Background & Aims: To investigate the effect of vitamin A supplementation on parameters of the immune system of vitamin A-deficient children. Methods: The study was carried out in four phases: 1) determination of serum retinol in 631 children from 36 to 83 months of age; 2) assessment of immunological markers [immunoglobulins and complement fractions, immunophenotyping of T and B lymphocytes, and natural killer (NK) cells], blood count, and serum ferritin of 52 vitamin A-deficient children (serum retinol <0.70 mu mol/L); 3) supplementation of the 52 deficient children with 200,000 IU of vitamin A; 4) determination of serum retinol and the immunological parameters 2 months after vitamin A supplementation. Results: Before vitamin A supplementation, 24.0% of the children were anemic and 4.3 %had reduced ferritin concentrations. There was no significant difference between mean values of retinol according to the presence/absence of anemia. The mean values of the humoral and cellular immunological parameters did not show a statistically significant difference before and after supplementation with vitamin A. Children with concomitant hypovitaminosis A and anemia presented a significant increase in absolute CD4 and CD8 T-cell counts after vitamin A supplementation (p < 0.05). Conclusion: Vitamin A had an effect on the recruitment of T and B lymphocytes to the circulation of children with hypovitaminosis A and anemia.

Pteridium aquilinum (bracken fern) is one of the most common plants. Epidemiological studies have revealed a higher risk of certain types of cancers (i.e., esophageal, gastric) in people who consume bracken fern directly ( as crosiers or rhizomes) or indirectly through the consumption of milk from livestock that fed on the plant. In animals, evidence exists regarding the associations between chronic bracken fern intoxication, papilloma virus infection, and the development of carcinomas. While it is possible that some carcinogens in bracken fern could be responsible for these cancers in both humans and animals, it is equally plausible that the observed increases in cancers could be related to induction of an overall immunosuppression by the plant/its various constituents. Under the latter scenario, normal tumor surveillance responses against nascent (non-bracken-induced) cancers or responses against viral infections ( specifically those linked to induction of cancers) might be adversely impacted by continuous dietary exposure to this plant. Therefore, the overall objective of this study was to evaluate the immunomodulatory effects of bracken fern following daily ingestion of its extract by a murine host over a period of 14 ( or up to 30) days. In C57BL/6 mice administered ( by gavage) the extract, histological analyses revealed a significant reduction in splenic white pulp area. Among a variety of immune response parameters/functions assessed in these hosts and isolated cells, both delayed-type hypersensitivity (DTH) analysis and evaluation of IFN gamma. production by NK cells during T(H)1 priming were also reduced. Lastly, the innate response in these hosts-assessed by analysis of NK cell cytotoxic functionality-was also diminished. The results here clearly showed the immunosuppressive effects of P. aquilinum and that many of the functions that were modulated could contribute to the increased risk of cancer formation in exposed hosts.

The proteinase-activated receptor 2 (PAR(2)) is a putative therapeutic target for arthritis. We hypothesized that the early pro-inflammatory effects secondary to its activation in the temporomandibular joint (TMJ) are mediated by neurogenic mechanisms. Immunofluorescence analysis revealed a high degree of neurons expressing PAR(2) in retrogradely labeled trigeminal ganglion neurons. Furthermore, PAR(2) immunoreactivity was observed in the lining layer of the TMJ, co-localizing with the neuronal marker PGP9.5 and substance-P-containing peripheral sensory nerve fibers. The intra-articular injection of PAR(2) agonists into the TMJ triggered a dose-dependent increase in plasma extravasation, neutrophil influx, and induction of mechanical allodynia. The pharmacological blockade of natural killer 1 (NK(1)) receptors abolished PAR(2)-induced plasma extravasation and inhibited neutrophil influx and mechanical allodynia. We conclude that PAR(2) activation is proinflammatory in the TMJ, through a neurogenic mechanism involving NK(1) receptors. This suggests that PAR(2) is an important component of innate neuro-immune response in the rat TMJ.

The environmental chemical 1,2-naphthoquinone (1,2-NQ) is implicated in the exacerbation of airways diseases induced by exposure to diesel exhaust particles (DEP), which involves a neurogenic-mediated mechanism. Plasma extravasation in trachea, main bronchus and lung was measured as the local (125)I-bovine albumin accumulation. RT-PCR quantification of TRPV1 and tachykinin (NK(1) and NK(2)) receptor gene expression were investigated in main bronchus. Intratracheal injection of DEP (1 and 5 mg/kg) or 1,2-NQ (35 and 100 nmol/kg) caused oedema in trachea and bronchus. 1,2-NQ markedly increased the DEP-induced responses in the rat airways in an additive rather than synergistic manner. This effect that was significantly reduced by L-732,138, an NK(1) receptor antagonist, and in a lesser extent by SR48968, an NK(2) antagonist. Neonatal capsaicin treatment also markedly reduced DEP and 1,2-NQ-induced oedema. Exposure to pollutants increased the TRPV1, NK(1) and NK(2) receptors gene expression in bronchus, an effect was partially suppressed by capsaicin treatment. In conclusion, our results are consistent with the hypothesis that DEP-induced airways oedema is highly influenced by increased ambient levels of 1,2-NQ and takes place by neurogenic mechanisms involving up-regulation of TRPV1 and tachykinin receptors.

Stings by Polistes wasps can cause life-threatening allergic reactions, pain and inflammation. We examined the changes in microvascular permeability and neutrophil influx caused by the venom of Polistes lanio a paper wasp found in southeastern Brazil. The intradermal injection of wasp venom caused long-lasting paw oedema and dose-dependently increased microvascular permeability in mouse dorsal skin. SR140333, an NK(1) receptor antagonist, markedly inhibited the response, but the NK(2) receptor antagonist SR48968 was ineffective. The oedema was reduced in capsaicin-treated rats, indicating a direct activation of sensory fibres. Dialysis of the venom partially reduced the oedema and the remaining response was further inhibited by SR140333. Mass spectrometric analysis of the venom revealed two peptides (QPPTPPEHRFPGLM and ASEPTALGLPRIFPGLM) with sequence similarities to the C-terminal region of tachykinin-like peptides found in Phoneutria nigniventer spider venom and vertebrates. Wasp venom failed to release histamine from mast cells in vitro and spectrofluorometric assay of the venom revealed a negligible content of histamine in the usual dose of P.l. lanio venom (1 nmol of histamine/7 mu g of venom)that was removed by dialysis. The histamine H(1) receptor antagonist pyrilamine, but not bradykinin B(1) or B(2) receptor antagonists, inhibited venom-induced oedema. In conclusion, P. l. lanio venom induces potent oedema and increases vascular permeability in mice, primarily through activation of tachykinin NK(1) receptors by substance P released from sensory C fibres, which in turn releases histamine from dermal mast cells. This is the first description of a neurovascular mechanism for P. l. lanio venom-mediated inflammation. The extent to which the two tachykinin-like peptides identified here contribute to this neurogenic inflammatory response remains to be elucidated. (c) 2008 Elsevier Ltd. All rights reserved.

Primary sensory afferent neurons modulate the hyperdynamic circulation in Cirrhotic rats with portal hypertension.The stomach of cirrhotic rats is prone to damage induced by ethanol, a phenomenon associated with reduced gastric hyperemic response to acid-back diffusion. The aim of this study was to examine the impact of ablation of capsaicin-sensitive neurons and the tachykinin NK(1) receptor antagonist A5330 on the susceptibility of the portal hypertensive gastric mucosa, to ethanol-induced injury and its effects on gastric cyclooxygenase (COX) and nitric oxide synthase (NOS) mRNA expression. Capsaicin was administered to neonatal, male, Wistar rats and the animals were allowed to grow. Cirrhosis was then induced by bile duct ligation in adult rats while controls had sham operation. Ethanol-induced gastric damage was assessed using ex vivo gastric chamber experiments. Gastric blood flow was measured as well as COX/NOS mRNA expression. Topical application of ethanol produced significant gastric damage in cirrhotic rats compared to controls, which was reversed in capsaicin- and A5330-treated animals. Mean arterial and portal pressure was normalized in capsaicin-treated cirrhotic rats. Capsaicin and A5330 administration restored gastric blood flow responses to topical application of ethanol followed by acid in cirrhotic rats. Differential COX and NOS mRNA expression was noted in bile duct ligated rats relative to controls. Capsaicin treatment significantly modified gastric eNOS/iNOS/COX-2 mRNA expression in cirrhotic rats. Capsaicin-sensitive neurons modulate the susceptibility of the portal hypertensive gastric mucosa to injury induced by ethanol via tachykinin NK(1) receptors and signalling of prostaglandin and NO production/release. Innate immunity is based in pre-existing elements of the immune system that directly interact with all types of microbes leading to their destruction or growth inhibition. Several elements of this early defense mechanism act in concert to control initial pathogen growth and have profound effect on the adaptative immune response that further develops. Although most studies in paracoccidioidomycosis have been dedicated to understand cellular and humoral immune responses, innate immunity remains poorly defined. Hence, the main purpose of this review is to present and discuss some mechanisms of innate immunity developed by resistant and susceptible mice to Paracoccidioides brasiliensis infection, trying to understand how this initial host-pathogen interface interferes with the protective or deleterious adaptative immune response that will dictate disease outcome. An analysis of some mechanisms and mediators of innate immunity such as the activation of complement proteins, the microbicidal activity of natural killer cells and phagocytes, the production of inflammatory eicosanoids, cytokines, and chemokines among others, is presented trying to show the important role played by innate immunity in the host response to P. brasiliensis infection.

A hipertensão é uma doença multifatorial pela interação de vários mecanismos fisiopatológicos, sendo considerada um importante fator de risco para outras doenças cardiovasculares. Uma vez que o exercício tem sido recomendado com uma forma de tratamento antihipertensivo, buscou-se avaliar os efeitos do treinamento físico nos sistemas reguladores da pressão arterial (PA) e no seu controle reflexo, e sua correlação com o estresse oxidativo de ratos espontaneamente hipertensos (SHR). Assim, este estudo foi realizado com ratos machos Wistar-Kyoto (NK) e SHR, com 15 semanas, divididos em dois grupos: SHR sedentário (HS) e SHR treinado (HT). Após o estabelecimento da hipertensão nesses animais, o protocolo de exercício foi realizado por 10 semanas com a intensidade determinada pelo limiar de lactato (20 m/min). Foram avaliadas as respostas bradicárdicas e taquicárdicas do barorreflexo, bem como a sensibilidade do reflexo cardiopulmonar de Bezold-Jarisch. O estresse oxidativo foi medido no sangue, coração e aorta dos animais em estudo. O treinamento físico diminuiu a PAM, PAS e PAD dos animais do grupo HT, quando comparado com o grupo HS, apesar de não ter igualado aos valores dos NK. No entanto, a FC não foi diferente entre os grupos hipertensos. O barorreflexo esteve atenuado no grupo HS comparado com o NK, porém, o exercício mostrou aumentar essa sensibilidade no grupo HT comparado com o HS. O exercício também melhorou a sensibilidade dos receptores cardiopulmonares à serotonina. Pelo bloqueio não foi observada diferença significativa na atividade dos sistemas arginina-vasopressina, renina-angiotensina e simpático. O sistema regulador endotelial, avaliado pela administração de L-NAME, um bloqueador da síntese do óxido nítrico, esteve significativamente aumentado no grupo HT quando comparado com o HS. O estresse oxidativo diminuiu significativamente no grupo HT comparado com o HS. Verificou-se nesse estudo diminuição da lipoperoxidação em eritrócitos, coração e aorta induzida pelo exercício. Além disso, a atividade das enzimas antioxidantes esteve aumentada no grupo HT comparado com o HS. Foi observada forte correlação negativa entre os valores de lipoperoxidação em eritrócitos e aorta e a sensibilidade barorreflexa, bem como entre a lipoperoxidação em eritrócitos e a sensibilidade do reflexo de Bezold-Jarisch. Estes resultados demonstram o efeito benéfico do exercício moderado em SHR, tanto no controle reflexo da PA, como no estresse oxidativo. Conclui-se também, que o estresse oxidativo desempenha importante papel na alteração da sensibilidade barorreflexa e do reflexo cardiopulmonar na hipertensão.

O Brasil é um dos maiores produtores e exportadores mundiais de tabaco em folha; entretanto, o beneficiamento industrial gera aproximadamente 35.000 t anuais de resíduos sem valor comercial. O objetivo deste trabalho foi avaliar o potencial de reciclagem agrícola de dois resíduos de agroindústria fumageira (RAF´s), sua mineralização no solo e liberação de nutrientes às plantas. O experimento de aplicação dos resíduos foi conduzido a céu aberto, utilizando vasos com capacidade de 32 litros de solo (PVAd), com um tubo coletor de lixiviado e cultivados com três plantas de milho. Foram utilizados 18 tratamentos com adubo mineral, cama-de-aviário, composto orgânico e com adições de doses crescentes (0, 7,5, 15, 30 e 60 t ha-1) dos resíduos RAF P (pó) e RAF T (talos) além de tratamentos com resíduos (15 t ha-1) e complementação com nutrientes minerais dois a dois (NP, NK e PK), em quatro repetições. O experimento foi iniciado em 15/01/2004, tendo sido a parte aérea das plantas colhida em 16/03/2004. O efeito residual da aplicação dos resíduos foi estudado em duas das repetições, enquanto que em outras duas foram reaplicadas doses equivalentes a 33,3% da dose inicial dos resíduos, utilizando-se o sorgo como planta teste. O sorgo foi semeado em 26/03/2004 e a parte aérea colhida em 14/05/2004. Foram determinados as curvas de resposta de rendimento de matéria seca e o teor de macronutrientes no tecido, no solo e na água de lixiviação, além de outros atributos de fertilidade do solo. Os resultados indicaram que os RAF´s são boas fontes de biomassa vegetal e de potássio e possuem potencial para serem reciclados no solo, permitindo a liberação de parte dos macronutrientes necessários ao desenvolvimento das plantas. As curvas de resposta mostraram que a adição de doses entre 15 e 20 t ha-1 dos resíduos são as mais recomendadas agronomicamente, proporcionando rendimentos de matéria seca equivalentes à fertilização mineral. A mineralização dos resíduos, avaliada pela evolução de C-CO2, foi de aproximadamente 40% para o RAF P e de 49% para o RAF T. Os resíduos RAF P e RAF T apresentaram valores de neutralização (médias de três solos) de 7,9% e 9,5%, respectivamente. A adição dos resíduos ao solo deve ser calculada para manter o pH do solo próximo a 6,0.

O câncer de colo uterino consiste em um problema de grande relevância social, visto que se trata da segunda malignidade ginecológica mais freqüente no mundo. O principal precursor do câncer da cérvice uterina consiste na infecção pelo Papiloma vírus humano (HPV). O HPV tem despertado grande interesse na comunidade científica, devido à relação estabelecida entre a sua epidemiologia e o câncer de cérvice uterina. Apesar do grande avanço na biologia do HPV, pouco se sabe acerca da resposta imunológica a este vírus. O Papilomavírus humano (HPV) é um DNA vírus epiteliotrófico, que está vinculado à carcinogênese do colo uterino por meio de evidências epidemiológicas e laboratoriais. As infecções pelo HPV ocorrem em mulheres em todo o mundo. Existe consenso na literatura quanto à associação do DNA HPV com as neoplasias intra epiteliais cervicais e o câncer cervical. Por este motivo é de grande importância a detecção precoce deste tipo de infecção para que seja efetuado um tratamento específico. Esse trabalho está caracterizando o perfil imunológico de pacientes portadoras de HPV através da quantificação linfocitária (TCD4, TCD8, células NKT e células NK), e correlacionou esse material à técnica de captura híbrida, para poder comprovar a infecção pelo HPV, esperando assim contribuir precocemente para o diagnóstico do câncer de colo uterino.

Chronic lymphoproliferative disorders (DLPC) are lymphoid system diseases characterized by the abnormal proliferation of mature lymphocytes that affect B cells, T lymphocytes and NK cells. The aim of the study was to demonstrate the relevance of immunophenotyping by flow cytometry in patients with prolonged lymphocytosis and / or cytomorphological changes compatible with lymphoproliferative diseases. In this study 460 patients (244 men and 216 women) with DLPC were evaluated. Were analyzed by flow cytometry with a panel of monoclonal antibodies consisting of CD3, CD4, CD5, CD8, CD10, CD19, CD22, CD23, CD25, CD38, CD45, CD16/CD56, and HLADR heavy and light chains of immunoglobulins. It also examines information regarding age, gender of patients and laboratory data as leucocytes, cytomorphological analysis, platelet count and hemoglobin determination. The results showed 398 cases of chronic lymphoproliferative disorders and 62 of DLPC B cell lymphoproliferative diseases T. B showed the following distribution : 253 cases of chronic lymphocytic leukemia (CLL), 42 cases of multiple myeloma ( MM ), 37 cases of lymphoma non - Hodgkin lymphoma in leukemic phase (NHL) , 17 cases of pro- B lymphocytic leukemia ( B -PLL), 15 cases of mantle cell lymphoma (MCL ), 12 cases of plasma cell leukemia ( PCL), 9 cases of lymphoma Burkitt (Linf B), 8 cases of leukemia villous cells ( LCV), 3 cases of splenic lymphoma with villous cells (LECV), a case of follicular lymphoma (LF) and a Waldenströn macroglobulinemia ( MW). The diseases source NK / T were 23 cases of peripheral T cell lymphoma (LCTP), 14 cases of T prolymphocytic leukemia (T -PLL), 10 cases of leukemia T of large granular lymphocytes (LGL -T) 9 cases of leukemia cells of adult T (LCTA), 5 cases of Sezary syndrome (SS) and a case of large granular NK leukemia (LGL -NK) lymphocytes. In conclusion, the combined use of the monoclonal antibody panel careful cytomorphological analysis was shown to be essential in immune diagnosis and classification of chronic lymphoproliferative disorders. O presente trabalho teve como objetivo avaliar a influência da adubação da cultura do sorgo Sorghum bicolor (L.) Moench, na biologia da broca da cana-de-açúcar Diatraea saccharalis (Fabr., 1794) (Lepidoptera: Pyralidae). Para isso foi utilizado sorgo da cultivar Rubi-Asgrow, plantado em vasos utilizando-se diferentes doses de fertilizantes e mantidos em casa-de-vegetação. Os tratamentos utilizados (doses de NK) foram: N1 = 0-200 ppm; N2 = 50-200 ppm; N3 = 100-200 ppm; N4 = 200-200 ppm; N5 = 400-200 ppm; K1 = 200-0 ppm; K2 = 200-50 ppm; K3 = 200-100 ppm; K4 = 200-200 ppm; e K5 = 200-400 ppm. de modo geral, pode-se concluir que doses de 50 a 200 ppm de N promoveram o desenvolvimento normal das larvas de D. saccharalis, sendo que as menores porcentagens de dano foram verificadas nas menores doses; para o potássio, quanto maior a dose, menor foi o dano causado pelas lagartas, apesar de favorecer o desenvolvimento da mesma.

Objetivando-se estudar a influência da adubação na manifestação da resistência de feijoeiro (Phaseolus vulgaris L.) (cultivares Rosinha G-2, ESAL-564 - Carioca e Diamante Negro) ao ataque de Acanthoscelides obtectus (Say, 1831) (Coleoptera: Bruchidae) foram conduzidos, na época das águas, testes com e sem chance de escolha, utilizando-se grãos obtidos de parcelas adubadas com N, P, K, NP, NK, PK, NPK, e sem adubo, totalizando 24 tratamentos. Os testes foram realizados em condições controladas de temperatura, umidade e luz. Foram avaliados o número de insetos atraídos e de ovos, em cada tratamento. Concluiu-se, no teste com chance de escolha, que o número de ovos de A. obtectus por recipiente foi reduzido pela aplicação do nitrogênio. A aplicação de nitrogênio em adubação resultou em menor porcentagem de insetos atraídos e menor número de ovos no genótipo Rosinha G-2. A manifestação da resistência nos genótipos ESAL-564 e Rosinha G-2 ao ataque de A. obtectus ficou evidente quando utilizados nitrogênio e potássio. Nos testes sem chance de escolha o consumo dos insetos foi reduzido nos grãos produzidos com a aplicação do nitrogênio. A aplicação de nitrogênio em adubação resultou no aumento do número de ovos de A. obtectus no genótipo ESAL-564. Não ficou evidente a manifestação da resistência nos genótipos ESAL-564 e Rosinha G-2 ao ataque do caruncho, pela aplicação dos macronutrientes N, P e K. Na ausência de fósforo o ciclo biológico do inseto foi maior na presença de potássio.
