In an effort to control fugitive dust on a gravel surfaced roadway in Boone County, a cationic asphalt emulsion was blended with warm water and applied with an asphalt distributor. The test included various application procedures. After visual observations, it was concluded that this procedure utilizing a dilute asphalt emulsion was not an effective method of dust control.

A study of hydrochemical characteristics of the Arbúcies river (a seminatural mediterranean basin) has been undertaken during 1991. Mean solute yield for each ion has been determined using the correlation curves between discharge and concentration, and the flow duration curve. The hydrochemical budget of the Arbúcies drainage basin shows that the basin is a net source for al1 the elements. The cationic denudation rate obtained in this granitic basin is 5.24 keqhalyear. This high rate is due to the intensive chemical weathering of the granodionte, that delivers an important amount of cations which will release to be transported. The results demostrate clearly the strength of lithological control on stream loads, particulary on the dissolved component.

Membrane-active antimicrobial peptides, such as polymyxin B (PxB), are currently in the spotlight as potential candidates toovercome bacterial resistance. We have designed synthetic analogs ofPxB in order to determine the structural requirements for membraneaction. Since the mechanism of action of PxB involves interaction withboth the outer membrane and the cytoplasmic membrane of Gramnegative bacteria, we have used an approach based on mimicking theouter layers of these membranes using monolayers, Langmuir-Blodgettfilms and unilamelar vesicles, and applying a battery of biophysicalmethods in order to dissect the different events of membraneinteraction. Collectively, results indicate that the PxB analogues act inthe bacterial membrane by the same mechanism than PxB, and that cationic amphipathicity determines peptide activity.

Iron deficiency is generally investigated when faced with anemia, or with symptoms that could be related to iron deficiency without anemia. This simple disorder is easy to treat, provided that the diagnosis is correct. Several biological tests are available, but their interpretation is oftentimes problematic. Pre-analytical factors can interfere with measurements, normal values can change depending on suppliers, and, above all, results from different markers can be contradictory in some clinical situations. The aim of this article is to evaluate how the evolution of scientific knowledge and clinical trials can contribute to a better understanding and greater reliability in the diagnosis of iron deficiency.

Two common lung-related complications in the neonate are respiratory distress syndrome, which is associated with a failure to generate low surface tension at the air-liquid interface because of pulmonary surfactant insufficiency, and bronchopulmonary dysplasia (BPD), a chronic lung injury with reduced alveolarization. Surfactant phosphatidylcholine (PC) molecular species composition during alveolarization has not been examined. Mass spectrometry analysis of bronchoalveolar lavage fluid of rodents and humans revealed significant changes in surfactant PC during alveolar development and BPD. In rats, total PC content rose during alveolarization, which was caused by an increase in palmitoylmyristoyl-PC (16:0/14:0PC) concentration. Furthermore, two animal models of BPD exhibited a specific reduction in 16:0/14:0PC content. In humans, 16:0/14:0PC content was specifically decreased in patients with BPD and emphysema compared with patients without alveolar pathology. Palmitoylmyristoyl-PC content increased with increasing intrinsic surfactant curvature, suggesting that it affects surfactant function in the septating lung. The changes in acyl composition of PC were attributed to type II cells producing an altered surfactant during alveolar development. These data are compatible with extracellular surfactant 16:0/14:0PC content being an indicator of alveolar architecture of the lung.

Asphalt is used as a binder for thin maintenance surface (TMS) applications because of two key properties, it is waterproof and it adheres relatively well to the aggregate. Since asphalt is too stiff at room temperature to apply to the road surface, it is usually applied as either a cutback asphalt or an asphalt emulsion. The asphalt emulsions can be further divided into high float emulsions, cationic emulsions or polymer-modified binders, which are emulsions with polymers added to them. These types of binders are discussed further below.

Road dust is caused by wind entraining fine material from the roadway surface and the main source of Iowa road dust is attrition of carbonate rock used as aggregate. The mechanisms of dust suppression can be considered as two processes: increasing particle size of the surface fines by agglomeration and inhibiting degradation of the coarse material. Agglomeration may occur by capillary tension in the pore water, surfactants that increase bonding between clay particles, and cements that bind the mineral matter together. Hygroscopic dust suppressants such as calcium chloride have short durations of effectiveness because capillary tension is the primary agglomeration mechanism. Somewhat more permanent methods of agglomeration result from chemicals that cement smaller particles into a mat or larger particles. The cements include lignosulfonates, resins, and asphalt products. The duration of the cements depend on their solubility and the climate. The only dust palliative that decreases aggregate degradation is shredded shingles that act as cushions between aggregate particles. It is likely that synthetic polymers also provide some protection against coarse aggregate attrition. Calcium chloride and lignosulfonates are widely used in Iowa. Both palliatives have a useful duration of about 6 months. Calcium chloride is effective with surface soils of moderate fine content and plasticity whereas lignin works best with materials that have high fine content and high plasticity indices. Bentonite appears to be effective for up to two years and works well with surface materials having low fines and plasticity and works well with limestone aggregate. Selection of appropriate dust suppressants should be based on characterization of the road surface material. Estimation of dosage rates for potential palliatives can be based on data from this report, from technical reports, information from reliable vendors, or laboratory screening tests. The selection should include economic analysis of construction and maintenance costs. The effectiveness of the treatment should be evaluated by any of the field performance measuring techniques discussed in this report. Novel dust control agents that need research for potential application in Iowa include; acidulated soybean oil (soapstock), soybean oil, ground up asphalt shingles, and foamed asphalt. New laboratory evaluation protocols to screen additives for potential effectiveness and determine dosage are needed. A modification of ASTM D 560 to estimate the freeze-thaw and wet-dry durability of Portland cement stabilized soils would be a starting point for improved laboratory testing of dust palliatives.

Luminescence spectroscopy has been used to characterize MgO films prepared by rf-sputtering. A clear correlation is found between the appearance of an emission peak centered at approximately 460 nm and the detection of ferromagnetic ordering in the samples. We suggest that cationic vacancies are responsible for the blue-light emission by introducing p states into the electronic band-gap. In accordance with this, our results strongly indicate that cationic vacancies are at the heart of the appearance of long-range magnetic ordering in MgO films.

Non-viral vectors for potential gene replacement and therapy have been developed in order to overcome the drawbacks of viral vectors. The diversity of non-viral vectors allows for a wide range of various products, flexibility of application, ease of use, low-cost of production and enhanced "genomic" safety. Using non-viral strategies, oligonucleotides (ODNs) can be delivered naked (less efficient) or entrapped in cationic lipids, polymers or peptides forming slow release delivery systems, which can be adapted according to the organ targeted and the therapy purposes. Tissue and cell internalization can be further enhanced by changing by physical or chemical means. Moreover, a specific vector can be selected according to disease course and intensity of manifestations fulfilling specific requirements such as the duration of drug release and its level along with cells and tissues specific targeting. From accumulating knowledge and experience, it appears that combination of several non-viral techniques may increase the efficacy and ensure the safety of these evolving and interesting gene therapy strategies.

Laktoosi eli maitosokeri on tärkein ainesosa useimpien nisäkkäiden tuottamassa maidossa. Sitä erotetaan herasta, juustosta ja maidosta. Laktoosia käytetään elintarvike- ja lääketeollisuuden raaka-aineena monissaeri tuotteissa. Lääketeollisuudessa laktoosia käytetään esimerkiksi tablettien täyteaineena. Hapettamalla laktoosia voidaan valmistaa laktobionihappoa, 2-keto-laktobionihappoa ja laktuloosia. Laktobionihappoa käytetään biohajoavien pintojen ja kosmetiikkatuotteiden valmistuksessa, sekä sisäelinten säilöntäliuoksissa, joissa laktobionihappo estää happiradikaalien aiheuttamien kudosvaurioiden syntymistä. Tässä työssä laktoosia hapetettiin laktobionihapoksi sekoittimella varustetussa laboratoriomittakaavaisessa panosreaktorissa käyttäenkatalyyttinä palladiumia aktiivihiilellä. Muutamissa kokeissa katalyytin promoottorina käytettiin vismuttia, joka hidastaa katalyytin deaktivoitumista. Työn tarkoituksena oli saada lisää tietoa laktoosin hapettamisen kinetiikasta. Laktoosin hapettumisessa laktobionihapoksi havaittiin selektiivisyyteen vaikuttavan muunmuassa reaktiolämpötila, paine, pH ja käytetyn katalyytin määrä. Katalyyttiä kierrättämällä eri kokeiden välillä saatiin paremmat konversiot, selektiivisyydet ja saannot. Parhaat koetulokset saatiin hapetettaessa synteettisellä ilmalla 60 oC lämpötilassa ja 1 bar paineessa. Tehdyissä kokeissa pH:n säätö tehtiin manuaalisesti, joten pH ei pysynyt koko ajan haluttuna. Laktoosin konversio oli parhaimmillaan 95 %. Laktobionihapon suhteellinen selektiivisyys oli 100% ja suhteellinen saanto 100 %. Kinetiikan matemaattinen mallinnus tehtiin Modest-ohjelmalla käyttäen kokeista saatuja mittaustuloksia.Ohjelman avulla estimoitiin parametreja ja saatiin matemaattinen malli reaktorille. Tässä työssä tehtiin kineettinen mallinnus myös ravistelureaktorissa tehdyille laktoosin hapetuskokeille, missä pH pysyi koko ajan haluttuna 'in-situ' titrauksen avulla. Työn yhteydessä selvitettiin myös mahdollisuutta käyttää monoliittikatalyyttejä laktoosin hapetusreaktiossa.

Kirjallisuusosassa perehdyttiin retentioaineisiin ja täyteaineisiin sekä retentioaineiden ja rainanmuodostusolosuhteiden vaikutukseen retentioon, vedenpoistoon ja paperin ominaisuuksiin. Tarkemmin kirjallisuusosassa keskityttiin täyteaineiden esiflokkaukseen, retentiopolymeerin adsorptioon sekä retentiopolymeerien ja täyteaineiden annostelutapoihin. Kokeellisessa osassa tutkittiin sarjaa retentiopolymeerejä, joiden varaustiheys ja moolimassa muuttuivat. Yksi polymeereistä oli kahdesta polymeeristävalmistettu suoladispersio ja yksi modifioitu kationinen PAM. Näillä polymeereillä käytiin läpi koesarjoja, joissa muutettiin täyteaineen annosteluaikaa retentiopolymeerin annosteluajan pysyessä vakiona. Lähinnä vertailtiin keskenään perinteistä annostelua, jossa täyteaine annosteltiin paljon ennen retentiopolymeeriä,ja yhtäaikaista annostelua, jossa molemmat annosteltiin yhtä aikaa lähellä perälaatikkoa. Kokeet tehtiin MBF-laitteella, jolla pystytään paperikonetta vastaaviin pulsaatiotaajuuksiin ja sillä voidaan valmistaa tasoviirakoneella valmistetunpaperin kaltaisia laboratorioarkkeja. Valmistetuista arkeista tutkittiin retentioita ja paperiteknisiä ominaisuuksia. Laboratoriokokeiden perusteella yhtäaikainen annostelu antoi paremmat täyteaineretentiot verrattaessa perinteiseen annosteluun lähes kaikissa koesarjoissa. Varsinkin lyhytketjuiset polymeerit näyttivättoimivan hyvin yhtäaikaisannostelulla, mikä saattaisi johtua siitä, että lyhyt reagointiaika sulpun kanssa on lyhytketjuisille polymeereille edullinen, sillä silloin polymeeriketjun konformaatio ei ehdi asettua liian alhaiseksi ja ketjun toimintakyky säilyy parempana. Polymeerin varaustiheyden kasvaessa riittävästi laski täyteaineretentio seuraavissa tapauksissa: SC-massa + kaoliini ja SC-massa +GCC kummallakin annostelulla sekä SC-massa + PCC A perinteisellä annostelulla. Hienopaperimassalla samaa trendiä noudatti täyteaine GCC kummallakin annostelulla, kun taas PCC H:ta käytettäessä paranivat täyteaineretentiot molemmilla annosteluilla. Retentiopolymeerin moolimassan kasvaessa riittävästi kääntyi täyteaineretentio laskuun täyteaineilla GCC ja kaoliini, kun käytettiin SC-massaa. Hienopaperimassalla GCC noudatti tätä samaa taipumusta. Sen sijaan SC-massalla PCC A:takäytettäessä täyteaineretentio puolestaan nousi hieman moolimassan kasvaessa. Näin kävi myös hienopaperimassalla, kun täyteaineena käytettiin PCC H:ta. Käytettäessä SC-massaa, perinteisellä annostelulla saatiin parempi tai yhtä hyvä valonsironta kuin yhtäaikaisella annostelulla kaikilla täyteaineilla. Tämä saattaisi johtua siitä, että yhtäaikaisannostelulla on muodostunut suurempia täyteaineflokkeja, mikä on alentanut valoa sirottavia pintoja. Täyteaineista korkeimmat valonsirontakertoimet antoi PCC A ja alhaisimmat kaoliini. PCC A:lla oli kapein partikkelikokojakauma, mikä korottaa paperin valonsirontaa. Hienopaperimassalla valonsirontakerroin ja opasiteetti suurenivat GCC-pitoisuuden kasvaessa kummallakin annostelulla, mikä voisi johtua täyteainepartikkelien antamasta paremmasta sironnasta. Yhtäaikaisella annostelulla saavutettiin huomattavasti paremmat valonsironnan arvot perinteiseen annosteluun verrattuna. PCC H-pitoisuuden kasvaessa suurenivat myös valonsirontakerroin ja opasiteetti kummallakin annostelulla. PCC H antoi korkeammat valonsirontakertoimet kuin GCC. PCC omaa suuremman valonheijastusluvun kuin GCC, minkä vuoksi se antaa paremmat valonsirontakertoimen arvot. PCC H:n partikkelikokojakauma oli myös kapeampi kuin GCC:n, mikä mahdollisti paremman valonsironnan ja opasiteetin saavuttamisen.
