Human oral cavity is colonized by a wide range of microorganisms, often organized in biofilms. These biofilms are responsible for the pathogenesis of caries and most periodontal diseases. A possible alternative to reduce biofilms is the photodynamic inactivation (PDI). The success of the PDI depends on different factors. The time required by the PS to remain in contact with the target cells prior to illumination is determinant for the technique's efficacy. This study aimed to assess the interaction between the PS and the biofilm prior to the PDI. We used confocal microscopy and FLIM to evaluate the interaction between the PS and the biofilm's microorganism during the pre-irradiation time (PIT). The study of this dynamics can lead to the understanding of why only some PSs are effective and why is necessary a long PIT for some microorganisms. Our results showed that are differences for each PIT. These differences can be the determinate for the efficacy of the PDI. We observed that the microorganism needs time to concentrate and/or transport the PS within the biofilm. We presented preliminary results for biofilms of Candida albicans and Streptococcus mutans in the presence of Curcumin and compared it with the literature. We observed that the effectiveness of the PDI might be directly correlated to the position of the PS with the biofilm. Further analyses will be conducted in order to confirm the potential of FLIM to assess the PS dynamics within the biofilms. © 2013 SPIE.

The clinical use of topical photodynamic therapy in herpes simplex lesions in Sao Paulo is presented and discussed. Although previous attempts utilising this type of approach in the USA were discontinued in the early 1970s due to several presentations of post-treatment Bowen's disease, none of the cases from the clinic presented here have displayed any complications on follow-up. In addition, lesion recrudescence periods are generally much longer than with conventional approaches. This is thought to be due to improvements in the treatment protocol, viz, use of the non-toxic photosensitisers methylene blue and Hypericum perforatum extract in place of proflavine and neutral red in the original trials, differences in photosensitisation pathway and illumination of the treatment site with red rather than fluorescent/UV light. A nanotecnologia tem sido aplicada para o desenvolvimento de materiais para diversas aplicações inclusive na inativação de patógenos. As nanopartículas de sílica (npSi) destacam-se pela alta área superficial, facilidade na alteração da superfície para aumento da eficiência adsortiva, penetrabilidade e toxicidade para bactérias gram-negativas sendo biocompatíveis para células de mamíferos e mais foto-estáveis que a maioria dos compostos orgânicos. Devido as suas vantagens, as npSi podem ser usadas para veicular fotossensibilizadores (FSs) uma vez que permitem sua utilização em solução aquosa em que os FSs geralmente são insolúveis. Além disso, o uso de FSs em vez de antibióticos, permite a inativação microbiológica pela Terapia Fotodinâmica sem que as bactérias adquiram resistência por mecanismos genéticos. Esse processo ocorre pela interação entre um FS, luz e oxigênio molecular produzindo oxigênio singleto que é extremamente reativo danificando estruturas celulares. O objetivo desse estudo foi otimizar a fotoinativação dinâmica de E .coli utilizando Azul de Metileno (AM) e Azul de Toluidina O (ATO) veiculados por npSi. As npSi foram preparadas pela metodologia sol-gel, caracterizadas por microscopia eletrônica de varredura (MEV) e submetidas à adsorção de AM e ATO em sua superfície. A presença de AM e ATO na superfície das npSi foram analisadas por espectroscopia no infravermelho; espectroscopia de fluorescência por raio-X e análise termogravimétrica. O planejamento experimental, iniciado pelo fatorial 23 e modelado ao composto central em busca das condições ótimas foi adotado pela primeira vez nessa aplicabilidade, visando a fotoinativação de E. coli empregando AM e ATO em solução e em seguida com npSi. AM e ATO veiculados por npSi permitem a fotoinativação em concentrações mais baixas de FS (20 e 51% respectivamente), causando desestruturação da integridade bacteriana demonstrada por MEV. Os resultados sugerem que a veiculação de AM e ATO por npSi é extremamente efetiva para a fotoinativação dinâmica de E. coli e que o planejamento composto central pode levar à completa inativação das bactérias.

Die zunehmende Luftverschmutzung aufgrund des steigenden Energiebedarfs und Mobilitätsanspruchs der Bevölkerung, insbesondere in urbanen Gebieten, erhöht das Gefährdungspotential für die Gesundheit und verschlechtert so die Lebensqualität. Neben der Vermeidung von Emissionen toxischer Gase als mittel- und langfristig optimale Maßnahme zur Verbesserung der Luftqualität, stellt der Abbau emittierter Luftschadstoffe ein geeignetes und kurzfristig wirksames Mittel dar. Ein solcher Abbau kann durch Photokatalyse erzielt werden, allerdings nutzen Photokatalysatoren, die auf dem Halbleiter Titandioxid (TiO2) basieren, das solare Emissionsspektrum nur geringfüfig aus und sind in Innenräumen und anderen UV-schwachen Bereichen nicht wirksam. Um diese Nachteile zu überwinden, wurde ein Photokatalysator entwickelt und hergestellt, der aus TiO2 (P25) als UV-aktiver Photokatalysator und als Trägermaterial sowie einem seinerseits im Vis-Bereich photoaktiven Porphyrazin-Farbstoff als Beschichtung besteht. Die sterisch anspruchsvollen und in der Peripherie mit acht Bindungsmotiven für TiO2 versehenen Farbstoffmoleküle wurden zu diesem Zweck auf der Halbleiteroberfläche immobilisiert. Die so gebildeten Porphyrazin-Titandioxid-Hybride wurde ausführlich charakterisiert. Dabei wurden unter anderem die Bindung der Farbstoffe auf der Titandioxidoberfläche mittels Adsorptionsisothermen und die UV/Vis-spektroskopischen Eigenschaften des Hybridmaterials untersucht. Zur Bestimmung der photokatalytischen Aktivitäten der Einzelkomponenten und des Hybridmaterials wurden diese auf die Fähigkeit zur Bildung von Singulett-Sauerstoff, Wasserstoffperoxid und Hydroxylradikalen hin sowie in einem an die ISO-22197-1 angelehnten Verfahren auf die Fähigkeit zum Abbau von NO hin jeweils bei Bestrahlung in drei Wellenlängenbereichen (UV-Strahlung, blaues Licht und rotes Licht) geprüft. Darüber hinaus konnte die Aktivität des Hybridmaterials bei der Photodynamischen Inaktivierung (PDI) von Bakterien unter UV- und Rotlichtbestrahlung im Vergleich zum reinen Ttandioxid bestimmt werden. Die Charakterisierung des Hybridmaterials ergab, dass die Farbstoffmoleküle in einer neutralen Suspension nahezu irreversibel in einer monomolekularen Schicht mit einer Bindungsenergie von -41.43 kJ/mol an die Oberfläche gebunden sind und das Hybridmaterial mit hohen Extinktionskoeffizienten von bis zu 105 M-1cm-1 in großen Bereichen des UV/Vis-Spektrums Photonen absorbiert. Das Spektrum des Hybridmaterials setzt sich dabei additiv aus den beiden Einzelspektren zusammen. Die Auswirkungen der Charakterisierungsergebnisse auf die Bildung reaktiver Sauerstoffspezies wurden ausführlich diskutiert. Der Vergleich der Aktivitäten in Bezug auf die Bildung der reaktiven Sauerstoffspezies zeigte, dass die Aktivität des Hybridmaterials bis auf die bei der Bildung von Hydroxylradikalen unter UV-Bestrahlung in allen Versuchen deutlich höher war als die Aktivität des reinen Titandioxids. Im Gegensatz zu reinem Titandioxid erzeugte das Hybridmaterial in allen untersuchten Wellenlängenbereichen Mengen an Singulett-Sauerstoff, die photophysikalisch eindeutig detektierbar waren. Zur Erklärung und Deutung dieser Beobachtungen wurde eine differenzierte Diskussion geführt, die die Ergebnisse der Hybridpartikelcharakterisierung aufgreift und implementiert. Der Vergleich der NO-Abbaueffizienzen ergab bei allen Experimenten durchgängig deutlich höhere Werte für das Hybridmaterial. Zudem wurden durch das Hybridmaterial nachgewiesenermaßen wesentlich geringere Mengen des unerwünschten Nebenprodukts des Abbaus (NO2) gebildet. Im Zuge der Diskussion wurden verschiedene mögliche Mechanismen der „sauberen“ Oxidation zu Nitrat durch das Hybridmaterial vorgestellt. Untersuchungen zur Photodynamischen Inaktivierung verschiedener Bakterien ergaben, dass das Hybridmaterial neben einer zu P25 ähnlichen Aktivität unter UV-Bestrahlung, anders als P25, auch eine PDI verschiedener Bakterien unter Rotlichtbestrahlung erreicht.

A cavidade oral é um habitat favorável ao desenvolvimento de microrganismos, alguns dos quais podem causar doenças, sendo Enterococcus faecalis uma bactéria frequentemente encontrada em biofilmes instalados em diferentes nichos da cavidade oral. Este trabalho teve como objetivo testar a aplicabilidade da inativação fotodinâmica (PDI), usando porfirinas como fotossensibilizadores, como estratégia de controlo de biofilmes da cavidade oral, tomando E. faecalis como microrganismo modelo. Como fotossensibilizadores, foram testadas as porfirinas catiónicas Tetra-Py+-Me, Tri-Py+-Me-PF, PCat 2, PCat 3, PCat 4 e o corante azul de toluidina O (TBO), incluído como fotossensibilizador de referência. Os biofilmes de E. faecalis foram irradiados com luz branca (270 J.cm-2) a uma intensidade de 150 mW.cm-2, na presença de até 50 µM de porfirina ou até 20 µM de TBO. A cinética de inativação foi caracterizada pela variação da concentração de células viáveis ao longo da experiência. Foi também testada a inativação de células na forma livre, em condições equivalentes. Os biofilmes de E. faecalis mostraram-se muito resistentes à PDI com qualquer dos PS testados, não tendo sido conseguidos fatores de inativação superiores a 2 log com a concentração máxima de PS (50 µM) e a dose máxima de luz (270 J.cm-2). Na forma livre as células foram inativadas até ao limite de quantificação com concentrações de PS de 0,5 µM e doses de luz até 108 J.cm-2, com uma intensidade de 10 mW.cm-2. No entanto, a eficiência de ligação dos PS às células livres não foi maior do que aos biofilmes. Embora os fatores de inativação obtidos não permitam ainda considerar que a PDI com os compostos testados seja uma abordagem antimicrobiana eficiente contra biofilmes de E. faecalis, o facto de se confirmar uma relação entre as propriedades químicas e físicas do PS e a sua eficiência, bem como os resultados muito promissores obtidos com uma das famílias de porfirinas testadas apenas em células livres, justifica a prossecução do desenvolvimento de novos PS para o controle de biofilmes bacterianos na cavidade oral.

This study describes the association of curcumin with light emitting diode (LED) for the inactivation of Candida albicans. Suspensions of Candida were treated with nine curcumin concentrations and exposed to LED at different fluences. The protocol that showed the best outcomes for Candida inactivation was selected to evaluate the effect of the preirradiation time (PIT) on photodynamic therapy (PDT) effectiveness, the uptake of curcumin by C. albicans cells and the possible involvement of singlet oxygen in the photodynamic action. Curcumin-mediated PDT was also assessed against biofilms. In addition to the microbiological experiments, similar protocols were tested on a macrophage cell line and the effect was evaluated by Methyltetrazolium assay (MTT) and SEM analysis. The optical properties of curcumin were investigated as a function of illumination fluence. When compared with the control group, a statistically significant reduction in C. albicans viability was observed after PDT (P < 0.05), for both planktonic and biofilm cultures. Photodynamic effect was greatly increased with the presence of curcumin in the surrounding media and the PIT of 20 min improved PDT effectiveness against biofilms. Although PDT was phototoxic to macrophages, the therapy was more effective in inactivating the yeast cell than the defense cell. The spectral changes showed a high photobleaching rate of curcumin.

P>Although photodynamic therapy (PDT) has shown great promise for the inactivation of Candida species, its effectiveness against azole-resistant pathogens remains poorly documented. This in vitro study describes the association of Photogem (R) (Photogem, Moscow, Russia) with LED (light emitting diode) light for the photoinactivation of fluconazole-resistant (FR) and American Type Culture Collection (ATCC) strains of Candida albicans and Candida glabrata. Suspensions of each Candida strain were treated with five Photogem (R) concentrations and exposed to four LED light fluences (14, 24, 34 or 50 min of illumination). After incubation (48 h at 37 degrees C), colonies were counted (CFU ml-1). Single-species biofilms were generated on cellulose membrane filters, treated with 25.0 mg l-1 of Photogem (R) and illuminated at 37.5 J cm-2. The biofilms were then disrupted and the viable yeast cells present were determined. Planktonic suspensions of FR strains were effectively killed after PDT. It was observed that the fungicidal effect of PDT was strain-dependent. Significant decreases in biofilm viability were observed for three strains of C. albicans and for two strains of C. glabrata. The results of this investigation demonstrated that although PDT was effective against Candida species, fluconazole-resistant strains showed reduced sensitivity to PDT. Moreover, single-species biofilms were less susceptible to PDT than their planktonic counterparts.

The ability of Staphylococcus aureus to develop multidrug resistance is well documented, and the antibiotic resistance showed by an increasing number of bacteria has shown the need for alternative therapies to treat infections, photodynamic therapy (PDT) being a potential candidate. The aim of this study was to determine the effect of photodynamic therapy as a light-based bactericidal modality to eliminate Staphylococcus aureus. The study investigated a technique based on a combination of light and a photosensitizer that is capable of producing oxidative species to induce a cytotoxic effect. A Staphylococcus aureus suspension was exposed to a light emitting diode (LED) emitting at 628 nm, 14.6 mW/cm(2), and energy density of 20J/cm(2), 40J/cm(2), or 60 J/cm(2) in the presence of different porphyrin concentrations (PhotogemA (R)). Three drug concentrations were employed: 12 mu l/ml, 25 mu l/ml, and 50 mu l/ml. The treatment response was evaluated by the number of bacterial colony forming units (CFU) after light exposure. The results indicated that exposure to 60 J/cm(2) eliminated 100% (10 log(10) scales) of bacteria, on average. The best PDT response rate to eliminate Staphylococcus aureus was achieved with exposure to LED light in combination with the photosensitizer at concentrations ranging from 25 mu l/ml to 50 mu l/ml. These data suggest that PDT has the potential to eliminate Staphylococcus aureus in suspension and indicates the necessary drug concentration and light fluency.

To evaluate the cytotoxicity of PDT (photodynamic therapy) with Photogem (R) associated to blue LED (light-emitting diode) on L929 and MDPC-23 cell cultures, 30000 cells/cm(2) were seeded in 24-well plates for 48 h, incubated with Photogem (R) (10, 25 or 50 mg/l) and irradiated with an LED source (460 +/- 3 nm; 22 mW/cm(2)) at two energy densities (25.5 or 37.5 J/cm(2)). Cell metabolism was evaluated by the MTT (methyltetrazolium) assay (Dunnet`s post hoc tests) and cell morphology by SEM (scanning electron microscopy). Flow cytometry analysed the type of PDT-induced cell death as well and estimated intracellular production of ROS (reactive oxygen species). There was a statistically significant decrease of mitochondrial activity (90% to 97%) for all Photogem (R) concentrations associated to blue LED, regardless of irradiation time. It was also demonstrated that the mitochondrial activity was not recovered after 12 or 24 h, characterizing irreversible cell damage. PDT-treated cells presented an altered morphology with ill-defined limits. In both cell lines, there was a predominance of necrotic cell death and the presence of Photogem (R) or irradiation increased the intracellular levels of ROS. PDT caused severe toxic effects in normal cell culture, characterized by the reduction of the mitochondrial activity, morphological alterations and induction of necrotic cell death.
