Determinou-se à demanda hÃdrica e o coeficiente de cultivo (Kc) do maracujá amarelo (Passiflora edulis Sims f. flavicarpa Deg.), seleção Sul-Brasil, cultivado sob irrigação localizada, no municÃpio de Botucatu-SP (22o 51’ S, 48o 26’ W). A evapotranspiração máxima da cultura (ETc) e a evapotranspiração de referência (ETo) foram medidas em lisÃmetros de nÃvel de lençol freático constante, durante o perÃodo de 29 de setembro de 2000 a 20 de julho de 2001. Os valores da ETc e ETo foram de 954,98 mm e 1.069,21 mm, respectivamente, durante todo o perÃodo medido. Os valores de Kc variaram de 0,42 a 1,12, com os valores máximos registrados entre 150 e 210 dias após o transplantio das mudas no campo (DAT), perÃodo correspondente aos estádios fenológicos de florescimento e formação dos frutos.

This greenhouse study investigated the efficacy of acibenzolar-S-methyl (Bion®) treatment of lower leaves of passionfruit, (Passiflora edulis f. sp. flavicarpa), on Passionfruit woodiness disease and activities of two pathogenesis-related proteins, chitinase and β-1,3-glucanase after inoculation with passionfruit woodiness virus (PWV). All Bion® concentrations reduced disease symptoms, but the concentration of 0.025 g active ingredient (a.i.)/l was the most effective, reducing disease severity in systemic leaves by 23, 29 and 30 compared with water-treated controls at 30, 40 and 50 days post inoculation (dpi) with PWV, respectively. Correspondingly, relative virus concentration as determined by DAS-ELISA in the upper, untreated leaves (new growth) above the site of inoculation at 50 dpi was reduced by 17 and 22 in plants treated with 0.025 and 0.05 g a.i./l, respectively. Bion® treatment and subsequent inoculation with PWV increased chitinase and β-1,3-glucanase activities in the new leaves above the site of inoculation at 30 dpi with PWV. It was concluded that optimal protective Bion® treatment concentrations were 0.025 and 0.05 g a.i./l.

A method is reported for the quantification of isoorientin (using a standard addition method) and total flavonoids (expressed as rutin, using the external standard method) in passion fruit pulp (Passiflora edulis Sims f. flavicarpa Degener, Passifloraceae). Extraction of flavonoids was optimized by experimental design methodology, and quantitative analysis was performed by high-performance liquid chromatography with photo-diode array detection (HPLC-UV/DAD). The method was developed and validated according to ICH requirements for specificity, linearity, accuracy, precision (repeatability and intermediate precision). LOD and LOQ. Rutin was chosen as standard for the quantification of total flavonoids in order to propose a HPLC method feasible for routine analysis of the flavonoids in the passion fruit pulp. The passion fruit pulp contained 16.226 +/- 0.050 mg L(-1) of isoorientin and 158.037 +/- 0.602 mg L(-1) of total flavonoid, suggesting that P. edulis fruits may be comparable with other flavonoid food sources such as orange juice or sugarcane juice. The antioxidant activity of methanol extracts from Passiflora edulis and Passiflora alata pulp, and P. edulis rinds, healthy or infected with the passion fruit woodiness virus (PWV), was investigated using the oxidant activities of the neutrophil and the neutrophil granule enzyme myeloperoxidase (MPO), both playing key roles in inflammation. The reactive oxygen species produced by stimulated neutrophils were evaluated by lucigenin-enhanced chemiluminescence (CL) and the activity of purified MPO was measured by SIEFED (Specific Immunological Extraction Followed by Enzymatic Detection), a technique for studying the direct interaction of a compound with the enzyme. The rind extracts of P. edulis possessed higher and dose-dependent inhibitory effects on CL response and on the peroxidase activity of MPO than total pulp extracts from both passion fruit species. The quantification of isoorientin in the extracts showed a correlation with their antioxidant activity, suggesting the potential of P. edulis rinds as functional food or as a possible source of natural flavonoids. This study aimed to characterize the anatomical events and ultrastructural aspects of direct and indirect in vitro organogenesis in Passiflora edulis. Root explants were cultured on induction medium, supplemented with 4.44 mu M 6-benzyladenine. Roots at different stages of development were collected and processed for observation by light microscopy and scanning and transmission electron microscopy. Patterns of direct and indirect regeneration were observed in the explants. During direct organogenesis, the organogenic buds and nodules, formed from meristemoids, originated from the pericycle regions distant from the cut surface. Completely differentiated buds were observed after 20 days of culture. During indirect organogenesis, bud formation occurred via meristemoids at the periphery of the calli, which differentiated from the cortical region of the initial explant. Regardless of the regeneration pattern, the meristemoids had similar ultrastructural characteristics; however, differences were reported in the nuclear shape of the cells of the meristemoids formed directly and indirectly. This study provides important information for enhancing the understanding and characterization of the organogenic process in non-meristematic explants and provides information on the use of roots as explants in genetic transformation protocols for this important tropical species.

The content of isoorientin in passion fruit rinds (Passiflora Mulls fo. flavicarpa O. Degener) was determined by HPTLC (high performance thin layer chromatography) with densitometric analysis. The results revealed a higher amount of isoorientin in healthy rinds of P edulis (92.275 +/- 0.610 mg L-1) than in rinds with typical symptoms of PWV (Passion fruit Woodiness Virus) infection (28.931 +/- 0.346 mg L-1). The HPTLC data, allied to assays of radical scavenging activity, suggest the potential of P Mulls rinds as a natural source of flavonoids or as a possible functional food.

O presente estudo investigou as atividades antioxidantes in vitro, ex vivo e in vivo dos extratos de folhas Passiflora alata e Passiflora edulis, plantas usadas na medicina popular e ricas em polifenóis, compostos com reconhecida atividade antioxidante. No modelo experimental in vitro, ambos os extratos demonstraram atividade antioxidante e proteção contra dano protéico induzido por glicose. Fatias de fÃgado de ratos foram utilizadas como modelo ex vivo. Tanto o extrato de P. alata quanto o extrato de P. edulis protegeram de forma significativa o dano protéico e a morte celular induzidos por FeSO4. Como a P. alata é uma droga oficial da Farmacopéia Brasileira, os efeitos antioxidantes deste extrato foram investigados in vivo. Ratos machos Wistar receberam tratamento intragástrico de extrato de folhas de P. alata (1 e 5 mg/kg), trolox (0,18 mg/kg) ou água (controle) durante 30 dias, seguido de uma dose de CCl4 (3 ml/kg, i.p.) no 30º dia. O dano hepático e os efeitos antioxidantes do pré-tratamento com extrato de P. alata foram avaliados em vários órgãos. Quando comparados ao grupo controle, os ratos pré-tratados com o extrato demonstraram dano hepático menor, evidenciado por um grau menor de necrose, nÃveis menores de lipoperoxidação e maior atividade das enzimas superóxido dismutase e catalase. Adicionalmente, nÃveis menores de lipoperoxidação cardÃaca foram observados com o pré-tratamento de P. alata (5mg/kg). Os resultados obtidos neste estudo indicam que os extratos de Passiflora são fontes potenciais de antioxidantes naturais. Estudos adicionais investigando o papel de compostos isolados destes extratos em patologias humanas onde o estresse oxidativo está envolvido são necessários.

Objetivou-se analisar economicamente a cultura do maracujá-amarelo (Passiflora edulis) na região de MarÃlia-SP, safra de 2010/2011. Foram utilizadas estruturas do custo operacional efetivo e custo operacional total referente à fase de implantação e condução da lavoura por unidade de área e cinco indicadores de rentabilidade. Identificou-se um custo total de produção de R$ 37.751,67 por hectare ou R$ 1,89 por quilo da fruta. Os itens do custo operacional efetivo que mais oneraram o sistema de produção corresponderam à s operações de máquinas (31,1%) e mão de obra (23,5%). Os indicadores de rentabilidade mostraram-se desfavoráveis para o sistema produtivo analisado, em decorrência, principalmente, do alto preço dos insumos e práticas para controle de doenças. Há necessidade de adequações técnicas relacionadas com o manejo fitossanitário da cultura para a redução do custo total de produção de modo a tornar a atividade rentável.

This work was carried out in the city of Maringa, located in the Northwest region of Parana, with the aim of analyzing economically the yellow passion fruit crop (Passiflora edulis Sims. f. flavicarpa Deg.), comparing organic and conventional production systems. To estimate technical coefficients, planting and production costs, and profitability indicators of culture, the data were surveyed from technicians of the region and based in experimental work, carried out at the Experimental Farm of lguaterm from the Maringa State University, in the period of june/2002 to july/2004. The total production costs of the culture in two agricultural years reached a value 12.94% higher for the conventional system, and in this same period the medium index of profitability was 21.39% higher for the organic system. The organic production system was economically feasible and provided a larger index of profitability.
