Ozone, first discovered in the mid 1800’s, is a triatomic allotrope of oxygen that is a powerful oxidant. For over a century, research has been conducted into the synthetic application and mechanism of reactions of ozone with organic compounds. One of the major areas of interest has been the ozonolysis of alkenes. The production of carbonyl compounds is the most common synthetic application of ozonolysis. The generally accepted mechanism developed by Rudolf Criegee for this reaction involves the 1,3-electrocyclic addition of ozone to the π bond of the alkene to form a 1,2,3-trioxolane or primary ozonide. The primary ozonide is unstable at temperatures above -100 °C and undergoes cycloreversion to produce the carbonyl oxide and carbonyl intermediates. These intermediates then recombine in another 1,3-electrocyclic addition step to form the 1,2,4-trioxolane or final ozonide. While the final ozonide is often isolable, most synthetic applications of ozonolysis require a subsequent reductive or oxidative step to form the desired carbonyl compound. During investigations into the nucleophilic trapping of the reactive carbonyl oxide, it was discovered that when amines were used as additives, an increased amount of reaction time was required in order to consume all of the starting material. Surprisingly, significant amounts of aldehydes and a suppression of ozonide formation also occurred which led to the discovery that amine N-oxides formed by the ozonation of the amine additives in the reaction were intercepting the carbonyl oxide. From the observed production of aldehydes, our proposed mechanism for the in situ reductive ozonolysis reaction with amine N-oxides involves the nucleophilic trapping of the carbonyl oxide intermediate to produce a zwitterionic adduct that fragments into 1O2, amine and the carbonyl thereby avoiding the formation of peroxidic intermediates. With the successful total syntheses of peroxyacarnoates A and D by Dr. Chunping Xu, the asymmetric total synthesis of peroxyplakorate A3 was investigated. The peroxyplakoric acids are cyclic peroxide natural products isolated from the Plakortis species of marine sponge that have been found to exhibit activity against malaria, cancer and fungi. Even though the peroxyplakorates differ from the peroxyacarnoates in the polyunsaturated tail and the head group, the lessons learned from the syntheses of the peroxyacarnoates have proven to be valuable in the asymmetric synthesis of peroxyplakorate A3. The challenges for the asymmetric synthesis of peroxyplakorate A3 include the stereospecific formation of the 3-methoxy-1,2-dioxane core with a propionate head group and the introduction of oxidation sensitive dienyl tail in the presence of a reduction sensitive 1,2-dioxane core. It was found that the stereochemistry of two of the chiral centers could be controlled by an anti-aldol reaction of a chiral propionate followed by the stereospecific intramolecular cyclization of a hydroperoxyacetal. The regioselective ozonolysis of a 1,2-disubstituted alkene in the presence of a terminal alkyne forms the required hydroperoxyacetal as a mixture of diastereomers. Finally, the dienyl tail is introduced by a hydrometallation/iodination of the alkyne to produce a vinyl iodide followed by a palladium catalyzed coupling reaction. While the coupling reaction was unsuccessful in these attempts, it is still believed that the intramolecular cyclization to introduce the 1,2-dioxane core could prove to be a general solution to many other cyclic peroxides natural products.

Black fungi are able to adapt to extreme environmental conditions, such as: high temperatures, the presence of toxic chemical substances and lack of nutrients. Besides, they are also potential pathogens to humans. The natural environment of many black fungi is still unknown and some studies are being conducted to evaluate the biodiversity of this group and their different habitats. This study aimed to isolate black fungi in domestic environments and facilities, such as toothbrushes, fridge sealing rubbers, bathroom strainers and divisions, windows, wall tiles and bath sponge. For the collection, material surfaces were scratched with a scalpel and the resulting fragments were sewed in Mycosel agar (DifcoTM), supplemented with actidione to inhibit the growth of highly-sporulating fungi. Plates were incubated at 25ºC for three weeks. The 46 isolated fungi were maintained on MA2% slants at 8ºC and cryopreserved at -80ºC. Fungal identification was performed through the analysis of macro and microscopic features and ITS rDNA sequencing. The following black fungi taxa were found: Ascomycota sp., Cladosporium spp., Dothideomycete sp., Exophiala alcalophila, Ochroconis mirabilis and Rhinocladiella atrovirens. Non-melanized fungi were also found, such as Geosmithia sp., Penicillium sp. and Rhodotorula mucilaginosa. The temperature tests showed that isolated black fungi were not able to grow at 37°C, however, this temperature proved to be fungistatic to 43% of them. According to literature, all black fungi isolated in this study are opportunistic pathogens and additional studies are necessary to evaluate the risk that these micro-organisms offer to health, once they were isolated from domestic environments

This work investigated the effect of gallium arsenide (GaAs) irradiation (power: 5 mW; intensity: 77.14 mW/cm(2), spot: 0.07 cm(2)) on regenerating skeletal muscles damaged by crotoxin (CTX). Male C57Bl6 mice were divided into six groups (n = 5 each): control, treated only with laser at doses of 1.5 J or 3 J, CTX-injured and, CTX-injured and treated with laser at doses of 1.5 J or 3 J. The injured groups received a CTX injection into the tibialis anterior (TA) muscle. After 3 days, TA muscles were submitted to GaAs irradiation at doses of 1.5 or 3 J (once a day, during 5 days) and were killed on the eighth day. Muscle histological sections were stained with hematoxylin and eosin (H&E) in order to determine the myofiber cross-sectional area (CSA), the previously injured muscle area (PIMA) and the area density of connective tissue. The gene expression of MyoD and myogenin was detected by real-time PCR. GaAs laser at a dose of 3 J, but not 1.5 J, significantly increased the CSA of regenerating myofibers and reduced the PIMA and the area density of intramuscular connective tissue of CTX-injured muscles. MyoD gene expression increased in the injured group treated with GaAs laser at a dose of 1.5 J. The CTX-injured, 3-J GaAs laser-treated, and the CTX-injured and treated with 3-J laser groups showed an increase in myogenin gene expression when compared to the control group. Our results suggest that GaAs laser treatment at a dose of 3 J improves skeletal muscle regeneration by accelerating the recovery of myofiber mass.

Background: The conventional methods of maxillary alveolar reconstruction in patient with cleft are the periosteoplasty and autologous bone grafting. As an important alternative of bone substitution, there is the recombinant human bone morphogenetic protein-2 (rhBMP-2). This study compares the rhBMP-2 with periosteoplasty and autologous bone grafting. Methods: Patients with cleft and alveolar defect were divided into 3 groups of 6 patients who underwent to autologous iliac crest bone grafting, resorbable collagen sponge with rhBMP2, and periosteoplasty, respectively. The analysis was performed through computed tomographic scan preoperatively and at months 3, 6, and 12 postoperatively. The variables analyzed were the alveolar defect volume, formed bone volume, bone formation rate, maxillary height repair rate, and the formed bone density mean. Results: The formed bone volume was similar comparing the bone graft and BMP groups at 1-year postoperative analysis (P = 0.58). Both of them had the formed bone volume significantly larger than the periosteoplasty group at 3 and 6 months postoperatively. In this last group, the 1-year follow-up was canceled because the bone formation was insufficient. The bone formation rate, the maxillary height repair rate, and the mean of density of the formed bone were similar in the bone graft and BMP groups at 1-year follow-up with P values of 0.93, 0.90, and 0.81, respectively. Conclusions: The amount of formed bone in the periosteoplasty group was insufficient. There was no difference among the bone graft and rhBMP-2 therapy considering the parameters analyzed.

Approximately 370 brachyuran species have so far been recorded from the Brazilian coast, 123 of which have had their larval stages fully or partially described. The pictorial guide allows the identification of the first zoea of 110 species. The remaining 13 species with known larval stages are treated to the genus level because of difficulties in the morphological differentiation of closely related species.

We have cloned and characterized for the first time an allograft inflammatory factor 1 (Sn-AIF-1) from the Antarctic sea urchin. We report the cloning of Sn-AIF-1 cDNA and the characterization of its expression in coelomocytes after a bacterial challenge. The cDNA Sn-AIF-1 has a size of 608 bp and encodes a polypeptide of 151 aa. The deduced amino acid sequence has a putative size of 17.430 Da, an isoelectric point of 4.92, and shows 2 elongation factor handlike motifs that normally bind calcium ions. BLAST analysis revealed close matches with other known AIF-1. The deduced amino acid sequence of Sn-AIF-1 showed high homology with AIF-1 in vertebrates such as fish, mice, and humans; and in the case of invertebrates, the major degree of identity (55%) was with a predicted sequence of the purple sea urchin AIF-1, and 52% corresponded to a sponge. Expression of Sn-AIF-1 mRNA was analyzed by qPCR. Sn-AIF-1 mRNA expression was measured from coelomocytes after a bacterial challenge using RT-PCR and revealed that the gene was upregulated after 24 h. Sn-AIF-1 could participate in the inflammatory response, particularly in the activation of coelomocytes and their survival.

An efficient and concise synthesis of nine populene D analogues was performed using an iodine-catalyzed Prins cyclization as the key transformation. The antiproliferative activity of these new pyrans against several cancer cell lines was then investigated. Among them, an isochromene with moderate activity (mean logGI(50) = 0.91) was found. Additionally, compounds with selectivity toward the tumor cell lines NCI-ADR/RES, OVCAR-3, and HT29 were discovered.

Different species of Laurencia have proven to be a rich source of natural products yielding interesting bioactive halogenated secondary metabolites, such as terpenoids and acetogenins. It is shown that such compounds are accumulated in the spherical, reniform to claviform refractive inclusions called corps en cerise (CC), which are intensively osmiophilic and located mainly in the cortical cells of the thalli and also in trichoblast cells. Up to now, it was believed that CC were present only in these two kinds of cells. Recently, however, a species of Laurencia, L. marilzae, with CC in all cells of the thallus, i.e., cortical, medullary, including the pericentral and axial cells, as well as in the trichoblasts, was described from the Canary Islands, and subsequently also reported to Brazil and Mexico. Within the Laurencia complex, only Laurencia species produce CC. Since the species of Laurencia are targets of interest for the prospection of bioactive substances due to their potential antibacterial, antifungal, anticholinesterasic, antileishmanial, cytotoxic, and antioxidant activities, the present paper carries out a comparative analysis of the corps en cerise in several species of Laurencia from the Atlantic Ocean to obtain basic information that can support natural product bioprospection projects. Our results show that the number and size of the CC are constant within a species, independent of the geographical distribution, corroborating their use for taxonomical purposes to differentiate groups of species that present a lower number from those that have a higher number. In this regard, there was a tendency for the number of CC to be higher in some species of Laurencia from the Canary Islands. The presence of CC can also be used to distinguish species in which these organelles are present in all cells of the thallus from those in which CC are restricted to the cortical cells. Among the species analyzed, L. viridis displayed the most varied secondary metabolites composition, such as sesquiterpenes, diterpenes, triterpenes, all of which showed potent antiviral, cytotoxic, and antitumoral activities, including protein phosphatase type 2A (PP2A) inhibitory effects.

The efficacy of estrus synchronization using short-term protocol was evaluated by ultrasound exams in Suffolk ewes during the pre-breeding season. The control Group (n = 12) was synchronized by treatment for 12 days with vaginal sponges impregnated with medroxyprogesterone acetate, and 400 IU eCG at sponge withdrawal. Experimental groups I, II and III kept the sponge in place for 4 days, and 100 µg of PGF2a was administered at sponge withdrawal. Additionally, Group I (n = 12) had 0.1 mg of estradiol benzoate (EB) administered during sponge placement and 50 µg of GnRH 48 hours after sponge removal. Group II (n = 6) had 35 mg of progesterone (P4) injected, and 0.1 mg of EB administered during sponge placement, 400 IU eCG at withdrawal and 48 hours after, 50 µg GnRH were administrated. Group III (n = 12) had 35 mg of P4 and 0.2 mg of EB administered at sponge placement, 400 IU eCG at withdrawal, and 50 µg of GnRH was administrated after 56 hours. Ovaries were monitored through ultrasound scanning. Concerning the first wave, no difference was detected between the control group and the experimental groups. However, the characteristics of ovulatory wave were significantly different between the groups. The duration of the follicular wave was shorter for Group III than for Group II. The follicle in Group I reached its maximum diameter before the Group II. The diameter of the follicle at the sponge withdrawal in the control group was larger than in Group I. After sponge withdrawal, the follicular growth rate was smaller in the control group than in Group III. The maximum diameter of the follicle in Group II was larger than in the other groups. The short-term protocol in which estrogen was used did not synchronize the emergence of the wave of follicular development.

Bread dough and particularly wheat dough, due to its viscoelastic behaviour, is probably the most dynamic and complicated rheological system and its characteristics are very important since they highly affect final products’ textural and sensorial properties. The study of dough rheology has been a very challenging task for many researchers since it can provide numerous information about dough formulation, structure and processing. This explains why dough rheology has been a matter of investigation for several decades. In this research rheological assessment of doughs and breads was performed by using empirical and fundamental methods at both small and large deformation, in order to characterize different types of doughs and final products such as bread. In order to study the structural aspects of food products, image analysis techniques was used for the integration of the information coming from empirical and fundamental rheological measurements. Evaluation of dough properties was carried out by texture profile analysis (TPA), dough stickiness (Chen and Hoseney cell) and uniaxial extensibility determination (Kieffer test) by using a Texture Analyser; small deformation rheological measurements, were performed on a controlled stress–strain rheometer; moreover the structure of different doughs was observed by using the image analysis; while bread characteristics were studied by using texture profile analysis (TPA) and image analysis. The objective of this research was to understand if the different rheological measurements were able to characterize and differentiate the different samples analysed. This in order to investigate the effect of different formulation and processing conditions on dough and final product from a structural point of view. For this aim the following different materials were performed and analysed: - frozen dough realized without yeast; - frozen dough and bread made with frozen dough; - doughs obtained by using different fermentation method; - doughs made by Kamut® flour; - dough and bread realized with the addition of ginger powder; - final products coming from different bakeries. The influence of sub-zero storage time on non-fermented and fermented dough viscoelastic performance and on final product (bread) was evaluated by using small deformation and large deformation methods. In general, the longer the sub-zero storage time the lower the positive viscoelastic attributes. The effect of fermentation time and of different type of fermentation (straight-dough method; sponge-and-dough procedure and poolish method) on rheological properties of doughs were investigated using empirical and fundamental analysis and image analysis was used to integrate this information throughout the evaluation of the dough’s structure. The results of fundamental rheological test showed that the incorporation of sourdough (poolish method) provoked changes that were different from those seen in the others type of fermentation. The affirmative action of some ingredients (extra-virgin olive oil and a liposomic lecithin emulsifier) to improve rheological characteristics of Kamut® dough has been confirmed also when subjected to low temperatures (24 hours and 48 hours at 4°C). Small deformation oscillatory measurements and large deformation mechanical tests performed provided useful information on the rheological properties of samples realized by using different amounts of ginger powder, showing that the sample with the highest amount of ginger powder (6%) had worse rheological characteristics compared to the other samples. Moisture content, specific volume, texture and crumb grain characteristics are the major quality attributes of bread products. The different sample analyzed, “Coppia Ferrarese”, “Pane Comune Romagnolo” and “Filone Terra di San Marino”, showed a decrease of crumb moisture and an increase in hardness over the storage time. Parameters such as cohesiveness and springiness, evaluated by TPA that are indicator of quality of fresh bread, decreased during the storage. By using empirical rheological tests we found several differences among the samples, due to the different ingredients used in formulation and the different process adopted to prepare the sample, but since these products are handmade, the differences could be account as a surplus value. In conclusion small deformation (in fundamental units) and large deformation methods showed a significant role in monitoring the influence of different ingredients used in formulation, different processing and storage conditions on dough viscoelastic performance and on final product. Finally the knowledge of formulation, processing and storage conditions together with the evaluation of structural and rheological characteristics is fundamental for the study of complex matrices like bakery products, where numerous variable can influence their final quality (e.g. raw material, bread-making procedure, time and temperature of the fermentation and baking).

Der evolutionär älteste Tierstamm der Metazoa, die Schwämme (Porifera), bietet sich als Modellorganismus zur Untersuchung der Selbst/Fremd-Erkennung an. Diese Unterscheidung wurde bei der Evolution von einzelligen zu mehrzelligen Tieren wichtig. Schwämme besitzen ein angeborenes Immunsystem. Ob sie auch Vorläufer der Immunrezeptoren aufweisen, sollte in dieser Arbeit untersucht werden. Einen ersten Schritt stellte die Identifizierung weiterer Gene mit Immunglobulin-ähnlichen Domänen dar. Mit der Isolierung der beiden 'Schwamm-Adhäsionsmoleküle' GCSAML und GCSAMS durch ein Homologiescreening ist dies gelungen. Die Beteiligung dieser Moleküle an einer Immunantwort wurde mit Hilfe von Transplantationsexperimenten untersucht. Die Gewebe von Autotransplantaten fusionierten, während es bei Allotransplantaten zu einer Abstoßung des eingesetzten Gewebes kam. Durch Immunfluoreszenz-Untersuchungen sowohl an Geodia cydonium- als auch an Suberites domuncula-Zellen konnte gezeigt werden, daß Moleküle mit Immunglobulin-ähnlichen Domänen an den Transplantationsprozessen beteiligt sind. Northern Blot-Analysen zeigten die Beteiligung speziell von GCSAML an der Autotransplantation. Western Blot-Analysen bestätigten diese Ergebnisse und weiteten sie auf Allotransplantationen aus. Beide GCSAM-Gene können der Immunglobulin-Superfamilie zugeordnet werden. GCSAML kann durch die Existenz des sogenannten ITIM-Motives weiter in die spezielle Klasse der inhibitorischen Rezeptoren gruppiert werden und stellt damit einen Vorläufer der Immunrezeptoren dar.

Die Schwämme (Porifera) sind eine reiche Quelle bioaktiver Naturstoffe. Viele dieser Naturstoffe besitzen das Potential, als Pharmazeutika, molekulare Sonden usw. eingesetzt oder weiterentwickelt zu werden. Die Beschaffung dieser Naturstoffe in ausreichenden Mengen stellt jedoch eines der größten Probleme bei der Testung und Produktion vielversprechender Substanzen dar. Der Transfer von DNA in Schwammzellen bzw. in komplette Organismen wäre ein vielversprechender Ansatz, dieses Problem zu lösen. Das Ziel dieser Arbeit war es deshalb, die Funktion und Struktur homologer Promotoren zu untersuchen und eine Methode des Gentransfers in Schwammzellen auszuarbeiten. Zu diesem Zweck wurde zusätzlich zu der bereits vorhandenen 5'-flankierenden Region des conventional PKC-Gens aus Geodia cydonium eine genomische Bibliothek von Suberites domuncula konstruiert, um diese mit Hilfe des DNA-Homologiescreenings nach den 5'-flankierenden Regionen des cPKC- und des SNZ (SnooZe)-Gens (SD_SNZG) zu durchsuchen. Die Klonierung und Sequenzierung sowohl des 5'-Bereichs als auch die Charakterisierung der Exon-Intron Struktur beider Gene wurde erfolgreich durchgeführt. In der 5'-Region des SNZ-Gens konnte dabei ein weiteres Gen (SD_SNO; SNZ proximal Open Reading Frame) identifiziert werden, das in einer 'Kopf-an-Kopf' Anordnung zu SD_SNZG orientiert ist. Sowohl SD_SNZG als auch SD_SNO wurden hochkonservierten Genfamilien zugeordnet, deren Vorkommen in Metazoen hier erstmals beschrieben wird.Funktionelle Studien mit Hilfe der Reportergene Luciferase und Enhanced Green Fluorescent Protein (EGFP) im heterologen System der NIH 3T3 Zellen wiesen sowohl dem cPKC-Promotor aus G. cydonium als auch dem SNZ-Promotor aus S. domuncula eine starke Promotoraktivität im Verhältnis zum SV40-Promotor nach. Die Aktivität des cPKC-Promotors aus S. domuncula dagegen war relativ schwach. Darüber hinaus konnte geklärt werden, daß die 5'-flankierende Region des SNZ-Gens bidirektionale Promotoraktivität aufweist und daß der G. cydonium cPKC-Promotor keine TATA-Box besitzt, sondern eine GC-Box für die basale Funktion benötigt.Als geeignete Methode zur Transfektion von Zellen des Schwamms S. domuncula erwies sich der ballistische Gentransfer mit Hilfe der Gene Gun. Homologe Promotoren konnten die sichtbare Expression des Reportergens EGFP jedoch nicht bewirken. Nur der virale CMV-Promotor erwies sich als hierfür geeignet.

Zusammenfassung der Dissertation von Markus Böhm 'Klonierung, Sequenzierung und Funktion der beiden SAPK-Mitglieder JNK und p38 MAPK des marinen Schwamms S. domuncula' am Fachbereich Biologie der Johannes Gutenberg-Universität Mainz: Da Schwämme zu den einfachsten Metazoen gehören, eignen sie sich gut zur Erforschung von Signaltransduktionsprozessen. Die SAPKs stellen hoch konservierte Signalmoleküle dar, die durch viele Zellstress-auslösende Faktoren aktiviert werden und in zahlreichen biologischen Prozessen involviert sind.Im Rahmen dieser Arbeit wurden zwei SAPK-Gene aus S. domuncula isoliert. Ihre abgeleiteten Aminosäuresequenzen wiesen die höchsten Homologien zu den Mitgliedern der SAPK1/JNK- und SAPK2/p38 MAPK-Subfamilie der Metazoen auf. Die geringste Übereinstimmung existierte gegenüber der einzigen SAPK der Hefe (HOG1). Beide Gene des Schwamms besaßen zudem eine außerordentlich hohe Übereinstimmung hinsichtlich ihrer Exon/Intron-Strukturen. Diese Ergebnisse deuten daraufhin, dass die SAPKs der multizellulären Tiere durch Duplikation eines HOG1-verwandten Vorläufergens entstanden sind. Durch den Vergleich der Intronpositionen mit denen von SAPK-Genen aus D. melanogaster, C. elegans und H. sapiens wurde ersichtlich, dass die Positionen der nichtkodierenden Sequenzbereiche dieser Gene hoch konserviert sind. Western Blot-Analysen demonstrierten, dass beide Schwamm-Kinasen durch hyperosmotischen Stress, LPS und den Phosphataseinhibitor Okadainsäure aktiviert werden. Außerdem wurde durch Versuche mit HOG1-defizienten Hefemutanten gezeigt, dass sie die Funktion des HOG1-Proteins in S. cerevisiae vollständig übernehmen können. Da die aktivierten Kinasen des Schwamms wie HOG1 im Nukleus der Hefezellen akkumuliert werden, müssen die Kerntransportmechanismen der SAPKs ebenfalls evolutionär erhalten sein.

ZusammenfassungAus dem Schwamm Geodia cydonium konnte die vollständige cDNA-Sequenz eines mutmaßlichen Bestandteiles des Aggregationsfaktors kloniert werden. Durch einen Northern-Blot konnte gezeigt werden, daß der gefundene Klon das vollständige Transkript repräsentiert. Das entsprechende Protein wurde in E. coli als Fusionsprotein rekombinant hergestellt. Mit einem Western-Blot-Experiment wurde der Nachweis geführt, daß es sich bei dem gefundenen Protein tatsächlich um einen Bestandteil des Aggregationsfaktors handelt. Der in diesem Western-Blot eingesetzte Antikörper wurde verwendet, um das Protein in histologischen Schnitten nachzuweisen. Das rekombinante Protein wurde in einem Aggregationsassay auf seine Funktionalität hin untersucht. Es stellte sich heraus, daß es einen Einfluß auf die Zellaggregation hat. Die Bindung des rekombinanten Aggregationsfaktors an das Lektin aus Geodia cydonium konnte gezeigt werden. Aus dem Schwamm Suberites domuncula wurde ein cDNA-Klon isoliert. Das durch diese cDNA kodierte Protein zeigt eine hohe Übereinstimmung mit einem in Vertebraten und in Limulus polyphemus vorkommendem Protein der extrazellulären Matrix, welches dort eine Rolle bei der Zellaggregation spielt. Die Vollständigkeit des Klons konnte anhand eines Northern-Blot gezeigt werden. Das Protein wurde in E. coli rekombinant hergestellt. Das rekombinante Protein führt in vitro zu einer verstärkten Aggregation von dissoziierten Schwammzellen.

'Bcl-2-homologe Proteine aus dem Schwamm Geodia cydonium: Klonierung, Charakterisierung und Funktionsanalyse von Apoptose-Regulatoren der Porifera'. Auf der Suche nach der molekularbiologischen Grundlage der Apoptose in den Porifera, dem phylogenetisch ältesten Metazoen-Tierstamm, wurden Mitglieder der apoptoseregulatorischen Bcl?2 Familie unter Anwendung diverser Techniken im Schwamm G. cydonium identifiziert: GCBHP1 und GCBHP2. Detaillierte Analysen offenbarten Bcl-2-charakteristische Signaturen sowie eine durch apoptotische Stimuli induzierbare Expression. Die parallele HSP70-Induktion zeugte von der GCBHP2-Expression als Teil einer antiapoptotischen Streßantwort zum Schutz des Organismus'. Die kontinuierliche Transkription des im Rahmen dieser Arbeit gleichfalls klonierten Proliferationsmarkers SEP1 veranschaulichte zudem die Effektivität dieser Streßreaktion. Mit der Herstellung eines rekombinanten Proteins und der Gewinnung eines Antiserums konnte auch die streßinduzierte Proteinexpression des GCBHP2 verfolgt werden. Zum Zweck der Funktionsstudie wurden Säugetierzellen (HEK-293, NIH/3T3) mit einem GCBHP2-Konstrukt stabil transfiziert und auf die Expression des Schwammproteins untersucht. Diese Zellen unterschieden sich bereits phänotypisch von mock-transfizierten Zellen. Immunzytochemische Untersuchungen enthüllten eine für antiapoptotische Bcl-2 Proteine charakteristische Assoziation mit Mitochondrien. Unter dem Einfluß zweier apoptotischer Stimuli wurde für GCBHP2-transfizierte Zellen eine vierzehn-/sechsmal höhere Vitalität und eine reduzierte Aktivierung der Caspase-Kaskade registriert (im Vergleich zu mock-transfizierten Kontrollen). Somit wurde der antiapoptotische Charakter des GCBHP2 und die Existenz apoptotischer Mechanismen in den Porifera bestätigt.
