En raison de l’utilisation d’un mode de communication totalement différent de celui des entendants, le langage des signes, et de l’absence quasi-totale d’afférences en provenance du système auditif, il y a de fortes chances que d’importantes modifications fonctionnelles et structurales s’effectuent dans le cerveau des individus sourds profonds. Les études antérieures suggèrent que cette réorganisation risque d’avoir des répercussions plus importantes sur les structures corticales situées le long de la voie visuelle dorsale qu’à l’intérieur de celles situées à l’intérieur de la voie ventrale. L’hypothèse proposée par Ungerleider et Mishkin (1982) quant à la présence de deux voies visuelles dans les régions occipitales, même si elle demeure largement acceptée dans la communauté scientifique, s’en trouve aussi relativement contestée. Une voie se projetant du cortex strié vers les régions pariétales postérieures, est impliquée dans la vision spatiale, et l’autre se projetant vers les régions du cortex temporal inférieur, est responsable de la reconnaissance de la forme. Goodale et Milner (1992) ont par la suite proposé que la voie dorsale, en plus de son implication dans le traitement de l’information visuo-spatiale, joue un rôle dans les ajustements sensori-moteurs nécessaires afin de guider les actions. Dans ce contexte, il est tout à fait plausible de considérer qu’un groupe de personne utilisant un langage sensori-moteur comme le langage des signes dans la vie de tous les jours, s’expose à une réorganisation cérébrale ciblant effectivement la voie dorsale. L’objectif de la première étude est d’explorer ces deux voies visuelles et plus particulièrement, la voie dorsale, chez des individus entendants par l’utilisation de deux stimuli de mouvement dont les caractéristiques physiques sont très similaires, mais qui évoquent un traitement relativement différent dans les régions corticales visuelles. Pour ce faire, un stimulus de forme définie par le mouvement et un stimulus de mouvement global ont été utilisés. Nos résultats indiquent que les voies dorsale et ventrale procèdent au traitement d’une forme définie par le mouvement, tandis que seule la voie dorsale est activée lors d’une tâche de mouvement global dont les caractéristiques psychophysiques sont relativement semblables. Nous avons utilisé, subséquemment, ces mêmes stimulations activant les voies dorsales et ventrales afin de vérifier quels pourraient être les différences fonctionnelles dans les régions visuelles et auditives chez des individus sourds profonds. Plusieurs études présentent la réorganisation corticale dans les régions visuelles et auditives en réponse à l’absence d’une modalité sensorielle. Cependant, l’implication spécifique des voies visuelles dorsale et ventrale demeure peu étudiée à ce jour, malgré plusieurs résultats proposant une implication plus importante de la voie dorsale dans la réorganisation visuelle chez les sourds. Suite à l’utilisation de l’imagerie cérébrale fonctionnelle pour investiguer ces questions, nos résultats ont été à l’encontre de cette hypothèse suggérant une réorganisation ciblant particulièrement la voie dorsale. Nos résultats indiquent plutôt une réorganisation non-spécifique au type de stimulation utilisé. En effet, le gyrus temporal supérieur est activé chez les sourds suite à la présentation de toutes nos stimulations visuelles, peu importe leur degré de complexité. Le groupe de participants sourds montre aussi une activation du cortex associatif postérieur, possiblement recruté pour traiter l’information visuelle en raison de l’absence de compétition en provenance des régions temporales auditives. Ces résultats ajoutent aux données déjà recueillies sur les modifications fonctionnelles qui peuvent survenir dans tout le cerveau des personnes sourdes, cependant les corrélats anatomiques de la surdité demeurent méconnus chez cette population. Une troisième étude se propose donc d’examiner les modifications structurales pouvant survenir dans le cerveau des personnes sourdes profondes congénitales ou prélinguales. Nos résultats montrent que plusieurs régions cérébrales semblent être différentes entre le groupe de participants sourds et celui des entendants. Nos analyses ont montré des augmentations de volume, allant jusqu’à 20%, dans les lobes frontaux, incluant l’aire de Broca et d’autres régions adjacentes impliqués dans le contrôle moteur et la production du langage. Les lobes temporaux semblent aussi présenter des différences morphométriques même si ces dernières ne sont pas significatives. Enfin, des différences de volume sont également recensées dans les parties du corps calleux contenant les axones permettant la communication entre les régions temporales et occipitales des deux hémisphères.

Au cours des 25 dernières années, les recherches sur le développement visuel chez l’humain à l’aide de l’électrophysiologie cérébrale et des potentiels évoqués visuels (PEV) ont permis d’explorer plusieurs fonctions associées au cortex visuel. Néanmoins, le développement de certaines d’entre elles (p. ex. segmentation des textures), tout comme les effets de la prématurité sur celles-ci, sont des aspects qui nécessitent d’être davantage étudiés. Par ailleurs, compte tenu de l’importance de la vision dans le développement de certaines fonctions cognitives (p. ex. lecture, visuomotricité), de plus en plus de recherches s’intéressent aux relations entre la vision et la cognition. Les objectifs généraux de la présente thèse étaient d’étudier le développement visuel chez les enfants nés à terme et nés prématurément à l’aide de l’électrophysiologie, puis de documenter les impacts de la prématurité sur le développement visuel et cognitif. Deux études ont été réalisées. La première visait à examiner, chez des enfants nés prématurément, le développement des voies visuelles primaires durant la première année de vie et en début de scolarisation, ainsi qu’à documenter leur profil cognitif et comportemental. À l’aide d’un devis semi-longitudinal, dix enfants nés prématurément ont été évalués à l’âge de six mois (âge corrigé) et à 7-8 ans en utilisant des PEV, et des épreuves cognitives et comportementales à l’âge scolaire. Leurs résultats ont été comparés à ceux de 10 enfants nés à terme appariés pour l’âge. À six mois, aucune différence de latence ou d’amplitude des ondes N1 et P1 n’a été trouvée entre les groupes. À l’âge scolaire, les enfants nés prématurément montraient, comparativement aux enfants nés à terme, une plus grande amplitude de N1 dans la condition P-préférentielle et dans celle co-stimulant les voies M et P, et de P1 (tendance) dans la condition M-préférentielle. Aucune différence n’a été trouvée entre les groupes aux mesures cognitives et comportementales. Ces résultats suggèrent qu’une naissance prématurée exerce un impact sur le développement des voies visuelles centrales. L’objectif de la seconde étude était de documenter le développement des processus de segmentation visuelle des textures durant la petite enfance chez des enfants nés à terme et nés prématurément à l’aide des PEV et d’un devis transversal. Quarante-cinq enfants nés à terme et 43 enfants nés prématurément ont été évalués à 12, 24 ou 36 mois (âge corrigé pour les prématurés à 12 et 24 mois). Les résultats indiquaient une diminution significative de la latence de la composante N2 entre 12 et 36 mois en réponse à l’orientation, à la texture et à la segmentation des textures, ainsi qu’une diminution significative d’amplitude pour l’orientation entre 12 et 24 mois, et pour la texture entre 12 et 24 mois, et 12 et 36 mois. Les comparaisons entre les enfants nés à terme et ceux nés prématurément démontraient une amplitude de N2 réduite chez ces derniers à 12 mois pour l’orientation et la texture. Bien que ces différences ne fussent plus apparentes à 24 mois, nos résultats semblent refléter un délai de maturation des processus visuel de bas et de plus haut niveau chez les enfants nés prématurément, du moins, pendant la petite enfance. En conclusion, nos résultats indiquent que la prématurité, même sans atteinte neurologique importante, altère le développement des fonctions visuelles à certaines périodes du développement et mettent en évidence l’importance d’en investiguer davantage les impacts (p. ex. cognitifs, comportementaux, scolaires) à moyen et long-terme.

The visual recognition of complex movements and actions is crucial for communication and survival in many species. Remarkable sensitivity and robustness of biological motion perception have been demonstrated in psychophysical experiments. In recent years, neurons and cortical areas involved in action recognition have been identified in neurophysiological and imaging studies. However, the detailed neural mechanisms that underlie the recognition of such complex movement patterns remain largely unknown. This paper reviews the experimental results and summarizes them in terms of a biologically plausible neural model. The model is based on the key assumption that action recognition is based on learned prototypical patterns and exploits information from the ventral and the dorsal pathway. The model makes specific predictions that motivate new experiments.

The endocannabinoid system has been implicated in several neurobiological processes, including neurodegeneration and neuro protection. The aim of this study was to evaluate the effects of unilateral retinal ablation on the expression of the cannabinoid receptor subtype 1 (CB1) at both protein and mRNA levels in the optic tectum of the adult chick brain. After different survival times postlesion (2-30 days), the chick brains were subjected to immunohistochemical, immunoblotting, and real-time PCR procedures to evaluate CB1 expression. TUNEL and Fluoro-Jade B were used to verify the possible occurrence of cell death, and immunostaining for the microtubule-associated protein MAP-2 was performed to verify possible dendritic remodeling after lesions. No cell death could be observed in the deafferented tectum, at least up to 30 days postlesion, although Fluoro-Jade B could reveal degenerating axons and terminals. Retinal ablation seems to generate an increase of CB1 protein in the optic tectum and other retinorecipient visual areas, which paralleled an increase in MAP-2 staining. On the other hand, CB, mRNA levels were not changed after retinal ablation. Our results reveal that CB, expression in visual structures of the adult chick brain may be negatively regulated by the retinal innervation. The increase of CB1 receptor expression observed after retinal removal indicates that these receptors are not presynaptic in retinal axons projecting to the tectum and suggests a role of the cannabinoid system in plasticity processes ensuing after lesions. Muitos laboratórios de eletrofisiologia visual não possuem seus próprios valores de normalidade para o eletrorretinograma de campo total. Isto prejudica a confiabilidade dos diagnósticos de diversas doenças que afetam as vias visuais. Desta forma, o objetivo deste trabalho foi estabelecer os valores normativos para o teste Eletrorretinograma de Campo Total para o Laboratório de Neurologia Tropical (LNT) da Universidade Federal do Pará (UFPA). Realizaram o eletrorretinograma 68 indivíduos saudáveis e sem queixas visuais divididos em três grupos de acordo com a faixa etária: 36 indivíduos pertenceram ao grupo 1 (entre 17 e 30 anos), 21 indivíduos ao grupo 2 (entre 31 e 45 anos) e 11 indivíduos ao grupo 3 (entre 46 e 60 anos). O protocolo de realização do teste seguiu as recomendações da ISCEV, com a utilização de seis tipos de estimulação. Quatro após adaptação escotópica e estimulação com intensidades de: 0,01 cd.s/m2 (resposta de bastonetes), 3,0 cd.s/m2 (resposta mista de cones e bastonetes e potenciais oscilatórios) e 10,0 cd.s/m2 (resposta mista adicional). Dois após adaptação fotópica em fundo de 30 cd/m2: 3,0 cd.s/m2 (resposta de cones e Flicker 30Hz). Para a análise dos resultados foram calculados os valores de amplitude e tempo implícito das ondas a e b obtidas em resposta a cada um dos seis tipos de estimulação utilizados. Estes valores foram descritos estatisticamente através da mediana, intervalos de confiança, 1º e 3º quartis, coeficiente de variação, média, desvio padrão e valores mínimos e máximos. Os grupos de maior faixa etária apresentaram menores valores de amplitude e atraso no tempo implícito. A utilização da transformada wavelet permitiu a melhor visualização das ondas sem alteração de amplitude e tempo implícito. Portanto, os valores normativos obtidos podem servir como parâmetros de normalidade confiáveis para auxiliar o diagnóstico de doenças retinianas.

As contribuições dos mecanismos de detecção de contraste ao potencial cortical provocado visual (VECP) têm sido investigadas com o estudo das funções de resposta ao contraste e de resposta à frequência espacial. Anteriormente, o uso de sequências-m para o controle da estimulação era restrito à estimulação eletrofisiológica multifocal que, em alguns aspectos, se diferencia substancialmente do VECP convencional. Estimulações únicas com contraste espacial controlado por sequências-m não foram extensivamente estudadas ou comparadas às respostas obtidas com as técnicas multifocais. O objetivo deste trabalho foi avaliar a influência da frequência espacial e do contraste de redes senoidais no VECP gerado por estimulação pseudoaleatória. Nove sujeitos normais foram estimulados por redes senoidais acromáticas controladas por uma sequência-m binária pseudoaleatória em 7 frequências espaciais (0,4 a 10 cpg) em 3 tamanhos diferentes (4º, 8º e 16º de ângulo visual). Em 8º, foram testados adicionalmente seis níveis de contraste (3,12% a 99%). O kernel de primeira ordem não forneceu respostas consistentes com sinais mensuráveis através das frequências espaciais e dos contrastes testados – o sinal foi muito pequeno ou ausente – enquanto o primeiro e o segundo slice do kernel de segunda ordem exibiram respostas bastante confiáveis para as faixas de estímulo testadas. As principais diferenças entre os resultados obtidos com o primeiro e o segundo slice do kernel de segunda ordem foram o perfil das funções de amplitude versus contraste e de amplitude versus frequência espacial. Os resultados indicaram que o primeiro slice do kernel de segunda ordem foi dominado pela via M, porém para algumas condições de estímulo, pôde ser percebida a contribuição da via P. Já o segundo slice do kernel de segunda ordem refletiu contribuição apenas da via P. O presente trabalho estende achados anteriores sobre a contribuição das vias visuais ao VECP gerado por estimulação pseudoaleatória para uma grande faixa de frequências espaciais.

A esquistossomose é uma doença tropical causada, principalmente, pelo trematódeo Schistosoma mansoni, sendo que sua ocorrência afeta, mundialmente, 110 milhões de pessoas. A deposição dos ovos do parasita pode ocorrer, de forma ectópica, no sistema nervoso central (SNC) o qual leva à formação de granulomas com consequente produção do Fator de Crescimento Neuronal (NGF). Uma vez que muitos estudos demonstram a importância do NGF no desenvolvimento das vias corticais visuais, nosso estudo visou avaliar a possível alteração dos níveis de NGF no sistema visual assim como o impacto deste sobre a morfologia de células piramidais em dois modelos animais. A alteração na concentração do fator de crescimento assim como a morfometria neuronal foram avaliadas em animais permissíveis (camundongos) e não permissíveis (ratos) à infecção. Foram utilizados 174 ratos (Hooded Lister) e 135 camundongos albinos criados e mantidos em gaiolas e alimentados ad libitum. Esses animais foram inoculados, logo após o nascimento, com 50 cercárias. Setenta e sete ratos e 73 camundongos foram inoculados com solução salina e constituíram o grupo controle do estudo. Os períodos de infecção abrangeram uma a 48 semanas. Amostras do fígado e córtex visual foram retiradas, extraídas e quantificadas com kit de imunoensaio (ChemiKineTM Nerve Growth Factor (NGF) Sandwich ELISA Kit – Chemicon International). Para a análise morfométrica utilizamos células piramidais da camada IV do córtex visual marcadas através de injeção extracelular com Dextrana-Biotinilada (10.000 kDa). Os resultados foram expressos como média ± desvio padrão. Utilizamos teste t de Student para determinar diferenças estatísticas entre os grupos estudados. O valor médio de NGF encontrado no córtex visual de ratos infectados foi 39,2% maior do que no grupo controle (infectados: 400,9 ± 143,1 pg/mL; controle: 288 ± 31,9 pg/mL; p < 0,0001). Nas amostras de fígado, o aumento foi 28,9% maior no grupo infectado (infectados: 340,9 ± 103,9 pg/mL; p < 0,01; controle: 264,4 ± 38,6 pg/mL). Nenhum aumento significativo foi detectado antes de uma semana de infecção. Entre os camundongos, o aumento de NGF na área visual foi de 94,1% (infectados: 478,4 ± 284 pg/mL; p < 0,01; controle: 246,5 ± 76,8 pg/mL). No fígado destes animais o aumento foi de 138,7% (infectados: 561,8 ± 260,7 pg/mL; p < 0,01; controle: 301,3 ± 134,6 pg/mL). Em camundongos encontramos diferenças significativas quanto aos parâmetros dendríticos avaliados. A quantidade de dendritos foi 11,41% maior no grupo infectado do que no controle (controle: 25,28 ± 5,19; infectados: 28,16 ± 7,45; p < 0,05). O comprimento total dos dendritos também foi afetado (controle: 4.916,52 ± 1.492,65 μm; infectados: 5.460,40 ± 1.214,07 μm; p < 0,05) correspondendo a um aumento de 11,06%. A área total do campo receptor dendrítico sofreu um aumento de 12,99% (controle: 29.346,69 ± 11.298,62 μm2; infectados: 33.158,20 ± 7.758,31; p < 0,05) enquanto que a área somática teve uma redução de 13,61% (controle: 119,38 ± 19,68 μm2; infectados: 103,13 ± 24,69 μm2; p < 0,001). Quando foram avaliados os efeitos do aumento de NGF em ratos infectados não observamos diferenças significativas quanto aos parâmetros dendríticos analisados, em comparação ao grupo controle, com exceção de um aumento na área do corpo neuronal da ordem de 21,18% (controle: 132,20 ± 28,46 μm2; infectados: 160,20 ± 31,63 μm2; p < 0,00001). Este trabalho mostrou que a reação de produção de NGF no SNC durante a infecção por Schistosoma mansoni ocorre em maior magnitude no modelo permissível do que no modelo não permissível. Também demonstramos que, em camundongos, os efeitos sobre a morfologia neuronal é drasticamente afetada quando o organismo é submetido a um aumento na concentração de NGF em decorrência da infecção por Schistosoma mansoni. Diante destes dados, estudos avaliando as possíveis repercussões visuais e também dos efeitos na fisiologia celular causados pela infecção mansônica torna-se necessário para avaliar o real dano causado por este aumento patológico do fator de crescimento neuronal nas vias visuais de mamíferos.

We assessed chromatic discrimination in multiple sclerosis (MS) patients both with (ON) and without (no ON) a history of optic neuritis using the Cambridge color test (CCT). Our goal was to determine the magnitude and chromatic axes of any color vision losses in both patient groups, and to evaluate age-related changes in chromatic discrimination in both patient groups compared to normals. Using the CCT, we measured chromatic discrimination along the protan, deutan and tritan axes in 35 patients with MS (17 ON eyes) and 74 age matched controls. Color thresholds for both patient groups were significantly higher than controls` along the protan and tritan axes (P < 0.001). In addition, the ON and no-ON groups differed significantly along all three-color axes (p < 0.001). MS patients presented a progressive color discrimination impairment with age (along the deutan and tritan axes) that was almost two times faster than controls, even in the absence of ON. These findings suggest that demyelinating diseases reduce sensitivity to color vision in both red-green and blue-yellow axes, implying impairment in both parvocellular and koniocellular visual pathways. The CCT is a useful tool to help characterize vision losses in MS and the relationship between these losses and degree of optic nerve involvement.

Ocular enucleation produces significant morphological and physiological changes in central visual areas. However, our knowledge of the molecular events resulting from eye enucleation in visual brain areas remains elusive. We characterized here the transcription nuclear factor kappa-B (NF-kappa B) activation induced by ocular enucleation in the rat superior colliculus (SC). We also tested the effectiveness of the synthetic glucocorticoid dexamethasone in inhibiting its activation. Electrophoretic mobility shift assays to detect NF-kappa B indicated that this transcription factor is activated in the SC from 1 h to day 15 postlesion. The expression of p65 and p50 proteins in the nuclear extracts was also increased. Dexamethasone treatment was able to significantly inhibit NF-kappa B activation. These findings suggest that this transcriptional factor is importantly involved in the visual system short-term processes that ensue after retinal lesions in the adult brain. Acquired cataract and cognitive impairment are both common age-related problems, and ophthalmologists are increasingly likely to encounter patients who have both. Patients with dementia types who display early visuoperceptual impairment may present first to ophthalmology services. When these patients have coexisting cataract, it may be difficult to distinguish visual complaints due to cataract from those due to dementia. The interaction between visual impairment due to cataract and neurodegenerative disorders affecting the central visual pathways, is not fully understood. Visual impairment due to cataract may stress impaired attentional mechanisms and cataract extraction may improve cognitive performance in some patients with early cognitive impairment; however, the benefits of cataract surgery in established dementia are less clear. In this study, the literature on this subject was reviewed and the implications for practice were considered.

Small bistratified cells (SBCs) in the primate retina carry a major blue-yellow opponent signal to the brain. We found that SBCs also carry signals from rod photoreceptors, with the same sign as S cone input. SBCs exhibited robust responses under low scotopic conditions. Physiological and anatomical experiments indicated that this rod input arose from the AII amacrine cell-mediated rod pathway. Rod and cone signals were both present in SBCs at mesopic light levels. These findings have three implications. First, more retinal circuits may multiplex rod and cone signals than were previously thought to, efficiently exploiting the limited number of optic nerve fibers. Second, signals from AII amacrine cells may diverge to most or all of the approximately 20 retinal ganglion cell types in the peripheral primate retina. Third, rod input to SBCs may be the substrate for behavioral biases toward perception of blue at mesopic light levels.

V2 has long been recognized to contain functionally distinguishable compartments that are correlated with the stripelike pattern of cytochrome oxidase activity. Early electrophysiological studies suggested that color, direction/disparity, and orientation selectivity were largely segregated in the thin, thick, and interstripes, respectively. Subsequent studies revealed a greater degree of homogeneity in the distribution of response properties across stripes, yet color-selective cells were still found to be most prevalent in the thin stripes. Optical recording studies have demonstrated that thin stripes contain both color-preferring and luminance-preferring modules. These thin stripe color-preferring modules contain spatially organized hue maps, whereas the luminance-preferring modules contain spatially organized luminance-change maps. In this study, the neuronal basis of these hue maps was determined by characterizing the selectivity of neurons for isoluminant hues in multiple penetrations within previously characterized V2 thin stripe hue maps. The results indicate that neurons within the superficial layers of V2 thin stripe hue maps are organized into columns whose aggregated hue selectivity is closely related to the hue selectivity of the optically defined hue maps. These data suggest that thin stripes contain hue maps not simply because of their moderate percentage of hue-selective neurons, but because of the columnar and tangential organization of hue selectivity.

Ocular dominance (OD) plasticity is a robust paradigm for examining the functional consequences of synaptic plasticity. Previous experimental and theoretical results have shown that OD plasticity can be accounted for by known synaptic plasticity mechanisms, using the assumption that deprivation by lid suture eliminates spatial structure in the deprived channel. Here we show that in the mouse, recovery from monocular lid suture can be obtained by subsequent binocular lid suture but not by dark rearing. This poses a significant challenge to previous theoretical results. We therefore performed simulations with a natural input environment appropriate for mouse visual cortex. In contrast to previous work, we assume that lid suture causes degradation but not elimination of spatial structure, whereas dark rearing produces elimination of spatial structure. We present experimental evidence that supports this assumption, measuring responses through sutured lids in the mouse. The change in assumptions about the input environment is sufficient to account for new experimental observations, while still accounting for previous experimental results.

The correlation between cholinergic sensitivity and the level of stratification for ganglion cells was examined in the rabbit retina. As examples, we have used ON or OFF alpha ganglion cells and ON/OFF directionally selective (DS) ganglion cells. Nicotine, a cholinergic agonist, depolarized ON/OFF DS ganglion cells and greatly enhanced their firing rates but it had modest excitatory effects on ON or OFF alpha ganglion cells. As previously reported, we conclude that DS ganglion cells are the most sensitive to cholinergic drugs. Confocal imaging showed that ON/OFF DS ganglion cells ramify precisely at the level of the cholinergic amacrine cell dendrites, and co-fasciculate with the cholinergic matrix of starburst amacrine cells. However, neither ON or OFF alpha ganglion cells have more than a chance association with the cholinergic matrix. Z -axis reconstruction showed that OFF alpha ganglion cells stratify just below the cholinergic band in sublamina a while ON alpha ganglion cells stratify just below cholinergic b . The latter is at the same level as the terminals of calbindin bipolar cells. Thus, the calbindin bipolar cell appears to be a prime candidate to provide the bipolar cell input to ON alpha ganglion cells in the rabbit retina. We conclude that the precise level of stratification is correlated with the strength of cholinergic input. Alpha ganglion cells receive a weak cholinergic input and they are narrowly stratified just below the cholinergic bands.
