A short and efficient approach to a range of new chiral and achiral functionalized (E)-enaminopyran-2,4-diones starting with commercially available dehydroacetic acid is described. The phytotoxic properties of these (E)-enaminopyran-2,4-diones were evaluated by their ability to interfere with the growth of Sorghum bicolor and Cucumis sativus seedlings. A different sensitivity of the two crops was evident with the (E)-enaminopyran-2,4-diones. The most active compounds were also tested against two weeds, Ipomoea grandifolia and Brachiaria decumbens. To the best of our knowledge, this is the first report describing enaminopyran-2,4-diones as potential plant growth regulators.

In vitro morphogenesis and cell suspension culture establishment in Piper solmsianum C. DC. (Piperaceae)). Piper solmsianum is a shrub from Southeast Brazil in which many biologically active compounds were identified. The aim of this work was to establish a cell suspension culture system for this species. With this in mind, petiole and leaf explants obtained from in vitro plantlets were cultured in the presence of different plant growth regulator combinations (IAA, NAA, 2,4-D and BA). Root and indirect shoot adventitious formation, detected by histological analysis, was observed. Besides the different combinations of plant growth regulators, light regime and the supplement of activated charcoal (1.5 mg.l(-1)) were tested for callus induction and growth. Cultures maintained in light, on a 0.2 mg.l(-1) 2,4-D and 2 mg.l(-1) BA supplemented medium, and in the absence of activated charcoal, showed the highest calli fresh matter increment. From a callus culture, cell suspension cultures were established and their growth and metabolite accumulation studied. The achieved results may be useful for further characterization of the activated secondary metabolites pathways in in vitro systems of P. solmsianum.

Fiber surface morphologies and associated internal structures are closely related to its properties. Unlike other fibers including cotton, bast fibers possess transverse nodes and fissures in cross-sectional and longitudinal directions. Their morphologies and associated internal structures were anatomically examined under the scanning electron microscope. The results showed that the morphologies of the nodes and the fissures of bast fibers varied depending on the construction of the inner fibril cellular layers. The transverse nodes and fissures were formed by the folding and spiralling of the cellular layers during plant growth. The dimensions of nodes and fissures were determined by the dislocations of the cellular layers. There were also many longitudinal fissures in bast fibers. Some deep longitudinal fissures even opened the fiber lumen for a short way along the fiber. In addition, the lumen channel of the bast fibers could be disturbed or disrupted by the nodes and the spirals of the internal cellular layers. The existence of the transverse nodes and fissures in the bast fibers could degrade the fiber mechanical properties, whereas the longitudinal fissures may contribute to the very rapid moisture absorption and desorption.

The authors have been involved with a group of primary school children since 1998, exploring their science learning in the first years of school. This article describes an activity run with two classes in grade 1, focussing on their ideas about plants. The activity involves constructing a terrarium and using it to challenge children's ideas about the conditions for plant growth.

Abscisic acid (ABA)-mediated gene expression is a critical component of plant responses to this important hormone, which affects plant growth, development, and responses to environmental stresses. Plant responses to ABA are mediated by a number of factors including PKABA1, an ABA induced protein kinase involved in ABA-suppressed gene expression in cereal grains, and TaWD40, which has previously been shown to physically interact with PKABA1. A full-length 1.9 kb TaWD40 cDNA, CK210682, was sequenced as part of this project. Based on the deduced protein sequence, it is thought that TaWD40 may belong to the family of E3 ubiquitin ligases, possibly targeting PKABA1 for destruction. Construction of expression plasmids for overproduction of the TaWD40 polypeptide in E. coli is currently underway. The TaWD40 cDNA has been successfully amplified from the source plasmid and inserted into an intermediate plasmid, pCR2.1. The TaWD40 cDNA is currently being cloned from the pCR2.1 intermediate plasmid into two different expression vectors, pRSET-A and pMAL-c2x, for future protein production and purification.

Although soils are generally considered to wet readily, some are actually water repellent at the surface and in the rhizosphere. This phenomenon occurs at low to moderate moisture contents and has been reported from soils under a range of vegetation types and from many regions around the globe. Water repellency in soils can have serious environmental implications including reduced seed germination and plant growth as well as irrigation efficiency, accelerated soil erosion, and enhanced leaching of agrochemicals through preferential flow. it has been proposed that water repellency is caused by the accumulation of hydrophobic organic compounds released as root exudates, microbial byproducts or from decomposing organic matter, which are deposited on mineral or aggregate surfaces, or are present as interstitial matter, Few studies to date have attempted to isolate and characterize these compounds and their structure is therefore only poorly understood, These studies have generally focussed on only a single soil or a small range of samples, have not included non-repellent soils as a control and have not always been able to demonstrate that the substances isolated are indeed responsible for repellency formation.

This study reports on the first part (extraction procedures) of a research programme addressing these gaps in current knowledge by investigating a wide range of severely repellent and wettable ‘control’ samples from different countries, and by including assessments of extraction efficiency and ability of extracts to cause repellency. Analytical methods include DRIFT (Diffuse Reflectance Infrared Fourier Transform Spectroscopy) of soils and IR (Infrared) analysis of extracts.

Key findings are that (i) soil sample heating after extraction is valuable in assessing the effectiveness of the extraction procedure, (ii) Soxhlet extraction using isopropanol/ ammonia (70/30 v/v) was the most effective method in extracting hydrophobic compounds, while leaving the ability of extracted compounds to induce water repellency virtually unaffected, (iii) wettable control soils also contain hydrophobic substances capable of inducing water repellency, (iv) the amount of organic compounds extracted was poorly related to sample repellency, indicating that compounds responsible for repellency may only represent a small fraction of the extract, (v) differences in extraction efficiency between different samples indicate that the compounds responsible may differ generically and/or in terms of their bonding to minerals, and (vi) the combination of repellency assessments with DRIFT on soils and JR on extracts used with internal standards has considerable potential to allow quantification of CH bearing organic matter in the soil, the efficiency of extraction processes for its removal, and its significance in causing water repellency in soils.

This work suggests how food storing corvids use spatial memory to relocate caches, and how they can do this after some landmarks surrounding caches have become hidden due to leaf fall, snow fall or plant growth. Experiments involved training European jays (Garrulus glandarius) to find buried food, the location of which was specified by an array of 12 landmarks. Tests were then performed with the array rotated, or with certain landmarks removed from the array. The.main findings were: (1) birds primarily remembered the position of the goal using the near tall landmarks (15-30 cm from the goal and 20 cm high); (2) birds obtained a sense of direction both from the landmark array and something external to the array; (3) birds did not use smell or marks in the surface of the ground to find the goal. Memory of near tall landmarks is likely to be functional for these birds since (a) nearer landmarks provide a more accurate fix, and (b) taller landmarks are less likely to be completely obscured by snow fall, leaf fall or intervening vegetation. The work also demonstrates the use of G.I.S. software for the analysis and representation of animal search patterns.

Decisions taken during migration can have a large effect on the fitness of birds. Migration must be accurately timed with food availability to allow efficient fueling but is also constrained by the optimal arrival date at the breeding site. The decision of when to leave a site can be driven by energetics (sufficient body stores to fuel flight), time-related cues (internal clock under photoperiodic control), or external cues (temperature, food resources). An individual based model (IBM) that allows a mechanistic description of a range of departure decision rules was applied to the spring migration of pink-footed geese (Anser brachyrhynchus) from wintering grounds in Denmark to breeding grounds on Svalbard via 2 Norwegian staging sites. By comparing predicted with observed departure dates, we tested 7 decision rules. The most accurate predictions were obtained from a decision rule based on a combination of cues including the amount of body stores, date, and plant phenology. Decision rules changed over the course of migration with the external cue decreasing in importance and the time-related cue increasing in importance for sites closer to breeding grounds. These results are in accordance with descriptions of goose migration, following the “green-wave”: Geese track the onset of plant growth as it moves northward in spring, with an uncoupling toward the end of the migration if time is running out. We demonstrate the potential of IBMs to study the possible mechanisms underlying stopover ecology in migratory birds and to serve as tools to predict consequences of environmental change.

Wetlands are among the most important ecosystems on Earth both in terms of productivity and biodiversity, but also as a source of the greenhouse gas CH4. Microbial processes catalyzing nutrient recycling and CH4 production are controlled by sediment physico-chemistry, which is in turn affected by plant activity and the foraging behaviour of herbivores. We performed field and laboratory experiments to evaluate the direct effect of herbivores on soil microbial activity and their indirect effects as the consequence of reduced macrophyte density, using migratory Bewick’s swans (Cygnus columbianus bewickii Yarrell) feeding on fennel pondweed (Potamogeton pectinatus L.) tubers as a model system. A controlled foraging experiment using field enclosures indicated that swan bioturbation decreases CH4 production, through a decrease in the activity of methanogenic Archaea and an increased rate of CH4 oxidation in the bioturbated sediment. We also found a positive correlation between tuber density (a surrogate of plant density during the previous growth season) and CH4 production activity. A laboratory experiment showed that sediment sterilization enhances pondweed growth, probably due to elimination of the negative effects of microbial activity on plant growth. In summary, the bioturbation caused by swan grazing modulates CH4 cycling by means of both direct and indirect (i.e. plant-mediated) effects with potential consequences for CH4 emission from wetland systems.

Bioactive brassinosteroids have been localized in developing and mature pollen of anhydrously fixed rye-grass (Lolium perenne) by immunocytochemistry using polyclonal antibodies to castasterone generated in rabbits. Tricellular pollen fixed by freeze-substitution was also labelled in the starch granules. Study of the developmental sequence of the pollen through the microsporocyte, microspore, bicellular and tricellular stages showed that the brassinosteroids were increasingly sequestered in starch granules as the amyloplasts matured, supporting the view that these are storage organelles for these potent plant growth promoters. In bicellular pollen, heavy labelling was seen in the zone within 0.5 μm of the starch granule, where stromal tissue remains. Thus, the stroma may be the site of synthesis of these compounds. During aqueous fixation, the brassinosteroids leached from the starch granules of tricellular pollen, indicating that they would be quickly available after imbibition to influence the physiology of germinating pollen. The results from high-performance liquid chromatography of dansylaminophenylboronates from partially purified extracts of freshly dehisced tricellular pollen of rye-grass showed 25-methylcastasterone may be a minor component, together with two unknown peaks. No specific binding of brassinolide to any soluble proteins extracted from tricellular rye-grass pollen was observed using the antibodies in gel electrophoresis or enzyme-linked immunosorbent assays.

The obligate soil-borne biotroph Plasmodiophora brassicae has a significant economic impact on Brassicaceae crops. The pathogen severely disrupts the roots by inducing the production of galls which leads to malformation and reduced growth of the roots and a reduced ability to take up water and nutrients. Control of P. brassicae is difficult because it has a number of survival and dissemination strategies that involve both motile and resting stages that need to be targeted by any control agent. We investigated, under controlled conditions and in glasshouse and field experiments, the potential of salicylic acid (SA), a key phytohormone, that is required for defence against certain biotic and abiotic stresses, to reduce infection by P. brassicae in broccoli (Brassicae oleracea var. italica). Under controlled conditions in a growth cabinet exogenous application of SA to roots resulted in its transport systemically to the leaves where it promoted the up-regulation of the pathogenesis related genes PR-1 and PR-2 in an SAR-like response as early as 24 h post-treatment. Concentrations of SA >20 mM reduced significantly both shoot and root weights when applied exogenously but lower concentrations had little measureable effect on plant growth. When SA was applied to plants above 5 mM there was a significant reduction (25-65 %) in gall formation 6 weeks post-inoculation with P. brassicae, indicating that the pathogen was being controlled by the addition of SA. A combination of SA and JA was also shown to reduce severity (25-35 %) of disease associated with P. brassicae. These findings indicate that there may be SA inducible mechanisms in B. oleracea that if fine-tuned could provide enhanced resistance to clubroot disease.

Objetivou-se, neste trabalho, avaliar o efeito da aplicação de herbicidas em pós-emergência sobre a eficiência fotoquímica de duas cultivares de soja (M-SOY 7908 RR e M-SOY 8001). O delineamento experimental utilizado foi em blocos ao acaso, com quatro repetições. Os tratamentos constaram da aplicação de herbicidas isolados e em misturas: lactofen, glyphosate, lactofen + chlorimuron-ethyl, chlorimuron-ethyl + imazethapyr, chlorimuron-ethyl + bentazon, glyphosate + imazethapyr, lactofen + chlorimuron-ethyl + imazethapyr e lactofen + chlorimuron-ethyl + imazethapyr/haloxyfop-methyl. Além disso, foram mantidas duas testemunhas sem aplicação de herbicida. As avaliações foram realizadas aos 4; 11; 18; 25 e 32 dias após aplicação dos herbicidas (DAA) no primeiro ano, e aos 6, 14, 21, 28 e 35 DAA, no segundo. A cultivar M-SOY 8001 foi suscetível à aplicação dos herbicidas, principalmente às misturas contendo lactofen + chlorimuron-ethyl e lactofen + chlorimuron-ethyl + imazethapyr + haloxifop-methyl, os quais provocaram redução no rendimento quântico máximo do PSII (Fv/Fm). As duas cultivares apresentaram suscetibilidade aos herbicidas quando tratados com lactofen aplicado isolado e nas misturas lactofen + chlorimuron-ethyl, chlorimuron-ethyl + imazethapyr, lactofen + chlorimuron-ethyl + imazethapyr e lactofen + chlorimuron-ethyl + imazethapyr + haloxifop-methyl com redução nos teores de clorofila, estimados pelo Medidor Portátil de Clorofilas. Os danos foram reversíveis, desaparecendo gradativamente com a idade das plantas.

A área foliar é uma das principais características usadas para avaliar o crescimento vegetal. O objetivo desta pesquisa foi determinar uma equação matemática para estimar a área foliar de Synedrellopsis grisebachii - uma importante planta daninha no Brasil - a partir de dimensões lineares dos limbos foliares. Duzentas folhas foram medidas em comprimento (C), largura máxima (L) e área foliar real (AF). Os dados de AF e CxL foram submetidos à análise de regressão linear, determinando-se uma equação matemática para estimar a área foliar da espécie. A correlação entre os valores de área foliar real e estimada foi significativa. Portanto, a área foliar de S. grisebachii pode ser estimada satisfatoriamente pela equação: AF = 0,730829(C×L).

It is well-documented that phytochromes can control plant growth and development from germination to flowering. Additionally, these photoreceptors have been shown to modulate both biotic and abiotic stress. This has led to a series of studies exploring the molecular and biochemical basis by which phytochromes modulate stresses, such as salinity, drought, high light or herbivory. Evidence for a role of phytrochromes in plant stress tolerance is explored and reviewed.
