Mon mémoire de maîtrise a été réalisé dans le cadre du projet Génorem (www.genorem.ca), un projet multidisciplinaire qui réunit différents chercheurs de l'Université de Montréal et de l'Université McGill dans le but d'améliorer les techniques utilisées en bioremédiation. Dans le cadre de l'étude, des saules à croissance rapide (Salix sp.) ont été utilisés comme plantes modèles dans l'étude. Ainsi, 11 cultivars de saule ont été suivis afin de déterminer leur potentiel à produire un bon rendement de biomasse, à tolérer des conditions de stress sévère causé par la présence de HAPs (hydrocarbures aromatiques polycycliques) , BPCs (biphényles polychlorés) et d'hydrocarbures pétroliers C10-C50. L'expérimentation consistait en une plantation de saule à forte densité qui a été mise en place en 2011 sur le site d'une ancienne industrie de pétrochimie à Varennes, dans le sud du Québec. Les boutures des génotypes sélectionnés ont été plantées sur une superficie d'environ 5000 m2. Les plantes ont été suivies pendant les deux saisons de croissance suivant le recépage et une série de paramètres de croissance et de mesures physiologiques ont été récoltés (surface foliaire, taux de chlorophylle, conductance stomatique et statut nutritionnel) dans le but d'évaluer et de comparer les performances de chaque génotype sur un sol pollué. Les analyses statistiques ont démontré que le cultivar S. miyabeana (SX61) était le meilleur producteur de biomasse sur le site contaminé, tandis que S. nigra (S05) et S. acutifolia (S54) présentaient la meilleure capacité photosynthétique. S. dasyclados (SV1), S. purpurea (‘Fish Creek’) et S. caprea (S365) ont semblé particulièrement affectés par la présence de contaminants. La capacité d'établissement et la croissance de S. nigra (S05), S. eriocephala (S25) and S. purpurea x S. miyabeana (‘Millbrook’) indiquent une tolérance globale supérieure à la pollution . Cette analyse comparative des différentes réponses physiologiques des saules cultivés sur un sol contaminé pourra guider le processus de sélection de plantes et les techniques de bioremédiation dans les futurs projets de phytoremédiation.

There exists a need for potential microorganism that could facilitate effective bioremediation of crude oil pollutants in the environment. Hence it was desired to isolate a potential bacterium from marine sediment, which often experiences oil pollution and develop a bioprocess for crude oil biodegradation. In the present study the sediment deposits in the beach of Munakkal, Trichur dist, Kerala, collected immediately after the major event Tsunami in 2004 was collected and analyzed by enrichment culture technique towards isolation of potential strains that could degrade crude oil and its fractions. From the results obtained it was found that the sediment deposits harbor several bacteria with potential for degrading hydrocarbons. However, among the strains obtained, isolate no. BTTS 10 showed capabilities for utilizing both alkanes and aromatic hydrocarbons and hence the same was selected for further studies.

An understanding of the primary pathways of plant uptake of organic pollutants is important to enable the risks from crops grown on contaminated soils to be assessed. A series of experiments were undertaken to quantify the importance of the pathways of contamination and the Subsequent transport within the plant using white clover plants grown in solution culture. Root uptake was primarily an absorption process, but a component of the contamination was a result of the transpiration flux to the shoot for higher Solubility compounds. The root contamination can be easily predicted using a simple relationship with K-OW, although if a composition model was used based on lipid content, a significant under prediction of the contamination was observed. Shoot uptake was driven by the transpiration stream flux which was related to the solubility of the individual PAH rather than the K-OW. However, the experiment was over a short duration, 6 days, and models based on K-OW may be better for crops grown in the field where the vegetation will approach equilibrium and transpiration cannot easily be measured, A significant fraction of the shoot contamination resulted from aerial deposition derived from volatilized PAH. This pathway was more significant for compounds approaching log K-OA > 9 and log K-AW < -3. The shoot uptake pathways need further investigation to enable them to be modeled separately, There was no evidence of significant systemic transport of the PAR so transfer outside the transpiration stream is likely to be limited.

Here we present the catalytic hydrogenation of polycyclic aromatic hydrocarbons (PAHs) to less toxic mixtures of saturated and partial unsaturated polycyclic hydrocarbons under mild reaction conditions using a magnetically recoverable rhodium catalyst and molecular hydrogen as the exclusive H source. The catalyst is easily recovered after each reaction by placing a permanent magnet on the reactor wall and it can be reused in successive runs without any significant loss of catalytic activity. As an example, anthracene was totally converted into the saturated polycyclic hydrocarbon form (ca. 60%) and the partially hydrogenated form, 1,2,3,4,5,6,7,8-octahydroanthracene (ca. 40%). The catalyst operates in a broad range of temperature and H(2) pressure in both organic and aqueous/organic solutions of anthracene and it also exhibits significant activity at low substrate concentrations (20 ppm). This can be an efficient recycling process for hydrogenation of PAHs present in contaminated fluid waste streams. (C) 2009 Elsevier B.V. All rights reserved.

The [Ru(3)O(H(3)CCO(2))(6)(py)(2)(L)]PF(6) clusters, where L=methanol or dimethyl sulfoxide, can be activated by peroxide or oxygen donor species, such as tert-butyl hydroperoxide (TBHP) or iodosylbenzene (PhIO), respectively, generating reactive intermediates of the type [Ru(3)(IV,IV,III)=0](+). In this way, they catalyse the oxidation of cyclohexane or cyclohexene by TBHP and PhIO, via oxygen atom transfer, rather than by the alternative oxygen radical mechanism characteristic of this type of complexes. In addition to their ability to perform efficient olefin epoxydation catalysis, these clusters also promote the cleavage of the C-H bond in hydrocarbons, resembling the oxidation catalysis by metal porphyrins. A simple and fast multiresidue method has been developed to determine 48 pesticides within the major groups of pesticides (organohalogen, organophosphorous, pyrethroids and organonitrogen) in representative samples of locally produced honey, in Bauru (State of São Paulo, Brazil) during 2003-2004. The recovery results found ranged from 76% to 95% and the limits of detection were lower than 0.01 mg/kg for gas chromatography with electron impact mass spectrometric detection in the selected ion monitoring mode (GC-MS-SIM). The results indicated that most pesticides found in the samples belonged to the organohalogen and organophosphorous groups and lower levels of residues of some organonitrogen and pyretroids were also detected. Malathion residues were detected in all the samples, in a high concentration, owing to its applications to control dengue mosquitoes in the area studied. (c) 2005 Elsevier Ltd. All rights reserved.

Chromosomal aberration (CA) assays have been widely used, not only to assess the genotoxic effects of chemical agents, but also to evaluate their action mechanisms on the genetic material of exposed organisms. This is of particular interest, since such analyses provide a better knowledge related to the action of these agents on DNA. Among test organisms, Allium cepa is an outstanding species due to its sensitivity and suitable chromosomal features, which are essential for studies on chromosomal damage or disturbances in cell cycle. The goal of the present study was to analyze the action mechanisms of chemical agents present in petroleum polluted waters. Therefore, CA assay was carried out in A. cepa meristematic cells exposed to the Guaeca river waters, located in the city of Sao Sebastiao, SP, Brazil, which had its waters impacted by an oil pipeline leak. Analyses of the aberration types showed clastogenic and aneugenic effects for the roots exposed to the polluted waters from Guaeca river, besides the induction of cell death. Probably all the observed effects were induced by the petroleum hydrocarbons derived from the oil leakage. (C) 2008 Elsevier B.V. All rights reserved.

Groundwater contamination with benzene, toluene, ethylbenzene and xylene (BTEX) has been increasing, thus requiring an urgent development of methodologies that are able to remove or minimize the damages these compounds can cause to the environment. The biodegradation process using microorganisms has been regarded as an efficient technology to treat places contaminated with hydrocarbons, since they are able to biotransform and/or biodegrade target pollutants. To prove the efficiency of this process, besides chemical analysis, the use of biological assessments has been indicated. This work identified and selected BTEX-biodegrading microorganisms present in effluents from petroleum refinery, and evaluated the efficiency of microorganism biodegradation process for reducing genotoxic and mutagenic BTEX damage through two test-systems: Allium cepa and hepatoma tissue culture (HTC) cells. Five different non-biodegraded BTEX concentrations were evaluated in relation to biodegraded concentrations. The biodegradation process was performed in a BOO Trak Apparatus (HACH) for 20 days, using microorganisms pre-selected through enrichment. Although the biodegradation usually occurs by a consortium of different microorganisms, the consortium in this study was composed exclusively of five bacteria species and the bacteria Pseudomonas putida was held responsible for the BTEX biodegradation. The chemical analyses showed that BTEX was reduced in the biodegraded concentrations. The results obtained with genotoxicity assays, carried out with both A. cepa and HTC cells, showed that the biodegradation process was able to decrease the genotoxic damages of BTEX. By mutagenic tests, we observed a decrease in damage only to the A. cepa organism. Although no decrease in mutagenicity was observed for HTC cells, no increase of this effect after the biodegradation process was observed either. The application of pre-selected bacteria in biodegradation processes can represent a reliable and effective tool in the treatment of water contaminated with BTEX mixture. Therefore, the raw petroleum refinery effluent might be a source of hydrocarbon-biodegrading microorganisms. (c) 2010 Elsevier B.A. All rights reserved.
