The authors repport in this paper, the results of trials by means of which were determined the composition and nutritive value of three grasses, that are cultivated in Piracicaba, in State of São Paulo, Brazil: Capim jaragaá, Hyparrhenia rufa, (Nees.) Stapf. (hay); capim de Rhodes, Chloris gayana, Kunth. (hay), and milho Santa Rosa, Zea mays, L. (silage). The digestion trials were carried out with sheep, following the technique of MEAD and GUILBERT. The chemical analysis of the forages were made by methods usually employed. The results are in the tables I and II, of this paper.

Um experimento sobre adubação de milho (Zea maysL.), variedades "Agroceres - 13", foi conduzido no Município de Oliveira, Estado de Minas Gerais, com a finalidade de se estudar o efeito de adubos nitrogenado, fosfatado e potássico sobre a produção desse importante cereal. Os resultados obtidos a partir de um ensaio fatorial 2³ mostraram que a adubação nitrogenada foi a que proporcionou maior produção e renda líquida. Para a adubação fosfatada, houve um pequeno aumento de produção. Não houve resposta ao potássio.

Estudou-se efeito residual de oito fontes de fósforo em dois solos de baixo teor em fósforo, na cultura de milho (Zea mays L.). Os experimentos foram realizados na casa de vegetação. Pela avaliação do peso de matéria seca e quantidade de nitrogênio, fósforo e potássio absorvidos, constatou-se que os fertilizantes com melhor efeito residual foram metafosfato de cálcio e metafosfato de potássio, seguidos por termofosfato e fosforita de Olinda, no regosol. No latosol não se observou diferenças estatisticamente significativas para peso de materia seca, mas de uma maneira geral o metafosfato de cálcio, fosforita de Olinda e metafosfato de potássio foram os melhores. Os fertilizantes com menor efeito residual foram os que contem alto teor em fósforo solúvel em água: superfosfato triplo, superfosfato simples, fosfato diamonico e polifosfato de amonio.

Reguladores e estimulantes vegetais tem sido utilizados nas condições dos trópicos sem a verificação de seus efeitos sob condições controladas, levando frequentemente ao mascaramento de seus efeitos pelas condições do agroecossistema. Com a finalidade de observar os efeitos dessas substâncias sob condições de laboratório, sementes de Zea mays cv. C-525 e Lycopersicon esculentum cv. Kada foram submetidas aos efeitos de giberelina 100 ppm, chlormequat 2000 ppm, hidrazida maleica 1000 ppm, Agrostemin 1,25 g. l-1 e Atonik 0,5 ml.1-1, além do controle, em caixas de Petri. Observações realizadas 7 e 15 dias após a instalação do ensaio mostraram que giberelina e Agrostemin promoveram aumento no crescimento da radícula e do hipocótilo de milho e tomateiro. Hidrazida maleica inibiu o desenvolvimento da radícula das plântulas de milho. Hidrazida maleica e Atonik tenderam a reduzir o crescimento da radícula e do hipocótilo de tomateiro.

O presente trabalho teve como objetivo estudar os efeitos da aplicação de zinco em duas unidades de solos originalmente cobertas com vegetação de cerrado, classificadas como Latossolo Vermelho Amarelo-fase arenosa e Areias Quartzosas, que ocorrem no Município de São Simão no Estado de São Paulo Foram feitas determinações químicas do zinco solúvel em HCl 0,1 N, em amostras de solos antes e depois de terem recebido a calagem. As determinações de zinco foram complementadas com o teste microbiológico do Aspergilus niger Simultaneamente, foram conduzidos ensaios de campo com milho (Zea mays L.) e feijão Phaseolus vulgaris L.) para estudar os efeitos da aplicação de zinco nas duas unidades de solo. Outros experimentos com milho foram desenvolvidos sob condições controladas, em casa de vegetação, para estudos de aplicação de zinco. As conclusões do trabalho foram as seguintes: 1. Os resultados dos ensaios microbiológicos mostraram boa concordância com os testes químicos e com a resposta do milho a aplicação de zinco em condições de campo. 2. A cultura do milho deu boas respostas a aplicação de zinco quando cultido nos solos Areias Quartzosas, correspondendo a uma dose econômica de 2,4 kg/ha de zinco. 3. Não houve efeito do zinco para o milho quando este foi cultivado no Latossolo Vermelho Amarelo-fase arenosa. 4. Não houve efeitos do zinco na produção do feijoeiro nas duas unidades de solos. 5. O ensaio de vasos usando o milho como planta teste não foi eficiente na resposta dos solos as aplicações de zinco.

Novos estimulantes vegetais têm merecido estudos por possuírem alto potencial para melhorarem a produtividade de culturas. Triacontanol, um álcool alifático primário de cadeia longa isolado de vegetais encontra-se dentre essas substâncias. Com o objetivo de verificar o efeito deste estimulante vegetal sob condições de laboratório, sementes de Zea mays cv. C-525 e Lycopersicon esculentum cv. Kada foram submetidas aos efeitos de 10 ml de triacontanol 0,01, 0,1 e 1 mg. 1-1, além do controle, em caixas de Petri. Observações realizadas 7 e 14 dias após a instalação do ensaio mostraram que concentrações crescentes de triacontanol tendem a aumentar o crescimento da radícula e do hipocótilo do tomateiro, 14 dias após a aplicação. Nesta data notou-se também que concentrações crescentes de triacontanol tendem a incrementar o desenvolvimento do hipocótilo do milho.

De uma plantação bem conduzida de milho doce (Zea mays var. saahirata) cv. Contibrasil situada sobre um Latossolo Vermelho Escuro Orto, série "Luiz de Queiroz" de alta fertilidade natural no município de Piracicaba, SP, foram coletadas plantas aos 45, 60, 75, 90 e 105 dias após a germinação. As plantas foram divididas em folhas novas, velhas, colmo, pendão e espigas. O material após sofrer os processos necessários foi seco, pesado e analisado para N, P, K, Ca, Mg, S, B, Cu, Fe, Mn e Zn por métodos convencionais de laboratório. Os autores concluíram que: aumento de peso de matéria seca total é contínua até o final do ciclo; a acumulação de macronutrientes pela planta inteira obedece a seguinte ordem decrescente: N, K, P,Ca=S e Mg; o maior acúmulo de macronutrientes no final do ciclo ocorre no colmo, com exceção do N que é acumulado em maior quantidade nas espigas; o acúmulo de micronutrientes pela planta inteira obedece a seguinte ordem decrescente: Fe, Mn, Cu, Zn e B; o maior acúmulo de micronutrientes no final do ciclo ocorre no colmo; a exportação de nutrientes pela espiga representa 26,15% do total de nutrientes contidos na plantação.

Whiteflies and whitefly-transmitted viruses are some of the major constraints on European tomato production. The main objectives of this study were to: identify where and why whiteflies are a major limitation on tomato crops; collect information about whiteflies and associated viruses; determine the available management tools; and identify key knowledge gaps and research priorities. This study was conducted within the framework of ENDURE (European Network for Durable Exploitation of Crop Protection Strategies). Two whitefly species are the main pests of tomato in Europe: Bemisia tabaci and Trialeurodes vaporariorum. Trialeurodes vaporariorum is widespread to all areas where greenhouse industry is present, and B. tabaci has invaded, since the early 1990’s, all the subtropical and tropical areas. Biotypes B and Q of B. tabaci are widespread and especially problematic. Other key tomato pests are Aculops lycopersici, Helicoverpa armigera, Frankliniella occidentalis, and leaf miners. Tomato crops are particularly susceptible to viruses causingTomato yellow leaf curl disease (TYLCD). High incidences of this disease are associated to high pressure of its vector, B. tabaci. The ranked importance of B. tabaci established in this study correlates with the levels of insecticide use, showing B. tabaci as one of the principal drivers behind chemical control. Confirmed cases of resistance to almost all insecticides have been reported. Integrated Pest Management based on biological control (IPM-BC) is applied in all the surveyed regions and identified as the strategy using fewer insecticides. Other IPM components include greenhouse netting and TYLCD-tolerant tomato cultivars. Sampling techniques differ between regions, where decisions are generally based upon whitefly densities and do not relate to control strategies or growing cycles. For population monitoring and control, whitefly species are always identified. In Europe IPM-BC is the recommended strategy for a sustainable tomato production. The IPM-BC approach is mainly based on inoculative releases of the parasitoids Eretmocerus mundus and Encarsia formosa and/or the polyphagous predators Macrolophus caliginosus and Nesidiocoris tenuis. However, some limitations for a wider implementation have been identified: lack of biological solutions for some pests, costs of beneficials, low farmer confidence, costs of technical advice, and low pest injury thresholds. Research priorities to promote and improve IPM-BC are proposed on the following domains: (i) emergence and invasion of new whitefly-transmitted viruses; (ii) relevance of B. tabaci biotypes regarding insecticide resistance; (iii) biochemistry and genetics of plant resistance; (iv) economic thresholds and sampling techniques of whiteflies for decision making; and (v) conservation and management of native whitefly natural enemies and improvement of biological control of other tomato pests.

DNA based techniques have proved to be very useful methods to study trophic relationships 17 between pests and their natural enemies. However, most predators are best defined as omnivores, 18 and the identification of plant-specific DNA should also allow the identification of the plant 19 species the predators have been feeding on. In this study, a PCR approach based on the 20 development of specific primers was developed as a self-marking technique to detect plant DNA 21 within the gut of one heteropteran omnivorous predator (Macrolophus pygmaeus) and two 22 lepidopteran pest species (Helicoverpa armigera and Tuta absoluta). Specific tomato primers 23 were designed from the ITS 1-2 region, which allowed the amplification of a tomato DNA 24 fragment of 332 bp within the three insect species tested in all cases (100% of detection at t = 0) 25 and did not detect DNA of other plants nor of the starved insects. Plant DNA half-lives at 25ºC 26 ranged from 5.8h, to 27.7h and 28.7h within M. pygmaeus, H. armigera and T. absoluta, 27 respectively. Tomato DNA detection within field collected M. pygmaeus suggests dietary mixing 28 in this omnivorous predator and showed a higher detection of tomato DNA in females and 29 nymphs than males. This study provides a useful tool to detect and to identify plant food sources 30 of arthropods and to evaluate crop colonization from surrounding vegetation in conservation 31 biological control programs.

Membranes of maize (Zea mays L., cv LG 11) roots were fractionated by sucrose (in presence or absence of Mg2+) or dextran density gradient centrifugations and the locations of organelles were determined using marker enzymes. Latent UDPase was used as a Golgi marker, catalase for the peroxysomes, cytochrome c oxidase for the mitochondria, UDP-Gal-galactosyltransferase for the amyloplast membranes and NADH-cytochrome c reductase for the ER. Two markers were selected for the plasmalemma, the vanadate-sensitive ATPase and UDP-Glc-sterolglucosyltransferase. The distributions of the PPase and vacuolar ATPase were found to be similar after density gradient centrifugation. The PPase and vacuolar ATPase activities were clearly separated from almost all the other markers tested, however, a partial association of both activities with the ER cannot be completely ruled out. The PPase of maize roots is more active and easier to measure than the vacuolar ATPase and is therefore an excellent candidate for use as a tonoplast marker.

Calcium uptake by tonoplast enriched membrane vesicles from maize (Zea mays L. cv. LG 11) primary roots was studied. A pH gradient, measured by the fluorescence quenching of quinacrine, was generated across sealed vesicles driven by the pyrophosphate-dependent proton pump. The fluorescence quenching was strongly inhibited by Ca2+; moreover, when increasing Ca2+ concentrations were added to vesicles at steady-state, a concomitant decrease in the proton gradient was observed. Ca2+ uptake using Ca-45(2+) was linear from 10 min when oxalate (10 mM) was present, while Ca2+ uptake was completely inhibited with proton ionophores (FCCP and monensin), indicating a Ca2+/H+ antiport. Membranes were further fractionated using a linear sucrose density gradient (10-45%) and were identified with marker enzymes. Ca2+ uptake co-migrated with the tonoplast pyrophosphate-dependent proton pumping, pyrophosphatase and ATPase activities: the Ca2+/H+ antiport is consequently located at the tonoplast.

The subcellular localization of a calmodulin-stimulated calcium (Ca2+)-ATPase activity from maize roots (Zea mays L., cv LG 11) was studied. For this purpose, an efficient procedure was developed to prepare sealed plasma membrane vesicles allowing the measurement of proton and Ca2+ transport activities. Two-day-old root membranes were fractionated by sucrose and dextran density gradient centrifugation. Marker enzymes were used to study the distribution of the different membranes in the gradients and a filtration technique was developed to measure Ca-45(2+) transport in sealed vesicles. Most of the ATP-dependent Ca2+ transport activity was associated with the ER. However, a small part of this activity was associated with the tonoplast (corresponding to the activity of the H+/Ca2+ antiport) and the plasma membrane. When the Ca2+ transport was measured in the presence of exogenous calmodulin (1 muM), a 3-5-fold increase of uptake was measured. The calmodulin-stimulated activity was associated with the tonoplast vesicles only. This activity was insensitive to monensin, a proton ionophore, ruling out a direct effect of calmodulin on the H+/Ca2+ antiport. In conclusion, four different Ca2+ transporters are present in young maize root cells. A Ca2+/H+ antiport system is present on the tonoplast, whereas, the plasma membrane and the ER possess each a calmodulinin-sensitive Ca2+-ATPase. Finally, a calmodulin-stimulated Ca2+-ATPase is associated with the tonoplast.

The tonoplast calcium Ca2+/H+ antiport system of maize (Zea mays L. cv LG 11) roots was characterized using the ''pH jump'' technique in order to avoid interference from the tonoplast proton and Ca2+ pumps. Ca2+ uptake was recorded in the presence of different inhibitors and divalent ions. Chemical modification of amino acid residues of the antiport was used to elucidate the amino acid residues participating in the Ca2+ transport activity. The Ca2+/H+ antiport activity was found to be strongly inhibited by ruthenium red and verapamil, whereas diethylstilbestrol was less effective. Vanadate, erythrosin B, cyclopiazonic acid, bafilomycin, thapsigargin, N,N'-dicyclohexylcarbodiimide (DCCD) and 4,4'-diisothiocyanatostilbene-2,2'-disulfonate (DIDS) were without effect. Lanthanum and divalent ions were strongly inhibitory (Cd2+ > Mn2+ > Sr2+ > Ba2+). While reagents modifying sulfhydryl groups (N-ethylmaleimide and 5,5'-dithio-bis(2-nitrobenzoate)) did not affect the antiport activity, modification of trytophan residues (N-bromosuccinimide) was strongly inhibitory. We conclude that ruthenium red, verapamil, lanthanum and divalent cations directly inhibit Ca2+ uptake independent of the function of the proton and Ca2+ pumps. Moreover, the results of chemically modified amino acid residues suggest that sulfhydryl groups are not involved in Ca2+ transport, while tryptophan residues seem important for this translocation.

BACKGROUND To assess and compare the effectiveness and costs of Phototest, Mini Mental State Examination (MMSE), and Memory Impairment Screen (MIS) to screen for dementia (DEM) and cognitive impairment (CI). METHODS A phase III study was conducted over one year in consecutive patients with suspicion of CI or DEM at four Primary Care (PC) centers. After undergoing all screening tests at the PC center, participants were extensively evaluated by researchers blinded to screening test results in a Cognitive-Behavioral Neurology Unit (CBNU). The gold standard diagnosis was established by consensus of expert neurologists. Effectiveness was assessed by the proportion of correct diagnoses (diagnostic accuracy [DA]) and by the kappa index of concordance between test results and gold standard diagnoses. Costs were based on public prices and hospital accounts. RESULTS The study included 140 subjects (48 with DEM, 37 with CI without DEM, and 55 without CI). The MIS could not be applied to 23 illiterate subjects (16.4%). For DEM, the maximum effectiveness of the MMSE was obtained with different cutoff points as a function of educational level [k = 0.31 (95% Confidence interval [95%CI], 0.19-0.43), DA = 0.60 (95%CI, 0.52-0.68)], and that of the MIS with a cutoff of 3/4 [k = 0.63 (95%CI, 0.48-0.78), DA = 0.83 (95%CI, 0.80-0.92)]. Effectiveness of the Phototest [k = 0.71 (95%CI, 0.59-0.83), DA = 0.87 (95%CI, 0.80-0.92)] was similar to that of the MIS and higher than that of the MMSE. Costs were higher with MMSE (275.9 ± 193.3€ [mean ± sd euros]) than with Phototest (208.2 ± 196.8€) or MIS (201.3 ± 193.4€), whose costs did not significantly differ. For CI, the effectiveness did not significantly differ between MIS [k = 0.59 (95%CI, 0.45-0.74), DA = 0.79 (95%CI, 0.64-0.97)] and Phototest [k = 0.58 (95%CI, 0.45-0.74), DA = 0.78 (95%CI, 0.64-0.95)] and was lowest for the MMSE [k = 0.27 (95%CI, 0.09-0.45), DA = 0.69 (95%CI, 0.56-0.84)]. Costs were higher for MMSE (393.4 ± 121.8€) than for Phototest (287.0 ± 197.4€) or MIS (300.1 ± 165.6€), whose costs did not significantly differ. CONCLUSION MMSE is not an effective instrument in our setting. For both DEM and CI, the Phototest and MIS are more effective and less costly, with no difference between them. However, MIS could not be applied to the appreciable percentage of our population who were illiterate.

BACKGROUND Illiteracy, a universal problem, limits the utilization of the most widely used short cognitive tests. Our objective was to assess and compare the effectiveness and cost for cognitive impairment (CI) and dementia (DEM) screening of three short cognitive tests applicable to illiterates. METHODS Phase III diagnostic test evaluation study was performed during one year in four Primary Care centers, prospectively including individuals with suspicion of CI or DEM. All underwent the Eurotest, Memory Alteration Test (M@T), and Phototest, applied in a balanced manner. Clinical, functional, and cognitive studies were independently performed in a blinded fashion in a Cognitive Behavioral Neurology Unit, and the gold standard diagnosis was established by consensus of expert neurologists on the basis of these results. Effectiveness of tests was assessed as the proportion of correct diagnoses (diagnostic accuracy [DA]) and the kappa index of concordance (k) with respect to gold standard diagnoses. Costs were based on public prices at the time and hospital accounts. RESULTS The study included 139 individuals: 47 with DEM, 36 with CI, and 56 without CI. No significant differences in effectiveness were found among the tests. For DEM screening: Eurotest (k = 0.71 [0.59-0.83], DA = 0.87 [0.80-0.92]), M@T (k = 0.72 [0.60-0.84], DA = 0.87 [0.80-0.92]), Phototest (k = 0.70 [0.57-0.82], DA = 0.86 [0.79-0.91]). For CI screening: Eurotest (k = 0.67 [0.55-0.79]; DA = 0.83 [0.76-0.89]), M@T (k = 0.52 [0.37-0.67]; DA = 0.80 [0.72-0.86]), Phototest (k = 0.59 [0.46-0.72]; DA = 0.79 [0.71-0.86]). There were no differences in the cost of DEM screening, but the cost of CI screening was significantly higher with M@T (330.7 ± 177.1 €, mean ± sd) than with Eurotest (294.1 ± 195.0 €) or Phototest (296.0 ± 196. 5 €). Application time was shorter with Phototest (2.8 ± 0.8 min) than with Eurotest (7.1 ± 1.8 min) or M@T (6.8 ± 2.2 min). CONCLUSIONS Eurotest, M@T, and Phototest are equally effective. Eurotest and Phototest are both less expensive options but Phototest is the most efficient, requiring the shortest application time.
