The location of extracellular enzymes within the soil architecture and their association with the various soil components affects their catalytic potential. A soil fractionation study was carried out to investigate: (a) the distribution of a range of hydrolytic enzymes involved in C, N and P transformations, (b) the effect of the location on their respective kinetics, (c) the effect of long-term N fertilizer management on enzyme distribution and kinetic parameters. Soil (silty clay loam) from grassland which had received 0 or 200 kg N ha(-1) yr(-1) was fractionated, and four particle-size fractions (> 200, 200-63, 63-2 and 0. 1-2 mum) were obtained by a combination of wet-sieving and centrifugation, after low-energy ultrasonication. All fractions were assayed for four carbohydrases (beta-cellobiohydrolase, N-acetyl-beta-glucosammidase, beta-glucosidase and beta-xylosidase), acid phosphatase and leucine-aminopeptidase using a microplate fluorimetric assay based on MUB-substrates. Enzyme kinetics (V-max and K-m) were estimated in three particle-size fractions and the unfractionated soil. The results showed that not all particle-size fractions were equally enzymatically active and that the distribution of enzymes between fractions depended on the enzyme. Carbohydrases predominated in the coarser fractions while phosphatase and leucine-aminopeptidase were predominant in the clay-size fraction. The Michaelis constant (K.) varied among fractions, indicating that the association of the same enzyme with different particle-size fractions affected its substrate affinity. The same values of Km were found in the same fractions from the soil under two contrasting fertilizer management regimes, indicating that the Michaelis constant was unaffected by soil changes caused by N fertilizer management. White clover (Trifolium repens) is an important pasture legume but is often difficult to sustain in a mixed sward because, among other things, of the damage to roots caused by the soil-dwelling larval stages of S. lepidus. Locating the root nodules on the white clover roots is crucial for the survival of the newly hatched larvae. This paper presents a numerical model to simulate the movement of newly hatched S. lepidus larvae towards the root nodules, guided by a chemical signal released by the nodules. The model is based on the diffusion-chemotaxis equation. Experimental observations showed that the average speed of the larvae remained approximately constant, so the diffusion-chernotaxis model was modified so that the larvae respond only to the gradient direction of the chemical signal but not its magnitude. An individual-based lattice Boltzmann method was used to simulate the movement of individual larvae, and the parameters required for the model were estimated from the measurement of larval movement towards nodules in soil scanned using X-ray microtomography. The model was used to investigate the effects of nodule density, the rate of release of chemical signal, the sensitivity of the larvae to the signal, and the random foraging of the larvae on the movement and subsequent survival of the larvae. The simulations showed that the most significant factors for larval survival were nodule density and the sensitivity of the larvae to the signal. The dependence of larval survival rate on nodule density was well fitted by the Michealis-Menten kinetics. (c) 2005 Elsevier B.V All rights reserved.

Enzymes are versatile biocatalysts with major advantages of ultrahigh reaction selectivity and specificity under mild conditions, which currently find increasing applications. However, their applications are often hampered by difficulties in recovery and recycling. As a result, we carried out detailed investigations on the synthesis and characterization of silica-encapsulated iron oxide magnetic nanoparticles of controlled dimension as an enzyme carrier. It is shown that the relatively smaller sized silica-coated magnetic nanoparticle prepared by the microemlusion technique can a carry bulky enzyme, beta-lactamase, via chemical linkages on the silica overlayer without severely blocking the enzymatic active center ( which is commonly encountered in conventional solid supports). An activity study by Michalis-Menten kinetics reflects that this new type of immobilization allows enzyme isolation with accessibility as good as free enzyme. The recovery and reusability of the nanoparticle-supported enzyme upon application of magnetic separation are also demonstrated.

Neutron diffraction at 11.4 and 295 K and solid-state 67Zn NMR are used to determine both the local and average structures in the disordered, negative thermal expansion (NTE) material, Zn(CN)2. Solid-state NMR not only confirms that there is head-to-tail disorder of the C≡N groups present in the solid, but yields information about the relative abundances of the different Zn(CN)4-n(NC)n tetrahedral species, which do not follow a simple binomial distribution. The Zn(CN)4 and Zn(NC)4 species occur with much lower probabilities than are predicted by binomial theory, supporting the conclusion that they are of higher energy than the other local arrangements. The lowest energy arrangement is Zn(CN)2(NC)2. The use of total neutron diffraction at 11.4 K, with analysis of both the Bragg diffraction and the derived total correlation function, yields the first experimental determination of the individual Zn−N and Zn−C bond lengths as 1.969(2) and 2.030(2) Å, respectively. The very small difference in bond lengths, of ~0.06 Å, means that it is impossible to obtain these bond lengths using Bragg diffraction in isolation. Total neutron diffraction also provides information on both the average and local atomic displacements responsible for NTE in Zn(CN)2. The principal motions giving rise to NTE are shown to be those in which the carbon and nitrogen atoms within individual Zn−C≡N−Zn linkages are displaced to the same side of the Zn···Zn axis. Displacements of the carbon and nitrogen atoms to opposite sides of the Zn···Zn axis, suggested previously in X-ray studies as being responsible for NTE behavior, in fact make negligible contribution at temperatures up to 295 K.

The present study describes the enzymatic properties and molecular identification of 5`-nucleotidase in soluble and microsomal fractions from rat cardiac ventricles. Using AMP as a substrate, the results showed that the cation and the concentration required for maximal activity in the two fractions was magnesium at a final concentration of 1 mM. The pH optimum for both fractions was 9.5. The apparent K-m (Michaelis constant) values calculated from the Eadie-Hofstee plot were 59.7 +/- 10.4 mu M and 134.8 +/- 32.1 mu M, with V-max values of 6.7 +/- 0.4 and 143.8 +/- 23.8 nmol P-i/min/mg of protein (means +/- S.D., n = 4) from soluble and microsomal fractions respectively. Western blotting analysis of ecto-5`-nucleotidase revealed a 70 kDa protein in both fractions, with the major proportion present in the microsomal fraction. The presence of these enzymes in the heart probably has a physiological function in adenosine signalling. Furthermore, the presence of ecto-5`-nucleotidase in the microsomal fraction could have a role in the modulation of the excitation-contraction-coupling process through involvement of the Ca2+ influx into the sarcoplasmic reticulum. The measurement of maximal enzyme activities in the two fractions highlights the potential capacity of the different pathways of purine metabolism in the heart.

A different and improved procedure for the preparation of [bis(2,2,2-trifluoroethyl)phosphono]acetic acid in just one step from bis(2,2,2-trifluoroethyl)phosphonate is described. The protocol employs a Michaelis-Becker reaction between commercially available bis(2,2,2-trifluoroethyl) phosphonate and bromoacetic acid, furnishing [bis(2,2,2-trifluoroethyl)phosphono]acetic acid in 50-54% yield.

O tratamento de sementes com fungicida é um seguro de baixo custo para os produtores de sementes e plantadores de amendoim. O uso de fungicidas adequados pode contribuir para o melhor desempenho do material de propagação, proporcionando maiores rendimentos na colheita. Esta pesquisa teve por objetivo avaliar a eficiência do fungicida carboxin + thiram e a viabilidade do uso de óleo vegetal e de surfactante à base de organosilicone como agentes veiculadores do fungicida, no controle de fungos associados às sementes de amendoim. Foram comparadas duas doses de carboxin + thiram (50 + 50 e 70 + 70 g.i.a./100 kg de sementes) em mistura com água e a dose de 50 + 50 g.i.a./100 kg de sementes em mistura com os agentes veiculadores, avaliando-se os efeitos na germinação e vigor das sementes e estabelecimento da cultura no campo. Verificou-se que o principal efeito benéfico dos tratamentos é a redução significativa de sementes mortas e/ou da ocorrência de damping-off de pré-emergência. Independentemente da dose, o fungicida associado à água, proporciona controle eficiente dos patógenos presentes nas sementes, exceto no caso de Rhizopus sp. O óleo vegetal e o surfactante proporcionam melhor cobertura e aderência do fungicida às sementes, aumentando a eficiência do mesmo no controle de Aspergillus spp. e de Penicillium sp. Não se constata efeito fitotóxico do fungicida e dos agentes veiculadores sobre as sementes de amendoim.

O presente trabalho teve como objetivo verificar se os sintomas incitados na bananeira cv. Nanicão, do subgrupo Cavendish, na região do Vale do Ribeira, estão relacionados com níveis de nutrientes no solo e nas folhas. Foram separadas 16 áreas na região, sendo a metade com plantas sintomáticas e a outra com plantas sadias. Nessas áreas, coletou-se a terceira folha de cinco plantas e o solo junto a essas mesmas plantas, nas profundidades de 0 a 20 cm e de 20 a 40 cm. em ambas as profundidades do solo amostrado, níveis de Ca, Mg, PO(-3)4, S e da capacidade de trocas catiônicas (CTC) foram significativamente diferentes entre as áreas, sendo que os valores baixos destes elementos estavam presentes nas áreas contendo plantas sintomáticas. em ambas as profundidades, o Mg, o Al e o H em relação à CTC foram significativamente diferentes entre as áreas, sendo que o valor baixo de Mg e alto de Al e H estava presente nas áreas com plantas sintomáticas. O N, K e S nas folhas foram significativamente diferentes entre as áreas. Estes elementos apresentaram valores baixos nas áreas contendo plantas sintomáticas. Apesar de algumas quantidades de macronutrientes do solo e das folhas estarem presentes somente nas áreas contendo plantas de Nanicão com sintomas semelhantes aos de fusariose, deve-se buscar comprovação de uma possível ocorrência da raça 4 do patógeno no Vale do Ribeira.

Both P-i-repressible acid phosphatases, IIb (mycelial) and IIc (extracellular), synthesized by Neurospora crassa and purified to apparent homogeneity by 7.5% PAGE, are monomers, are inhibited by 2 mM ZnCl2 and are nonspecifically stimulated by salts. However, the IIc form is activated by p-nitrophenylphosphate (in a negative cooperativity effect with a K-0.5 of 2.5 mM) whereas form IIb shows Michaelis kinetics, with a K-m of 0.5 mM. Thus, since both enzymatic forms may be expressed by the same gene (pho-3), it is possible that post-translational modifications lead to the excretion of an enzymatic form with altered Michaelis kinetics compared with the enzymatic form retained by the mycelium.

Background: Treatment of deep-vein thrombosis (DVT) with a once-daily regimen of enoxaparin, rather than a continuous infusion of unfractionated heparin (UFH) is more convenient and allows for home care in some patients. This study was designed to compare the efficacy and safety of these two regimens for the treatment of patients with proximal lower limb DVT. Methods: 201 patients with proximal lower limb DVT from 13 centers in Brazil were randomized in an open manner to receive either enoxaparin [1.5 mg/kg subcutaneous (s.c.) OD] or intravenous (i.v.) UFH (adjusted to aPTT 1.5-2.5 times control) for 5-10 days. All patients also received warfarin (INR 2-3) for at least 3 months. The primary efficacy endpoint Was recurrent DVT (confirmed by venography or ultrasonography), and safety endpoints included bleeding and serious adverse events. The rate of pulmonary embolism (PE) was also collected. Hospitalization was at the physician's discretion. Results: Baseline patient characteristics were comparable between groups. The duration of hospital stay was significantly shorter with enoxaparin than with UFH (3 versus 7 days). In addition, 36% of patients receiving enoxaparin did not need to be hospitalized, whereas all of the patients receiving UFH were! hospitalized. The treatment duration was slightly longer with enoxaparin (8 versus 7 days). There was a nonsignificant trend toward a reduction in the rate of recurrent DVT with enoxaparin versus UFH, and similar safety. Conclusions: A once-daily regimen of enoxaparin 1.5 mg/kg subcutaneous is at least as effective and safe as conventional treatment with a continuous intravenous infusion of UFH. However, the once daily enoxaparin regimen is easier to administer (subcutaneous versus intravenous), does not require aPTT monitoring, and leads to both a reduced number of hospital admissions and an average 4-day-shorter hospital stay. The common beans (Phaseolus vulgaris L.) is a fabacea sufficiently spread out in all domestic territory. However, the quality of its seeds represents one of the main causes of low productivity in the beans farmings in Brazil. The objective of this work was to evaluate physiological and sanitary seed qualities of eleven bean cultivars. The physiological seed quality was evaluated trough standard germination and vigor tests. The sanitary seed quality was evaluated through two tests: blotter test was employed to evaluate fungi incidence and Koch & Menten method was employed to observe Sclerotinia sclerotiorum (Lib) de Bary occurrence. Xamego, BRS Valente, Bambu and Pérola had the best results of physiological tests. Jalo Precoce, Roxo 90, Corrente and Aporé had no good results of vigor and germination, besides presenting the lowest indices of died seeds. Fusarium sp., Aspergillus spp., Penicillium sp., Phoma sp., Rhizopus sp. and Botrytis sp. were the fungi detected in the sanity tests.

The cotton disease known as angular leaf spot, caused by Xanthomonas axonopodis pv. malvacearum (Xam) has been causing cotton losses in several producing regions around the world. Xam is transmitted by seeds, which may be infected both externally and internally. Infected seeds constitute the main long-distance dissemination mode of the pathogen. In view of this, the use of healthy seeds is a must. To accomplish that, detection methodologies for the bacteria must be developed be used in seed health analysis laboratories. This study aimed to develop a semi-selective medium for Xam detection in cotton seeds. The semi-selective culture medium was named MSSXAN and it was consisted of peptone (5.0 g), beef extract (3 g), sucrose (5 g), soluble starch (10 g), agar (15 g), CaCl 2 (0.25 g), Tween 80 (10 mL), distilled water (1,000 mL), crystal violet solution at 1% (150 μL), cephalexin (50 mg 1*), methyl thyophanate (10 mg*) and chlorothalonil (10 mg*) - *added after culture medium autoclaving. This MSSXAN medium shows low repressiveness to Xam and it be used for isolation of this bacteria in cotton seeds health analysis. © 2009 Academic Journals Inc.
