Leishmania amazonensis causes a wide spectrum of leishmaniasis. There are no vaccines or adequate treatment for leishmaniasis, therefore there is considerable interest in the identification of new targets for anti-leishmania drugs. The central role of telomere-binding proteins in cell maintenance makes these proteins potential targets for new drugs. In this work, we used a combination of purification chromatographies to screen L. amazonensis proteins for molecules capable of binding double-stranded telomeric DNA. This approach resulted in the purification of a 38 kDa polypeptide that was identified by mass spectrometry as Rbp38, a trypanosomatid protein previously shown to stabilize mitochondrial RNA and to associate with nuclear and kinetoplast DNAs. Western blotting and supershift assays confirmed the identity of the protein as LaRbp38. Competition and chromatin immunoprecipitation assays confirmed that LaRbp38 interacted with kinetoplast and nuclear DNAs in vivo and suggested that LaRbp38 may have dual cellular localization and more than one function. The phylogeny of Celastraceae subfamily Salacioideae (ca. 255 species in the Old and New World tropics) and tribe Lophopetaleae (ca. 29 species in southern Asia and the Austral-Pacific) was inferred using morphological characters together with plastid (matK, trnL-F) and nuclear (ITS and 26S rDNA) genes. Brassiantha, a monotypic genus endemic to New Guinea, is inferred to be more closely related to the clade of Dicarpellum (New Caledonia) and Hypsophila (Queensland, Australia) than it is to Hippocrateoideae or Salacioideae. This unambiguously supported resolution indicates that a nectary disk positioned outside the stamens has been convergently derived in these two lineages. The clade of Kokoona and Lophopetalum is resolved as more closely related to Breria and Elaeodendron than it is to Hippocrateoideae or Salacioideae. Sarawakodendron, a monotypic genus endemic to Borneo, is resolved as sister to Salacioideae. Salacioideae are inferred to have an Old World origin that was followed by a single successful radiation within Central and South America. We infer that capsular fruits are primitive within the clade of Hippocrateoideae + Sarawakodendron + Salacioideae, with berries a synapomorphy for Salacioideae. Based on the resolution of Sarawakodendron as sister to Salacioideae, we hypothesize that the filaments of Sarawakodendron arils are homologous to the spiral filaments in the mucilagenous pulp of Salacioideae.

The 3-isobutyl-1-methylxanthine (IBMX) is able to prevent resumption of meiosis by maintaining elevated cyclic AMP (cAMP) concentrations in the oocyte, and roscovitine, a purine known to specifically inhibit MPF kinase activity, maintains bovine oocytes at the germinal vesicle (GV) stage. The present study was conducted to analyze whether cytoplasmic maturation (examined by the pattern of cortical granule (CG) distribution) of bovine oocytes is improved during meiotic arrest with IBMX and roscovitine. Oocytes were matured in vitro in a 10% Knockout(SR) supplemented TCM-199 medium (Control) with either 0.5 mM IBMX or 25 mu M roscovitine (ROSC). Oocytes were stained with fluorescein isothiocyanate conjugated Lens culinaris agglutinin (FITC-LCA) for CG evaluation and with Hoechst 33342 for nuclear stage assessment. At 16 h of culture, the percentage of oocytes remaining in the GV stage was higher (P < 0.05) in the ROSC group (32.41%) compared with the Control and IBMX groups (8.61% and 9.73%, respectively). At 24h of culture, progression of meiosis to M II stage was retarded (P < 0.05) in the ROSC group (24.05%) compared to the Control (60.20%), whereas the IBMX group (33.88%) showed no significant difference to the other two groups. At 16h of maturation, the proportion of oocytes with CG in clusters (immature cytoplasm) was similar between the groups, as was the percentage of peripheral CG (mature) at 24h of maturation. The results of the present study demonstrated that the meiotic inhibitors IBMX and roscovitine delay the progression of nuclear maturation without affecting cytoplasmic maturation, assessed by the analysis of CG repositioning. [ES]Durante la campaña La Bocaina 0497 se llevaron a cabo una serie de 14 lances con una red de arrastre pelágico de tipo comercial entre 20 y 700 m. de profundidad y una porspección acústica con una ecosonda SIMRAD EK-500 en aguas neríticas y oceánicas adyacentes a Lanzarote, Fuerteventura y Gran Canaria. Además se obtuvieron imágenes SST para obtener información de las condiciones hidrológicas en el área de estudio. Los resultados mostraron que la caballa Scomber japonicus presentaba gran variabilidad espacial en biomasa entre las diferentes localidades muestreadas.

This event study investigates the impact of the Japanese nuclear disaster in Fukushima-Daiichi on the daily stock prices of French, German, Japanese, and U.S. nuclear utility and alternative energy firms. Hypotheses regarding the (cumulative) abnormal returns based on a three-factor model are analyzed through joint tests by multivariate regression models and bootstrapping. Our results show significant abnormal returns for Japanese nuclear utility firms during the one-week event window and the subsequent four-week post-event window. Furthermore, while French and German nuclear utility and alternative energy stocks exhibit significant abnormal returns during the event window, we cannot confirm abnormal returns for U.S. stocks.
