目的 克隆和分析荧光素再生酶基因(LRE).方法 通过GeneBank中已知的荧光素再生酶基因保守区段设计引物,利用5'RACE(rapid-amplification of cDNA ends)和3'RACE技术克隆了来自云南省两双版纳州的卵黄萤(Luciola ovalis)荧光素再生酶基因cDNA和全基因序列.通过GeneBank、National Center for Bioteclmology Information和ProDom at the ExPASy Server软件和数据库进行序列分析.结果 卵黄萤荧光素再牛酶的cDNA序列和基因序列存在2个不同碱基位点,但是它们编码的荧光素再生酶是相同的.卵黄萤荧光素再生酶基因全长(从起始密码子到终止密码子)为1131 bp,包含5个外显子4个内含子,其cDNA 序列为1008 bp,包含924bp的荧光素酶基因开放阅读框和84 bp的3'UTR序列.卵黄萤荧光素酶基因的开放阅读框编码1个307个氨基酸的蛋白质.它与北美萤火虫(Photinus pyralis)荧光素再生酶在碱基序列和氨基酸序列上分别有61.8%和53.3%的相似性.结论 成功地克隆了荧光素再生酶的cDNA和基因序列,为其在基因工程中的应用奠定了基础.

目的 研究中华眼镜蛇毒因子(CVF)消耗补体对大鼠心脏移植急性排斥反应的影响.方法 以近交系BN大鼠为供者,Lewis大鼠为受者,建立腹腔异位心脏移植模型.实验分为2组,每组8只.CVF组:心脏移植术前3 d、2 d时,经受者尾静脉注射CVF 50 μg/kg;术前12 h至移植心停跳时,经尾静脉注射CVF 20 μg/kg,每2 d注射1次.对照组:术前、术后不给予受者任何特殊处理.术后观察移植心的存活时间,测定术后第1、3、5和6d及移植心停跳时的血清总补体活性(CH50法)、移植心C3沉积及CD3+T细胞浸润情况,并观察移植心的病理变化.结果 CVF组和对照组的移植心存活时间分别为(11.69±0.72)d和(6.65±0.35)d,两组比较,差异有统计学意义(P<0.01).CVF组移植心组织内C3沉积和CD3+T细胞浸润程度均较对照组同期明显减轻,病理损害程度也较对照组同期明显减轻.结论 CVF消耗补体对大鼠心脏移植急性排斥反应起到了明显的抑制作用,从而延长移植心存活时间.

本文比较了3种臭蛙,即云南臭蛙(R. andersonii)、无指盘臭蛙(R.grahami)和滇南臭蛙(R.tiennanensis)的核型、C-带和银染核仁组织者区(Ag-NORs)。结果表明,3种臭蛙的核型均为2n=26(NF=52),由5对大型染色体和8对小型染色体组成,与蛙科大多数种类的核型相似,但是染色体形态和次缢痕的位置和数目有差异。其中,云南臭蛙和无指盘臭蛙的核型相似程度比较大,与其他已报道的臭蛙类的核型相差不大,而滇南臭蛙的核型比较特殊,与其他臭蛙类的核型存在显著差异。 C-带技术显示的结果表明,3种臭蛙所有染色体的着丝点都有比较显著的C-带,而居间区C-带和端点C-带却存在明显的差异。3种中,滇南臭蛙的C-带带型比较特殊。有趣的是在研究中还发现,云南臭蛙的早中期细胞(染色体很长)染色体上发现了很多结构异染色质区,除着丝点C-带外,几乎所有的染色体上都有居间区或端点C-带,而在晚中期细胞中所观察到的C-带数目却较少,除着丝点外,只在少数染色体上发现了C-带。 云南臭蛙和无指盘臭蛙的Ag-NORs位于第10对染色体的长臂上,与该染色体上的次缢痕位置相对应,而滇南臭蛙的Ag-NORs位于第6...

We identified a new class of human immunodeficiency virus type 1 (HIV-1) recombinants (00CN-HH069 and 00CN-HH086) in which further recombination occurred between two established circulating recombinant forms (CRFs). These two isolates were found among 57 HIV-1 samples from a cohort of injecting drug users in eastern Yunnan Province of China. Informative-site analysis in conjunction with bootscanning plots and exploratory tree analysis revealed that these two strains were closely related mosaics comprised of CRF07_BC and CRF08_BC, which are found in China. The genotype screening based on gag-reverse transcriptase sequences if 57 samples from eastern Yunnan identified 47 CRF08_BC specimens (82.5%), 5 CRF07_BC specimens (8.8%), and 3 additional specimens with the novel recombinant structure. These new "second-generation" recombinants thus constitute a substantial proportion (5 of 57; 8.8%) of HIV-1 strains in this population and may belong to a new but yet-undefined class of CRF. This might be the first example of CRFs recombining with each other, leading to the evolution of second-generation inter-CRF recombinants.

Sodium rutin sulfate (SRS) is a sulfated rutin modified from the natural flavonol glycoside rutin. Here, we investigated its in vitro anti-HIV and -HSV activities and its cytotoxic profile. Fifty percent inhibitory concentration (IC50) values of SRS against HIV-1 X4 virus IIIB, HIV-1 R5 isolates Ada-M and Ba-L were 2.3 +/- 0.2, 4.5 +/- 2.0 and 8.5 +/- 3.8 mu M with a selectivity index (SI) of 563, 575 and 329, respectively. Its IC50 against primary R5 HIV-1 isolate from Yunnan province in China was 13.1 +/- 5.5 mu M, with a Sl of 197. In contrast, unsulfated rutin had no activity against any of the HIV-1 isolates tested. Further study indicated that SRS blocked viral entry and virus-cell fusion likely through interacting with the HIV- I envelope glycoprotein. SRS also demonstrated some activity against human herpes simplex virus (HSV) with an IC50 of 88.3 +/- 0.1 mu M and a Sl of 30. The 50% cytotoxicity concentration (CC50) of SRS was >3.0 mM, as determined in human genital ME 180, HeLa and primary human foreskin fibroblast cells. Minimum inhibitory concentration of SRS for vaginal lactobacilli was >3.0 mM. These results collectively indicate that SRS represents a novel candidate for anti-HIV-1/HSV microbicide development. 利用在信阳农村养猪场中的2个3.3 m深的污水氧化塘进行光合细菌和水浮莲净化污水的研究。1#氧化塘加入8×108mL-1光合细菌50 L,2#氧化塘加入水浮莲(100株.m-2)。为期1 a的试验结果表明:经光合细菌和水浮莲的共同作用,总磷下降了41.8%~76.0%,总氮下降了61.8%~81.0%,COD下降了66.8%~79.1%,BOD5下降了64.8%~82.3%。显示光合细菌和水浮莲能够明显改善养猪场污水水质。

To extract gas from hydrate reservoirs, it has to be dissociated in situ. This endothermic dissociation process absorbs heat energy from the formation and pore fluid. The heat transfer governs the dissociation rate, which is proportional to the difference between the actual temperature and the equilibrium temperature. This study compares three potential gas production schemes from hydrate-bearing soil, where the radial heat transfer is governing. Cylindrical samples with 40% pore-filling hydrate saturation were tested. The production tests were carried out over 90 min by dissociating the hydrate from a centered miniature wellbore, by either lowering the pressure to 6, 4, or 6 MPa with simultaneous heating of the wellbore to 288 K. All tests were replicated by a numerical simulation. With additional heating at the same wellbore pressure, the gas production from hydrates could, on average, be increased by 1.8 and 3.6 times in the simulation and experiments, respectively. If the heat influx from the outer boundary is limited, a simulation showed that the specific heat of the formation is rapidly used up when the wellbore is only depressurized and not heated. © 2012 American Chemical Society.

以鳞片作为鉴定年龄的材料,对龙溪河厚颌鲂(Megalobrama pellegrini)种群开展年龄结构与生长特性的研究。结果表明,厚颌鲂鳞片年轮结构呈疏密切割型,年轮特征显著,可用于年龄鉴定,3~4月是年轮形成高峰时间。种群由0~7龄共8个龄组组成,以低龄个体为主,1~3龄个体占83.14%,体长分布主要集中在130~250 mm间(77.71%)。体质量分布主要集中在50~250 g(64.33%)。群体总性比为♀∶♂=1∶1.03,符合1∶1理论比值。体长和鳞径呈直线关系,体长和体质量呈幂函数关系且

VP2 8是对虾白斑综合症病毒 (Whitespotsyndromevirus ,WSSV)的一个重要的囊膜蛋白。为了便于研究VP2 8在和宿主细胞相互作用过程中扮演的角色 ,将VP2 8基因克隆到一个原核表达系统 ,对原核表达的VP2 8的特性进行了研究 ,并制备了抗VP2 8的多克隆抗体和单链抗体 ,对原核表达的和天然病毒的VP2 8蛋白免疫原性进行了比较。结果表示 ,原核表达的VP2 8与天然蛋白具有相似的免疫原性。
