血管生成是肿瘤生长、发展的必经之路,并且与实体瘤的发生、转移有着密切的关系,抑制肿瘤新生血管生成具有特异性高、疗效好、不易产生耐药性以及毒副作用低等特点,因此抑制肿瘤血管形成可望成为治疗癌症的一个突破点,以抗血管生成为主的肿瘤生物治疗研究已成为近十年的研究热点。玻璃海鞘(Coina intestinalis)属于内性目、玻璃海鞘科,因其进化上的独特地位常作为研究神经发育、免疫系统进化的材料。在其它种属的海鞘中分离发现了多种具有抗肿瘤活性的多肽,但关于玻璃海鞘抗血管生成活性多肽的分离纯化和活性研究未见报道。本文利用多种分离纯化手段,采用活性追踪的方法首次从玻璃海鞘中分离得到具有抗新生血管生成作用的多肽,并对其抗血管生成活性做了初步研究。 本研究利用冷丙酮分级沉淀,超滤截留分子量小于5kDa的蛋白,再经SephadexG25、Superdex75柱层析,µRPC C2/C18反相柱层析等分离手段,采用活性追踪的方法,由玻璃海鞘(Coina intestinalis)中分离纯化出抗血管生成多肽PCI,据保留时间计算其分子量为1.8 kDa。MTT检测表明其对人脐静脉血管内皮细胞(HUVEC)具有强烈的抑制作用,IC50为7.5 μg/ml,并对多种肿瘤细胞有直接的抑制作用。斑马鱼胚胎体内实验进一步表明PCI在40 μg/ml的浓度下作用12h,斑马鱼胚胎新生血管生成受到显著抑制,肠下静脉血管长度为正常组的30%,斑马鱼胚胎血管生成率为正常组的45%。

Background: Small molecule inhibitors of the zinc finger domain (ZFI) in the nucleocapsid protein (NCp7) of HIV-1 are potent inhibitors of HIV and SIV
replication and may have utility as topical products to prevent infection. Furthermore, intravaginal rings (IVRs) were developed as coitally-independent,
sustained release devices which could be used for administration of HIV microbicides. The aims of these studies were to demonstrate that IVRs sized for
macaques are practical and compatible with the current generation of thioester-based NCp7 inhibitors.
Methods: Non-medicated silicone elastomer vaginal rings of various sizes thought to be applicable for macaques were prepared and tested for vaginal fit in Pigtailed and Chinese Rhesus macaques. Macaques were monitored for 8 weeks for mucosal disruption by colposcopy and proinflammatory cytokine markers in cervical vaginal lavages (CVL) using Luminex bead-based technology. Three different ZFIs (compounds 52, 89 and 122, each derived from an N-substituted S-acyl-2-mercaptobenzamide thioester scaffold) were loaded at 50 mg into an optimal matrix-type ring design. In vitro continuous release studies were then conducted over 28 days and analyzed by HPLC. Rate of release was determined by linear regression analysis.
Results: Qualitative evaluation at the time of ring insertion suggested that the 25 mm ring provided optimal fit in both macaque species. All rings remained in
place during the study period (2 to 4 weeks), and the animals did not attempt to remove the rings. No tissue irritation was observed, and no signs of physical
discomfort were noted. Also, no significant induction of cervicovaginal proinflammatory markers was observed during the 8-week period during and following ring insertion. One Pigtailed macaque showed elevated IL-8 levels in the CVL during the period when the ring was in place; however, these levels were comparable to those observed in two control macaques. In vitro release of the ZFIs peaked at day 1 and then continually declined to near steady-state rates between 20-30 mcg/day. The percent release after 14 days was 2.9, 2.0 and 0.9 for ZFI 89, 52 and 122, respectively.
Conclusions: IVRs of 25mm diameter, determined to be the optimal size for macaques, were well tolerated and did not induce inflammation. Release of all ZFI compounds followed t 0.5 kinetics. These findings suggest that efficacy testing in primate models is warranted to fully evaluate the potential to prevent
transmission.

Cardiogenesis is a delicate and complex process that requires the coordination of an intricate network of pathways and the different cell types. Therefore, understanding heart development at the morphogenetic level is an essential requirement to uncover the causes of congenital heart disease and to provide insight for disease therapies. Mouse Cerberus like 2 (Cerl2) has been defined as a Nodal antagonist in the node with an important role in the Left-Right (L/R) axis establishment, at the early embryonic development. As expected, Cerl2 knockout mice (Cerl2-/-) showed multiple laterality defects with associated cardiac failure. In order to identify the endogenous role of Cerl2 during heart formation independent of its described functions in the node, we accurately analyzed animals where laterality defects were not present. We thereby unravel the consequences of Cerl2 lossof- function in the heart, namely increased left ventricular thickness due to hyperplasia of cardiomyocytes and de-regulated expression of cardiac genes. Furthermore, the Cerl2 mutant neonates present impaired cardiac function. Once that the cardiac expression of Cerl2 is mostly observed in the left ventricle until around midgestration, this result suggest a specific regulatory role of Cerl2 during the formation of the left ventricular myoarchitecture. Here, we present two possible molecular mechanisms underlying the cardiac Cerl2 function, the regulation of Cerl2 antagonist in activation of the TGFßs/Nodal/Activin/Smad2 signaling identified by increased Smad2 phosphorilation in Cerl2-/- hearts and the negative feedback between Cerl2 and Wnt/ß-catenin signaling in heart formation. In this work and since embryonic stem cells derived from 129 mice strain is extensively used to produce targeted mutants, we also present echocardiographic reference values to progressive use of juveniles and young adult 129/Sv strain in cardiac studies. In addition, we investigate the cardiac physiology of the surviving Cerl2 mutants in 129/Sv background over time through a follow-up study using echocardiographic analysis. Our results revealed that Cerl2-/- mice are able to improve and maintain the diastolic and most of systolic cardiac physiologic parameters as analyzed until young adult age. Since Cerl2 is no longer expressed in the postnatal heart, we suggest that an intrinsic and compensatory mechanism of adaptation may be active for recovering the decreased cardiac function found in Cerl2 mutant neonates. Altogether, these data highlight the role of Cerl2 during embryonic heart development in mice. Furthermore, we also suggest that Cerl2-/- may be an interesting model to uncover the molecular, cellular and physiological mechanisms behind the improvement of the cardiac function, contributing to the development of therapeutic approaches to treat heart failures.

SAMHD1 has recently been identified as an HIV-1 restriction factor operating in myeloid cells. As a countermeasure, the Vpx accessory protein from HIV-2 and certain lineages of SIV have evolved to antagonize SAMHD1 by inducing its ubiquitin-proteasome-dependent degradation. Here, we show that SAMHD1 experienced strong positive selection episodes during primate evolution that occurred in the Catarrhini ancestral branch prior to the separation between hominoids (gibbons and great apes) and Old World monkeys. The identification of SAMHD1 residues under positive selection led to mapping the Vpx-interaction domain of SAMHD1 to its C-terminal region. Importantly, we found that while SAMHD1 restriction activity toward HIV-1 is evolutionarily maintained, antagonism of SAMHD1 by Vpx is species-specific. The distinct evolutionary signature of SAMHD1 sheds light on the development of its antiviral specificity.

Les chimiokines et leurs récepteurs respectifs jouent un rôle important dans l’immunité innée et adaptative. Les récepteurs de chimiokines identifient des cellules T CD4+ avec potentiel de migration dans des tissus spécifiques et à fonctionnalité distincte du point de vue de la spécificité antigénique et de la production de cytokines. L’identité de la population des cellules T CD4+ susceptibles versus résistantes à l’infection par le virus de l’immunodéficience humaine (VIH) reste mal définie. Le recrutement dans les muqueuses intestinales d’un excès de cellules T effectrices (CD8+) comparé aux cellules cibles (CD4+) représente un bon pronostic de l’infection par le virus de l’immunodéficience simienne (VIS), tandis que la déplétion des cellules Th17 dans les tissus lymphoïdes associés au tractus gastro-intestinal (GALT) est un marqueur de la progression de l’infection à VIH. L’effet régulateur des chimiokines sur l’activation de la réplication virale dans différentes sous-populations cellulaires T CD4+ reste peu étudié. Ce projet de maîtrise est divisé en 3 parties: (1) l’identification des récepteurs de chimiokines CCR4, CXCR3 et CCR6 comme marqueurs de surfaces des sous populations T CD4+ avec susceptibilité distincte à l’infection par le VIH; (2) la caractérisation phénotypique et fonctionnelle des cellules T CD4+ et T CD8+ spécifiques au VIH de sujets à progression lente vers le stade sida (LTNP); et (3) les effets des chimiokines ligands de CCR4, CXCR3 et CCR6 sur l’activation cellulaire et la réplication virale in vitro. Nos résultats démontrent que les cellules T CD4+ CCR4+CCR6+ (profile cytokinique Th17) et CXCR3+CCR6+ (profile cytokinique Th1/Th17) sont hautement permissives à l’infection par le VIH. Nous proposons également de nouveaux corrélats de protection immunitaire contre le VIH chez les sujets LTNP: (i) le potentiel de co-localisation dans les muqueuses intestinales des cellules T CD4+ et CD8+ spécifiques au VIH via l’intégrine β7, (ii) le ratio élevé entre les cellules T effectrices (CD8+) versus les cellules cibles (CD4+) spécifiques au VIH, (iii) le profil cytokinique Th17 et (iv) la capacité des cellules T CD4+ et CD8+ spécifiques au VIH à produire des ligands de CCR5 bloquant l’entrée virale. Finalement, nos résultats sur l’effet co-stimulateur des chimiokines sur les cellules T et leurs effets opposés sur la réplication virale démontrent l’implication du réseau des chimiokines dans la régulation de la pathogenèse de l’infection à VIH.

La protéine Nef du VIH-1 joue un rôle important dans la pathogenèse du VIH-1 en modulant les voies de signalisation de la cellule hôte. La signalisation par le TcR est essentielle à la sélection positive pour générer les cellules simples positives (SP) CD4+ et simples positives (SP) CD8+, processus largement dépendant de l’activité de la Src kinase Lck et de son habileté à lier la queue cytoplasmique des corécepteurs CD4 et CD8. Nous avons précédemment trouvé que l’expression de Nef dans le VIH ou VIS peut induire une sévère déplétion des thymocytes et une baisse d’expression du corécepteur CD4 à la membrane. Nous avons également montré que Nef bloque la génération des thymocytes doubles positifs (DP) CD4+ CD8+ en plus d’altérer la transition des cellules DP vers CD4+ SP. Par contre, ce phénotype est récupérable par plusieurs approches dont le croisement d’une souris transgéniques exprimant Nef avec une souris exprimant la forme constitutivement active de Lck Y505F. Les résultats indiquent que la maturation des cellules CD4+ est altérée par le dysfonctionnement de la signalisation CD4-Lck. Toutefois, les mécanismes moléculaires par lesquels Nef contribue au bloc de la génération des cellules CD4+ dans le thymus demeurent très imprécis. Dans cette étude, en utilisant des approches biochimiques et de microscopie confocale, nous avons trouvé que les thymocytes transgéniques Nef+ expriment plus de Lck que les thymocytes Nef-. Malgré cette augmentation, une partie significative de Lck est incapable d’atteindre la membrane plasmique. Cette fraction était significativement accumulée dans un compartiment intracellulaire des thymocytes transgéniques exprimant Nef. Également, en utilisant la technique d’essai kinase in vitro, nous avons trouvé que l’activité kinase de Lck est significativement augmentée dans les thymocytes transgéniques mais demeure stable suite à une stimulation par un α-CD3ε + α-CD4. Également, comparativement aux thymocytes Nef-, la kinase Lck dans les thymocytes transgéniques était résistante à la dégradation suite à une stimulation. En examinant le statut de c-Cbl, le principal régulateur négatif de Lck, nous avons montré que c-Cbl colocalise faiblement avec Lck, malgré son hyperphosphorylation constitutive. Ceci pourrait expliquer l’échec de la dégradation de Lck. En plus, nous avons trouvé que suite à une stimulation par un α-CD3ε + α-CD4, la phosphorylation de Zap-70 en tyrosine 493 par Lck est diminuée, résultant d’une importante baisse de l’activité kinase de Zap-70 et d’un bloc des premiers évènements de la voie de signalisation par le TcR. Ces données indiquent que la signalisation CD4-Lck est interrompue par la présence de Nef.

Structure is an important physical feature of the soil that is associated with water movement, the soil atmosphere, microorganism activity and nutrient uptake. A soil without any obvious organisation of its components is known as apedal and this state can have marked effects on several soil processes. Accurate maps of topsoil and subsoil structure are desirable for a wide range of models that aim to predict erosion, solute transport, or flow of water through the soil. Also such maps would be useful to precision farmers when deciding how to apply nutrients and pesticides in a site-specific way, and to target subsoiling and soil structure stabilization procedures. Typically, soil structure is inferred from bulk density or penetrometer resistance measurements and more recently from soil resistivity and conductivity surveys. To measure the former is both time-consuming and costly, whereas observations made by the latter methods can be made automatically and swiftly using a vehicle-mounted penetrometer or resistivity and conductivity sensors. The results of each of these methods, however, are affected by other soil properties, in particular moisture content at the time of sampling, texture, and the presence of stones. Traditional methods of observing soil structure identify the type of ped and its degree of development. Methods of ranking such observations from good to poor for different soil textures have been developed. Indicator variograms can be computed for each category or rank of structure and these can be summed to give the sum of indicator variograms (SIV). Observations of the topsoil and subsoil structure were made at four field sites where the soil had developed on different parent materials. The observations were ranked by four methods and indicator and the sum of indicator variograms were computed and modelled for each method of ranking. The individual indicators were then kriged with the parameters of the appropriate indicator variogram model to map the probability of encountering soil with the structure represented by that indicator. The model parameters of the SIVs for each ranking system were used with the data to krige the soil structure classes, and the results are compared with those for the individual indicators. The relations between maps of soil structure and selected wavebands from aerial photographs are examined as basis for planning surveys of soil structure. (C) 2007 Elsevier B.V. All rights reserved.

Scope: The use of biomarkers in the objective assessment of dietary intake is a high priority in nutrition research. The aim of this study was to examine pentadecanoic acid (C15:0) and heptadecanoic acid (C17:0) as biomarkers of dairy foods intake. Methods and results: The data used in the present study were obtained as part of the Food4me Study. Estimates of C15:0 and C17:0 from dried blood spots and intakes of dairy from an FFQ were obtained from participants (n=1,180) across 7 countries. Regression analyses were used to explore associations of biomarkers with dairy intake levels and receiver operating characteristic (ROC) analyses were used to evaluate the fatty acids. Significant positive associations were found between C15:0 and total intakes of high-fat dairy products. C15:0 showed good ability to distinguish between low and high consumers of high-fat dairy products. Conclusion: C15:0 can be used as a biomarker of high-fat dairy intake and of specific high-fat dairy products. Both C15:0 and C17:0 performed poorly for total dairy intake highlighting the need for caution when using these in epidemiological studies.

Waldorfpedagogiken har utvecklats från antroposofin och används idag i skolor över hela världen. Antroposofin uppfyller flera av de viktigaste kriterierna i de vanligaste definitionerna av religion. Â I denna artikel behandlas frågan om huruvida waldorfpedagogiken ska uppfattas som religiös eller inte. Mitt material består av skriftliga och elektroniska källor av olika slag, intervjuer samt observationer på en waldorfskola. Frågan kommer att diskuteras utifrån tre aspekter: ideologisk frikoppling, lärarens roll samt potentiellt religiösa inslag relaterade till morgonspråket och religionsundervisning. Jag föreslår att waldorfpedagogiken ska ses som en hybrid i den terminologi som Siv-Ellen Kraft använder. En hybrid är en företeelse där en förÂståelse med religiösa inslag utgör en av flera möjligheter (Kraft 2011: 78). Trots tillämpandet av detta perspektiv kvarstår dock andra problematiska områden gällande relationen mellan waldorfpedagogik och religion.

Os granodioritos Água Azul (GrdAA) e Água Limpa (GrdAL) afloram no extremo sul do Domínio Carajás como dois corpos alongados segundo o trend regional E-W, anteriormente inseridos no Complexo Xingu. O GrdAL é formado essencialmente por biotita-anfibólio granodioritos e muscovita-biotita granodioritos, além de anfibólio-biotita tonalitos subordinados; no GrdAA, epídoto-anfibólio-biotita granodioritos são dominantes, epídoto-anfibólio-biotita tonalitos e (anfibólio)-epídoto-biotita monzogranitos, subordinados. Essas rochas mostram assinaturas geoquímicas afins dos sanukitoides arqueanos. O estudo de suscetibilidade magnética (SM) mostrou valores relativamente baixos para o GrdAL (média de 17,54 × 10-4 SIv) e o GrdAA (média de 4,19 × 10-4 SIv). Os estudos dos minerais opacos mostram que a magnetita e a hematita são as fases comuns e que a ilmenita está ausente nessas rochas. O GrdAL contém titanita associada à magnetita, enquanto o GrdAA contém pirita, calcopirita e goethita. No GrdAL, a magnetita é mais abundante e desenvolvida que no GrdAA, justificando, assim, sua SM mais elevada. A martitização da magnetita e a oxidação dos sulfetos, gerando goethita, ocorreram a baixas temperaturas. A correlação positiva entre os valores de SM e os conteúdos modais de opacos, anfibólio, epídoto + allanita e quartzo + K-feldspato, assim como a correlação negativa de SM com biotita e máficos observadas nessas unidades, denunciam uma tendência no aumento de SM no sentido anfibólio tonalitos/anfibólio granodioritos à biotita granodioritos/biotita monzogranitos. Os dados geoquímicos corroboram esse comportamento, com correlação negativa entre os valores de SM e Fe2O3T, FeO e MgO, refletindo para as duas unidades uma tendência de aumento nos valores de SM paralelamente à diferenciação magmática. As afinidades geoquímicas e mineralógicas entre essas rochas e os sanukitoides do Domínio Rio Maria sugerem condições de fugacidade de oxigênio entre os tampões HM e FMQ para os granitoides estudados.

T-cell based vaccine approaches have emerged to counteract HIV-1/AIDS. Broad, polyfunctional and cytotoxic CD4(+) T-cell responses have been associated with control of HIV-1 replication, which supports the inclusion of CD4(+) T-cell epitopes in vaccines. A successful HIV-1 vaccine should also be designed to overcome viral genetic diversity and be able to confer immunity in a high proportion of immunized individuals from a diverse HLA-bearing population. In this study, we rationally designed a multiepitopic DNA vaccine in order to elicit broad and cross-clade CD4(+) T-cell responses against highly conserved and promiscuous peptides from the HIV-1 M-group consensus sequence. We identified 27 conserved, multiple HLA-DR-binding peptides in the HIV-1 M-group consensus sequences of Gag, Pol, Nef, Vif, Vpr, Rev and Vpu using the TEPITOPE algorithm. The peptides bound in vitro to an average of 12 out of the 17 tested HLA-DR molecules and also to several molecules such as HLA-DP, -DQ and murine IA(b) and IA(d). Sixteen out of the 27 peptides were recognized by PBMC from patients infected with different HIV-1 variants and 72% of such patients recognized at least 1 peptide. Immunization with a DNA vaccine (HIVBr27) encoding the identified peptides elicited IFN-gamma secretion against 11 out of the 27 peptides in BALB/c mice; CD4(+) and CD8(+) T-cell proliferation was observed against 8 and 6 peptides, respectively. HIVBr27 immunization elicited cross-clade T-cell responses against several HIV-1 peptide variants. Polyfunctional CD4(+) and CD8(+) T cells, able to simultaneously proliferate and produce IFN-gamma and TNF-alpha, were also observed. This vaccine concept may cope with HIV-1 genetic diversity as well as provide increased population coverage, which are desirable features for an efficacious strategy against HIV-1/AIDS.
