consists of a straight duct of varying length and diameter, subdivided in three main regions: the foregut formed by the oral cavity, pharynx, esophagus, and crop; the midgut that is the largest portion of the digestive tract without noticeable morphological differentiation along its length; and the hindgut that is morphologically differentiated in pylorus, ileum, colon, and rectum. Although the general morphology of the A. gemmatalis digestive tract is quite similar to the other Lepidoptera species, the anatomical array of the crop muscular layers is quite different comparing with the description for other larval insect.

The metabolic switch From C-3-photosynthesis to crassulacean acid metabolism (CAM),and the antioxidative response of Mesembryanthemum crystallinum L. plants cultured under severe salt stress and high light intensities, and a combination of booth stress conditions, were studied. High light conditions led to a more rapid CAM induction than salinity. The induction time was still shortened when both stress factors were combined. A main pattern observed in CAM plants was a decrease in mitochondrial Mn-superoxide dismutase (SOD) activity during the day. The activities of the chloroplastic Fe-SOD and cytosolic CuZn-SOD were increased due to salt treatment after a lag phase, while catalase activity was decreased. Combination of salt and light stress did not lead to a higher SOD activity as found after application of one stress factor alone, indicating that there is a threshold level of the oxidative stress response. The fact that salt-stressed plants grown under high light conditions showed permanent photoinhibition and lost the ability for nocturnal malate storage after 9 d of treatment indicate serious malfunction of metabolism, leading to accelerated senescence. Comparison of CuZn-SOD activity with CuZn-SOD protein amount, which was determined immunologically, indicates that the activity of the enzyme is at least partially post-translationally regulated.

P>The use of enzymes in juice industry has contributed in increasing the yield and production of various types of juices. The addition pectinases aims in particular to degrade the pectic substances, in the cell wall and middle lamella of the cells of plants, aiming to minimise the impacts of these compounds on the characteristics of the final product, such as colour, turbidity and viscosity. Enzymes able to remove bitterness of citrus juice, extract pigments, among other applications, have also had great interest in the juice industry.

Aim: Studies on feeding of early life stages are very important to the understanding of the biology and trophic ecology of fish species. Therefore, the aim of this work is to describe the development of the digestive tube, and to characterize the diet of larvae and juveniles of Bryconamericus stramineus Eigenmann, 1908 of the upper Parana River floodplain; Methods: Larvae were obtained from, monthly samples during nychthemeral cycles with four-hour interval between samplings from February/91 to February/92, utilizing a conical-cylindrical plankton net; Results: At the preflexion stage, larvae at approximately 4.00 mm SL, showed a morphologically undifferentiated straight tube, with the anterior region more dilated. At the flexion stage a differentiation in the anterior region of the digestive tube occurs, with the intestine wall getting thicker (8.30 mm SL). The formation of pyloric caeca occurs at 9.00 mm SL. At the postflexion stage the first loop is formed at 9.25 mm SL and the second loop at about 10.00 mm SL. Bryconamericus stramineus consumed mainly cladocerans, also ingesting copepods, rotifers, nematodes, algae, insects and inorganic particles. Along the development, there was an increase in the number of food items and a diversification in the number of consumed taxa; Conclusions: Changes in the diet of B. stramineus larvae were not observed, being them zooplanktivores during all the initial development. However, an increase of large preys, such as insects larvae, was observed at the end of the larval period and in juveniles, suggesting a tendency towards invertivory. Larvae and juveniles preferentially fed during the night.
