In functional dairy products, polyunsaturated fatty acids such as, conjugated linoleic acid (CLA) and alpha-linolenic acid (ALA) have been highlighted for their benefits related to prevention of some chronic diseases. In order to study the effect of type of milk (conventional vs. organic, characterized by a specific fatty acid composition), Bifidobacterium animalis subsp. lactis (BB12, B94, BL04 and HN019) counts, acidification activity and chemical composition (pH, lactose, lactic acid contents and fatty acids profile) were investigated before fermentation and after 24 h of products stored at 4 degrees C. Organic and conventional milk influenced acidification performance and bacteria counts, which was strain-dependent. Higher counts of BB12 were observed in organic milk, whereas superior counts of BL04 were found in conventional milk. Organic fermented milk showed lower levels in saturated fatty acids (FA) and higher in monounsaturated FA contents. Similarly, among bioactive FA, organic fermented milks have higher amounts of trans vaccenic acid (TVA-C18:1t), conjugated linoleic acid (CLA) and slightly higher contents of alpha-linoleic acid (ALA). In der vorliegenden Arbeit wurden Essigsäure-, Propionsäure und Buttersäure-bildende Bakterien aus einer thermophilen und drei mesophilen Biogasanlagen sowie aus zwei Hochdruck-Biogas-Laborfermentern isoliert. Die Fermenter waren mit dem nachwachsenden Rohstoff Maissilage, teilweise mit Rinder- oder Schweinegülle und weiteren festen Inputstoffen gefüttert. Für die Isolierung von Säure-bildenden Bakterien wurde ein Mineralsalzmedium verwendet, welchem als Kohlenstoffquelle Na-DL-Laktat, Succinat, Ethanol, Glycerin, Glucose oder eine Aminosäuremischung (Alanin, Serin, Threonin, Glutaminsäure, Methionin und Cystein) hinzugefügt wurde. Hierbei handelt es sich um Substrate, welche beim anaeroben Abbau während der Hydrolyse oder der primären Gärung entstehen können. Die erhaltenen Isolate waren in der Lage, aus diesen Substraten Essigsäure, Propionsäure oder Buttersäure zu bilden. Insgesamt wurden aus den beprobten Anlagen 49 Isolate gewonnen, welche zu den Phyla Firmicutes, Tenericutes oder Thermotogae gehörten. Mit Hilfe von 16S rDNA-Sequenzen konnten die meisten Isolate als Clostridium sporosphaeroides, Defluviitoga tunisiensis und Dendrosporobacter sp. identifiziert werden. Die Bildung von Essigsäure, Propionsäure oder Buttersäure wurde in Kulturen von Isolaten festgestellt, welche als folgende Arten identifiziert wurden: Bacillus thermoamylovorans, Clostridium aminovalericum, Clostridium cochlearium/Clostridium tetani, Clostridium sporosphaeroides, Dendrosporobacter sp., Proteiniborus sp., Selenomonas bovis und Tepidanaerobacter sp. Zwei Isolate, verwandt mit Thermoanaerobacterium thermosaccharolyticum, konnten Buttersäure und Milchsäure bilden. In Kulturen von Defluviitoga tunisiensis wurde Essigsäurebildung festgestellt. Ein Vergleich der 16S rDNA-Sequenzen mit Datenbanken und die Ergebnisse der PCR-Amplifikationen mit Isolat-spezifischen Primerpaaren ergaben zusätzlich Hinweise, dass es sich bei einigen Isolaten um neue Arten handeln könnte (z. B. Stamm Tepidanaerobacter sp. AS34, Stamm Proteiniborus sp. ASG1.4, Stamm Dendrosporobacter sp. LG2.4, Stamm Desulfotomaculum sp. EG2.4, Stamm Gallicola sp. SG1.4B und Stamm Acholeplasma sp. ASSH51). Durch die Entwicklung Isolat-spezifischer Primerpaare, abgeleitet von 16S rDNA-Sequenzen der Isolate oder Referenzstämmen, konnten die Isolate in Biogasanlagen detektiert und mittels qPCR quantifiziert werden (hauptsächlich im Bereich zwischen 1000 bis 100000000 Kopien der 16S rDNA/g BGA-Probe). Weiterhin konnten die Isolate mit Hilfe physiologischer Versuche charakterisiert und deren Rolle in der anaeroben Abbaukette diskutiert werden. Die Art Defluviitoga tunisiensis scheint eine große Bedeutung in Biogasanlagen zu spielen. Defluviitoga tunisiensis wurde am häufigsten in Untersuchungen im Rahmen der vorliegenden Arbeit isoliert und konnte auch mit Hilfe des entwickelten Primerpaares in hohen Abundanzen in den beprobten Biogasanlagen detektiert werden (10000 - 100000000 Kopien der 16S rDNA/g BGA-Probe). Die manuelle Annotation des Gesamtgenoms sowie die Substratverwertungsversuche haben gezeigt, dass Defluviitoga tunisiensis ein sehr breites Substratspektrum in der Verwertung von Kohlenhydraten besitzt und dadurch möglicherweise eine wichtige Rolle bei der Verwertung von Biomasse in Biogasanlagen einnimmt. Mit Hilfe der Ergebnisse der vorliegenden Arbeit konnten somit neue Einblicke in die zweite Stufe des anaeroben Abbaus, die Acidogenese, in Biogasanlagen gegeben werden. rn

In temperate, subpolar and polar marine systems, the classical perception that bacteria are carbon limited by end of winter and respond in activity and abundance to the production of new carbon during the diatom spring bloom and post bloom. Contrary to this view, we here document an strong increase in bacterial abundance and activity (latter measured by increasing high nuclei acid (HNA) to low nuclei acid (LNA) bacteria ratio) during the winter-spring transition, where phytoplankton smaller than 10 µm dominate. Further DNA-virus were enumerated and revealed the virus to bacteria ratio (VBR) to be decreasing during winter-spring transition, indicating that the virus did not increase in number accordingly to bacteria. During repeated visits to stations in the deep Icelandic and the Norwegian Basins and the shallow Shetland Shelf (26 March to 29 April 2012), we investigated the abundance of bacteria and the succession of HNA:LNA bacteria and VBR. Water samples were collected from CTD rosette .10 L Niskin bottles and fixed in glutaraldehyde (final conc. 5%), flash frozen in liquid Nitrogen and stored at -80°C until analysis.

This study aimed to evaluate the e ect of diets containing increasing levels of citrus pulp on the physic-chemical and microbiological characteristics of horses feces. Five mares, at an average age of 3.5 years old and body weight of 492 ± 44.5 kg were arranged in a 5 x 5 Latin Square. The experimental diet consisted of 60% coast-cross hay and 40 % of concentrate with increasing levels of citrus pulp (0, 7, 14, 21, and 28 %). To determine the fecal pH, samples were collected directly from the oor, immediately after defecation, in the rst feces the day at 07:00 a.m., and color and fecal consistency were evaluated. For microbiological analysis, an aliquot was reserved in plastic bags, frozen, and sent to the microbiological laboratory for further analysis. Lactic acid bacteria were counted for Lactobacillus spp. and Streptococcus spp. from fecal samples under anaerobic conditions. The diet produced di erences (P<0.05) in feces consistency 98% had normal and rm stools, while 2% had loose ruminant-type feces. We observed no di erence (P<0.05) for color, verifying 100% of greenish feces, normal for equines. There was no e ect (P>0.05) on pH and on the number of Lactobacillus spp. and Streptococcus spp. The inclusion of up to 28% citrus pulp concentrates for horses did not promote change in the physio-chemical characteristics and on the population of lactic acid-producing bacteria in feces.

The removal of sulfate and organic matter was assessed in an ASBR, which treated wastewater containing 500 mg COD L(-1) (3 g COD L(-1) d(-1)) in 8 h-cycles at 30 degrees C. The wastewater was enriched with sulfate at [COD/SO(4)(2-]) ratios of 1.34, 0.67 and 0.34 (8.8,4.5 and 2.2 gSO(4)(2-) L(-1) d(-1)). For each COD/[SO(4)(2-)] ratio fill times used were: 10 min (batch), 3 and 6 h (fed-batch), achieving sulfate reduction of 30%, 72% and 72% (COD/[SO(4)(2-)] of 1.34); 25%, 58% and 55% (COD/[SO(4)(2-)] of 0.67) and 23%, 37% and 27% (COD/[SO(4)(2-)] of 0.34), respectively, and organic matter removal of 87%, 68% and 80% (COD/[SO(4)(2-)] of 1.34); 78%, 75% and 69% (COD/[SO(4)(2-)] of 0.67) and 85%, 84% and 83% (COD/[SO(4)(2-)] of 0.34), respectively. The results showed that fed-batch operation improved sulfate reduction, whereas organic matter removals were similar for batch and fed-batch operation. The aims of this study were: (1) to correlate surface (SH) and cross-sectional hardness (CSH) with microradiographic parameters of artificial enamel lesions; (2) to compare lesions prepared by different protocols. Fifty bovine enamel specimens were allocated by stratified randomisation according to their initial SH values to five groups and lesions produced by different methods: MC gel (methylcellulose gel/lactic acid, pH 4.6, 14 days); PA gel (polyacrylic acid/lactic acid/hydroxyapatite, pH 4.8, 16 h); MHDP (undersaturated lactate buffer/methyl diphosphonate, pH 5.0, 6 days); buffer (undersaturated acetate buffer/fluoride, pH 5.0, 16 h), and pH cycling (7 days). SH of the lesions (SH(1)) was measured. The specimens were longitudinally sectioned and transverse microradiography (TMR) and CSH measured at 10- to 220-mu m depth from the surface. Overall, there was a medium correlation but non-linear and variable relationship between mineral content and root CSH. root SH(1) was weakly to moderately correlated with surface layer properties, weakly correlated with lesion depth but uncorrelated with integrated mineral loss. MHDP lesions showed the highest subsurface mineral loss, followed by pH cycling, buffer, PA gel and MC gel lesions. The conclusions were: (1) CSH, as an alternative to TMR, does not estimate mineral content very accurately, but gives information about mechanical properties of lesions; (2) SH should not be used to analyse lesions; (3) artificial caries lesions produced by the protocols differ, especially considering the method of analysis. Os iogurtes simbióticos, que combinam microrganismos probióticos e substâncias prebióticas, adicionados de polpa de frutas são uma tendência crescente no mercado. O fruto açaí (Euterpe edulis) se destaca pela presença de compostos bioativos, como as antocianinas. Neste contexto, o presente trabalho teve como objetivo caracterizar e avaliar os parâmetros físico-químicos e a viabilidade microbiológica de iogurte simbiótico de açaí enriquecido com inulina e adicionado de cultura probiótica de Bifidobacterium animallis subsp. lactis BB-12. As formulações de iogurte atenderam aos requisitos físico-químicos exigidos pela IN no 46/2007 do MAPA apresentando teor de cinzas de 0,86 % ± 0,10, extrato seco total de 23,18 % ± 2,59, teor de gordura de 4,16 % ± 0,31, acidez de 0,70 % ± 0,05 e pH de 4,45 ± 0,10. Entre as formulações o conteúdo fenólico total variou de 18,17 a 117,84 mg de AGE/100 g, teor de antocianinas de 1,92 a 47,88 mg/100 g e atividade antioxidante de 0,71 a 6,95 μmol Trolox/g, observando-se um aumento de acordo com o aumento do teor de polpa de açaí adicionada. Ao final de 28 dias de armazenamento a 5 °C, observou-se uma redução no teor de antocianinas e da atividade antioxidante. Verificou-se a contribuição positiva da polpa de açaí na viabilidade das bactérias láticas totais, cujas contagens variaram de 4,56 a 7,04 log UFC.g-1 e de B. lactis BB-12 que variou de 3,17 a 6,34 log UFC.g-1, favorecendo a multiplicação dessas bactérias nos iogurtes. Nas formulações com 20 e 25 % de polpa de açaí as contagens das bactérias láticas totais e probiótica mantiveram-se viáveis de acordo com a IN no 46/2007 do MAPA e a Lista de Alegação de Propriedade Funcional (Anvisa), durante os 28 dias de armazenamento a 5 oC. Concluiu-se que a adição de polpa de açaí E. edulis, inulina e B. lactis BB-12 foi tecnologicamente viável na elaboração de iogurte simbiótico de açaí, sendo uma excelente alternativa de diversificação do produto no mercado.

The bacterial insertion sequence IS21 shares with many insertion sequences a two-step, reactive junction transposition pathway, for which a model is presented in this review: a reactive junction with abutted inverted repeats is first formed and subsequently integrated into the target DNA. The reactive junction occurs in IS21-IS21 tandems and IS21 minicircles. In addition, IS21 shows a unique specialization of transposition functions. By alternative translation initiation, the transposase gene codes for two products: the transposase, capable of promoting both steps of the reactive junction pathway, and the cointegrase, which only promotes the integration of reactive junctions but with higher efficiency. This review also includes a survey of the IS21 family and speculates on the possibility that other members present a similar transpositional specialization.

The objective of this work was to assess the time-related action of probiotic Lactobacillus plantarum in the bacterial microbiota of the digestive tract of Litopenaeus vannamei, and the relation of total haemocyte count and serum phenol oxidase activity of shrimp challenged with Vibrio harveyi. Shrimps were fed with a probiotic-supplemented diet, for eight days, then shifted to a commercial diet. Shrimps fed only with the commercial diet served as control. Evaluations were made on the 8th day of experiment and repeated two, four, six and eight days later. Total lactic bacteria in the digestive tract was higher until the 4th day of evaluation in the probiotic-supplemented group. Vibrio spp. counts were higher in the control at days zero and two. Until the 4th day of evaluation, the total haemocyte counts in shrimps after challenge with V. harveyi were higher in probiotic-supplemented group than in control group. Significant difference was not observed in phenol oxidase activity. On the 6th day after shifting from supplemented to control diet, all parameters were equal in both groups, suggesting that the time-related action of L. plantarum in shrimp is short.

La concentración de ácido láctico en LCR en pacientes con sospecha de meningitis postquirúrgica luego de clipaje de aneurisma cerebral y hemorragia subaracnoidea espontánea se midió prospectivamente por un período de tres años. Se analizaron un total de 32 muestras de líquido cefalorraquídeo, se midió la concentración de ácido láctico y se comparó con el cultivo de LCR. Los cultivos fueron positivos en cinco pacientes, con una prevalencia de infección del 15%. Se utilizó un valor umbral de ácido láctico de 4 mmol/L. y se encontró una sensibilidad del 80%, especificidad del 52%, VPP del 23%, VPN del 93%, y likelihood ratio (LHR) positivo de 1,66 con una probabilidad post test de 15% de la concentración del ácido láctico en el diagnóstico de meningitis postquirúrgica en pacientes con hemorragia subaracnoidea aneurismática. La concentración de ácido láctico en LCR tiene un desempeño limitado en el diagnóstico de meningitis postquirúrgica en pacientes con hemorragia subaracnoidea aneurismática.

The number and the main groups of microorganisms present in samples of different nonalcoholic carbonated beverages (lemon, orange and guaraná soft drinks) obtained from a small factory were analyzed. The samples were obtained at the end of the processing line. They were then divided into two lots: one was sent to immediate analysis, the other was stored at environmental temperature for 90 d thereafter it was submitted to the same analysis. Aliquots of 1 mL were drawn from the various samples and the corresponding decimal dilutions were prepared. They were then grown in culture media and counts of mesophilic aerobic bacteria, molds and yeasts, acid-producing bacteria, total and fecal coliforms were taken. It was observed that, of all the analyzed samples, at time 0 or after storage sample C (orange) was the best, since it conformed to the microbiological standards established by legislation. The guaraná type could also be consumed on day zero; the lemon type was inadequate for consumption of all the analyzed samples, the orange type was the only one that could be consumed within 3 months of storage.

This research aims to measure the energy spending in parturient women of low gestation risk. Participants were selected randomly and submitted to fasting (n=15; Group I) or honey ingestion (n = 15; Group II). Data were collected by means of capillary blood values and heart frequency monitoring. The paired t-test with a 5% significance level and Tukey's method were used in statistical analysis. The results showed that honey ingestion did not promote an overload in the mother's glucose; the lactate response demonstrated that the substrate offered was well used; the cardiorespiratory rate demonstrated good performance for both groups; the total energy spent during labor demonstrated that carbohydrate ingestion exerts significant influence, improving maternal anaerobic performance; the group which remained in fasting presented, immediately after labor, higher levels of lactate, showing the organism's efforts to compensate for the energy spent.

Several beetle luciferases have been cloned and sequenced. However, most studies on structure and function relationships and bioanalytical applications were done with firefly luciferases, which are pH sensitive. Several years ago we cloned Pyrearinus termitilluminans larval click beetle luciferase, which displays the most blue-shifted bioluminescence among beetle luciferases and is pH insensitive. This enzyme was expressed in E. coli, purified, and its properties investigated. This luciferase shows slower luminescence kinetics, KM values comparable to other beetle luciferases and high catalytic constant. Fluorescence studies with 8-anilino-1-naphtalene-sulfonic acid (1,8-ANS) and modeling studies suggest that the luciferin binding site of this luciferase is very hydrophobic, supporting the solvent and orientation polarizability effects as determining mechanisms for bioluminescence colors. Although pH insensitive in the range between pH 6-8, at pH 10 this luciferase displays a remarkable red-shift and broadening of the bioluminescence spectrum. Modeling studies suggest that the residue C312 may play an important role in bioluminescence color modulation. Compared to other beetle luciferases, Pyrearinus termitilluminans luciferase also displays higher thermostability and sustained luminescence in a bacterial cell environment, which makes this luciferase particularly suitable for in vivo cell analysis and bioimaging. © The Royal Society of Chemistry and Owner Societies 2009.
