Nitric Oxide (NO) has been known for long to regulate vessel tone. However, the close proximity of the site of NO production to “sinks” of NO such as hemoglobin (Hb) in blood suggest that blood will scavenge most of the NO produced. Therefore, it is unclear how NO is able to play its physiological roles. The current study deals with means by which this could be understood. Towards studying the role of nitrosothiols and nitrite in preserving NO availability, a study of the kinetics of glutathione (GSH) nitrosation by NO donors in aerated buffered solutions was undertaken first. Results suggest an increase in the rate of the corresponding nitrosothiol (GSNO) formation with an increase in GSH with a half-maximum constant EC50 that depends on NO concentration, thus indicating a significant contribution of ∙NO2 mediated nitrosation in the production of GSNO. Next, the ability of nitrite to be reduced to NO in the smooth muscle cells was evaluated. The NO formed was inhibited by sGC inhibitors and accelerated by activators and was independent of O2 concentration. Nitrite transport mechanisms and effects of exogenous nitrate on transport and reduction of nitrite were examined. The results showed that sGC can mediate nitrite reduction to NO and nitrite is transported across the smooth muscle cell membrane via anion channels, both of which can be attenuated by nitrate. Finally, a 2 – D axisymmetric diffusion model was constructed to test the accumulation of NO in the smooth muscle layer from reduction of nitrite. It was observed that at the end of the simulation period with physiological concentrations of nitrite in the smooth muscle cells (SMC), a low sustained NO generated from nitrite reduction could maintain significant sGC activity and might affect vessel tone. The major nitrosating mechanism in the circulation at reduced O2 levels was found to be anaerobic and a Cu+ dependent GSNO reduction activity was found to deliver minor amounts of NO from physiological GSNO levels in the tissue.

This dissertation examines the quality of hazard mitigation elements in a coastal, hazard prone state. I answer two questions. First, in a state with a strong mandate for hazard mitigation elements in comprehensive plans, does plan quality differ among county governments? Second, if such variation exists, what drives this variation? My research focuses primarily on Florida’s 35 coastal counties, which are all at risk for hurricane and flood hazards, and all fall under Florida’s mandate to have a comprehensive plan that includes a hazard mitigation element. Research methods included document review to rate the hazard mitigation elements of all 35 coastal county plans and subsequent analysis against demographic and hazard history factors. Following this, I conducted an electronic, nationwide survey of planning professionals and academics, informed by interviews of planning leaders in Florida counties. I found that hazard mitigation element quality varied widely among the 35 Florida coastal counties, but were close to a normal distribution. No plans were of exceptionally high quality. Overall, historical hazard effects did not correlate with hazard mitigation element quality, but some demographic variables that are associated with urban populations did. The variance in hazard mitigation element quality indicates that while state law may mandate, and even prescribe, hazard mitigation in local comprehensive plans, not all plans will result in equal, or even adequate, protection for people. Furthermore, the mixed correlations with demographic variables representing social and disaster vulnerability shows that, at least at the county level, vulnerability to hazards does not have a strong effect on hazard mitigation element quality. From a theory perspective, my research is significant because it compares assumptions about vulnerability based on hazard history and demographics to plan quality. The only vulnerability-related variables that appeared to correlate, and at that mildly so, with hazard mitigation element quality, were those typically representing more urban areas. In terms of the theory of Neo-Institutionalism and theories related to learning organizations, my research shows that planning departments appear to have set norms and rules of operating that preclude both significant public involvement and learning from prior hazard events.

County jurisdictions in America are increasingly exercising self-government in the provision of public community services through the context of second order federalism. In states exercising this form of contemporary governance, county governments with “reformed” policy-making structures and professional management practices, have begun to rival or surpass municipalities in the delivery of local services with regional implications such as environmental protection (Benton 2002, 2003; Marando and Reeves, 1993). The voter referendum, a form of direct democracy, is an important component of county land preservation and environmental protection governmental policies. The recent growth and success of land preservation voter referendums nationwide reflects an increase in citizen participation in government and their desire to protect vacant land and its natural environment from threats of over-development, urbanization and sprawl, loss of open space and farmland, deterioration of ecosystems, and inadequate park and recreational amenities. The study’s design employs a sequential, mixed method. First, a quantitative approach employs the Heckman two-step model. It is fitted with variables for the non-random sample of 227 voter referendum counties and all non-voter referendum counties in the U.S. from 1988 to 2009. Second, the qualitative data collected from the in-depth investigation of three South Florida county case studies with twelve public administrator interviews is transformed for integration with the quantitative findings. The purpose of the qualitative method is to complement, explain and enrich the statistical analysis of county demographic, socio-economic, terrain, regional, governance and government, political preference, environmentalism, and referendum-specific factors. The research finds that government factors are significant in terms of the success of land preservation voter referendums; more specifically, the presence of self-government authority (home rule charter), a reformed structure (county administrator/manager or elected executive), and environmental interest groups. In addition, this study concludes that successful counties are often located coastal, exhibit population and housing growth, and have older and more educated citizens who vote democratic in presidential elections. The analysis of case study documents and public administrator interviews finds that pragmatic considerations of timing, local politics and networking of regional stakeholders are also important features of success. Further research is suggested utilizing additional public participation, local government and public administration factors.

This presentation was given at the Panhandle Library Access Network's (PLAN) Innovation Conference: Digitization- Preserving the Past for the Future Conference on August 14th, 2015. The presentation uses a specific collection of directories as a case study of the complications librarians and archivists face in digitizing older materials that may also be quite large, such as a directory. Prime OCR and Abbyy Fine Reader are discussed and their pros and cons covered. Troubleshooting and editing with Adobe Photoshop is also discussed.

In recent years, there has been an enormous growth of location-aware devices, such as GPS embedded cell phones, mobile sensors and radio-frequency identification tags. The age of combining sensing, processing and communication in one device, gives rise to a vast number of applications leading to endless possibilities and a realization of mobile Wireless Sensor Network (mWSN) applications. As computing, sensing and communication become more ubiquitous, trajectory privacy becomes a critical piece of information and an important factor for commercial success. While on the move, sensor nodes continuously transmit data streams of sensed values and spatiotemporal information, known as ``trajectory information". If adversaries can intercept this information, they can monitor the trajectory path and capture the location of the source node. This research stems from the recognition that the wide applicability of mWSNs will remain elusive unless a trajectory privacy preservation mechanism is developed. The outcome seeks to lay a firm foundation in the field of trajectory privacy preservation in mWSNs against external and internal trajectory privacy attacks. First, to prevent external attacks, we particularly investigated a context-based trajectory privacy-aware routing protocol to prevent the eavesdropping attack. Traditional shortest-path oriented routing algorithms give adversaries the possibility to locate the target node in a certain area. We designed the novel privacy-aware routing phase and utilized the trajectory dissimilarity between mobile nodes to mislead adversaries about the location where the message started its journey. Second, to detect internal attacks, we developed a software-based attestation solution to detect compromised nodes. We created the dynamic attestation node chain among neighboring nodes to examine the memory checksum of suspicious nodes. The computation time for memory traversal had been improved compared to the previous work. Finally, we revisited the trust issue in trajectory privacy preservation mechanism designs. We used Bayesian game theory to model and analyze cooperative, selfish and malicious nodes' behaviors in trajectory privacy preservation activities.

A estética é algo que interfere no bem-estar das pessoas e o sorriso não é excepção, sendo até considerado um factor estético essencial nos dias de hoje, perante uma sociedade cada vez mais exigente relativamente aos padrões de beleza. A área da estética dentária, encontra-se em constantes mudanças devido à evolução da qualidade dos materiais e das técnicas utilizadas, levando a melhorias na reprodução das características naturais dos dentes. O médico dentista tem a responsabilidade de adquirir conhecimento e habilidades profissionais para a elaboração de tratamentos estéticos dentários que satisfaçam as expectativas dos pacientes quanto ao seu sorriso. Actualmente, o médico dentista possui várias opções para planear os tratamentos, entre as quais, o planeamento digital através do Digital Smile Design. O Digital Smile Design (DSD), criado pelo Doutor Christian Coachman, veio responder à procura elevada por tratamentos cada vez mais personalizados por parte dos pacientes. O DSD amplia a visão relativamente aos diagnósticos, melhora a comunicação entre as diferentes especialidades na área da medicina dentária e cria planos previsíveis durante o tratamento dentário. Trata-se de um programma onde são trabalhadas imagens fotográficas do paciente para a elaboração de um tratamento estético que responda as necessidades biológicas, funcionais e emocionais do paciente. Este poderá acompanhar e visualizar todos os passos do tratamento e deste modo, torna-se parte integrante do processo. O paciente expressa a sua opinião e as suas expectativas quanto ao resultado final. Neste trabalho realizou-se uma revisão narrativa da literatura sobre a técnica Digital Smile Design utilizando as palavras-chaves: Digital Smile Design; Visagism; dental planning; meaning of smile; mock-up. Os objetivos deste trabalho foi o de conhecer a técnica Digital Smile Design, os princípios do visagismo e a importância do planeamento nestes contextos. O sucesso de um tratamento está dependente de um correcto planeamento e de uma execução clínica e laboratorial cuidadosa.

Metadata that is associated with either an information system or an information object for purposes of description, administration, legal requirements, technical functionality, use and usage, and preservation, plays a critical role in ensuring the creation, management, preservation and use and re-use of trustworthymaterials, including records. Recordkeeping1 metadata, of which one key type is archival description, plays a particularly important role in documenting the reliability and authenticity of records and recordkeeping systemsas well as the various contexts (legal-administrative, provenancial, procedural, documentary, and technical) within which records are created and kept as they move across space and time. In the digital environment, metadata is also the means by which it is possible to identify how record components – those constituent aspects of a digital record that may be managed, stored and used separately by the creator or the preserver – can be reassembled to generate an authentic copy of a record or reformulated per a user’s request as a customized output package.Issues relating to the creation, capture, management and preservation of adequate metadata are, therefore, integral to any research study addressing the reliability and authenticity of digital entities, regardless of the community, sector or institution within which they are being created. The InterPARES 2 Description Cross-Domain Group (DCD) examined the conceptualization, definitions, roles, and current functionality of metadata and archival description in terms of requirements generated by InterPARES 12. Because of the needs to communicate the work of InterPARES in a meaningful way across not only other disciplines, but also different archival traditions; to interface with, evaluate and inform existing standards, practices and other research projects; and to ensure interoperability across the three focus areas of InterPARES2, the Description Cross-Domain also addressed its research goals with reference to wider thinking about and developments in recordkeeping and metadata. InterPARES2 addressed not only records, however, but a range of digital information objects (referred to as “entities” by InterPARES 2, but not to be confused with the term “entities” as used in metadata and database applications) that are the products and by-products of government, scientific and artistic activities that are carried out using dynamic, interactive or experiential digital systems. The nature of these entities was determined through a diplomatic analysis undertaken as part of extensive case studies of digital systems that were conducted by the InterPARES 2 Focus Groups. This diplomatic analysis established whether the entities identified during the case studies were records, non-records that nevertheless raised important concerns relating to reliability and authenticity, or “potential records.” To be determined to be records, the entities had to meet the criteria outlined by archival theory – they had to have a fixed documentary format and stable content. It was not sufficient that they be considered to be or treated as records by the creator. “Potential records” is a new construct that indicates that a digital system has the potential to create records upon demand, but does not actually fix and set aside records in the normal course of business. The work of the Description Cross-Domain Group, therefore, addresses the metadata needs for all three categories of entities.Finally, since “metadata” as a term is used today so ubiquitously and in so many different ways by different communities, that it is in peril of losing any specificity, part of the work of the DCD sought to name and type categories of metadata. It also addressed incentives for creators to generate appropriate metadata, as well as issues associated with the retention, maintenance and eventual disposition of the metadata that aggregates around digital entities over time.

La empresa Agentur es una agencia de viajes con más de 60 años de experiencia que recientemente ha incursionado en las ventas on-line por medio de un portal web. Actualmente, la empresa no cuenta con un plan de mercadeo que le permita aumentar exponencialmente las ventas por este canal, por lo que el desempeño de este no ha sido el esperado. Debido a este problema es de suma importancia determinar ¿Cuáles son las estrategias de posicionamiento y mercadeo on-line que debe usar la empresa para aumentar las ventas por su portal web? Para realizar este plan de mercadeo, se utilizó la segmentación propuesta por la empresa AMADEUS, llamada “FUTURE TRAVELLER TRIBES 2030 UNDERSTANDING TOMORROW’S TRAVELLER” en las que se explora los diferentes tipos de personas que viajaran en el futuro dependiendo de sus hábitos de compra, personalidad y necesidades específicas.. A partir de esta segmentación, se escogieron los segmentos que más se ajustan a lo que ofrece la página web de Agentur, y se diseñó una estrategia de posicionamiento que le permitirá a la empresa llegar a sus consumidores potenciales y aumentar las ventas de su portal. Se realizó una investigación de cuáles eran las mejores estrategias de publicidad y mercadeo que se tenían que utilizar para llegar a cada segmento de forma individual además del costo que tendría esta estrategia, y de esta forma lograr el volumen de ventas que desea alcanzar la empresa con este portal.

El interés de este caso de estudio es reflexionar sobre los procesos participativos en la formulación de instrumentos de planeamiento urbano y sus efectos en la gestión de las ciudades, particularmente en el espacio público. Esto se realiza partir de una revisión normativa en el presente y de algunos aspectos históricos de los procesos de “Participación Ciudadana” y del concepto de “Espacio Público” como escenarios propicios para que la ciudadanía aporte en las decisiones de planificación. Además, se plantea una revisión de cómo estos se constituyen en elementos estructurantes de la ciudad que la dotan de identidad, referentes simbólicos y apropiación ciudadana. Para este efecto, se realizó un balance de la estrategia de gestión social y participativa propuesta por el Plan Maestro de Espacio Público de 2005, con el fin de resaltar la importancia de la participación ciudadana en la gestión de los instrumentos de planificación con los que cuenta la ciudad y sus resultados en la gestión de espacio público en Bogotá.

Este estudio de caso presenta el rol de la cooperación internacional para avanzar hacia un modelo de desarrollo sostenible en Colombia, analizando los ámbitos en que el Programa 21 permeó la agenda ambiental del país durante el periodo de diseño e implementación del Plan Nacional de Desarrollo 2010-2014 (PND). Se analizan los logros y retos del PND a la luz del Programa 21 desde dos de los pilares que representan un mayor desafío para la construcción del desarrollo sostenible en el país: (1)crecimiento sostenible y competitivo y (2) la sostenibilidad ambiental

Actualmente, la competitividad entre empresas es cada vez más fuerte. Por tanto, las empresas deben innovar en sus procesos, para mantenerse en el mercado. Sin embargo, en la era actual y con la revolución en la tecnología, lo digital se ha vuelto tendencia y una necesidad para los negocios, lo cual, lo lleva a mejorar su posicionamiento, abarcar y conseguir nuevos clientes. De esta forma, el ambiente digital no es una opción al momento de definir las estrategias de una compañía, es una necesidad. El objetivo de este trabajo es aplicar los fundamentos teóricos del marco digital, para así plantear un plan de mercadeo que se ajuste mejor, teniendo en cuenta el sector de empresa y mercado en el que se piensa laborara, además de mostrar los impactos que este tipo de estrategia tiene para el presupuesto y los costos dentro de la misma.

Cartonería Mosquera S.A. es una empresa familiar, que se encarga de la producción y distribución de diferentes empaques de cartón. La compañía pertenece al sector de artes gráficas colombiano, que se desagrega en las siguientes categorías: empaques y etiquetas; publicidad y comercial; editoriales, periódicos y revistas. Cartonería Mosquera S.A. se encuentra en la categoría de empaques y etiquetas. A 2016 la empresa cuenta con un área que administra tanto el mercadeo como la parte comercial conjuntamente y que tiene como objetivo final la gestión de la publicidad, la satisfacción del cliente y la fidelización del mismo. Aunque desde sus inicios, la compañía ha venido trabajando con esta estructura y ha cumplido hasta ahora con algunos de los resultados esperados, los nuevos retos del mercado tales como; el compromiso con el medio ambiente, la necesidad de realizar registros que avalen la calidad de la compañía, la entrada de competidores internos como externos al mercado y el desarrollo de nuevos insumos, ha creado la necesidad de buscar nuevas herramientas que mejoren el desempeño del área comercial y promover así el incremento de las ventas. Este Trabajo surgió inicialmente con la idea de realizar un plan de mercadeo a fin de incentivar y proponer una estrategia que generara un impacto positivo en las ventas de la empresa. Sin embargo, en el proceso de desarrollo, se vio la necesidad de realizar primero un plan de mejoramiento con una orientación a la parte comercial de la compañía. Es importante resaltar que el enfoque de este documento investigativo no solo proveerá herramientas para el mejoramiento de las ventas y financiero, sino también pretende mejorar la percepción de los asesores comerciales hacia su trabajo, sus jefes y la compañía.

El planteamiento inicial del plan de negocios para Tamarindo Seco tiene como objetivo encontrar oportunidades de negocio con los productos actuales para involucrase y aportar al sector de consumo masivo en lo que se refiere a la alimentación saludable, con el único fin de cumplir un propósito superior, promover una cultura medio ambiental sostenible a través de la modificación en el manejo que se le da a los desperdicios que genera el consumo masivo de alimentos en Colombia y la región. Teniendo una trazabilidad de los productos que permitan tener políticas de comercio justo con cada uno de los implicados en el proceso productivo como comercial.

Esta investigación describe la situación de cómo Youtube se ha convertido a partir de sus estrategias y plan de mercadeo en la plataforma número uno en variedad de clips de películas, vídeos musicales, video de blogs, entre otros; llegando a popularizarse como una red social. Las redes sociales han desarrollado una nueva forma de comunicar y son una herramienta fundamental para la creación de conocimiento colectivo, es el caso de YouTube buscador de contenido audiovisual y red social que permite a millones de usuarios conectarse alrededor del mundo. Esta plataforma rompe las barreras culturales y de comunicación que anteriormente existían a falta de internet. En este sentido se pretende analizar a YouTube desde una perspectiva administrativa enfocada en el área de mercadeo.
