The ligand-binding region of the low-density lipoprotein (LDL) receptor is formed by seven N-terminal, imperfect, cysteine-rich (LB) modules. This segment is followed by an epidermal growth factor precursor homology domain with two N-terminal, tandem, EGF-like modules that are thought to participate in LDL binding and recycling of the endocytosed receptor to the cell surface. EGF-A and the concatemer, EGF-AB, of these modules were expressed in Escherichia coli. Correct protein folding of EGF-A and the concatemer EGF-AB was achieved in the presence or absence of calcium ions, in contrast to the LB modules, which require them for correct folding. Homonuclear and heteronuclear H-1-N-15 NMR spectroscopy at 17.6 T was used to determine the three-dimensional structure of the concatemer. Both modules are formed by two pairs of short, anti-parallel beta -strands. In the concatemer, these modules have a fixed relative orientation, stabilized by calcium ion-binding and hydrophobic interactions at the interface. N-15 longitudinal and transverse relaxation rates, and {H-1}-N-15 heteronuclear NOEs were used to derive a model-free description of the backbone dynamics of the molecule. The concatemer appears relatively rigid, particularly near the calcium ion-binding site at the module interface, with an average generalized order parameter of 0.85 +/- 0.11. Some mutations causing familial hypercholesterolemia may now be rationalized. Mutations of D41, D43 and E44 in the EGF-B calcium ion-binding region may affect the stability of the linker and thus the orientation of the tandem modules. The diminutive core also provides little structural stabilization, necessitating the presence of disulfide bonds. The structure and dynamics of EGF-AB contrast with the N-terminal LB modules, which require calcium ions both for folding to form the correct disulfide connectivities and for maintenance of the folded structure, and are connected by highly mobile linking peptides. During the postnatal development of cat visual cortex and corpus callosum the molecular composition of tau proteins varied with age. In both structures, they changed between postnatal days 19 and 39 from a set of two juvenile forms to a set of at least two adult variants with higher molecular weights. During the first postnatal week, tau proteins were detectable with TAU-1 antibody in axons of corpus callosum and visual cortex, and in some perikarya and dendrites in the visual cortex. At later ages, tau proteins were located exclusively within axons in all cortical layers and in the corpus callosum. Dephosphorylation of postnatal day 11 cortical tissue by alkaline phosphatase strongly increased tau protein immunoreactivity on Western blots and in numerous perikarya and dendrites in all cortical layers, in sections, suggesting that some tau forms had been unmasked. During postnatal development the intensity of this phosphate-dependent somatodendritic staining decreased, but remained in a few neurons in cortical layers II and III. On blots, the immunoreactivity of adult tau to TAU-1 was only marginally increased by dephosphorylation. Other tau antibodies (TAU-2, B19 and BR133) recognized two juvenile and two adult cat tau proteins on blots, and localized tau in axons or perikarya and dendrites in tissue untreated with alkaline phosphatase. Tau proteins in mature tissue were soluble and not associated with detergent-resistant structures. Furthermore, dephosphorylation by alkaline phosphatase resulted in the appearance of more tau proteins in soluble fractions. Therefore tau proteins seem to alter their degree of phosphorylation during development. This could affect microtubule stability as well as influence axonal and dendritic differentiation.

Inhibitory control refers to the ability to suppress planned or ongoing cognitive or motor processes. Electrophysiological indices of inhibitory control failure have been found to manifest even before the presentation of the stimuli triggering the inhibition, suggesting that pre-stimulus brain-states modulate inhibition performance. However, previous electrophysiological investigations on the state-dependency of inhibitory control were based on averaged event-related potentials (ERPs), a method eliminating the variability in the ongoing brain activity not time-locked to the event of interest. These studies thus left unresolved whether spontaneous variations in the brain-state immediately preceding unpredictable inhibition-triggering stimuli also influence inhibitory control performance. To address this question, we applied single-trial EEG topographic analyses on the time interval immediately preceding NoGo stimuli in conditions where the responses to NoGo trials were correctly inhibited [correct rejection (CR)] vs. committed [false alarms (FAs)] during an auditory spatial Go/NoGo task. We found a specific configuration of the EEG voltage field manifesting more frequently before correctly inhibited responses to NoGo stimuli than before FAs. There was no evidence for an EEG topography occurring more frequently before FAs than before CR. The visualization of distributed electrical source estimations of the EEG topography preceding successful response inhibition suggested that it resulted from the activity of a right fronto-parietal brain network. Our results suggest that the fluctuations in the ongoing brain activity immediately preceding stimulus presentation contribute to the behavioral outcomes during an inhibitory control task. Our results further suggest that the state-dependency of sensory-cognitive processing might not only concern perceptual processes, but also high-order, top-down inhibitory control mechanisms.

Laser diffraction (LD) provides detailed analysis of particle size distribution. Its application to testing the stability of soil aggregates can assist studies on the aggregation of soils with contrasting electrochemical properties. The objectives of the present work were: (a) to propose a protocol for using LD to study soil aggregation, (b) to study the aggregation of an Acrisol under the influence of different doses and forms of lime. Samples were collected in 2005 from a Brazilian Acrisol that in 1994 had received 0.0; 2.0; 8.5 and 17.0 Mg ha-1 of lime, left on the soil surface or incorporated. Aggregates from 4.76 to 8.00 mm diameters were studied using the traditional method proposed by Kemper & Chepil (1965), with wet sieving, while aggregates from 1.00 to 2.00 mm were studied using a CILAS® laser diffractometer that distinguishes particles ranging from 0.04 to 2,500.00 μm. LD readings were made after six consecutive pre-treatments, using agitation times, a chemical dispersion agent and ultrasound. Mean Weighted Diameter (MWD) and the Aggregate Stability Index (ASI) calculated, using the traditional method does not discriminate the treatments. However, LD is able to produce detailed data on soil aggregation, resulting in indexes of stability of aggregates that are linearly related to the doses of lime applied (MWD: R² = 0.986 and ASI: R² = 0.876). It may be concluded that electrochemical changes in the Brazilian Acrisol resulting from incorporated lime affect the stability of aggregates, increasing stability with increased doses of lime.

This essay focuses on how Spielberg's film engages with and contributes to the myth of Lincoln as a super-natural figure, a saint more than a hero or great statesman, while anchoring his moral authority in the sentimental rhetoric of the domestic sphere. It is this use of the melodramatic mode, linking the familial space with the national through the trope of the victim-hero, which is the essay's main concern. With Tony Kushner, author of Angels in America, as scriptwriter, it is perhaps not surprising that melodrama is the operative mode in the film. One of the issues that emerge from this analysis is how the film updates melodrama for a contemporary audience in order to minimize what could be perceived as manipulative sentimental devices, observing for most of the film an aesthetic of relative sobriety and realism. In the last hour, and especially the final minutes of the film, melodramatic conventions are deployed in full force and infused with hagiographic iconography to produce a series of emotionally charged moments that create a perfect union of American Civil Religion and classical melodrama. The cornerstone of both cultural paradigms, as deployed in this film, is death: Lincoln's at the hands of an assassin, and the Civil War soldiers', poignantly depicted at key moments of the film. Finally, the essay shows how film melodrama as a genre weaves together the private and the public, the domestic with the national, the familial with the military, and links pathos to politics in a carefully choreographed narrative of sentimentalized mythopoesis.

Schizotypy, defined in terms of commonly occurring personality traits related to the schizophrenia spectrum, has been an important construct for understanding the neurodevelopment and stress-diathesis of schizophrenia. However, as schizotypy nears its sixth decade of application, it is important to acknowledge its impressively rich literature accumulating outside of schizophrenia research. In this article, we make the case that schizotypy has considerable potential as a conceptual framework for understanding individual differences in affective and social functions beyond those directly involved in schizophrenia spectrum pathology. This case is predicated on (a) a burgeoning literature noting anomalies in a wide range of social functioning, affiliative, positive and negative emotional, expressive, and social cognitive systems, (b) practical and methodological features associated with schizotypy research that help facilitate empirical investigation, and (c) close ties to theoretical constructs of central importance to affective and social science (eg, stress diathesis, neural compensation). We highlight recent schizotypy research, ie providing insight into the nature of affective and social systems more generally. This includes current efforts to clarify the neurodevelopmental, neurobiological, and psychological underpinnings of affiliative drives, hedonic capacity, social cognition, and stress responsivity systems. Additionally, we discuss neural compensatory and resilience factors that may mitigate the expression of stress-diathesis and functional outcome, and highlight schizotypy's potential role for understanding cultural determinants of social and affective functions.

Physical and psychological stress cause different patterns of changes in the fluorescence intensity of nigral and tuberoinfundibular DA neurons which point to changes in neuronal activity. In order to investigate possible interactions between alpha-MSH (alpha-melanotropin) and DA systems in stress, systemic and intraventricular injections of antiserum against alpha-MSH were made. The functional state of DA neurons was assessed by histochemical microfluorimetry and hormone levels were measured by radioimmunossay. Antiserum against alpha-MSH was found to affect the functional state of DA neurons, but only thorugh the intravenous route. Under physical stress i.v. injection of antiserum against alpha-MSH was accompanied by elevated levels of activity of the DA neurons of the substantia nigra. An intraventricular injection of the same antiserum was ineffective. In psychological stress, an effect was again seen only after intravenous injection of antiserum against alpha-MSH. In this situation, the activity in DA cell groups of the substantia nigra, ventral tegmental area and tubero-infundibular system was increased after antiserum injection. Possible influences from manipulations were checked; certain effects which depended upon experimental situation were noted. Our data suggest a modulatory influence of circulating alpha-MSH on the functional state of central DA systems.

Dyes and pigments are additives used in polymers to improve mainly the aesthetic properties of the material. However, the incorporation of these additives can directly affect polymer stability. The colorants can drastically decrease the lifetime and the performance of the material or can act as a stabilizer, improving significantly the stability of the polymer against degradation. Interaction between colorants and polymers is the cause of the stability changes. Some mechanisms are proposed to explain the action of colorants on polymers. However it is difficult to foresee this action without experiments. This work reviews the main mechanisms involved in the degradation and stabilization of polymers containing colorants.

Polarization curves experimentally obtained in the electro-dissolution of iron in a 1 M H2SO4 solution using a rotating disc as the working electrode present a current instability region within the range of applied voltage in which the current is controlled by mass transport in the electrolyte. According to the literature (Barcia et. al., 1992) the electro-dissolution process leads to the existence of a viscosity gradient in the interface metal-solution, which leads to a velocity field quantitatively different form the one developed in uniform viscosity conditions and may affect the stability of the hydrodynamic field. The purpose of this work is to investigate whether a steady viscosity profile, depending on the distance to the electrode surface, affects the stability properties of the classic velocity field near a rotating disc. Two classes of perturbations are considered: perturbations monotonically varying along the radial direction, and perturbations periodically modulated along the radial direction. The results show that the hydrodynamic field is always stable with respect to the first class of perturbations and that the neutral stability curves are modified by the presence of a viscosity gradient in the second case, in the sense of reducing the critical Reynolds number beyond which perturbations are amplified. This result supports the hypothesis that the current oscillations observed in the polarization curve may originate from a hydrodynamic instability.

Malgré l’engouement pour les neurosciences cognitives des émotions et les nombreuses publications des dernières décennies tentant d’élucider les bases neurobiologiques des émotions, nos connaissances sur le domaine restent embryonnaires. Plusieurs questions importantes restent toujours sans réponses incluant s’il existe ou non un système unique pour le traitement de stimuli émotionnels et s’il y a ou non des différences entre les hommes et les femmes pour le traitement de stimuli émotionnels. L’objectif de cette thèse est d’apporter certains éléments de réponses à ces questions à travers une caractérisation du substrat neurobiologique impliqué dans le traitement de stimuli émotionnels visuels et dynamiques. Ce travail a été mené via l’imagerie par résonance magnétique fonctionnelle (IRMf) cérébrale. Le premier chapitre, subdivisé en quatre sections, permet de présenter la perspective dans laquelle s’inscrit la thèse. La première section de ce chapitre sert à établir certaines balises définitionnelles liées aux émotions. La seconde section, basée sur une lecture des textes originaux, retrace les faits historiques saillants de la neurobiologie des émotions allant de Charles Darwin à Joseph Ledoux. La troisième section débute où la seconde s’arrête et continue l’histoire de la neurobiologie des émotions à travers un résumé de toutes les principales méta-analyses d’imagerie fonctionnelle cérébrale des émotions. La dernière section du chapitre permet de présenter la problématique de recherche. La recherche, à proprement parler, qui constitue le corps de la thèse est ensuite présentée sous forme de trois articles. Enfin, les résultats de cette recherche et la manière dont ils s’inscrivent dans la continuité de nos connaissances actuelles font l’objet d’une discussion générale. Le premier article (chapitre II) rapporte, chez les hommes et les femmes, les régions du cerveau qui sont plus activées lors du traitement de films érotiques que lors du traitement de films dits ‘neutres’. Un chevauchement manifeste est observé entre les hommes et les femmes. Par contre, une activation significativement plus grande est observée chez les hommes pour l’hypothalamus, une région importante pour le comportement sexuel à travers la phylogénie. De plus, chez les hommes seulement, l’activation hypothalamique est corrélée à l’excitation sexuelle subjective. Comme la recherche présentée dans le premier article se sert de conditions expérimentales relativement longues pour l’IRMf (i.e. extraits de films de 3 minutes) et que ceci peut induire une nette diminution de signal en lien avec certaines contraintes de l’IRMf, le second article (chapitre III) examine les corrélats du traitement de stimuli sexuels en utilisant, cette fois, un paradigme d’IRMf classique où plusieurs extraits de films de 33 secondes sont présentés à la place. Cette étude démontre que, pour le traitement de stimuli sexuels, ce paradigme classique d’IRMf est beaucoup plus sensible que celui du premier article. De plus, comme ce paradigme mène à une reproduction des résultats du premier papier, ce travail soutient la perspective selon laquelle les paradigmes à époques courtes sont une alternative valide aux longues époques comme méthode d’étude du traitement de stimuli émotionnels. Le troisième article (chapitre IV) capitalise sur le protocole du second article et démontre que les patrons d’activation associés au visionnement de courts extraits de films induisant du dégoût, de l’amusement, ou de l’excitation sexuelle, sont très étendus. Une analyse de conjonction formelle démontre un large chevauchent de ces patrons à travers les différents affects étudiés. Enfin, le cinquième chapitre sert de discussion générale. Les questions des différences entre les hommes et les femmes dans le traitement des émotions, de l’existence ou non d’un système général pour le traitement des émotions, ainsi que de la manière dont un tel système pourrait être conçu, sont des points saillants de la discussion. Ces points sont abordés à la lumières des connaissances actuelles et des résultats des trois articles.

La SLA est une maladie neurodégénérative fatale se déclenchant tardivement. Elle est caractérisée par la perte des neurones moteurs supérieurs et inférieurs. Jusqu’à présent, aucun traitement ne permet de ralentir ou de guérir la maladie de façon robuste. De récentes découvertes portant sur TDP-43 et hnRNP A1 y ont identifié des mutations reliées à des cas de SLA. Comme les deux possèdent de multiples fonctions dans le métabolisme de l’ARN, l’impact de ces mutations devient difficile à définir. Notre hypothèse est que TDP-43 régule hnRNP A1 et que les mutations causant la SLA dérégulent ce mécanisme, aboutissant ainsi à un impact majeur sur la vulnérabilité des neurones moteurs. Nos résultats démontrent que TDP-43 lie l’ARNm de hnRNP A1, mais n’affecte pas sa stabilité. En revanche, TDP-43 réprime l’expression de hnRNP A1. Ce mécanisme pourrait être appliqué in vivo où le ratio protéique hnRNP A1B/A1 augmente chez les souris âgées et davantage chez les TDP-43A315T dans la région cervicale et lombaire de la moelle épinière. Cette différence n’est pas causée par un défaut de l’épissage alternatif. Aussi, la mutation TDP-43A315T serait davantage responsable de cette différence que la surexpression de TDP-43 (résultats obtenus en culture). L’impact d’une telle augmentation sur la cellule pourrait être la formation d’agrégats puisque la forme hnRNP A1B possède quatre domaines de fibrillation de plus que hnRNP A1. Nos résultats pourraient donc fournir un mécanisme potentiel de la formation d’inclusions cytoplasmiques reconnues comme étant une des caractéristiques pathologiques principales de la SLA.

The cure characteristics of short fiber-polyurethane elastomer were studied with respect to different fiber-matrix bonding agents. A hexamethylenetetramine- resorcinol -hydrated silica based bonding agent was found to affect the stability of the composite. A new bonding agent, TP resin, based on polymeric toluenediisocyanate and polypropylene glycol has been developed. Cure characteristics of the composite with and without TP resin at different fiber loadings were also compared. Minimum torque, scorch time and optimum cure time increased with fiber content. Maximum torque was consistently higher with TP resin at all fiber loadings.

Este artigo propõe um novo teste para distinção entre modelos macroeconômicos de precificação. Onde testes antigos concluíram haver uma relação negativa entre inflação corrente e duração de um preço, indicando estado-dependência, nosso teste indica que a relação verdadeira é entre inflação esperada e duração do preço, indicando tempo-dependência-endógena. Argumentamos que os resultados previamente encontrados possivelmente sofreram de viés de variável omitida.
