Los cambios que ocurren durante el funcionamiento (normal o patológico) de biomembranas resultan de alteraciones de interacciones moleculares dinámicas. Éstas involucran a fenómenos supramoleculares críticos (cambios de fase, electrostáticos y topológicos) que regulan la organización, estabilidad, expresión y metabolismo de lípidos y proteínas de membrana. Para conocer los factores físico-químicos que controlan el comportamiento de biomembranas se deben utilizar técnicas que permitan un control preciso de la organización molecular. (...) Objetivos generales y específicos: El propósito a largo plazo de las investigaciones es establecer las bases moleculares y supramoleculares por las cuales interacciones entre esfingolípidos, fosfolípidos y sus derivados regulan la organización, reconocimiento y reactividad de biomembranas. Al presente se estudia: I) Control supramolecular de la catálisis enzimática interfacial por enzimas fosfohidrolíticas que median la transducción de señales a nivel de membrana. II) Modulación de la estabilidad de biomembranas por cambios críticos de composición de la interfase, interacciones y transiciones topológicas. III) Caracterización y transferencia a soportes sólidos y líquidos de capas monomoleculares de organización intermolecular controlada, formadas con membranas neuronales y gliales.

Uno de los grandes desafíos analíticos es resolver la complejidad del análisis de cantidades trazas de compuestos orgánicos debido a la baja sensibilidad analítica de las técnicas usuales que permiten una determinación específica como IR o RMN. El uso de espectrofotometría UV-Visible y espectroluminiscencia, técnicas que presentan mayor sensibilidad, se ve dificultada en muchos casos por el efecto matriz producido en el tratamiento de muestras reales y complejas o pérdida de la selectividad debido a la superposición de bandas.La interacción por formación de complejos entre determinados sustratos y receptores macrocíclicos que presentan poros o cavidades nanométricas, puede afectar las propiedades espectroscópicas de los sustratos. La respuesta de técnicas sensibles puede traducirse así en un análisis selectivo debido al reconocimiento molecular que se establece entre un dado receptor y el sustrato de interés. Por otra parte puede mejorar la sensibilidad debido a efectos de micropolaridad del medio, a efectos de restricciones de grados de libertad, por compartamentalización o protección de los estados excitados de los sustratos incluidos. El uso analítico de receptores selectivos es un área actualmente en desarrollo, que permite una rápida determinación de especies químicas, disminuyendo el efecto de interferentes, mejorando la sensibilidad y disminuyendo el tratamiento de la muestra.Se estudiarán los mecanismos involucrados en las interacciones y los factores que los modifican por técnicas espectroscópicas como UV-visible, RMN y luminiscencia. Se determinarán los parámetros analíticos por luminiscencia en los medios y condiciones en que la sensibilidad analítica muestre el mayor incremento. Se realizarán las pruebas de validación en las mejores condiciones para cada uno y mezclas de analitos relacionados en muestras reales.

To investigate the role of the coreceptor CD8 and lipid rafts in cytotoxic T lymphocyte (CTL) activation, we used soluble mono-and multimeric H-2Kd-peptide complexes and cloned S14 CTL specific for a photoreactive derivative of the Plasmodium berghei circumsporozoite (PbCS) peptide 252-260 [PbCS(ABA)]. We report that activation of CTL in suspension requires multimeric Kd-PbCS(ABA) complexes co-engaging TCR and CD8. Using TCR ligand photo-cross-linking, we find that monomeric Kd-PbCS(ABA) complexes promote association of TCR/CD3 with CD8/p56lck. Dimerization of these adducts results in activation of p56lck in lipid rafts, where phosphatases are excluded. Additional cross-linking further increases p56lck kinase activity, induces translocation of TCR/CD3 and other signaling molecules to lipid rafts and intracellular calcium mobilization. These events are prevented by blocking Src kinases or CD8 binding to TCR-associated Kd molecules, indicating that CTL activation is initiated by cross-linking of CD8-associated p56lck. They are also inhibited by methyl-beta-cyclodextrin, which disrupts rafts and by dipalmitoyl phosphatidylethanolamine, which interferes with TCR signaling. Because efficient association of CD8 and p56lck takes place in rafts, both reagents, though in different ways, impair coupling of p56lck to TCR, thereby inhibiting the initial and essential activation of p56lck induced by cross-linking of engaged TCR.

In contrast to some extensively examined food mutagens, for example, aflatoxins, N-nitrosamines and heterocyclic amines, some other food contaminants, in particular polycyclic aromatic hydrocarbons (PAH) and other aromatic compounds, have received less attention. Therefore, exploring the relationships between dietary habits and the levels of biomarkers related to exposure to aromatic compounds is highly relevant. We have investigated in the European Prospective Investigation into Cancer and Nutrition (EPIC) cohort the association between dietary items (food groups and nutrients) and aromatic DNA adducts and 4-aminobiphenyl-Hb adducts. Both types of adducts are biomarkers of carcinogen exposure and possibly of cancer risk, and were measured, respectively, in leucocytes and erythrocytes of 1086 (DNA adducts) and 190 (Hb adducts) non-smokers. An inverse, statistically significant, association has been found between DNA adduct levels and dietary fibre intake (P = 0·02), vitamin E (P = 0·04) and alcohol (P = 0·03) but not with other nutrients or food groups. Also, an inverse association between fibre and fruit intake, and BMI and 4-aminobiphenyl-Hb adducts (P = 0·03, 0·04, and 0·03 respectively) was observed. After multivariate regression analysis these inverse correlations remained statistically significant, except for the correlation adducts v. fruit intake. The present study suggests that fibre intake in the usual range can modify the level of DNA or Hb aromatic adducts, but such role seems to be quantitatively modest. Fibres could reduce the formation of DNA adducts in different manners, by diluting potential food mutagens and carcinogens in the gastrointestinal tract, by speeding their transit through the colon and by binding carcinogenic substances.

The strategy is based on the Pauson-Khand (PK) reaction of norbornadiene and N-Boc-propargylamine as alkyne with a masked leaving group, which can be eliminated at will. This approach to the synthesis of 4,5-disubstituted cyclopentenones overcomes the problem of using the alkylation to introduce the alpha-side-chain. As an example, prostane 13-epi-12-oxo-phytodienoic acid (13-epi-12-oxo-PDA) methyl ester was synthesized.

Gold metallogelators is an emerging area of research. The number of results published in the literature is still scarce. The majority of these gels is observed in organic solvents, and the potential applications are still to be explored. In this work, we present an overview about gold metallogelators divided in two different groups depending on the type of solvent used in the gelation process (organogelators and hydrogelators). A careful analysis of the data shows that aurophilic interactions are a common motif directly involved in gelation involving Au(I) complexes. There are also some Au(III) derivatives able to produce gels but in this case the organic ligands determine the aggregation process. A last section is included about the potential applications that have been reported until now with this new and amazing class of supramolecular assemblies.

In this work is presented and tested (for 106 adducts, mainly of the zinc group halides) two empirical equations supported in TG data to estimate the value of the metal-ligand bond dissociation enthalpy for adducts: <D> (M-O) = t i / g if t i < 420 K and <D> (M-O) = (t i / g ) - 7,75 . 10-2 . t i if t i > 420 K. In this empirical equations, t i is the thermodynamic temperature of the beginning of the thermal decomposition of the adduct, as determined by thermogravimetry, andg is a constant factor that is function of the metal halide considered and of the number of ligands, but is not dependant of the ligand itself. To half of the tested adducts the difference between experimental and calculated values was less than 5%. To about 80% of the tested adducts, the difference between the experimental (calorimetric) and the calculated (using the proposed equations) values are less than 15%.

Mimicking Nature, supramolecular chemistry represents the chemistry beyond the molecule, in view that intermolecularinteractions constitute the driving force for the preparation of molecular and supramolecular assemblies, using the chemicalinformation contained in molecular building blocks. Upon molecular recognition between discrete units, chemical processessuch as self-assembly and self-organisation start operating, and are the leading processes to build up supramolecular aggregates and materials. When those materials have dimensions on thenanometric scale, a recently emerging scientific discipline is defined,Nanoscience. Nanomaterials are promising tools for many applications, and their use in biomedical and clinical applicationsdefines the so-called Nanomedicine. In this review we present a few selected examples of nanomaterials designed for therapeutical purposes, emphasizing the importance of the preparation methodology in terms of their therapeutical use.

This work reports the synthesis and characterization of adducts of general formula MCl2.ngly, where M= Mn and Cu; n= 2 and 4, and gly= glycine. The manganese adducts were synthesized by dissolution of both, manganese chloride and glycine in water, whereas the copper adducts were obtained by using an alternative solid state synthesis approach. For all adducts, the obtained infrared data shows that the coordination involves the amine nitrogen atom, as well as an oxygen atom of the COO- group. The TG curves for the synthesized adducts exhibit only one mass loss step associated with the release of glycine molecules.

Brazilian science is evolving rapidly and steadly in the last 10 years, reaching the 15º place in the international ranking. Research in nanotechnology is following a similar way generating new scientific and technological knowledge in several frontiers but specially in the interfaces of two or more areas, where Chemistry is consolidating itself as a central science. In this context, the supramolecular approach is a very promissing one because it allows the build-up of a chemical inteligence using all the sistematized knowledge for the design and development of new nanomaterials and products. The great challenge of Chemistry is not decrease the dimensionality of the materials but instead find ways to increase the dimensionality and structural complexity keeping strict control on the interactions between the components, in order to generate materials with new properties and functionalities. Unfortunately, the current vigorous advancement of scientific research has not been followed by the transformation of such know-how into patents and produts. Therefore much efforts should be devoted to build a national science and technology program, joining all the segments of the society involved in the technological development (university, institutes of technological research, industry and government) in order to promote the furtherance of the Brazilian technological base. Only in this way it is possible to evolve to a technological society capable to transform the scientific knowledge into wealthy, thus sustaining the socioeconomic development of the country.

The interest in the chemistry of cobalt (II) tetrasulfonated phthalocyanine (PcTsCo) comes mainly from its macrocycle-ligand structure combined with their special chemical characteristics, such as high solubility, well-defined redox reactions and remarkable optical absorption in the visible region. In this work, we use layer-by-layer technique in order to assemble CoTsPc and poly(allylaminehydrochloride) (PAH) in hybrid supramolecular system. The electronic spectroscopy and cyclic voltammetry techniques were utilized to study PAH/CoTsPc multilayers growth and the cysteine catalytic oxidation. PAH/CoTsPc showed high electrochemical stability and worthwhile to mention is the remarkable influence of supramolecular arrangement on the final redox properties of the system.

Giant extracellular hemoglobins are considered the summit of complexity in systems that carry oxygen, constituting an extraordinary model system to the study of hemoproteins. This class includes the hemoglobin of the annelid Glossoscolex paulistus that presents high cooperativity, great oligomeric and redox stabilities and ability of oligomeric reassociation. These properties have motivated evaluations about its utilization as prototype of artificial blood and biosensor. Kinetic studies involving autoxidation and detailed spectroscopic characterizations of its ferrous and ferric species have propitiated information about the structure-activity relationship of this macromolecule. The present review analyzes several biochemical issues, evaluating the state-of-art of this subject.
