Plusieurs cibles thérapeutiques dans le développement de médicaments contre l’obésité visent une diminution de l’appétit et de la masse adipeuse et à augmenter la dépense énergétique. L’appétit et le métabolisme énergétique sont régulés par certains neuropeptides qui agissent au niveau du système nerveux central, notamment dans l’hypothalamus. Parmi ces neuropeptides, les peptides RF-amide ou QRFP (pyroglutamylated RF-amide peptides), ainsi nommés par la présence du motif conservé Arg-Phe-NH2 dans le domaine C-terminal, induisent une hyperphagie et une augmentation de la masse adipeuse lorsqu’administrés par voie centrale. Les formes bioactives de ces peptides comprennent principalement 43 (QRFP-43) et 26 (QRFP-26) acides aminés. Outre les peptides QRFP, leurs récepteurs, les GPR103 de la famille des récepteurs à 7 passages transmembranaires couplés aux protéines G, sont exprimés dans l’hypothalamus. Plus récemment, des études ont montré la sécrétion de ces neuropeptides, et la présence du GPR103, dans le tissu adipeux. Cependant, le rôle de la voie signalétique (QRFP/GPR103) dans la régulation du métabolisme lipidique au niveau périphérique est peu connu. Les travaux de cette thèse ont porté sur la caractérisation des effets adipogéniques périphériques des neuropeptides QRFP. En premier lieu, nos travaux ont montré que les adipocytes 3T3-L1 et les adipocytes murins isolés des dépôts adipeux blancs expriment le prépro-QRFP et uniquement le récepteur GPR103B, un des deux sous-types de récepteurs présents chez la souris. De plus, nous avons montré que l’expression du récepteur est régulée par une diète riche en lipides réduisant l’expression du prépro-QRFP, mais augmentant celle du GPR103B dans les dépôts lipidiques. Chez l’humain, les adipocytes de l’omentum expriment autant le GPR103 que le prépro-QRFP. Nous avons de plus étudié la fonctionnalité du GPR103B dans les adipocytes 3T3-L1 par l’utilisation d’ARN interférents. Nous avons observé que ce récepteur médie les effets adipogéniques des QRFPs en augmentant l’expression du récepteur nucléaire PPAR-gamma (peroxisome proliferator-activated receptor gamma) et le facteur de transcription C/EBP-alpha (CCAAT-enhancer binding protein alpha) résultant en une accumulation des triglycérides. Nous avons aussi mis en évidence les effets anti-lipolytiques des QRFPs. En effet, les QRFP inhibent fortement la lipolyse induite avec l’isoprotérénol. L’étude des mécanismes moléculaires à l’origine des effets anti-lipolytiques du QRFP-43 a montré l’activation de la voie de signalisation PI3-K/PKB (phosphatidylinositol 3-kinase/protéine kinase B) en réponse à la stimulation du GPR103B. La réponse anti-lipolytique induite par le QRFP-43 est associée à une diminution de la phosphorylation de la périlipine A (PLIN1a) et de la lipase hormono-sensible (HSL). Nos études ont élucidé les mécanismes conduisant à l’inhibition de la phosphorylation de la PLIN1a en réponse à l’activation du GPR103B, impliquant l’inhibition de la migration de la cavéoline 1 et de la sous unité catalytique de la protéine kinase A (PKA) au niveau des gouttelettes lipidiques, ainsi que l’inhibition de l’activité des Src kinases et de la protéine kinase C (PKC). En conclusion, nos travaux ont montré que les QRFP-43 et -26 exercent un effet adipogénique et anti-lipolytique dans les adipocytes, mettant ainsi en évidence le rôle des neuropeptides QRFPs dans la régulation du métabolisme lipidique au niveau adipocytaire.

Die Spezifität und Effizienz zellulärer Signalprozesse wird durch die intrazelluläre Kompartimentierung von Signalmolekülen erreicht. A-Kinase-Ankerproteine (AKAPs) bilden eine Familie aus Gerüstproteinen, die zeitliche und räumliche Lokalisation der cAMP-abhängigen Proteinkinase (PKA) übernehmen. Die direkte Interaktion wird dabei über die Dimerisierungs- und Dockingdomäne (DD-Domäne) der regulatorischen Untereinheiten von PKA vermittelt. Das charakteristische strukturelle Merkmal bei kanonischen AKAPs ist eine amphipathische Helix. Es existiert allerdings auch eine kleine Gruppe von nicht-kanonischen AKAPs, deren Bindung an die DD-Domäne nicht über eine amphipathische Helix vermittelt wird. In dieser Arbeit wurden die zwei potentiellen nicht-kanonischen AKAPs Neurochondrin (neurite-outgrowth promoting protein) und Rack1 (receptor of activated C-kinase 1) charakterisiert. Neurochondrin, dessen Expression mit dem Neuriten-Wachstum in jungen Neuronen korreliert ist und das vermutlich eine entscheidende Funktion bei der Langzeitpotenzierung im Hippocampus übernimmt, zeigt in SPR-Bindungsstudien eine hochaffine, nanomolare Interaktion mit der R-Untereinheit Typ IIalpha von PKA. Kompetitionsanalysen mit dem AKAP-Disruptor-Peptid Ht 31 und Untersuchungen mit der isolierten DD-Domäne von RIIalpha bestätigen eine spezifische Interaktion. Das nicht-kanonische RII-Bindemotiv von Neurochondrin ist aus zwei Domänen aufgebaut, die einen hohen alpha-helikalen Anteil besitzen, aber keine amphipathische Helix bilden. Peptidbasierte Interaktionsstudien der einzelnen Domänen zeigen dennoch ebenfalls nanomolare Affinitäten zu RIIalpha. Rack1 ist ein etabliertes Gerüstprotein mit einer propellerartigen beta-Faltblattstruktur, für das bereits über 100 verschiedene Interaktionspartner beschrieben werden konnten. Die Integration von Rack1 in unterschiedliche Signalprozesse ist äußerst vielfältig. Um dabei die Spezifität jeder einzelnen Interaktion zu gewährleisten, sind individuelle Bindungsstrategien nötig. Die niedrigaffine Interaktion zur RIbeta-Untereinheit von PKA wird daher über multiple Bindestellen vermittelt. Die DD-Domäne von RIbeta übernimmt dabei eine spezifische Funktion, wie unter anderem durch Kompetitionsanalysen mit dem RI-spezifischen AKAP-Disruptor-Peptid RIAD gezeigt werden konnte. Die einzigartige Struktur der DD-Domäne generiert zudem ein Bindemotiv für Rack1, das Ähnlichkeiten mit der „Rack1 interacting-Domäne“ (RAID) von PDE4D5 aufweist. Sowohl Neurochondrin als auch Rack1 besitzen essenzielle neuronale Funktionen. Daher erweitert die Identifizierung der beiden neuen nicht-kanonischen AKAPs nicht nur die strukturelle Diversität der AKAP-Familie, sondern trägt zudem zum Verständnis der neuronalen Signalintegration von PKA bei.

Analizar y seleccionar actividades de ense??anza para una posterior taxomizaci??n. Taxomizar las actividades de ense??anza para facilitar la cientificidad de la toma de decisiones en el aula y ofrecer mayor rigor y claridad al pensamiento de los profesores. Dejar patente la estructura did??ctica de las actividades de ense??anza y posibilitar su selecci??n cient??fica. El campo de estudio son las actividades de Ciencias Experimentales, incluidas en textos y curricula escritos para la etapa de 11 a 13 a??os. Se analizaron 14 textos distintos para la selecci??n de actividades. Se seleccionaron 15 actividades de cada texto, de forma aleatoria, a trav??s de la numeraci??n de las actividades y la utilizaci??n de las tablas de Sixto Rios (1972) y Barbancho. En total se analizaron 210 actividades. La metodolog??a de trabajo elegida ha sido la taxomizaci??n num??rica, pero el primer paso consisti?? en la selecci??n de las actividades o elementos a sistematizar. Una vez elegidas ??stas se seleccionaron los caracteres o categor??as a trav??s de las cuales se taxomizaron las actividades (emisor, receptor, canal, mensaje, c??digo verbal, c??digo ic??nico, referente, etc.). Posteriormente, se construy?? la matriz de datos y a partir de ??sta la matriz de semejanzas, hallando el coeficiente de distancias eucl??deas, despues de un an??lisis de cluster obtendremos un fenograma que nos permite hallar las relaciones y amalgamaciones entre las actividades y extraer conclusiones. Matriz de semejanzas. Mediante dos programas inform??ticos: P2M, que es un programa cuyo objeto es formar clusters con los datos o casos que se le impriman, a partir de una cualquiera de las medidas de distancia existente (en este caso eucl??dea); el otro es un programa inform??tico que complementa al anterior (ordena los datos obtenidos, agrupa los datos iguales, halla porcentajes, etc.). ??ste ??ltimo es un programa creado para esta investigaci??n. Resultados del primer tratamiento de datos: 8 tipos de actividades; todas ellas cumplen una funci??n formativa; las de conocimiento son aconsejables para fijar ideas b??sicas; las de comprensi??n son positivas por la personalizaci??n de los contenidos; las de aplicaci??n favorecen la relaci??n teor??a-pr??ctica. Es importante el estudio de la situaci??n contextual antes de proceder a la elecci??n de actividades de un tipo. El segundo programa permite la creaci??n de un banco de datos para archivar todas las actividades de modo que un profesor podr??a elegir los caracteres que m??s le convengan. Escasa variedad de caracteres. Hay que elaborar actividades lo m??s variadas posibles. Hay que adecuar las actividades a los objetivos pretendidos y a la situaci??n contextual del alumno.

Praseodymium oxide as a thin film of controllable layer is known to display many unique physiochemical properties, which can be useful to ceramic, semiconductive and sensor industries. Here in this short paper, we describe a new chemical method of depositing praseodymium oxide on tin-doped indium oxide (ITO) surface using a layer-by-layer approach. The process is carried out by dipping the ITO in solutions of adsorbable polycationic chitosan and alkaline praseodymium hydroxide Pr(OH)(3) alternatively in order to build up the well-defined multi-layers. XRD suggests that the predominant form of the oxide is Pr6O11, obtained after heat treatment of the deposited ITO in static air at 500 degrees C. Microscopic studies including AFM, TEM and SEM indicate that the deposited oxide particles are uniform in size and shape (cylindrical), mesoporous and the thickness of the film can be controlled. AC impedance measurements of the deposited materials also reveal that the oxide layers display a high electrical conductivity hence suitable for sensor uses. (c) 2006 Elsevier B.V. The present paper describes the catalytic oxidation of urea performed by nickel hydroxide and nickel/cobalt hydroxide modified electrodes by using both electrodeposited films and nanoparticles. The incorporation of Co foreign atoms leads to a slight increase in sensitivity besides the shift in redox process, avoiding the oxygen reaction. Nanostructured Ni80Co20(OH)(2) was synthesized by sonochemical route producing 5 nm diameter particles characterized by high-resolution transmission electron microscopy (HRTEM) being immobilized onto electrode by using the electrostatic Layer-by-layer technique, yielding attractive modified electrodes for sensor development. The electrochemical behaviour of multi-walled carbon nanotubes was compared with that of glassy carbon, and the differences were investigated by cyclic voltammetry and electrochemical impedance spectroscopy before and after acid pre-treatment. The electrochemical techniques showed that acid functionalisation significantly improves the electrocatalytic properties of carbon nanotubes. These electrocatalytic properties enhance the analytical signal, shift the oxidation peak potential to a less positive value, and the charge-transfers rate increase of both dopamine and K(4)[Fe(CN)(6)]. The functionalisation step and the resulting appearance of edge planes covered with different chemical groups were confirmed by FTIR measurements. Carbon nanotubes after acid pre-treatment are a potentially powerful analytical tool for sensor development. (C) 2010 Elsevier B.V. All rights reserved.

The deposition of thick film pastes by screen-printing is a relatively simple and convenient method to produce thicker layers with thickness up to 100 mum. In the present work, the barium titanate thick films were prepared from mechanically activated powders based on BaC03 and TiO2. After mixing, the powders were calcined at low temperature by slow heating and cooling rates. The thick films were deposited on to Al2O3 substrates through hybrid technology. The obtained films were fired at 850 degreesC together with electrode material (silver/palladium). The electrical properties of thick films: dielectric permittivity, dielectric losses, Curie temperature, hysteresis loop were reported. The obtained BT thick films can be applied in as multilayer capacitors or in gas sensor application. (C) 2003 Elsevier Ltd. All rights reserved.

Chronic and excessive alcohol consumption has been related to an increased risk of several cancers, including that of the liver; however, studies in animal models have yet to conclusively determine whether ethanol acts as a tumor promoter in hepatic tumorigenesis. We examined whether prolonged alcohol consumption could act as a hepatic tumor promoter after initiation by diethylnitrosamine (DEN) in a rat model. Male Sprague-Dawley rats were injected with 20 mg DEN/kg body weight 1 wk before introduction of either an ethanol liquid diet or an isoenergic control liquid diet. Hepatic pathological lesions, hepatocyte proliferation, apoptosis, PPARα and PPARγ, and plasma insulin-like growth factor 1 (IGF-1) levels were assessed after 6 and 10 mo. Mean body and liver weights, plasma IGF-1 concentration, hepatic expressions of proliferating cellular nuclear antigen and Ki-67, and cyclin D1 in ethanol-fed rats were all significantly lower after 10 mo of treatment compared with control rats. In addition, levels of hepatic PPARγ protein, not PPARα, were significantly higher in the ethanol-fed rats after prolonged treatment. Although ethanol feeding also resulted in significantly fewer altered hepatic foci, hepatocellular adenoma was detected in ethanol-fed rats at 10 mo, but not in control rats given the same dose of DEN. Together, these results indicate that chronic, excessive ethanol consumption impairs normal hepatocyte proliferation, which is associated with reduced IGF-1 levels, but promotes hepatic carcinogenesis. © 2011 American Society for Nutrition.

Since 2000 the use of the GPS navigation increases considerably. The reason for that was the deactivation of the selective availability in May 2000. However, these receivers do not register the observables, they just estimate and store them and that prevents the post-processing data. Based in this aspect some softwares were developed and are available for free. They allow recording the GPS observables, pseudorange and carrier phase. These programs are able to read in binary files and record information concerning the GPS observables and to convert binary format to a RINEX format. This study presents the GPS Garmin 12 XL evaluation using free programs. Two experiments were carried out in Presidente Prudente-SP region using the relative static survey. The processing was carried out with intervals of 30, 15, 10 and 5 minutes. The results were compared with the coordinates from a geodetic receiver and show that 98.9 % of the points, the values in relation to the planimetric accuracy were better than 0.50 m. The only baseline which the value was larger or equal to 0.50 m is the point M0001 (baseline lesser than 2 km) referred to the first experiment. In terms of precision the values did not exceeded 0.30 m.

A epilepsia do lobo temporal (ELT) é a principal forma de epilepsia, e a principal lesão desenvolvida por pacientes com ELT é a esclerose mesial temporal (EMT) apresentando perda neuronal, gliose e atrofia da região do hipocampo. Os íons cálcio são fundamentais para o funcionamento de inúmeras vias metabólicas em todos os níveis fisiológicos e desempenham papel fundamental nas respostas de despolarização neuronal. Foi comprovado que a ativação tanto de receptores ionotrópicos quanto metabotrópicos de glutamato promove o influxo anormal de Ca2+ para o meio intracelular, potencializando o efeito de excitação associado à EMT. O cálcio atua tanto como mensageiro celular secundário, quanto primário através da interação com um receptor específico na superfície celular, o Receptor Sensor de Cálcio (CASR). O CASR é expresso em diversas regiões do cérebro, incluindo o hipocampo, no entanto são desconhecidas muitas das possíveis funções desempenhadas por este receptor no cérebro. Ele pertence à família de receptores acoplados a proteína-G, e exibe homologia significativa com os receptores metabotrópicos de glutamato (mGluRs). Ademais, os mGluRs regulam a expressão da família de proteínas ligantes de cálcio neuronal, da qual faz parte a proteína VILIP-1, associada especificamente com o fenômeno da plasticidade sináptica no cérebro. O objetivo deste projeto foi determinar a expressão das proteínas CASR, mGluR1 e VILIP-1 em tecidos de hipocampo de pacientes com epilepsia do lobo temporal comparando com a expressão de tecidos de indivíduos normais. Os três genes encontraram-se subexpressos nos tecidos de pacientes com ELT, sendo que os tecidos dos pacientes, em grau crônico da doença, justificam tais graus de expressão. beta-Adrenoceptor (beta-AR)-mediated relaxation plays an important role in the regulation of vascular tone. beta-AR-mediated vascular relaxation is reduced in various disease states and aging. We hypothesized that beta-AR-mediated vasodilatation is impaired in DOCA-salt hypertension due to alterations in the cAMP pathway. beta-AR-mediated relaxation was determined in small mesenteric arteries from DOCA-salt hypertensive and control uninephrectomized (Uni) rats. To exclude nitric oxide (NO) and cyclooxygenase (COX) pathways, relaxation responses were determined in the presence of L-NNA and indomethacin, NO synthase inhibitor and COX inhibitors, respectively. Isoprenaline (ISO)-induced relaxation was reduced in arteries from DOCA-salt compared to Uni rats. Protein kinase A (PKA) inhibitors (H89 or Rp-cAMPS) or adenylyl cyclase inhibitor (SQ22536) did not abolish the difference in ISO-induced relaxation between the groups. Forskolin (adenylyl cyclase activator)-induced relaxation was similar between the groups. The inhibition of IKCa/SKCa channels (TRAM-34 plus UCL1684) or BKCa channels (iberiotoxin) reduced ISO-induced relaxation only in Uni rats and abolished the relaxation differences between the groups. The expression of SKCa channel was decreased in DOCA-salt arteries. The expression of BKCa channel a subunit was increased whereas the expression of BKCa channel p subunit was decreased in DOCA-salt arteries. The expression of receptor for activated C kinase 1 (RACK1), which is a binding protein for BKG, channel and negatively modulates its activity, was increased in DOCA-salt arteries. These results suggest that the impairment of beta-AR-mediated relaxation in DOCA-salt mesenteric arteries may be attributable to altered IKCa/SKCa and/or BKCa channels activities rather than cAMP/PKA pathway. Impaired beta-AR-stimulated BKCa channel activity may be due to the imbalance between its subunit expressions and RACK1 upregulation. (C) 2012 Elsevier Ltd. All rights reserved.

BACKGROUND/OBJECTIVES: Serum amyloid A (SAA) is an acute-phase protein that has been recently correlated with obesity and insulin resistance. Therefore, we first examined whether human recombinant SAA (rSAA) could affect the proliferation, differentiation and metabolism of 3T3-L1 preadipocytes. DESIGN: Preadipocytes were treated with rSAA and analyzed for changes in viability and [H-3-methyl]-thymidine incorporation as well as cell cycle perturbations using flow cytometry analysis. The mRNA expression profiles of adipogenic factors during the differentiation protocol were also analyzed using real-time PCR. After differentiation, 2-deoxy-[1,2-H-3]-glucose uptake and glycerol release were evaluated. RESULTS: rSAA treatment caused a 2.6-fold increase in cell proliferation, which was consistent with the results from flow cytometry showing that rSAA treatment augmented the percentage of cells in the S phase (60.9 +/- 0.54%) compared with the control cells (39.8 +/- 2.2%, ***P<0.001). The rSAA-induced cell proliferation was mediated by the ERK1/2 signaling pathway, which was assessed by pretreatment with the inhibitor PD98059. However, the exposure of 3T3-L1 cells to rSAA during the differentiation process resulted in attenuated adipogenesis and decreased expression of adipogenesis-related factors. During the first 72 h of differentiation, rSAA inhibited the differentiation process by altering the mRNA expression kinetics of adipogenic transcription factors and proteins, such as PPAR gamma 2 (peroxisome proliferator-activated receptor gamma 2), C/EBP beta (CCAAT/enhancer-binding protein beta) and GLUT4. rSAA prevented the intracellular accumulation of lipids and, in fully differentiated cells, increased lipolysis and prevented 2-deoxy-[1,2-H-3]-glucose uptake, which favors insulin resistance. Additionally, rSAA stimulated the secretion of proinflammatory cytokines interleukin 6 and tumor necrosis factor alpha, and upregulated SAA3 mRNA expression during adipogenesis. CONCLUSIONS: We showed that rSAA enhanced proliferation and inhibited differentiation in 3T3-L1 preadipocytes and altered insulin sensitivity in differentiated cells. These results highlight the complex role of SAA in the adipogenic process and support a direct link between obesity and its co-morbidities such as type II diabetes.
