In the attempt to find out catalytic potency and properties of the endoglucanase of green mussel, it could be highlighted that the enzyme is efficient in degrading carboxymethylcellulose to reducing sugars. The immobilized enzyme will find applications in the food industry, paper and pulp industry, wood preservation, alcohol and pharmaceutical industry. The purification method employed i.e. Sephadex G100 chromatography employing affinity and exclusion principles simplify the purification procedure.Addition of Mg2+ and Co2+ at 10mM concentrations enhances endoglucanase activity of green mussel. The immobilized endoglucanase can be used for deinking mixed office waste paper. The endoglucanase if supplemented with exoglucanase and B-glucosidase under appropriate conditions would help in the recycling of paper.

The present scientific investigation of the effects of copper, mercury and cadmium has focussed on their effects on two commercially important marine bivalve species, Perna indica (brown mussel) and Donax incarnatus (wedge clam), conspicuous representatives of the tropical intertidal areas. The investigation centred around delineating the cause and effects of heavy metal stress, individually and in combination on these species under laboratory conditions. A clear understanding of the cause and effect can be had only if laboratory experiments are conducted employing sub-lethal concentrations of the above toxicants. Therefore, during the course of the investigation, sub-lethal concentrations of copper, mercury and cadmium were employed to assess the concentration dependent effects on survival, ventilation rate, O:N ratio and tissues. The results obtained are compared with the already available information and partitioned in sections to make a meaningful presentation. The thesis is presented in five chapters comprising INTRODUCTION, ACUTE TOXICITY, VENTILATION RATE, OXYGEN : NITROGEN RATIO and HISTOPATHOLOGY. Each chapter has been divided into various sections such as INTRODUCTION, REVIEW OF LITERATURE, MATERIAL AND METHODS, RESULTS and DISCUSSION

In this thesis certain important aspects of heavy metal toxicity have been worked out. Recent studies have clearly shown that when experimental media contained more than one heavy metals, such metals could conspicuously influence the toxic reaction of the animals both in terms of quantity and nature. The experimental results available on individual metal toxicity show that, in majority of such results, unrealistically high concentrations of dissolved metals are involved. A remarkable number of factors have been shown to influence metal toxicity such as various environmental factors particularly temperature and salinity, the condition of the organism and the ability of some of the marine organisms to adapt to metallic contamination. Further, some of the more sensitive functions like embryonic and larval development, growth and fecundity, oxygen utilization and the function of various enzymes are found to be demonstrably sensitive in the presence of heavy metals. However, some of the above functions could be compensated for by adaptive process. If it is assumed that the presence of a single metal in higher concentrations could affect the life function of marine animals, more than one metal in the experimental media should manifest such effects in a greater scale. Commonly known as synergism or more than additivity, majority of heavy metals bring about synergistic reaction .The work presented in this thesis comprises lethal and sublethal toxicities of different salt forms of copper and silver on the brown mussel Perna indica. during the present investigation sublethal concentrations of copper and silver in their dent effects on survival, oxygen consumption, filtration, accumulation and depuration on Perna indica. The results are presented under different sections to make the presentation meaningful .

This thesis deals with the reproductive physiology of the two species of Indian sea mussels, namely the brown mussel perna indica and the green mussel p viridis .The major aspects of the study include ecophysiology of reproduction linking up the animal reproductive cycle of the animals with the ecological conditions of the natural mussel beds, biochemical and histochemical changes associated with reproduction and neurosecretory cycle in synchrony with the reproductive cycle. Some basic studies on gamete morphology and certain aspects of gamete physiology have been taken up. The experimental work deals with the influence of different feeding levels on gamatogenesis and maturation and the effect of ganglia ablation on spawning. The material for the investigation on perna indica were collected from the natural mussel beds at Vizhinjam near Trivandrum and on p.viridic from Elathur, near Calicut. The period of observation extended from October 1981 to December 1982.

No serious attempt has so far been made in India to make use of the ‘Mussel watch’ concept as a useful tool for pollution monitoring of the marine and estuarine environment. The recently conducted 'National seminar on mussel watch’ by the Cochin University of Science and Technology (13-14 Feb, 1986) discussed the technical aspects related to mussel watch programme and the application of sentinel organism concept to the coastal areas of India. It is well known that the biological and physiological characteristics of the organism inhabiting tropical waters such as those prevailing in India, and the ecological as well as the environmental characteristics of temperate areas, where mussel watch programmes are already in existence differ greatly. So it is essential to adopt the techniques and standards developed for temperate species to the situations and conditions in India. In this context it is a prerequisite to collect information on physiology and other biological indices of stress of possible sentinel organisms like P.viridis. In consideration of the above, P. viridis which is a potential sentinel organism, is selected for the present study

In the present study an endeavour has been made to analyse the acute toxicity of WAFs of Bombay High crude and Light Diesel oil on commercially important bivalve species Perna viridis and Perna indica by static bioassay methods. The toxic effects of chemicals in the WAF on the organisms ; their tissues and eventually on rate functions have been elucidated. Marine oil pollution not only affects productivity and quality of marine organisms but also eventually affects the health of human population due to a possible health risk by way of consumption of oil contaminated seafood

The effects of the Linear Alkylbenzene Sulphonate (LAS) were evaluated on the mussel Perna perna (Linnaeus, 1758), using a cellular level biomarker. The Neutral Red Retention Time (NRRT) assay was used to estimate effects at cellular levels. Significant effects were observed for the NRRT assay, even in low concentrations. The effects at cellular level were progressive, suggesting that the organisms are not capable to recover of such increasing effects. Additionally, the results show that the levels of LAS observed for Brazilian coastal waters may chronically affect the biota.

The effects of the Linear Alkylbenzene Sulphonate (LAS) were evaluated on the mussel Perna perna, using physiological and genotoxic biomarkers. The Micronuclei (MN) assay was used to estimate effects at nuclear level, whereas the physiological effects were evaluated by measuring the oxygen consumption and ammonia excretion rates. Significant effects were observed for the MN assay and the ammonia excretion rate, even in low concentrations. The oxygen consumption was not affected in the tested concentrations. For MN and ammonia excretion, the animals exposed to intermediate concentrations were not affected, but responded to the higher concentrations, indicating the existence of compensatory mechanisms at physiological level. However, parallel to this study other authors indicate the presence of progressive effects at the cellular level, suggesting that the organisms are not capable to recover of such increasing effects. Additionally, the results show that the levels of LAS observed for Brazilian coastal waters may chronically affect the biota.

The objective of the present work was evaluate two techniques of smoking, the hot traditional and with use of smoke natural aroma or "liquid smoke", having as control Perna perna mussels vapor cooked. Sensorial characteristics were appraised (appearance, color, aroma, texture, flavor and salt tenor) and yield processing. The experimental design was completely randomized with three treatments (T1 = smoked with liquid smoke; T2 = traditional smoked and T3 = just cooked, control), with nine repetitions. For sensorial analysis 30 people were used, where each one was considered a block. There was significant difference for processing performance among treatments (P<0.05), however there was not significant difference independent of smoked technique applied. Nevertheless, smoking technique using smoke natural aroma was more attractive, presenting better results with relationship to sensorial characteristics and final product packaged for commercialization.
